    Family Science as Translational Science: A History of the Discipline

    Family science has been a translational science since its inception. The history of family science began with an interdisciplinary group of scholars who came together to explore the complex nature of families during the discovery phase, paying particular attention to applying information to resolve family challenges. In the pioneering stage, family professionals struggled with naming the discipline and assembled professional groups that collected and applied information to benefit families. In the maturing stage, disciplinary leaders deemed that family science met the criteria of a bona fide discipline and the field\u27s identity became more pronounced, with a great deal of translational work occurring. During the current stage, evaluation and innovation, family science professionals need to assess programs and practices to refine and better articulate and distinguish the field. This historical account accentuates the central importance of the translational nature of family science to the discipline\u27s identity

    Subcutaneous immunoglobulin replacement therapy in the treatment of patients with primary immunodeficiency disease

    Antibody deficiency is the most frequently encountered primary immunodeficiency disease (PIDD) and patients who lack the ability to make functional immunoglobulin require life-long replacement therapy to prevent serious bacterial infections. Human serum immunoglobulin manufactured from pools of donated plasma can be administered intramuscularly, intravenously or subcutaneously. With the advent of well-tolerated preparations of intravenous immunoglobulin (IVIg) in the 1980s, the suboptimal painful intramuscular route of administration is no longer used. However, some patients continued to experience unacceptable adverse reactions to the intravenous preparations, and for others, vascular access remained problematic. Subcutaneously administered immunoglobulin (SCIg) provided an alternative delivery method to patients experiencing difficulties with IVIg. By 2006, immunoglobulin preparations designed exclusively for subcutaneous administration became available. They are therapeutically equivalent to intravenous preparations and offer patients the additional flexibility for the self-administration of their product at home. SCIg as replacement therapy for patients with primary antibody deficiencies is a safe and efficacious method to prevent serious bacterial infections, while maximizing patient satisfaction and improving quality of life

    Type IV Pili Can Mediate Bacterial Motility within Epithelial Cells.

    Pseudomonas aeruginosa is among bacterial pathogens capable of twitching motility, a form of surface-associated movement dependent on type IV pili (T4P). Previously, we showed that T4P and twitching were required for P. aeruginosa to cause disease in a murine model of corneal infection, to traverse human corneal epithelial multilayers, and to efficiently exit invaded epithelial cells. Here, we used live wide-field fluorescent imaging combined with quantitative image analysis to explore how twitching contributes to epithelial cell egress. Results using time-lapse imaging of cells infected with wild-type PAO1 showed that cytoplasmic bacteria slowly disseminated throughout the cytosol at a median speed of >0.05 μm s-1 while dividing intracellularly. Similar results were obtained with flagellin (fliC) and flagellum assembly (flhA) mutants, thereby excluding swimming, swarming, and sliding as mechanisms. In contrast, pilA mutants (lacking T4P) and pilT mutants (twitching motility defective) appeared stationary and accumulated in expanding aggregates during intracellular division. Transmission electron microscopy confirmed that these mutants were not trapped within membrane-bound cytosolic compartments. For the wild type, dissemination in the cytosol was not prevented by the depolymerization of actin filaments using latrunculin A and/or the disruption of microtubules using nocodazole. Together, these findings illustrate a novel form of intracellular bacterial motility differing from previously described mechanisms in being directly driven by bacterial motility appendages (T4P) and not depending on polymerized host actin or microtubules.IMPORTANCE Host cell invasion can contribute to disease pathogenesis by the opportunistic pathogen Pseudomonas aeruginosa Previously, we showed that the type III secretion system (T3SS) of invasive P. aeruginosa strains modulates cell entry and subsequent escape from vacuolar trafficking to host lysosomes. However, we also showed that mutants lacking either type IV pili (T4P) or T4P-dependent twitching motility (i) were defective in traversing cell multilayers, (ii) caused less pathology in vivo, and (iii) had a reduced capacity to exit invaded cells. Here, we report that after vacuolar escape, intracellular P. aeruginosa can use T4P-dependent twitching motility to disseminate throughout the host cell cytoplasm. We further show that this strategy for intracellular dissemination does not depend on flagellin and resists both host actin and host microtubule disruption. This differs from mechanisms used by previously studied pathogens that utilize either host actin or microtubules for intracellular dissemination independently of microbe motility appendages

    Using the Cultivating Learning with School Gardens Curriculum in Burundi, Africa

    University faculty and Extension educators sought to use school gardens in Burundi, Africa, as a means of reducing food insecurity, teaching positive youth development, and increasing experiential learning for Burundian students. Washington State University personnel used videoconferencing to provide training to Burundian nongovernmental organization (NGO) staff on the Cultivating Learning with School Gardens curriculum. The NGO staff then trained teachers in pilot programs at four Burundian schools, where first harvests occurred in May 2016. An agricultural consultant also helped with the gardens. Suggestions for implementation of this school garden curriculum in other countries are provided

    The Impact of Family Rituals and Maternal Depressive Symptoms on Child Externalizing Behaviors: An Urban–Rural Comparison

    Background The association between maternal depression and child negative behavior outcomes has been well established in the literature. However, understanding how maternal depression is associated with child behaviors will have important implications for research and intervention strategies. Objective We used samples from two distinct family contexts: urban, middle-class families (N = 454); and rural, low-income families (N = 240), to compare the impact of family rituals and maternal depressive symptoms on child externalizing behaviors. We also examined the impact of maternal depressive symptoms on family rituals. Methods Structural equation modeling and multiple group analyses were conducted. Child age, child gender, mother age, mother education, mother partner status, family income, and family size served as control variables. Indirect effect tests and incremental validity tests were performed. Results Depressive symptoms were linked to externalizing behaviors in both samples. The impacts of family rituals on child externalizing behaviors were more context-specific. For urban, middle-class families, vacations played a particularly helpful intermediate role. Also, in these families, maternal depressive symptoms were associated with less involvement in religious holidays. For rural, low-income families, involvement in annual celebrations was associated with less child externalizing behaviors, although maternal depressive symptoms did not impact involvement in any family rituals. Conclusions Results suggest that family context matters in determining the impacts of maternal depression on the practice of family rituals, as well as the benefits of family rituals for child behaviors, and that certain family rituals can do more than others to reduce child externalizing behaviors (e.g., vacation)

    Endogenous ROS Levels in C. Elegans Under Exogenous Stress Support Revision of Oxidative Stress Theory of Life-History Tradeoffs

    Background: The oxidative stress theory of life-history tradeoffs states that oxidative stress caused by damaging free radicals directly underpins tradeoffs between reproduction and longevity by altering the allocation of energetic resources between these tasks. We test this theory by characterizing the effects of exogenous oxidative insult and its interaction with thermal stress and diet quality on a suite of life-history traits and correlations in Caenorhabditis elegans nematodes. We also quantify demographic aging rates and endogenous reactive oxygen species (ROS) levels in live animals. Results: Our findings indicate a tradeoff between investment in reproduction and antioxidant defense (somatic maintenance) consistent with theoretical predictions, but correlations between standard life-history traits yield little evidence that oxidative stress generates strict tradeoffs. Increasing oxidative insult, however, shows a strong tendency to uncouple positive phenotypic correlations and, in particular, to reduce the correlation between reproduction and lifespan. We also found that mild oxidative insult results in lower levels of endogenous ROS accompanied by hormetic changes in lifespan, demographic aging, and reproduction that disappear in combined-stress treatments–consistent with the oxidative stress theory of aging. Conclusions: Our findings demonstrate that oxidative stress is a direct contributor to life-history trait variation and that traditional tradeoffs are not necessary to invoke oxidative stress as a mediator of relationships between life-history traits, supporting previous calls for revisions to theory

    Based on an exceptionally long modern ecological dataset (41 years), it has been possible to show that warm weather in England associated with a positive North Atlantic Oscillation (NAO) index causes the spring migration of the green spruce aphid (Elatobium abietinum), a pest species of spruce trees (Picea) to start earlier, continue for longer and contain more aphids. An upward trend in the NAO index during the period 1966-2006 is associated with an increasing population size of E. abietinum. It is important to understand the mechanisms behind the population fluctuations, because this aphid causes considerable damage to Picea plantations. Present day weather associated fluctuations in forest insect pests may be useful analogues in understanding past pest outbreaks in forests

    Zoom Around the World: Using Videoconferencing Technology for International Trainings

    This article describes an innovative model of educational programming used by our team of U.S. Extension educators for an international development project in Burundi, Africa. Our team designed a model to provide ongoing professional development trainings at a distance using Zoom, a videoconferencing platform. Over a 2-year period, we conducted 18 Zoom trainings with Burundian educators. On the basis of participant evaluation data and the literature, we present key principles for using distance technology in international development projects. Given the current economy and budget cuts in Extension, videoconferencing provides an opportunity for Extension to remain engaged internationally

    As our understanding of genetics has improved, genome-wide association studies (GWAS) have identified numerous variants associated with lifestyle behaviours and health outcomes. However, what is sometimes overlooked is the possibility that genetic variants identified in GWAS of disease might reflect the effect of modifiable risk factors as well as direct genetic effects. We discuss this possibility with illustrative examples from tobacco and alcohol research, in which genetic variants that predict behavioural phenotypes have been seen in GWAS of diseases known to be causally related to these behaviours. This consideration has implications for the interpretation of GWAS findings
