A Novel Approach to Guided Implant Surgery: A Technical Note
Computer-guided software and kits have significantly improved the clinical applications of implant surgery. Nonetheless, some technical problems are still in evidence during clinical procedures because of cumbersome surgical tools that can limit access to implant sites, mainly in posterior areas of the mouth in the presence of bulky anatomical structures and in patients with reduced mouth-opening capacity. The present paper aimed to present a novel approach to guided implant surgery, describing the technical characteristics of an innovative guided surgical kit made up of modified sleeves and modular surgical drills. The proposed guided surgical kit is based on a novel patented system of sleeves and modular burs, with an increased length of the metal sleeves and a reduced height of the drills. The innovative design of the proposed system would allow the clinician to position guided fixtures in all clinical situations; the reduced encumbrance would be particularly helpful to gain access to the posterior areas of both maxilla and mandible, which have limited inter-arch space, with an easy and user-friendly approach. The modular system could overcome anatomical limitations, such as reduced mouth-opening capacity, and permit clinicians to maintain the stability and integrity of the surgical templates, even in cases where there is very limited intermaxillary space
Transfer of H2O2 from Mitochondria to the endoplasmic reticulum via Aquaporin-11
Some aquaporins (AQPs) can transport H2O2 across membranes, allowing redox signals to proceed in and between cells. Unlike other peroxiporins, human AQP11 is an endoplasmic reticulum (ER)-resident that can conduit H2O2 to the cytosol. Here, we show that silencing Ero1α, an ER flavoenzyme that generates abundant H2O2 during oxidative folding, causes a paradoxical increase in luminal H2O2 levels. The simultaneous AQP11 downregulation prevents this increase, implying that H2O2 reaches the ER from an external source(s). Pharmacological inhibition of the electron transport chain reveals that Ero1α downregulation activates superoxide production by complex III. In the intermembrane space, superoxide dismutase 1 generates H2O2 that enters the ER channeled by AQP11. Meanwhile, the number of ER-mitochondria contact sites increases as well, irrespective of AQP11 expression. Taken together, our findings identify a novel interorganellar redox response that is activated upon Ero1α downregulation and transfers H2O2 from mitochondria to the ER via AQP11.This work was supported through grants from the Associazione Italiana Ricerca sul Cancro (IG 2019-23285 to R.S.) and Ministero dell Istruzione, dell Università e della Ricerca (MIUR)-PRIN (grant no. 2017XA5J5N). Clinical evaluation of cement-retained implant-supported CAD/CAM monolithic zirconia single crowns in posterior areas: results of a 6-year prospective clinical study.
Background: Different technologies and materials can be used for implant-supported restorations in posterior areas. Our study aimed to evaluate the clinical performance of CAD/CAM implant-supported monolithic zirconia single crowns (ZrSCs) cemented onto titanium abutments with a dual-curing resin cement after 6 years of use. Methods: Fifty patients in need of one single crown in posterior regions were enrolled. The patients were recalled for a follow-up after 6 months and then yearly for a total observation period of 6 years. The biological and technical complications were examined using criteria set by the United States Public Health Service (USPHS), whereas visual analogue scales (VASs) were used to evaluate the patients’ aesthetic and functional satisfaction. Results: No patient was lost at follow-up; both the survival and success rates were 100%. No fractures, decementation, peri-implantitis, or soft tissue inflammation were recorded. Forty-six restorations were rated Alpha in each evaluated parameter of the USPHS’s criteria. According to the patients’ VASs assessments, the overall function of the ZrSCs was 8.4 (±2.1), whereas the overall aesthetic was 8.7 (±0.7). Conclusions: Monolithic zirconia single crowns cemented with dual-curing resin cements onto titanium abutments can be considered an effective clinical option in posterior regions after 6 years of function
Localization of lipopolysaccharide from Escherichia Coli into human atherosclerotic plaque
Experimental studies showed that gut-derived lipopolysaccharide (LPS) is pro-atherogenic, however, its relationship with human atherosclerosis is still to be defined. We investigate if gut-derived LPS from Escherichia Coli localizes in human carotid plaque and its potential role as pro-inflammatory molecule in the atherosclerotic lesion. LPS from Escherichia Coli and Toll-like receptor 4 (TLR4) were studied in specimens from carotid and thyroid arteries of 10 patients undergoing endarterectomy and 15 controls matched for demographic and clinical characteristics. Blood LPS were significantly higher in patients compared to controls. Immunochemistry analysis revealed positivity for antibodies against LPS and TLR4 coincidentally with positivity for CD68 only in the atherosclerotic plaque of carotid arteries but not in thyroid arteries; the positivity for LPS and TLR4 was greater in the area with activated macrophages. LPS concentration similar to that detected in atherosclerotic plaque resulted in a dose-dependent TLR4-mediated Nox2 up-regulation by human monocytes. These data provide the first evidence that LPS from Escherichia Coli localizes in human plaque and may contribute to atherosclerotic damage via TLR4-mediated oxidative stress
Zirconia-reinforced lithium silicate (ZLS) mechanical and biological properties: A literature review
Objectives: This paper aimed to provide a literature review of the mechanical and biological properties of zirconia-reinforced lithium silicate glass-ceramics (ZLS) in Computer-aided design / Computer-aided manufacturing (CAD/CAM) systems.Data/Sources: An extensive search of the literature for papers related to ZLS was made on the databases of PubMed/Medline, Scopus, Embase, Google Scholar, Dynamed, and Open Grey. The papers were selected by 3 independent calibrated reviewers.Study selection: The search strategy produced 937 records. After the removal of duplicates and the exclusion of papers that did not meet the inclusion criteria, 71 papers were included. Conclusions: After reviewing the included records, it was found that two types of ZLS (Vita Suprinity PC; Vita Zahnfabrik and Celtra Duo; Dentsply Sirona) are nowadays available on the market for CAD/CAM systems, similar in their chemical composition, microstructure, and biological-mechanical properties. ZLS is reported to be a biocompatible material, whose fracture resistance can withstand physiological chewing loads. The firing process influences the improvements of strength and fatigue failure load, with a volumetric shrinkage. To date, ZLS can be considered a viable alternative to other glass-ceramics for fixed single restorations. Clinical Significance: . As to biocompatibility and mechanical properties of ZLS, data are still scarce, often controversial and limited to short-term observational periods. These promising ceramics require further in vitro/ in vivo studies to accurately define mechanical and biological properties, mainly in the long-term performance of restorations produced with such materials
A Transportable Reflectarray Antenna for Satellite Ku-band Emergency Communications
The design of a Ku-band reconfigurable reflectarray antenna for emergency satellite communications is presented. Bidirectional high data rate satellite links are needed in emergency conditions where other telecommunication infrastructures are not available. In order to operate in this type of scenario, an antenna should be deployable, transportable, and easily repointable. The need of an automatic and fast satellite location and pointing system leads to a completely electronic reconfigurable antenna. The operative bandwidth is from 10.7 to 12.5 GHz for reception and from 14 up to 14.5 GHz for transmission (30% of relative bandwidth). The selected antenna architecture is based on a dual reflectarray system comprising a passive subreflectarray and an active main reflectarray made of reconfigurable 1-bit elementary cells based on PIN diodes
Interfacial Morphology Addresses Performance of Perovskite Solar Cells Based on Composite Hole Transporting Materials of Functionalized Reduced Graphene Oxide and P3HT
The development of novel hole transporting materials (HTMs) for perovskite solar cells (PSCs) that can enhance device's reproducibility is a largely pursued goal, even to the detriment of a very high efficiency, since it paves the way to an effective industrialization of this technology. In this work, we study the covalent functionalization of reduced graphene oxide (RGO) flakes with different organic functional groups with the aim of increasing the stability and homogeneity of their dispersion within a poly(3-hexylthiophene) (P3HT) HTM. The selected functional groups are indeed those recalling the two characteristic moieties present in P3HT, i.e., the thienyl and alkyl residues. After preparation and characterization of a number of functionalized RGO@P3HT blends, we test the two containing the highest percentage of dispersed RGO as HTMs in PSCs and compare their performance with that of pristine P3HT and of the standard Spiro-OMeTAD HTM. Results reveal the big influence of the morphology adopted by the single RGO flakes contained in the composite HTM in driving the final device performance and allow to distinguish one of these blends as a promising material for the fabrication of highly reproducible PSCs
