    Transfer matrix solution of the Wako-Sait\^o-Mu\~noz-Eaton model augmented by arbitrary short range interactions

    The Wako-Sait{\^o}-Mu\~noz-Eaton (WSME) model, initially introduced in the theory of protein folding, has also been used in modeling the RNA folding and some epitaxial phenomena. The advantage of this model is that it admits exact solution in the general inhomogeneous case (Bruscolini and Pelizzola, 2002) which facilitates the study of realistic systems. However, a shortcoming of the model is that it accounts only for interactions within continuous stretches of native bonds or atomic chains while neglecting interstretch (interchain) interactions. But due to the biopolymer (atomic chain) flexibility, the monomers (atoms) separated by several non-native bonds along the sequence can become closely spaced. This produces their strong interaction. The inclusion of non-WSME interactions into the model makes the model more realistic and improves its performance. In this study we add arbitrary interactions of finite range and solve the new model by means of the transfer matrix technique. We can therefore exactly account for the interactions which in proteomics are classified as medium- and moderately long-range ones.Comment: 15 pages, 2 figure

    Theory of temperature dependence of the Fermi surface-induced splitting of the alloy diffuse-scattering intensity peak

    The explanation is presented for the temperature dependence of the fourfold intensity peak splitting found recently in diffuse scattering from the disordered Cu3Au alloy. The wavevector and temperature dependence of the self-energy is identified as the origin of the observed behaviour. Two approaches for the calculation of the self-energy, the high-temperature expansion and the alpha-expansion, are proposed.     Design and construction of new central and forward muon counters for CDF II

    New scintillation counters have been designed and constructed for the CDF upgrade in order to complete the muon coverage of the central CDF detector, and to extend this coverage to larger pseudorapidity. A novel light collection technique using wavelength shifting fibers, together with high quality polystyrene-based scintillator resulted in compact counters with good and stable light collection efficiency over lengths extending up to 320 cm. Their design and construction is described and results of their initial performance are reported.Comment: 20 pages, 15 figure

    Vitellogenin RNAi Halts Ovarian Growth and Diverts Reproductive Proteins and Lipids in Young Grasshoppers

    Reduced reproduction extends lifespan of females in many animals. To test the effects of reproduction on storage of macronutrients, we block reproductive output in the lubber grasshopper by injecting RNAi against the precursor to egg-yolk protein, vitellogenin, in early adulthood. Controls were injected with either buffer or RNAi against the major storage protein in the hemolymph, hexamerin-90. Vitellogenin RNAi greatly reduced both levels of mRNA for vitellogenin and ovarian growth, in comparison to both controls. Fat body mass was increased upon vitellogenin RNAi, but concentrations of the three hexameric storage proteins from the hemolymph were not. Surprisingly, hemolymph vitellogenin levels were increased upon vitellogenin RNAi. Total reproductive protein (hemolymph vitellogenin plus ovarian vitellin) was unchanged by vitellogenin RNAi, as reproductive protein was diverted to the hemolymph. Similarly, the increased lipid storage upon vitellogenin RNAi was largely attributable to the reduction in lipid in the ovary, due to decreased ovarian growth. A BLAST search revealed that the 515 bp sequence of vitellogenin used for RNAi had three 11 bp regions identical to the vitellogenin receptor of the cockroach Leucophaea maderae. This suggests that our treatment, in addition to reducing levels of vitellogenin transcript, may have also blocked transport of vitellogenin from the hemolymph to the ovary. This would be consistent with halted ovarian growth simultaneous with high levels of vitellogenin in the hemolymph. Nonetheless, the accumulation of vitellogenin, instead of hexameric storage proteins, is inconsistent with a simple model of the trade-off between reproduction and storage. This was observed in young females; future studies will address whether investment of proteins may shift to the soma as individuals age. Overall, our results suggest that blockage of reproduction in young grasshoppers redirects lipids to storage and reproductive proteins to the hemolymph

    Depot Medroxyprogesterone Acetate Use and Blood Lead Levels in a Cohort of Young Women

    BACKGROUND: Injectable contraceptive use is common, with 74 million users worldwide. Use of the injectable contraceptive depot medroxyprogesterone acetate (DMPA) is associated with bone mineral density loss. We hypothesize that increased bone resorption with DMPA use allows for mobilization of the toxic metal lead stored in bone to blood, presenting users with increased systemic exposure to lead. OBJECTIVE: The objective of our study was to investigate the association between current DMPA use and blood lead concentrations. METHODS: We conducted a cross-sectional analysis using enrollment data from the Study of Environment, Lifestyle & Fibroids (SELF), a cohort of 1,693 African-American women who were 23-35 years of age. Data on DMPA use were collected by computer-assisted telephone interview. Blood lead concentrations were measured in whole blood samples among 1,548 participants (91% of cohort). We estimated the adjusted percent difference in blood lead concentrations and 95% confidence intervals (CI) between current DMPA users and nonusers using multivariable linear regression. RESULTS: Geometric mean blood lead concentration was 0.69 μg/dL (95% CI: 0.67, 0.71). After adjustment, current DMPA users (7% of cohort) had blood lead concentrations that were 18% higher than those of nonusers (95% CI: 8%, 29%). Similar associations were observed with additional analyses to assess for potential bias from smoking, DMPA-induced amenorrhea, use of estrogen-containing contraceptives, having given birth in the prior year, and history of medical conditions or current medication use associated with bone loss./ DISCUSSION: Our results indicate that current DMPA use is associated with increased blood lead concentrations. Further research, particularly in populations highly exposed to lead, is warranted to consider tradeoffs between the adverse effects of lead on human health and the importance of DMPA as a contraceptive option to prevent unintended pregnancy. https://doi.org/10.1289/EHP7017

    Temperature and velocity measurements and predictive model comparisons in the near-field region of surface thermal discharges

    Simultaneous temperature and velocity measurements were made in the near- field region of the surface thermal discharge at the Point Beach Unit 1 and Palisades Nuclear Power Plants on Lake Michigan. Data collected include measurements of temperature and velocity at the 0.5-, 1.0-, 1.5-, 2.0-, 2.5-, and 3.0-m depths, along with measurements of ambient lake and metenrological conditions. Bottom depth was also measured at variouss locations. four such surveys were made at the Point Beach plant, three at the Palisades plant. To examine the jet features from the abpve surveys and facilitate a comparison with analytical model predictions, a computer program was written to smooth the data, extracting such jet characteristics as trajectory, centerline temperature decay and temperature half-widths, centerline velocitv decay and velocity half-widths, temperature and velocity half-depths, and isotherm areas. Four near-field analytical models often used in environmental impact evaluations of power-plant surface discharges are compared to the jet characteristics determined from the smoothed jet data. The Pritchard model compares rather well with these limited data and is often conservative when model-data discrepancies exist. The Stolzenbach -Harleman and Prych models predict too rapid a temperature and velocity decay accompanying too great a lateral spread. The Motz --Benedict model is too sensitive to an entrainment coefficient E with little consistent data available for its determination for accurate prediction.
