    Influence of hypokinesis on physiological functions in fowl

    The effects of hypokinesis and postincubation stress (which are characteristic for modern techniques of poultry cage keeping) on the endocrine functions, metabolic reactions, body weight growth and proteosynthesis in the muscle of cocks was investigated. The stress due to hypokinesis was observed in growing cocks housed in metallic cages in which they could hardly turn around. The findings obtained indicate that a 35-day hypokinesis did not exert any more significant influence both on physiological functions and body weight growth as well as on proteosynthesis in the muscle of cocks under study; however, it speeded up the protein metabolism in the muscle. The postincubation stress modified significantly the hypokinesis effect. Findings recorded in birds differed considerably from findings obtained in laboratory mammals, in which the hypokinesis induced significant changes in endocrine functions, body weight decrease and proteosynthesis disorders. A good tolerance of hypokinesis by fowl can be interpreted not only by the phylogenetic remoteness of the compared species but also by the domestication

    Mutations in PSEN1 and PSEN2 genes account for the majority of cases of early-onset familial Alzheimer disease. Since the first prediction of a genetic link between PSEN1 and PSEN2 with Alzheimer's disease, many research groups from both academia and pharmaceutical industry have sought to unravel how pathogenic mutations in PSEN cause presenile dementia. PSEN genes encode polytopic membrane proteins termed presenilins (PS1 and PS2), which function as the catalytic subunit of γ-secretase, an intramembrane protease that has a wide spectrum of type I membrane protein substrates. Sequential cleavage of amyloid precursor protein by BACE and γ-secretase releases highly fibrillogenic β-amyloid peptides, which accumulate in the brains of aged individuals and patients with Alzheimer's disease. Familial Alzheimer's disease-associated presenilin variants are thought to exert their pathogenic function by selectively elevating the levels of highly amyloidogenic Aβ42 peptides. In addition to Alzheimer's disease, several recent studies have linked PSEN1 to familiar frontotemporal dementia. Here, we review the biology of PS1, its role in γ-secretase activity, and discuss recent developments in the cell biology of PS1 with respect to Alzheimer's disease pathogenesis

    How tyramine β-hydroxylase controls the production of octopamine, modulating the mobility of beetles

    Biogenic amines perform many kinds of important physiological functions in the central nervous system (CNS) of insects, acting as neuromodulators, neurotransmitters, and neurohormones. The five most abundant types of biogenic amines in invertebrates are dopamine, histamine, serotonin, tyramine, and octopamine (OA). However, in beetles, an important group of model and pest insects, the role of tyramine beta-hydroxylase (T beta H) in the OA biosynthesis pathway and the regulation of behavior remains unknown so far. We therefore investigated the molecular characterization and spatiotemporal expression profiles of T beta H in red flour beetles (Triboliun castaneum). Most importantly, we detected the production of OA and measured the crawling speed of beetles after dsTcT beta H injection. We concluded that TcT beta H controls the biosynthesis amount of OA in the CNS, and this in turn modulates the mobility of the beetles. Our new results provided basic information about the key genes in the OA biosynthesis pathway of the beetles, and expanded our knowledge on the physiological functions of OA in insects
