    ANTISPASMODIC EFFECTS OF YARROW (ACHILLEA MILLEFOLIUM L.) EXTRACT IN THE ISOLATED ILEUM OF RAT

    Achillea millefolium L. is cultivated in Iran and widely used in traditional medicine for gastrointestinal disorders. The aim of this study was to determine the effect of hydroalcoholic extract of A. millefolium on the contraction and relaxation of isolated ileum in rat. In this experimental study, aerial parts of A. millefolium were extracted by maceration in ethanol 70% for 72h. Terminal portion of ileum in 100 male Wistar rats was dissected and its contractions were recorded isotonically in an organ bath containing Tyrode solution (37 degrees C, pH 7.4) under one gram tension. Acetylcholine (1mM) and KCl (60mM) were used to create isotonic contractions. Propranolol and N omega-Nitro-L-arginine methylester hydrochloride (L-NAME) were used to investigate the mechanisms of action prior to giving the extract to the relevant groups. Data were compared by ANOVA and Turkey's post hoc test.. The results showed that the ileum contraction was induced by KCl and acetylcholine induced contraction was significantly reduced by A. millefolium extract. The cumulative concentrations of A. millefolium relaxed the KCl and acetylcholine induced contractions (n=14, p<0.001). The inhibitory effect of extract on contraction induced by KCl and acetylcholine was not significantly affected neither by propranolol (1 mu M) nor by L-NAME (100 mu M). There was no significant difference in the rate of relaxation by propranolol and L-NAME between the two groups. In conclusion, A. millefolium can inhibit contraction of smooth muscle of ileum in rat, and it can be used for eliminating intestinal spasms. These results suggest that the relaxatory effect of A. millefolium on ileum contractions can be due to the blockade of voltage dependent calcium channels. In addition, the beta-adrenoceptors, cholinergic receptors and nitric oxide production are not powerful actors in inhibitory effect of A. millefolium. So, the nitric oxide and adrenergic systems may also be involved in the antispasmodic effect of A. millefolium

    Abstract: Background Homoeopathic mother tinctures and herbal extracts are used worldwide for medicinal purposes on the basis that the plant extract contains the active components essential for medicinal use. Quality analysis of samples ensures that the correct active components are present for medicinal use. Thin layer chromatography has been used extensively to determine the quality of plant extracts and could just as readily be applied to the assessment of quality of homoeopathic mother tinctures. The development of a multidimensional technique allows for greater quality analyses of these extracts. Materials and Methods The Salvia officinalis sample was extracted and the one dimensional thin layer chromatographic plate development of extracted sample was performed according to the German Homoeopathic Pharmacopoeia (GHP). Before the two dimensional thin layer chromatographic plates where run, different solvent systems were tested through a comparison of the resultant one dimensional thin layer chromatographic plates with that of the plate developed according to the GHP. The two dimensional thin layer chromatographic plate was developed in one direction using the eluent given in the GHP, the plate was then rotated 90 degrees and further developed using neat dichloromethane. Results and Conclusion The two dimensional thin layer chromatogram for the selected sample Salvia officinalis was successfully established. This results in a more extensive profile regarding the identification of the individual components present in the homoeopathic mother tincture. The further production of two dimensional chromatograms is essential to the expanded detailed analysis of individual products and potential raw material production of even higher standards

    Possible hypocholesterolemic effect of ginger and rosemary oils in rats

    Background: Hypercholesterolemia is a major risk factor for development of atherosclerosis. The present study was conducted to evaluate the potential effect of ginger oil alone or combined with rosemary oil as hypocholesterolemic agent in rats fed high fat diet.Materials and methods: Healthy female albino rats (n=80) weighting about (150-180 g) were included in this study divided into two equal groups; Group (I): were fed on the basal diet. Group (I) were divided into 4 subgroups each 10: Group (Ia): negative control. Group (Ib): Rats received i.p 2.5 g/Kg b.w of ginger oil. Group (Ic): rats received i.p 2.5 g/Kg b.w of rosemary oil. Group (Id): Rats received i.p 5 g/Kg b.w mixture of ginger oil and rosemary oil (1:1). The second main groups; Group (II): high fat diet (HFD) were fed on the basal diet plus cholesterol (1%), bile salt (0.25%) and animal fat (15%) to induce hypercholesterolemia for six weeks. Group (II) was divided into 4subgroups: Group (IIa): HFD. Group (IIb): HFD were treated with i.p 2.5 g/Kg b.w ginger oil. Group (IIc): (n=10) HFD were treated with i.p 2.5 g/Kg b.w rosemary oil. Group (IId): (n=10) HFD were treated with i.p 5 g/Kg b.w mixture of oils.Results: It was found that HFD rats showed a significant elevation in glucose, total cholesterol, triglyceride, GOT, GPT, alkaline phosphatase and a reduction in serum HDL-c compared with negative control. Treatment with ginger oil, rosemary oil and their mixture modulated the elevation of these parameters. Histopathological examination of the liver tissue of HFD rats showed a lipid deposition and macrophage infiltration and stenosis of hepatic vein. Treatment with mixture oils preserves normal structure of liver.Conclusion: It was concluded that, hypocholesterolemic effect was related to the active oil content as Rosemary oil contain - α-pinene, Camphor, cineole, borneol and Ginger oil contain Linalool, Terpineol ,Borneol , Eucalyptol.Keywords: Ginger Oil, Rosemary Oil, Rats, Hypocholesterolemi

    Herbal Remedies And Their Adverse Effects In Tem Tribe Traditional Medicine In Togo

    In Africa, up to 80% of the population relies on herbal concoctions for their primarily health care. In Togo, western Africa, Tem tribe is a population with old knowledge of medicinal plants, however, still very little is known about their medical practices. The present study was conducted to access for the apprehension of adverse effects of traditional remedies by Tem traditional healers (TH). Enquiry was performed by interviews with healers from August to October 2007 in Tchaoudjo prefecture (Togo). The study allowed us to interview 54 TH including 41(75.93%) males and 13(24.07%) females, who cited 102 recipes assumed to have adverse effects. The recipes were used alone to cure several diseases including haemorrhoids (22.55%), female sexual disorders and infertility (21.57%), gastrointestinal disorders (18.63%), and malaria (6.86%). A total of 34 plants belonging to 21 families were cited to be components of the recipes. Euphorbiaceae and Mimosaceae families were the most represented, however, Nauclea latifolia, Khaya senegalensis, Pseudocedrela kotschyi and Xeroderris stuhlmannii were the main components of recipes linked to adverse effects. A total of 20 adverse effects were linked to the administration of theses drugs, and among them; diarrhoea, abdominal pains, polyuria, general weakness and vomiting were the most frequently encountered. These findings were in accordance with several reports of the literature concerning medicinal plants, although they were based on empirical observations. Laboratory screenings are needed to access for the effectiveness as well as the possible toxic effects of the recipes. Keywords: traditional concoctions, medicinal plants, adverse effects, Togo

    Whole body vibration improves attention and motor performance in mice depending on the duration of the whole-body vibration session

    BACKGROUND: Whole body vibration (WBV) is a form of physical stimulation via mechanical vibrations transmitted to a subject. It is assumed that WBV induces sensory stimulation in cortical brain regions through the activation of skin and muscle receptors responding to the vibration. The effects of WBV on muscle strength are well described. However, little is known about the impact of WBV on the brain. Recently, it was shown in humans that WBV improves attention in an acute WBV protocol. Preclinical research is needed to unravel the underlying brain mechanism. As a first step, we examined whether chronic WBV improves attention in mice.MATERIAL AND METHODS: A custom made vibrating platform for mice with low intensity vibrations was used. Male CD1 mice (3 months of age) received five weeks WBV (30 Hz; 1.9 G), five days a week with sessions of five (n=12) or 30 (n=10) minutes. Control mice (pseudo-WBV; n=12 and 10 for the five and 30 minute sessions, respectively) were treated in a similar way, but did not receive the actual vibration. Object recognition tasks were used as an attention test (novel and spatial object recognition - the primary outcome measure). A Balance beam was used for motor performance, serving as a secondary outcome measure.RESULTS: WBV sessions of five (but not WBV sessions of 30 minutes) improved balance beam performance (mice gained 28% in time needed to cross the beam) and novel object recognition (mice paid significantly more attention to the novel object) as compared to pseudo WBV, but no change was found for spatial object performance (mice did not notice the relocation). Although 30 minutes WBV sessions were not beneficial, it did not impair either attention or motor performance.CONCLUSION: These results show that brief sessions of WBV improve, next to motor performance, attention for object recognition, but not spatial cues of the objects. The selective improvement of attention in mice opens the avenue to unravel the underlying brain mechanisms.</p

    EVALUATION OF ACUTE TOXICITY AND ANTI-INFLAMMATORY EFFECTS OF BACCHAROIDES SCHIMPERI (DC.) IN EXPERIMENTAL ANIMALS

    Background: Steroidal and non-steroidal anti-inflammatory drugs are most commonly used to treat inflammation, and shown to have severe side effects. In this study, we aimed at evaluating the anti-inflammatory and acute toxicity effects of Baccharoides schimperi (DC.) in order to get new anti-inflammatory agents of natural origin. Materials and methods: The aerial part of the plant was dried under shade, ground and extracted with 96% alcohol (BSE). It was further fractionated in sequence to n-hexane (BSH), chloroform (BSC) and methanol (BSM) soluble fractions. Acute toxicity was evaluated by oral administration of plant and hind paw induced-edema method in rats was used for the anti-inflammatory evaluation. Results: The BSE was found safe up to the dose level of 3 g/kg b.w. and showed LD50 value 7.250 g/kg body weight (b.w.) in mice. BSE showed significant anti-inflammatory effect (62.91%) at 500 mg/kg b.w. Further the n-hexane, chloroform and methanol fractions of BSE were tested for anti-inflammatory activity. The n-hexane fraction (BSH) exhibits significant activity (64.87%) at 400 mg/kg b.w. The methanol fraction (BSM) showed dose dependent activity, highest activity (60.42%) was observed at higher dose 400 of mg/kg b.w. In chloroform fraction (BSC) no significant activity was observed. Conclusion: The results of the study revealed that the plant is safe to the experimental model and recommended as a potential source of anti-inflammatory agent

    ACUTE AND SUB-ACUTE TOXICITY OF UTHULI LWEZICHWEâ„¢, A TRADITIONAL MEDICINE USED IN THE MANAGEMENT OF DIABETES MELLITUS IN KWAZULU NATAL, SOUTH AFRICA

    Background: The number of people using traditional medicines in the management of chronic diseases is increasing. The safety profile of some of the formulations, however, has not been scientifically demonstrated. This study assessed the acute and sub-acute toxicity of Uthuli Lwezichwe™ (UL), a traditional medicine used by a traditional healer in the management of diabetes mellitus. Materials and Methods: In the acute toxicity assay, one female Sprague dawley rat was administered 1000mg/kg body weight (BW) of Uthuli lwezichwe™ and observed for 14 days. In the sub-acute assay, 24 Sprague dawley rats were randomized into four groups. With one group as the control, the other groups were administered varying daily doses (100 mg, 300 mg, 1000 mg/kg BW) of UL for 28 days. Phytochemical analysis of UL was done using Liquid Chromatography – Quadrupole Time of Flight - Mass Spectrometer (LC-QTOF-MS). Results: There were no significant differences in liver function biomarkers and kidney function biomarkers between the control and all the treatment groups. Histological examination of the kidney however, showed enlarged bowman’s space and distal convoluted tubule diameter and area in the 1000mg/kg UL treated group compared to the control group. Similarly, histological examinations of the liver showed increased sinusoidal space and decreased central vein area in the 1000mg/kg UL treated group. A number of phytoconstituents were identified in UL. Conclusion: The maximum safe dose was determined to be between 300 – 1000 mg/kg BW. We recommend a chronic toxicity study to assess the long-term effects

    ANTIBACTERIAL EFFECTS OF SINGLE AND COMBINED CRUDE EXTRACTS OF SYNADENIUM GLAUCESCENS AND COMMIPHORA SWYNNERTONII

    Background: Synadenium glaucescens and Commiphora swynnertonii are among the reported plants used traditionally for treatment of bacterial infections. This study reports antibacterial effects of single and combined extracts from leaves, stem and root barks of Commiphora swynnertonii and Synadenium glaucescens. Materials and Methods: Plants were collected from Manyara and Njombe regions in Tanzania. Extraction was done using dichloromethane and methanol. The extracts were assessed for antibacterial activity against Gram-positive bacteria (Staphylococcus aureus and Enterococcus faecalis) and Gram-negative bacteria (Escherichia coli, Klebsiella pneumonia and Pseudomonas aeruginosa). Minimum Inhibitory Concentrations (MIC) was determined by broth microdilution, while Fractional Inhibitory Concentration (FIC) indices were calculated from MIC values of combined extracts to determine combination effects. Results: Strong antibacterial activities were demonstrated by all extracts of S. glaucescens (MIC 0.011-0.375mg/mL) against Gram-positive bacteria and methanol extracts of C. swynnertonii (MIC 0.047-0.375mg/mL). Synergistic effect was observed when combining methanol extracts of C. swynnertonii stem bark with S. glaucescens leaves against S. aureus (∑FIC 0.5), Other synergistic effects were observed against E. faecalis with dichloromethane extracts of C. swynnertonii stem bark and S. glaucescens stem bark (∑FIC 0.5), and C. swynnertonii root bark and S. glaucescens root bark (FIC index 0.3). For the remaining combinations, mainly additive effects were observed. Conclusion: Synergistic effects on bacteria were observed by combining different plant parts of S. glaucescens and C. swynnertonii suggesting that it could be beneficial to combine such extracts when used for antibacterial purposes

    TOOTHBRUSH AND TOWEL HANDLING AND THEIR MICROBIAL QUALITY: THE CASE OF STUDENTS OF UNIVERSITY FOR DEVELOPMENT STUDIES, NYANKPALA CAMPUS, GHANA

    sought to assess how students of the University for Development Studies handle their toothbrushes and towels and the consequence of that with&nbsp; regards to the microbial quality of these personnel hygiene materials. Materials and Methods: A total of 100 swap samples were collected (50 toothbrushes and 50 towels) for microbial analysis. Questionnaires were administered to students from whom samples were collected to ascertain information on how they handle toothbrushes and towels. MacConkey agar and Mannitol Salt agar were used to isolate E. coli and S. aureus respectively, and cefoxitin used to identify the methicillin-resistant S. aureus strains. Results: E. coli was present in all sampled towels, while 98% of the sampled toothbrushes contained E. coli. It was found that 2% of the respondents kept their toothbrushes in bathhouses, 44% kept them unenclosed in rooms and 54% kept them enclosed in rooms (54%). Also, 48% of the respondents washed their towels once a week, 24% washed once every two weeks, 20% once every month and 8% once a trimester. Moreover, 52% dried their towels in rooms while 48% dried them outside rooms. The occurrence of S. aureus was 96% and 94% respectively for the towels and toothbrushes. Of the S. aureus isolated, 33.3% of sampled towels and 12.8% of the toothbrushes contained methicillin-resistant S. aureus. Conclusion:&nbsp; This study found that, students are at risk of contracting infectious disease if their personal hygiene behaviours do not changed. &nbsp
