    Gene-to-metabolite network for biosynthesis of lignans in MeJA-elicited Isatis indigotica hairy root cultures.

    Root and leaf tissue of Isatis indigotica shows notable anti-viral efficacy, and are widely used as "Banlangen" and "Daqingye" in traditional Chinese medicine. The plants' pharmacological activity is attributed to phenylpropanoids, especially a group of lignan metabolites. However, the biosynthesis of lignans in I. indigotica remains opaque. This study describes the discovery and analysis of biosynthetic genes and AP2/ERF-type transcription factors involved in lignan biosynthesis in I. indigotica. MeJA treatment revealed differential expression of three genes involved in phenylpropanoid backbone biosynthesis (IiPAL, IiC4H, Ii4CL), five genes involved in lignan biosynthesis (IiCAD, IiC3H, IiCCR, IiDIR, and IiPLR), and 112 putative AP2/ERF transcription factors. In addition, four intermediates of lariciresinol biosynthesis were found to be induced. Based on these results, a canonical correlation analysis using Pearson's correlation coefficient was performed to construct gene-to-metabolite networks and identify putative key genes and rate-limiting reactions in lignan biosynthesis. Over-expression of IiC3H, identified as a key pathway gene, was used for metabolic engineering of I. indigotica hairy roots, and resulted in an increase in lariciresinol production.     Pharmacopoeias are official sources that contain national and international rules that must be complied with legally and scientifically regarding substances, materials, drugs, devices and methods used in pharmaceutical field and pharmaceutical production. The monographs are consisted of general titles such as definition, production, characters, identification, tests, assay, storage, labelling and impurities. The World Health Organization states that sixtyeight different pharmacopoeia continue to be used effectively in fifty six countries around the world.     In vitro antioxidant activities and anti-proliferative properties of the functional herb Abrus cantoniensis and its main alkaloid abrine

    Abrus cantoniensis is a common and popular vegetative food consumed as beverage, soup and folk medicine in the tropical and subtropical areas of Asia. It has been claimed valuable for cleansing toxicants in the liver. However, the functional effects of A. cantoniensis have not yet been scientifically explored. This study comprehensively evaluated the in vitro antioxidant and anti-proliferative capacities of the herbal extract and the main alkaloid abrine. Abrine was qualitatively and quantitatively determined in methanol extract (ME) using HPLC-DAD and LC-MS/MS. The results showed that ME, ethyl acetate fraction (EF) and abrine exhibited comparable ABTS radical cation scavenging activities and reducing power to two commercial antioxidants (BHT and Trolox). The EF exerted strong cellular antioxidant activity and selective cytotoxicity against three cancer cell lines in a dose-dependent manner. Biological

    Awareness of adverse drug reactions reporting among doctors in a tertiary care centre

    Background: Adverse drug reactions (ADR) are more commonly observed during treatment course in a substantial number of patients. Though we have a functioning pharmacovigilance programme in India, we have only fewer number of adverse drug reactions being reported indicating lack of awareness, the unsure attitude and reluctance in reporting adverse drug reactions.Methods: We conducted a cross sectional pretested questionnaire based study for a period of six months. 600 questionnaires were distributed. 521 responded back to the questionnaire of which 29.6% were interns, 56.8% were junior residents, 5.2% doctors were senior residents, and 8.4% were Assistant professors. Awareness on ADR reporting, ADRs encountered in practice, common causative drugs for ADR, and awareness on the functions of Pharmacovigilance programme of India were assessed.Results: 44.9% were not aware of the pharmacovigilance programme; 59.5% of the study population were not aware of the voluntary reporting of ADRs. 100% of the doctors who participated actively in the study responded that have not reported any ADR so far under pharmacovigilance programme of India. 88.1% of the doctors who participated in the study were willing to undergo a training on ADR reporting and pharmacovigilance.Conclusions: This study concludes that serious measures have to be taken to educate the doctors on ADR reporting and pharmacovigilance. By means of involving pharmacovigilance as a part of the undergraduate programme and by organising regular workshops, group discussion and continuous medical education, we can improve the awareness among doctors regarding ADR reporting and pharmacovigilance

    Simultaneous qualitative and quantitative analysis of Tangkening granule using thin layer chromatography (TLC) and high performance liquid chromatography (HPLC)

    This study was designed to establish the quality standard of Tangkening granule in order to effectively control the quality of this new Chinese traditional patent medicine in the process of production, in addition to ensure the good clinical application. Fructus gardenia, Herba gynostemmatis, Dioscorea batatas, and Polygonatum odoratum in Tangkening granule were identified by thin layer chromatography (TLC), and the content of geniposide was determined by high performance liquid chromatography (HPLC). A comprehensive validation of the method that included sensitivity, linearity, repeatability and recovery was conducted. The results show that this analytical method was simple and suitable for the original identification and quality control of this new Chinese traditional patent medicine.Key words: Tangkening granule, geniposide, quality standard, thin layer chromatography, high performance liquid chromatography

    Dose-related liver injury of Geniposide associated with the alteration in bile acid synthesis and transportation.

    Fructus Gardenia (FG), containing the major active constituent Geniposide, is widely used in China for medicinal purposes. Currently, clinical reports of FG toxicity have not been published, however, animal studies have shown FG or Geniposide can cause hepatotoxicity in rats. We investigated Geniposide-induced hepatic injury in male Sprague-Dawley rats after 3-day intragastric administration of 100 mg/kg or 300 mg/kg Geniposide. Changes in hepatic histomorphology, serum liver enzyme, serum and hepatic bile acid profiles, and hepatic bile acid synthesis and transportation gene expression were measured. The 300 mg/kg Geniposide caused liver injury evidenced by pathological changes and increases in serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and γ-glutamytransferase (γ-GT). While liver, but not sera, total bile acids (TBAs) were increased 75% by this dose, dominated by increases in taurine-conjugated bile acids (t-CBAs). The 300 mg/kg Geniposide also down-regulated expression of Farnesoid X receptor (FXR), small heterodimer partner (SHP) and bile salt export pump (BSEP). In conclusion, 300 mg/kg Geniposide can induce liver injury with associated changes in bile acid regulating genes, leading to an accumulation of taurine conjugates in the rat liver. Taurocholic acid (TCA), taurochenodeoxycholic acid (TCDCA) as well as tauro-α-muricholic acid (T-α-MCA) are potential markers for Geniposide-induced hepatic damage

    Rapid Determination of Saponins in the Honey-Fried Processing of Rhizoma Cimicifugae by Near Infrared Diffuse Reflectance Spectroscopy.

    ObjectiveA model of Near Infrared Diffuse Reflectance Spectroscopy (NIR-DRS) was established for the first time to determine the content of Shengmaxinside I in the honey-fried processing of Rhizoma Cimicifugae.MethodsShengmaxinside I content was determined by high-performance liquid chromatography (HPLC), and the data of the honey-fried processing of Rhizoma Cimicifugae samples from different batches of different origins by NIR-DRS were collected by TQ Analyst 8.0. Partial Least Squares (PLS) analysis was used to establish a near-infrared quantitative model.ResultsThe determination coefficient R² was 0.9878. The Cross-Validation Root Mean Square Error (RMSECV) was 0.0193%, validating the model with a validation set. The Root Mean Square Error of Prediction (RMSEP) was 0.1064%. The ratio of the standard deviation for the validation samples to the standard error of prediction (RPD) was 5.5130.ConclusionThis method is convenient and efficient, and the experimentally established model has good prediction ability, and can be used for the rapid determination of Shengmaxinside I content in the honey-fried processing of Rhizoma Cimicifugae
