    Perseguindo o sonho europeu: a experiência educacional de jovens Africano desacompanhadas de Centro de Recepção, nas Ilhas Canárias, e mais além.

    In the last two decades, Spain’s Canary Islands have received thousands of undocumented migrants arriving by boat from the coasts of North and West Africa. The sharp increase of unaccompanied minors has presented a particular challenge, as these minors fall under the State’s protection system and are entitled to an education and other rights, once in Spain. What economic and socio-cultural factors push these youth to seek a better life whilst endangering their own? What educational opportunities are available to them in Spain and how can these propel them into secure living situations? What can be said about their integration prospects?This ethnographic study, based on field research carried out in a reception centre for unaccompanied minors in Tenerife, focuses on the youth’s migration trajectories, and on the extent to which the education and support they receive in Spain relates to their socio-economic integration into European society and beyond. The findings reveal that although the centre provides the youth with a window of opportunity to be in Spain and gain an education, the ambiguousness of their legal situation brought about by immigration policies once they have left the centre, is not conducive to their leading a stable and productive life in the current context.En las dos últimas décadas, las Islas Canarias han recibido miles de inmigrantes indocumentados, que llegan a las costas en botes desde el Norte y el Oeste de África. El aumento de menores extranjeros no acompañados presenta un desafío para el gobierno, pues al tratarse de menores en territorio español, el estado es el responsable de su guarda y tutela, y tiene que facilitarles protección y educación como cualquier otro menor. ¿Qué factores económicos y socioculturales empujan a estos jóvenes a poner en peligro sus vidas?; ¿qué oportunidades educativas tienen ellos en España, y cómo pueden ellos aprovechar esas situaciones para alcanzar un nivel mayor de bienestar?; ¿qué posibilidades de integración tienen en la sociedad española? Este estudio etnográfico, basado en una investigación llevada a cabo en un Centro de Acogida de Menores No Acompañados de Tenerife, se centra en las trayectorias migratorias de estos jóvenes, y en analizar en qué medida la educación y apoyo que reciben en España influye en su posterior integración laboral y social. Los resultados reflejan que, aunque el centro ofrece a los jóvenes bienestar y oportunidades educativas, la ambigüedad de sus situaciones jurídicas al cumplir la mayoría de edad, dependientes de las políticas de migración, no facilita mucho su desarrollo educativo e integración social posterior en el presente contexto.Nas últimas duas décadas, as Ilhas Canárias têm recebido milhares de imigrantes ilegais que chegam à costa em barcos do Norte e África Ocidental. Que fatores econômicos e socioculturais empurrar esses jovens colocam suas vidas em perigo?; Que oportunidades de educação que têm em Espanha, e como eles podem tirar proveito dessas situações para alcançar um maior nível de bem-estar?; Que chance tem integração na sociedade espanhola?     Identification of Genes Regulating Gene Targeting by a High-Throughput Screening Approach

    Homologous gene targeting (HGT) is a precise but inefficient process for genome engineering. Several methods for increasing its efficiency have been developed, including the use of rare cutting endonucleases. However, there is still room for improvement, as even nuclease-induced HGT may vary in efficiency as a function of the nuclease, target site, and cell type considered. We have developed a high-throughput screening assay for the identification of factors stimulating meganuclease-induced HGT. We used this assay to explore a collection of siRNAs targeting 19,121 human genes. At the end of secondary screening, we had identified 64 genes for which knockdown affected nuclease-induced HGT. Two of the strongest candidates were characterized further. We showed that siRNAs directed against the ATF7IP gene, encoding a protein involved in chromatin remodeling, stimulated HGT by a factor of three to eight, at various loci and in different cell types. This method thus led to the identification of a number of genes, the manipulation of which might increase rates of targeted recombination

    miRNA Profiling: How to Bypass the Current Difficulties in the Diagnosis and Treatment of Sarcomas

    Sarcomas are divided into a group with specific alterations and a second presenting a complex karyotype, sometimes difficult to diagnose or with few therapeutic options available. We assessed if miRNA profiling by TaqMan low density arrays could predict the response of undifferentiated rhabdomyosarcoma (RMS) and osteosarcoma to treatment. We showed that miRNA signatures in response to a therapeutic agent (chemotherapy or the mTOR inhibitor RAD-001) were cell and drug specific on cell lines and a rat osteosarcoma model. This miRNA signature was related to cell or tumour sensitivity to this treatment and might be not due to chromosomal aberrations, as revealed by a CGH array analysis of rat tumours. Strikingly, miRNA profiling gave promising results for patient rhabdomyosarcoma, discriminating all types of RMS: (Pax+) or undifferentiated alveolar RMS as well as embryonal RMS. As highlighted by these results, miRNA profiling emerges as a potent molecular diagnostic tool for complex karyotype sarcomas

    A systems approach towards remote health-monitoring in older adults: Introducing a zero-interaction digital exhaust.

    Using connected sensing devices to remotely monitor health is a promising way to help transition healthcare from a rather reactive to a more precision medicine oriented proactive approach, which could be particularly relevant in the face of rapid population ageing and the challenges it poses to healthcare systems. Sensor derived digital measures of health, such as digital biomarkers or digital clinical outcome assessments, may be used to monitor health status or the risk of adverse events like falls. Current research around such digital measures has largely focused on exploring the use of few individual measures obtained through mobile devices. However, especially for long-term applications in older adults, this choice of technology may not be ideal and could further add to the digital divide. Moreover, large-scale systems biology approaches, like genomics, have already proven beneficial in precision medicine, making it plausible that the same could also hold for remote-health monitoring. In this context, we introduce and describe a zero-interaction digital exhaust: a set of 1268 digital measures that cover large parts of a person's activity, behavior and physiology. Making this approach more inclusive of older adults, we base this set entirely on contactless, zero-interaction sensing technologies. Applying the resulting digital exhaust to real-world data, we then demonstrate the possibility to create multiple ageing relevant digital clinical outcome assessments. Paired with modern machine learning, we find these assessments to be surprisingly powerful and often on-par with mobile approaches. Lastly, we highlight the possibility to discover novel digital biomarkers based on this large-scale approach

    European Language Grid: A Joint Platform for the European Language Technology Community

    Europe is a multilingual society, in which dozens of languages are spoken. The only option to enable and to benefit from multilingualism is through Language Technologies (LT), i.e., Natural Language Processing and Speech Technologies. We describe the European Language Grid (ELG), which is targeted to evolve into the primary platform and marketplace for LT in Europe by providing one umbrella platform for the European LT landscape, including research and industry, enabling all stakeholders to upload, share and distribute their services, products and resources. At the end of our EU project, which will establish a legal entity in 2022, the ELG will provide access to approx. 1300 services for all European languages as well as thousands of data sets

    Inhibition of Chondrosarcoma Growth by mTOR Inhibitor in an In Vivo Syngeneic Rat Model

    BACKGROUND: Chondrosarcomas are the second most frequent primary malignant type of bone tumor. No effective systemic treatment has been identified in advanced or adjuvant phases for chondrosarcoma. The aim of the present study was to determine the antitumor effects of doxorubicin and everolimus, an mTOR inhibitor on chondrosarcoma progression. METHODS AND FINDINGS: Doxorubin and/or everolimus were tested in vivo as single agent or in combination in the rat orthotopic Schwarm chondrosarcoma model, in macroscopic phase, as well as with microscopic residual disease. Response to everolimus and/or doxorubicin was evaluated using chondrosarcoma volume evolution (MRI). Histological response was evaluated with % of tumor necrosis, tumor proliferation index, metabolism quantification analysis between the treated and control groups. Statistical analyses were performed using chi square, Fishers exact test. Doxorubicin single agent has no effect of tumor growth as compared to no treatment; conversely, everolimus single agent significantly inhibited tumor progression in macroscopic tumors with no synergistic additive effect with doxorubicin. Everolimus inhibited chondrosarcoma proliferation as evaluated by Ki67 expression did not induce the apoptosis of tumor cells; everolimus reduced Glut1 and 4EBP1 expression. Importantly when given in rats with microscopic residual diseases, in a pseudo neoadjuvant setting, following R1 resection of the implanted tumor, everolimus significantly delayed or prevented tumor recurrence. CONCLUSIONS: MTOR inhibitor everolimus blocks cell proliferation, Glut1 expression and HIF1a expression, and prevents in vivo chondrosarcoma tumor progression in both macroscopic and in adjuvant phase post R1 resection. Taken together, our preclinical data indicate that mTOR inhibitor may be effective as a single agent in treating chondrosarcoma patients. A clinical trial evaluating mTOr inhibitor as neo-adjuvant and adjuvant therapy in chondrosarcoma patients is being constructed

    Dengue Virus Type 2 Infections of Aedes aegypti Are Modulated by the Mosquito's RNA Interference Pathway

    A number of studies have shown that both innate and adaptive immune defense mechanisms greatly influence the course of human dengue virus (DENV) infections, but little is known about the innate immune response of the mosquito vector Aedes aegypti to arbovirus infection. We present evidence here that a major component of the mosquito innate immune response, RNA interference (RNAi), is an important modulator of mosquito infections. The RNAi response is triggered by double-stranded RNA (dsRNA), which occurs in the cytoplasm as a result of positive-sense RNA virus infection, leading to production of small interfering RNAs (siRNAs). These siRNAs are instrumental in degradation of viral mRNA with sequence homology to the dsRNA trigger and thereby inhibition of virus replication. We show that although dengue virus type 2 (DENV2) infection of Ae. aegypti cultured cells and oral infection of adult mosquitoes generated dsRNA and production of DENV2-specific siRNAs, virus replication and release of infectious virus persisted, suggesting viral circumvention of RNAi. We also show that DENV2 does not completely evade RNAi, since impairing the pathway by silencing expression of dcr2, r2d2, or ago2, genes encoding important sensor and effector proteins in the RNAi pathway, increased virus replication in the vector and decreased the extrinsic incubation period required for virus transmission.     Standardization of in vitro digestibility and DIAAS method based on the static INFOGEST protocol

    Background: The FAO recommends the digestible indispensable amino acid score (DIAAS) as the measure for protein quality, for which the true ileal digestibility needs to be assessed in humans or pigs. However, due to high costs and ethical concerns, the FAO strongly encourages as well the development of validated in vitro methods, which complement the in vivo experiments. Method: Recently, an in vitro workflow, based on the validated static INFOGEST protocol, was developed and compared towards in vivo data. In parallel to the validation with in vivo data, the repeatability and reproducibility of the in vitro protocol were tested in an international ring trial (RT) with the aim to establish an international ISO standard method within the International Dairy Federation (IDF). Five different dairy products (skim milk powder, whole milk powder, whey protein isolate, yoghurt, and cheese) were analyzed in 32 different laboratories from 18 different countries, across 4 continents. Results: in vitro protein digestibilities based on Nitrogen, free R-NH2, and total amino acids as well as DIAAS values were calculated and compared to in vivo data, where available.
