    Parler and the Road to the Capitol Attack: Investigating Alt-Tech Ties to January 6

    The January 6, 2021 mob assault on the U.S. Capitol exposed deep fissures between Americans and shook the very foundations of the country. The violence that day and the tech industry's response to the tsunami of polarizing content triggered a major public debate over how social media and tech companies manage their platforms and services and the impact of content moderation policies on polarization, extremism, and political violence in the United States. That debate is also now playing out in Congress where the House Select Committee to Investigate the January 6th Attack on the United States Capitol is now underway. One big question is: How did niche social media sites geared toward far-right audiences, like Parler, contribute to polarization around the 2020 presidential election and to what extent did Parler and other platforms factor into the January 6 attack? The first in a series of investigations into the impact of the alt-tech movement on U.S. national security, this report provides an initial snapshot of observations culled from an ongoing analysis of open source data related to the Capitol attack.Based, in part, on an early assessment of a cache of an estimated 183 million Parler posts publicly archived after Parler was temporarily deplatformed, the analysis in this report offers unique insights into online and offline early warning signs of the potential for election-related violence in the year-long run up to the Capitol attack. On the streets and online, the networked effects of poor platform governance across the internet during the 2020 presidential election were notable on mainstream and fringe social media sites. Nevertheless, the combined impact of Parler's loose content moderation scheme as well as data-management practices and platform features—either by design or neglect, or both—may have made the social media startup especially vulnerable to strategic influence campaigns that relied heavily on inauthentic behavior like automated content amplification and deceptive techniques like astroturfing

    Postsecondary Completion in Rural Texas: A Statewide Overview

    Educate Texas, our client, is a partner and key player in postsecondary education in Texas. The nonprofit seeks to improve postsecondary completion statewide. Under their mission, our capstone was charged with assessing the state of postsecondary completion in Rural Texas. Using a mixed methods approach, the capstone studied institutional, attitudinal, and academic barriers that impede rural students from pursuing and obtaining a postsecondary credential. Why should policymakers and stakeholders focus specifically on the rural student population in Texas for postsecondary enrollment and completion? Texas ranks below the national average in postsecondary educational enrollment and completion. Texas needs to “enroll immediately 200,000 more students” to reach the national average, according to Closing the Gaps, the Texas Higher Education Coordinator Board strategic plan. Within the state, the issue also applies to rural students. Only 26% of the most recent Texas high school graduating class tracked as a cohort ended up earning a postsecondary credential. There are 834,000 rural K-12 students in Texas, which is 18% of the total K-12 enrollment. Approximately 43% of the rural K-12 student population is considered low income. With the largest population of rural students in the country, Texas has a unique opportunity to capitalize on the success of these youth completing a postsecondary credential.Educate Texa

    Trends in the perceived body size of adolescent males and females in Scotland, 1990–2014: changing associations with mental well-being

    Objectives: This paper explores trends in Scottish adolescents’ body size perceptions and associated mental well-being outcomes. Methods: Data were collected on Scottish 11, 13 and 15-year olds by the Health Behaviour in School-aged Children study between 1990 and 2014 (n=42,312). Logistic regression was used to examine changes in the prevalence of over- and underweight perceptions. Ordinal and linear regression was used to examine changes in the association between body perception and mental well-being. Results: Little change was observed in over- or under-weight perceptions between 1990 and 2014. However, relative to those perceiving their body as ‘about right’, those perceiving themselves as overweight reported decreasing confidence (all groups), decreasing happiness (11- and 13-year old girls) and increasing psychological symptoms (all girls and 15 year-old boys). Perceived underweight is associated with poor well-being, especially in males, but we present little evidence that this is a recent phenomenon. Conclusions: We present evidence suggesting that the influence of body image on adolescent mental health is increasing over time. This may play a role in the recently observed worsening of mental well-being in Scottish adolescents.Publisher PDFPeer reviewe

    Pan-Cancer Analysis of lncRNA Regulation Supports Their Targeting of Cancer Genes in Each Tumor Context

    Long noncoding RNAs (lncRNAs) are commonly dys-regulated in tumors, but only a handful are known toplay pathophysiological roles in cancer. We inferredlncRNAs that dysregulate cancer pathways, onco-genes, and tumor suppressors (cancer genes) bymodeling their effects on the activity of transcriptionfactors, RNA-binding proteins, and microRNAs in5,185 TCGA tumors and 1,019 ENCODE assays.Our predictions included hundreds of candidateonco- and tumor-suppressor lncRNAs (cancerlncRNAs) whose somatic alterations account for thedysregulation of dozens of cancer genes and path-ways in each of 14 tumor contexts. To demonstrateproof of concept, we showed that perturbations tar-geting OIP5-AS1 (an inferred tumor suppressor) andTUG1 and WT1-AS (inferred onco-lncRNAs) dysre-gulated cancer genes and altered proliferation ofbreast and gynecologic cancer cells. Our analysis in-dicates that, although most lncRNAs are dysregu-lated in a tumor-specific manner, some, includingOIP5-AS1, TUG1, NEAT1, MEG3, and TSIX, synergis-tically dysregulate cancer pathways in multiple tumorcontexts

    Pan-cancer Alterations of the MYC Oncogene and Its Proximal Network across the Cancer Genome Atlas

    Although theMYConcogene has been implicated incancer, a systematic assessment of alterations ofMYC, related transcription factors, and co-regulatoryproteins, forming the proximal MYC network (PMN),across human cancers is lacking. Using computa-tional approaches, we define genomic and proteo-mic features associated with MYC and the PMNacross the 33 cancers of The Cancer Genome Atlas.Pan-cancer, 28% of all samples had at least one ofthe MYC paralogs amplified. In contrast, the MYCantagonists MGA and MNT were the most frequentlymutated or deleted members, proposing a roleas tumor suppressors.MYCalterations were mutu-ally exclusive withPIK3CA,PTEN,APC,orBRAFalterations, suggesting that MYC is a distinct onco-genic driver. Expression analysis revealed MYC-associated pathways in tumor subtypes, such asimmune response and growth factor signaling; chro-matin, translation, and DNA replication/repair wereconserved pan-cancer. This analysis reveals insightsinto MYC biology and is a reference for biomarkersand therapeutics for cancers with alterations ofMYC or the PMN

    Spatial Organization and Molecular Correlation of Tumor-Infiltrating Lymphocytes Using Deep Learning on Pathology Images

    Beyond sample curation and basic pathologic characterization, the digitized H&E-stained images of TCGA samples remain underutilized. To highlight this resource, we present mappings of tumorinfiltrating lymphocytes (TILs) based on H&E images from 13 TCGA tumor types. These TIL maps are derived through computational staining using a convolutional neural network trained to classify patches of images. Affinity propagation revealed local spatial structure in TIL patterns and correlation with overall survival. TIL map structural patterns were grouped using standard histopathological parameters. These patterns are enriched in particular T cell subpopulations derived from molecular measures. TIL densities and spatial structure were differentially enriched among tumor types, immune subtypes, and tumor molecular subtypes, implying that spatial infiltrate state could reflect particular tumor cell aberration states. Obtaining spatial lymphocytic patterns linked to the rich genomic characterization of TCGA samples demonstrates one use for the TCGA image archives with insights into the tumor-immune microenvironment
