    Analysis of Carbohydrate Storage Granules in the Diazotrophic Cyanobacterium Cyanothece sp. PCC 7822.

    The unicellular diazotrophic cyanobacteria of the genus Cyanothece demonstrate oscillations in nitrogenase activity and H2 production when grown under 12 h light–12 h dark cycles. We established that Cyanothece sp. PCC 7822 allows for the construction of knock-out mutants and our objective was to improve the growth characteristics of this strain and to identify the nature of the intracellular storage granules. We report the physiological and morphological effects of reduction in nitrate and phosphate concentrations in BG-11 media on this strain. We developed a series of BG-11-derived growth media and monitored batch culture growth, nitrogenase activity and nitrogenase-mediated hydrogen production, culture synchronicity, and intracellular storage content. Reduction in NaNO3 and K2HPO4 concentrations from 17.6 and 0.23 to 4.41 and 0.06 mM, respectively, improved growth characteristics such as cell size and uniformity, and enhanced the rate of cell division. Cells grown in this low NP BG-11 were less complex, a parameter that related to the composition of the intracellular storage granules. Cells grown in low NP BG-11 had less polyphosphate, fewer polyhydroxybutyrate granules and many smaller granules became evident. Biochemical analysis and transmission electron microscopy using the histocytochemical PATO technique demonstrated that these small granules contained glycogen. The glycogen levels and the number of granules per cell correlated nicely with a 2.3 to 3.3-fold change from the minimum at L0 to the maximum at D0. The differences in granule morphology and enzymes between Cyanothece ATCC 51142 and CyanothecePCC 7822 provide insights into the formation of large starch-like granules in some cyanobacteria

    This paper attempts to reconcile an apparent contradiction between short-run and long-run movements in the price of gold. The theoretical model suggests a set of conditions under which the price of gold rises over time at the general rate of inflation and hence be an effective hedge against inflation. The model also demonstrates that short-run changes in the gold lease rate, the real interest rate, convenience yield, default risk, the covariance of gold returns with other assets and the dollar/world exchange rate can disturb this equilibrium relationship and generate short-run price volatility. Using monthly gold price data (1976-1999), and cointegration regression techniques, an empirical analysis confirms the central hypotheses of the theoretical model

    Linearized model Fokker-Planck collision operators for gyrokinetic simulations. II. Numerical implementation and tests

    A set of key properties for an ideal dissipation scheme in gyrokinetic simulations is proposed, and implementation of a model collision operator satisfying these properties is described. This operator is based on the exact linearized test-particle collision operator, with approximations to the field-particle terms that preserve conservation laws and an H-Theorem. It includes energy diffusion, pitch-angle scattering, and finite Larmor radius effects corresponding to classical (real-space) diffusion. The numerical implementation in the continuum gyrokinetic code GS2 is fully implicit and guarantees exact satisfaction of conservation properties.     Analysis of the thermomechanical inconsistency of some extended hydrodynamic models at high Knudsen number

    There are some hydrodynamic equations that, while their parent kinetic equation satisfies fundamental mechanical properties, appear themselves to violate mechanical or thermodynamic properties. This article aims to shed some light on the source of this problem. Starting with diffusive volume hydrodynamic models, the microscopic temporal and spatial scales are first separated at the kinetic level from the macroscopic scales at the hydrodynamic level. Then we consider Klimontovich’s spatial stochastic version of the Boltzmann kinetic equation, and show that, for small local Knudsen numbers, the stochastic term vanishes and the kinetic equation becomes the Boltzmann equation. The collision integral dominates in the small local Knudsen number regime, which is associated with the exact traditional continuum limit. We find a sub-domain of the continuum range which the conventional Knudsen number classification does not account for appropriately. In this sub-domain, it is possible to obtain a fully mechanically-consistent volume (or mass) diffusion model that satisfies the second law of thermodynamics on the grounds of extended non-local-equilibrium thermodynamics

    Triggering synchronized oscillations through arbitrarily weak diversity in close-to-threshold excitable media

    It is shown that arbitrarily weak (frozen) heterogeneity can induce global synchronized oscillations in excitable media close to threshold. The work is carried out on networks of coupled van der Pol-FitzHugh-Nagumo oscillators.     Approximate Analytic Solution for the Spatiotemporal Evolution of Wave Packets undergoing Arbitrary Dispersion

    We apply expansion methods to obtain an approximate expression in terms of elementary functions for the space and time dependence of wave packets in a dispersive medium. The specific application to pulses in a cold plasma is considered in detail, and the explicit analytic formula that results is provided. When certain general initial conditions are satisfied, these expressions describe the packet evolution quite well. We conclude by employing the method to exhibit aspects of dispersive pulse propagation in a cold plasma, and suggest how predicted and experimental effects may be compared to improve the theoretical description of a medium's dispersive properties.Comment: 17 pages, 4 figures, RevTe
