    Infection- and procedure-dependent effects on pulmonary gene expression in the early phase of influenza A virus infection in mice

    BACKGROUND: Investigating the host response in the early stage of influenza A virus (IAV) infection is of considerable interest. However, it is conceivable that effects due to the anesthesia and/or intranasal infection procedure might introduce artifacts. We therefore aimed to evaluate the effects of anesthesia and/or intranasal infection on transcription of selected pulmonary mRNAs in two inbred mouse strains with differential susceptibility to IAV infection. RESULTS: DBA/2J and C57BL/6J mice were evaluated in a time course experiment in which lung tissue was sampled after 6, 12, 18, 24, 48 and 120 h. After anesthesia with ketamine and xylazine, a suspension of mouse-adapted IAV strain PR8_Mun in 20 μl sterile buffer, or 20 μl sterile buffer only, was instilled intranasally. The mice receiving anesthesia and PBS only were designated the “mock treatment” group. Pulmonary expression of 10 host mRNAs (Fos, Retnla, Irg1, Il6, Il1b, Cxcl10, Stat1, Ifng, Ifnl2, and Mx1) and viral hemagglutinin (HA) mRNA were determined at the designated time points. As expected, weight loss and viral replication were greater in the DBA/2J strain (which is more susceptible to IAV infection). Four mRNAs (Retnla, Irg1, Il6, and Cxcl10) were procedure-dependently regulated in DBA/2J mice between 6 and 24 h, and two (Retnla and Il6) in C57BL/6J mice, although to a lesser extent. All 10 mRNAs rose after infection, but one (Fos) only in DBA/2J mice. These infection-dependent effects could be separated from procedure-dependent effects beginning around 12 h in DBA/2J and 18 h in C57BL/6J mice. The interferon-related mRNAs Stat1, Ifng, Infl2, and Mx1 were unaffected by mock treatment in either mouse strain. Mx1 and Infl2 correlated best with HA mRNA expression (r = 0.97 and 0.93, respectively, in DBA/2J). CONCLUSIONS: These results demonstrate effects of the anesthesia and/or intranasal infection procedure on pulmonary gene expression, which are detectable between approximately 6 and 24 h post procedure and vary in intensity and temporal evolution depending on the mouse strain used. Mock infection controls should be included in all studies on pulmonary gene expression in the early phase of infection with IAV and, likely, other respiratory pathogens

    Pramipexole protective effect on rotenone induced neurotoxicity in mice

    Introduction: 
Pramipexole is a new dopaminergic drug which has been approved for PD treatment. However, we tried to find a new capacity for this drug rather than symptomatic effect. 

Materials and Methods: 
A chronic rotenone model with daily oral dose of 30mg/kg was induced in mice. Pramipexole was tried in a new approach where the treatment began in the middle of rotenone course with oral dose 1mg/kg/day of pramipexole. 

Results: 
Further analysis of behavioral tests and immunohistochemistry revealed success of pramipexole in improving the rotenone intoxicated mice. 

Conclusion: 
These results showed possible beneficial effects of pramipexole against rotenone-induced neurotoxicity

    DNA Fingerprinting, Chemical Composition, Antitumor and Antimicrobial Activities of the Essential Oils and Extractives of four Annona Species from Egypt

    The leaf essential oils of four members of the Annonaceae grown in Egypt (namely; Annona cherimola, A. squamosa, A. muricata and A. glabra) have been obtained by hydrodistillation and analyzed by GC-MS in order to compare and contrast the volatile chemical compositions of these species.  The essential oils were screened for in-vitro cytotoxic activity against breast cancer (MCF-7), colon cancer (CACO) and liver cancer (HEPG2) cell lines and antimicrobial activity against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeuroginosa, Aspergillus flavus and Candida albicans. beta-elemene (25.02%), beta-caryophyllene (37.11%), bicycloelemene (23.58%) and and beta-gurjunene (42.49%), were the major constituents of Annona cherimola, A. squamosa, A. muricata and A. glabra, respectively. Ethanol extracts showed highly significant cytotoxic activities (low IC50) much more than results displayed by essential oils on breast (MCF-7), colon (CACO) and liver (HEPG2) carcinoma cell lines. Relative to breast carcinoma cell line (MCF-7), The IC50 values of ethanol extracts were 3.43, 3.89 and 4.34 ?g/ml for A. cherimola, A. squamosa and A. muricata ethanol extractives respectively. While colon carcinoma cell line (CACO) displayed IC50 values 2.82, 2.97, 3.58 and 3.89 ?g/ml for A. muricata, A. cherimola, A. glabra and A. squamosa, respectively. Liver carcinoma cell line (HEPG2) exhibited IC50 valus of 3.12, 3.43 and 3.73 for A. squamosa, A. muricata and A. cherimola, respectively. Three of the four leaf essential oils showed notable invitro cytotoxic activity. Essential oils of A. glabra, A. muricata and A. squamosa showed moderate cytotoxic activities with IC50 values ranging from 12.35 to 24.21?g/ml. While the essential oils of A. cherimola showed IC50 values ranging from 7.67 to 9.22?g/ml. Leaf oils and ethanol extractives showed appreciable antibmicobial activity with variable MIC ranging from 30 to 315µg/ml. These findings suggest that A. cherimola essential oil and ethanol extract have great potential as a natural medicine for cancers and microbial infections.     Practical Needs in Assessing Response to Therapy in Operated Brain High Grade Glioma According to Response Assessment in Neuro-Oncology Criteria

    Background: Tumors known as gliomas begin in the brain or spine's glial cells and might spread throughout the body.  An effort was made to improve tumor response evaluation and end point selection through the Response Assessment in Neuro-Oncology (RANO) working group. Objective: To standardize the assessment method and results reporting by applying MRI-RANO criteria in detecting glioma response to surgical treatment. Patients and Methods: This was a prospective cohort study included 12 patients (4 males and 8 females) whose mean age was 53.6 ± 15.1years. Included patients are those who were operated and pathologically proved as brain glioma grade IV (GBM). They underwent post-operative MRI within 48 hours and are available for follow up MRI after 3 months. Imaging modalities applied in both MRI exams were conventional magnetic resonance imaging, MRI diffusion weighted images, and contrast imaging. RANO criteria were applied. Results: According to RANO criteria, none of the lesions had pseudo response or pseudo progression. However, two thirds (66.7%) had progressive disease and one third (33.3%) had complete response. There was statistically significant association between MRI findings 3 months postoperative and RANO criteria as patients with compete response had statistically significant decrease on measurable solid components, diffusion restriction, post contrast enhancement and all lesions had grade I edema with no mass effect or midline shift compared to patients with progressive disease. 4 cases that showed complete response category had fulfilled all the RANO criteria, while 8 cases with category progressive disease had fulfilled more than one of the described RANO criteria. Conclusion: RANO criteria are an effective tool to be used in interpretation of MRI for follow-up of surgically operated glioma patients to detect their response

    Hemodialysis associated dysautonomia; Effect of optimization of dialysis and nutrition: A prospective study

    Background: Autonomic neuropathy is common among hemodialysis patients, even if they are asymptomatic. The aim of this study is to assess the effect of optimization of patients' dialysis and nutrition on the presence of autonomic dysfunction in these patients guided by sympathetic skin response (SSR) test. Methods: Fifteen patients on maintenance hemodialysis (12 males and 3 females) their age ranged from 24 to 67 years, most of them were on bicarbonate dialysis were included in our study. Initially, the patients were assessed clinically and by laboratory investigations and their dialysis was assessed by studying their urea kinetic modeling. Their nutrition was assessed by laboratory parameters and by calculating the normalized protein catabolic rate (nPCR). Their autonomic functions were assessed by clinical examination, hand grip and SSR test. Dialysis dose was readjusted to achieve a target kt/v value of 1.3/session thrice weekly. Also, their nutrition was reviewed to achieve nPCR of 1.2 gm/kg/day and caloric intake of 30-40 KCal/kg/day through diet manipulation and support. They were reassessed after 3 months. Results: Analysis of the data showed a statistically significant improvement of the observed subclinical autonomic neuropathy evidenced by a significant change in the hand grip (p = 0.044), and a high statistically significant improvement in the sympathetic skin response test parameters (both amplitude and latency) (p=0.001) after optimization of both hemodialysis and nutrition status. Conclusion: Improving dialysis and patients nutrition by using urea kinetic modeling is valuable in improving their autonomic functions as assessed by SSR test
