    Understanding Ambidexterity: Managing Contradictory Tensions Between Exploration and Exploitation in the Evolution of Digital Infrastructure

    Prior research on the evolution of digital infrastructure has paid considerable attention to effective strategies for resolving contradictory tensions, yet what we still do not understand is the role of higher-level organizational capabilities that help balance the contradictory tensions that emerge during this evolution. In addressing this gap, two related questions guided our investigation: (1) How do organizations experience and resolve contradictory tensions throughout the evolution of digital infrastructure? and (2) What can we learn about the organizational capabilities that drive strategic actions in resolving these contradictory tensions? We approach these questions using an in-depth case study at RE/MAX LLC, a global real estate franchise. Based on our findings, we propose a theoretical model of digital infrastructure ambidexterity. The model recognizes three pairs of capabilities (identifying and germinating, expanding and legitimizing, and augmenting and implanting) and two supporting factors (leadership and structure) that are key to resolving contradictory tensions during this evolution. This study responds to a recent research call for dynamic process perspectives at multiple levels of analysis. We discuss the implications of this model for research and practice and offer observations for future research

    Transition Program for Adolescents with Congenital Heart Disease Focusing on Self-Management Knowledge

    Introduction: The primary goal of this project was to implement a pilot transition education program to prepare adolescents with congenital heart defects (CHD) by increasing knowledge about self-management of their health condition. Method: An educational program was developed and included use of an interactive binder. A total of 20 adolescents with CHD between the ages of 16 – 18 years participated in transition education during a single outpatient cardiology visit with pre and post education knowledge assessment performed using the Leuven Knowledge Questionnaire for Congenital Heart Disease (LKQCHD). Results: Following participation in the transition education, the overall mean knowledge scores increased from 43.6% (+/-) to 89.4%. (+/-). All participants scored \u3e 50% post intervention. Discussion: A formal transition education program demonstrated increased patient knowledge regarding their CHD. Advanced practice nurses are able to implement a transition education program and provide the necessary education to make a significant impact on patient knowledge

    Work and Career Experiences of Men from Families without College Experience

    A dearth of research exists exploring the career and work development of adult men and the influence of family-of-origin on that development. In this qualitative study, the researchers used a phenomenological approach to examine the career and work experiences of men whose parents have no education beyond high school and the influences of family on these experiences. We describe five constituents: (a) what work is like; (b) preparation/education; (c) who or what influenced them; (d) how to act at work; and (e) challenges/opportunities/choices; and one essence: mottos from father, which articulate the meaning of the experience of work and career and the family influence on the experience. In conclusion, we provide implications for counselors working with adult men whose parents lack post-secondary education

    Saccharomyces cerevisiae Rad9 Acts as a Mec1 Adaptor to Allow Rad53 Activation

    SummaryBackground: The DNA damage checkpoint is a protein kinase-based signaling system that detects and signals physical alterations in DNA. Despite having identified many components of this signaling cascade, the exact mechanisms by which checkpoint kinases are activated after DNA damage, as well as the role of the checkpoint mediators, remain poorly understood.Results: To elucidate the mechanisms that underlie the MEC1 and RAD9-dependent activation of Rad53, the Saccharomyces cerevisiae ortholog of Chk2, we mapped and characterized in vivo phosphorylation sites present on Rad53 after DNA damage by mass spectrometry. We find that Rad53 requires for its activation multisite phosphorylation on a number of typical and atypical Mec1 phosphorylation sites, thus confirming that Rad53 is a direct target of Mec1, the mammalian ATR homolog. Moreover, by using biochemical reconstitution experiments, we demonstrate that efficient and direct phosphorylation of Rad53 by Mec1 is only observed in the presence of purified Rad9, the archetypal checkpoint mediator. We find that the stimulatory activity of Rad9 requires a phospho- and FHA-dependent interaction with Rad53, which allows Rad53 to be recognized as a substrate for Mec1.Conclusions: Our results indicate that Rad9 acts as a bona fide signaling adaptor that enables Rad53 phosphorylation by Mec1. Given the high degree of conservation of checkpoint signaling in eukaryotes, we propose that one of the critical functions of checkpoint mediators such as MDC1, 53BP1, or Brca1 is to act as PIKK adaptors during the DNA damage response

    The Physiologic Effects of Multiple Simultaneous Electronic Control Device Discharges

    Objectives: Law enforcement and military personnel use electronic control devices to control non-compliant and actively resistive subjects. The TASER® Shockwave is a new electronic control device designed specifically as an area denial device capable of delivering multiple simultaneous discharges. This is the first study to examine the effects of multiple simultaneous device discharges in humans.Methods: Volunteers were exposed to multiple (two to three), simultaneous 5-second discharges from the Shockwave device to the chest, back, chest to abdomen, or thighs. Blood was analyzed before and after discharge for pH, lactate, potassium, creatine kinase (CK), and troponin. Continuous spirometry was performed before, during, and after the discharge. In addition, electrocardiograms (ECGs) before and after discharge were recorded, and echocardiography was used to determine the rhythm during discharge.Results: Small elevations of lactate occurred. Moderate increases in CK at 24 hours occurred and appeared to be related to the number of simultaneous discharges. There was a trend to a decrease in minute ventilation in the volunteers exposed to two simultaneous discharges, but it did not reach statistical significance. ECG changes only reflected an increase in vagal tone, and there was no evidence of capture by echocardiography. Five-second, simultaneous, multiple exposures to the TASER Shockwave device were reasonably tolerated by our human volunteers.Conclusion: Our study suggests that this device may have a reasonable risk/benefit ratio when used to protect an area from a threat.     Differentiation of 3-O-Sulfated Heparin Disaccharide Isomers: Identification of Structural Aspects of the Heparin CCL2 Binding Motif

    The presence of 3-O-sulfated glucosamine residues in heparin or heparan sulfate plays a role in binding to antithrombin III and HSV infection. In this study, tandem mass spectrometry was used to differentiate between two heparin disaccharide isomers containing variable sulfate at C6 in a common disaccharide and C3 in a more rare one. The dissociation patterns shown by MS2 and MS3 were clearly distinguishable between the isomers, allowing their differentiation and quantitation. Using this technique, we show that an octasaccharide with 11 sulfate groups with high affinity for inflammatory chemokine CCL2 does not contain 3-O-sulfated disaccharides

    Genetic Susceptibility of Cultured Shrimp (\u3ci\u3ePenaeus vannamei\u3c/i\u3e) to Infectious Hypodermal and Hematopoietic Necrosis Virus and \u3ci\u3eBaculovirus penaei\u3c/i\u3e: Possible Relationship with Growth Status and Metabolic Gene Expression

    Offspring of four crosses (I, II, III, and IV) of Penaeus vannamei from known high- and low-growth families were challenged with infectious hypodermal and hematopoietic necrosis virus (IHHNV) and Baculovirus penaei (BP) to compare their susceptibility to these viral agents and examine the genetic component involved in disease resistance or susceptibility. Family crosses were made using broodstock from five families developed by the U.S. Marine Shrimp Farming Program. The prevalence of IHHNV infection was highest in cross I and lowest in cross III. Cross I was developed using male and female broodstock from the low-growth family 1.6, and cross III was developed using a female from the high-growth family 1.3 and a male from the low-growth family 1.6. The prevalence of BP infection at Day 4 was highest (100%) in cross IV, which was developed using a female from the low-growth family 1.4 and a male from the high-growth family 1.5. The reciprocal cross, cross III, had the lowest (68%) prevalence at Day 4 postexposure. Both crosses I and II had 88% prevalence of infection at Day 4. Despite 100% prevalence of BP infection in cross IV at 4 days, animals from this cross and cross II exhibited high survival by Day 18 (85 and 77%). On the other hand, crosses I and III (with 88 and 68% prevalence at Day 4, respectively) showed low survival at Day 18 (19 and 24%). On the basis of prevalence of infection and mortality rates, it was concluded that the susceptibility to BP in penaeid shrimp is governed by the genetic background of the parental crosses. The random amplified polymorphic DNA polymorphisms for crosses I, II, III, and IV, were 43, 45, 53, and 51%, respectively, showing no clear relationship between IHHNV and BP prevalence of infection and levels of nuclear genetic diversity. Though the mtDNA haplotypes in offspring from the different crosses were the same, major differences were observed in both steady-state levels and patterns of expression of the mitochondrial 12s rRNA in offspring obtained at various early developmental stages from each of the four crosses. The possible relationship among disease susceptibility, growth status, and expression of mitochondrial 12s rRNA is discussed in the context of a complex nuclear-cytoplasmic genetic system involved in the regulation of gene expression
