    Get PDF
For more than one decade the workshops and courses of graffiti proliferate in the primary and secondary school. From the point of view of the teacher, this type of activities is often seen as an attractive educational complement to offer to the students. The experience of having lived in Granada since 1999 has helped me to recapitulate and analyze a series of problems related to the appearance and rise of graffiti in the city and its impact on the heritage building of this one, recalling a series of events on which to extract questions about how the whole issue has been raised from the institutions involved, and from the educational field. The fanzines sustained principally by the documentary photo, personified the collaboration spirit in the Graffiti of the 90s through a process of exchange of specific and unpublished information until then. We analyzed all this at a time when the distance communication system among graffiti writers was based exclusively on postal mail. Our case study focuses on a very specific context, the Graffiti scene in the city of Granada, from its gestation as a movement (1992-1993) to the arrival of the Internet (1997-1998), covering a dynamic period in which graphic and visual material was produced. In this way, we will consider the existing parallelism between the ways of doing in Graffiti and methods of editing and diffusion, analyzing the concepts of traveling, exploring and sharing informationLos fanzines, sustentados principalmente por la fotografía documental, encarnaron el espíritu de colaboración en el graffiti de la década de los noventa a través de un proceso de intercambio de información específica e inexistente hasta ese momento.

Site-specificity of pea histone acetyltransferase B in vitro

    Get PDF
Histone acetyltransferase B from pea embryonic axes has been purified approximately 300-fold by a combination of chromatographic procedures, including affinity chromatography on histone-agarose. The enzyme preparation has been used for the in vitro transfer of acetyl groups from [1-14C]acetyl-CoA to non-acetylated pea histone H4. Up to three acetyl groups can be introduced into the histone. The resulting mono-, di-, and triacetylated H4 isoforms were separated and sequenced to determine the acetylated sites. Only sites 5, 12, and 16 were used by histone acetyltransferase B, but no clear preference among them was observed. The absence of modification of other potentially acetylatable sites is another indication that acetylation of the different lysine residues in the N-terminal H4 tail serves as a specific signal in different nuclear processes

Early ERK1/2 activation promotes DRP1-dependent mitochondrial fission necessary for cell reprogramming

During the process of reprogramming to induced pluripotent stem (iPS) cells, somatic cells switch from oxidative to glycolytic metabolism, a transition associated with profound mitochondrial reorganization. Neither the importance of mitochondrial remodelling for cell reprogramming, nor the molecular mechanisms controlling this process are well understood. Here, we show that an early wave of mitochondrial fragmentation occurs upon expression of reprogramming factors. Reprogramming-induced mitochondrial fission is associated with a minor decrease in mitochondrial mass but not with mitophagy. The pro-fission factor Drp1 is phosphorylated early in reprogramming, and its knockdown and inhibition impairs both mitochondrial fragmentation and generation of iPS cell colonies. Drp1 phosphorylation depends on Erk activation in early reprogramming, which occurs, at least in part, due to downregulation of the MAP kinase phosphatase Dusp6. Taken together, our data indicate that mitochondrial fission controlled by an Erk-Drp1 axis constitutes an early and necessary step in the reprogramming process to pluripotency

c‑MYC Triggers Lipid Remodelling During Early Somatic Cell Reprogramming to Pluripotency

Metabolic rewiring and mitochondrial dynamics remodelling are hallmarks of cell reprogramming, but the roles of the reprogramming factors in these changes are not fully understood. Here we show that c-MYC induces biosynthesis of fatty acids and increases the rate of pentose phosphate pathway. Time-course profiling of fatty acids and complex lipids during cell reprogramming using lipidomics revealed a profound remodelling of the lipid content, as well as the saturation and length of their acyl chains, in a c-MYC-dependent manner. Pluripotent cells displayed abundant cardiolipins and scarce phosphatidylcholines, with a prevalence of monounsaturated acyl chains. Cells undergoing cell reprogramming showed an increase in mitochondrial membrane potential that paralleled that of mitochondrial-specific cardiolipins. We conclude that c-MYC controls the rewiring of somatic cell metabolism early in cell reprogramming by orchestrating cell proliferation, synthesis of macromolecular components and lipid remodelling, all necessary processes for a successful phenotypic transition to pluripotency

    Get PDF
