Long Term and Short Term Effects of Perturbations in a Immune Network Model
In this paper we review the trajectory of a model proposed by Stauffer and
Weisbuch in 1992 to describe the evolution of the immune repertoire and present
new results about its dynamical behavior. Ten years later this model, which is
based on the ideas of the immune network as proposed by Jerne, has been able to
describe a multi-connected network and could be used to reproduce immunization
and aging experiments performed with mice. Besides its biological implications,
the physical aspects of the complex dynamics of this network is very
interesting {\it per se}. The immunization protocol is simulated by introducing
small and large perturbations (damages), and in this work we discuss the role
of both. In a very recent paper we studied the aging effects by using
auto-correlation functions, and the results obtained apparently indicated that
the small perturbations would be more important than the large ones, since
their cumulative effects may change the attractor of the dynamics. However our
new results indicate that both types of perturbations are important. It is the
cooperative effects between both that lead to the complex behavior which allows
to reproduce experimental results.Comment: 15 pages, 5 figure
Gastrointestinal symbionts of wild chimpanzees and sympatric colobus monkeys living in close proximity of humans in Liberia and Uganda
Dissertação de Mestrado Integrado em Medicina VeterináriaEsta dissertação teve como objetivo principal caracterizar a fauna de simbiontes de
chimpanzés (Pan troglodytes), e colobos pretos e brancos (Colobus sp.) de diferentes
localidades Africanas (Parque Nacional de Sapo, na Libéria e em Bulindi, no Uganda),
comparar resultados entre eles e estudos prévios e teorizar sobre as possíveis implicações
zoonóticas dos simbiontes encontrados.
Recorrendo a técnicas de coprologia como sedimentação MIF modificada e a flutuação de
Sheather, e um teste de imunofluorescência direta para deteção de oocistos de
Crypstosporidium e cistos de Giardia, 47 amostras fecais preservadas de chimpanzé e 15 de
colobos pretos e brancos foram examinadas.
Os protozoários detetados incluíram Troglodytella abrassarti, Entamoeba spp., Blastocystis
sp. e Iodamoeba buetschlii; e nematodes, Trichuris sp, Colobenterobius sp., strongylideos e
várias larvas de nematodes. Todas as amostras foram negativas no teste de
imunofluorescência directa.
Em geral, o espectro de simbiontes encontrado no presente estudo está de acordo com o que
é regularmente descrito em estudos que abordam chimpanzés e colobos pretos e brancos de
vida selvagem. Contudo, pequenos microrganismos aquáticos denominados Rotifera ou
rotíferos foram observados pela primeira vez em amostras fecais de chimpanzé e colobos
pretos e brancos.
Considerando o contato próximo entre os primatas estudados e seres humanos, é importante
realizar futuras pesquisas recorrendo a técnicas moleculares nos simbiontes assinalados por
este trabalho e avaliar o seu real potencial zoonótico.ABSTRACT - Gastrointestinal symbionts of wild chimpanzees and sympatric colobus monkeys living in close proximity of humans in Liberia and Uganda - This dissertation aimed to characterize the symbiont fauna of chimpanzees (Pan troglodytes) and black and white colobus (Colobus sp.) from different African localities, (Sapo National Park in Liberia and Bulindi in Uganda), compare results between them and previous studies, and theorize about possible zoonotic implications of the found symbionts.
Using coprologic techniques as modified MIF sedimentation and Sheather’s flotation, and a direct immunofluorescence teste for detection of Crypstosporidium oocysts and Giardia cysts, 47 chimpanzee and 15 black and white colobus preserved fecal samples were examined.
Detected protozoans included Troglodytella abrassarti, Entamoeba spp., Blastocystis sp. and Iodamoeba buetschlii; and nematodes, Trichuris sp., Colobenterobius sp., strongylids and various nematodes larvae. All samples were negative for the direct immunofluorescence test.
Overall, the spectrum of symbionts found in the present study is in accordance with what is regularly described in studies addressing the free ranging chimpanzees and black and white colobus. However, small aquatic microorganisms denominated Rotifera or rotifers were observed for the first time in chimpanzee and in black and white colobus faecal samples. Considering the close contact between the studied primates and humans, it is important to conduct further research using molecular techniques in the found symbionts to assess the real zoonotic potential.N/
Development of Optogenetic Tools for Manipulating Neuronal Activity and Behaviour in Zebrafish
Integration of sensory input and computation of behavioural output is a dynamic process involving diverse populations of cells often distributed throughout the brain.
To study this topic, monitoring neuronal activity from a large population of cells and manipulating targeted neuronal activity in a behaving animal is crucial. This is possible in zebrafish, due to its small and transparent larval brain and its genetic malleability, by making use of optogenetic tools that allow reversible light-dependent activation and inhibition of neuronal activity, and genetically encoded calcium indicators (GECI) that enable non-invasive activity recording.
State-of-the-art optogenetic tools with faster kinetics and higher sensitivity facilitate reliable manipulation of activity with high temporal precision during behaviour. Such tools have been developed to be compatible with better calcium indicators to successfully manipulate and optically record neuronal activity simultaneously.
In this project, the latest developed optogenetic tools - activators ChrimsonR, C1V1(t/t) and Chronos, inhibitor Jaws, red calcium sensor jRCaMP1b and nuclear markers H2B-RFP and H2B-mCherry - were optimized to be expressed in zebrafish. Behavioural assays to characterize the activating and inhibitory optogenetic tools ChrimsonR and Jaws were established. An escape response of short latency could reliably be evoked in transgenic animals with stable expression of ChrimsonR in trigeminal neurons. Combination of this fast, sensitive and red-shifted tool with GCaMP calcium imaging opens the possibility to simultaneously manipulate and record activity with high spatial and temporal precision from a large population of neurons to study their dynamic interactions during behaviour

Implementation of paratuberculosis control measures in an endemically infected dairy herd in Ireland
Dissertação de Mestrado Integrado em Medicina Veterinária, área científica de ClínicaABSTRACT - Implementation of Paratuberculosis Control Measures in an Endemically Infected Dairy Herd in Ireland - Johne´s disease, or paratuberculosis, is a chronic enteritis caused by Mycobacterium avium paratuberculosis (Map), endemic in several countries. Since this disease has a significant economic impact, voluntary control programmes were established to improve the paratuberculosis status in several affected countries, such as Ireland. This study aims to define the aspects responsible for limiting the Irish Johne’s Control Programme (IJCP) performance through the analysis of this programme implementation, for 3 years, in a dairy herd. The analysis that took course in this project could offer valuable insights for better understanding and enhancing the performance of voluntary control programs. The research was performed through the analysis of each requirement of the programme accomplished by the herd in question. This analysis was conducted for the 3 years of the herd’s participation in IJCP, resorting to the official programme’s database, the Irish Cattle Breeding Federation (ICBF), to gather the information required. The data collected was then compared from one year to another to investigate the relation between each year and elaborate a reasoned explanation for the observed outcome. The results indicate that the disease's apparent prevalence is increasing yearly, from 3.37% in 2020, to 5.21% in 2021, and 11.46% in 2022. The analysis of the Veterinary Risk Assessment and Management Plan (VRAMP) results indicates several issues, mainly related to the lack of compliance from the herd owner and farm workers, that affect hygiene and farm management measures crucial to achieving optimum programme performance. The increase in the apparent prevalence could also be influenced by the fact that the herd has only been on the IJCP for 3 years and this time frame might not be enough to observe significant changes in the disease’s development and prevalence. Through the analysis and reflection conducted over the course of this study, it is possible to identify the factors that compromise the performance of the IJCP, contributing to a better understanding and enhancing the implementation of voluntary control programmes. Uma vez que esta doença tem um impacto económico significativo, foram estabelecidos programas de controlo voluntário para melhorar o estado da paratuberculose em vários países afectados, como a Irlanda. Este estudo tem como objetivo definir os aspectos responsáveis pela limitação do desempenho do Irish Johne's Control Programme (IJCP) através da análise da implementação deste programa, durante 3 anos, num efetivo leiteiro. A análise que decorreu neste projeto poderá contribuir para melhorar a compreensão e o desempenho dos programas de controlo voluntários. A pesquisa foi realizada através da análise de cada requisito do programa cumprido pelo efetivo em questão. Esta análise foi efectuada para os 3 anos de participação do efetivo no IJCP, recorrendo à base de dados oficial do programa, a Irish Cattle Breeding Federation (ICBF), para recolher a informação necessária. Os dados recolhidos foram depois comparados de um ano para o outro para investigar a relação entre cada ano e elaborar uma explicação fundamentada para o resultado observado. Os resultados indicam que a prevalência aparente da doença está a aumentar anualmente, de 3,37% em 2020 para 5,21% em 2021 e 11,46% em 2022. A análise dos resultados do Plano de Avaliação e Gestão dos Riscos Veterinários (VRAMP) indica várias questões, principalmente relacionadas com a falta de cumprimento por parte do proprietário do rebanho e dos trabalhadores agrícolas, que afectam as medidas de higiene e de gestão das explorações agrícolas cruciais para alcançar um desempenho ótimo do programa. O aumento da prevalência aparente também pode ser influenciado pelo facto de o efetivo só ter estado no IJCP durante 3 anos e este período de tempo pode não ser suficiente para observar mudanças significativas no desenvolvimento e prevalência da doença. Através da análise e reflexão realizadas ao longo deste estudo, é possível identificar os factores que comprometem o desempenho do PCIJ, contribuindo para um melhor conhecimento e para uma melhor implementação de programas de controlo voluntários. The major conclusion was that the company should choose Nigeria and the best way to
enter is initiating a partnership with a local consulting company with deeper local
knowledge, which can be seen as a competitive advantage
The Portuguese energy system in 2030
Tese de Mestrado Integrado, Engenharia da Energia e do Ambiente, 2022, Universidade de Lisboa, Faculdade de CiênciasCom o objetivo de mitigação das alterações climáticas, os países definiram metas para a
diminuição das emissões de gases de efeitos de estuda (GEE). A aprovação do Acordo de Paris
em 2015 definiu os objetivos necessários para a limitação do aumento da temperatura global
abaixo dos 2 ºC e reunir esforços para que este aumento seja ainda inferior a 1.5 ºC.
Por esta razão, as nações signatárias elaboraram estratégias para redução de gases de efeito de
estufa a médio e a longo prazo. Estes esforços para redução de emissões com fim à neutralidade
carbórnica têm em perspetiva metas mais exequíveis a médio prazo, ou seja para 2030, e que nos
levam a um fim estipulado para 2050.
Esta dissertação foca-se no estudo dos cenários a médio prazo de 2030, através da simulação dos
sistemas de energia destes países com base nos dados estimados, que estão descritos nos
documentos dos planos nacionais de energia e clima de Portugal e de Espanha. Utilizou-se uma
ferramenta de modelação de sistemas de energia que nos permite simular os vários setores de um
sistema de energia e interligações entre diversos modelos de sistemas.
O objetivo destes planos é a definição de metas atingíveis a médio prazo que contribuam para a
diminuição das emissões dos gases de efeito de estufa (GEE) através de estratégias de
descarbonização do setor da energia que passam essencialmente pelo aumento da produção de
energia por fontes renováveis, e que vêm a substituir as tecnologias de geração de energia com
origem em combustíveis fósseis. Essas mudanças, no entanto, devem garantir a robustez e a
flexibilidade dos sistemas de energia, e devem ser um mix de tecnologias que muda
progressivamente com a integração de energia renovável e o surgimento de novos vetores de
energia, como o hidrogénio, considerados importantes por contribuírem para a descarbonização
e diminuição da dependência energética do país.
Os resultados obtidos neste estudo evidenciam se as medidas contempladas nos planos de 2030
contribuem para atingir os ambiciosos objetivos definidos para 2050, quais são os maiores
desafios para atingir estas metas, e o que poderá ter de ser diferente. A simulação dos sistemas de
energia de Portugal e de Espanha tendo em conta as várias tecnologias de energia renovável,
tecnologias de armazenamento, consideração de phase-out de centrais de produção utilizando
recursos não renováveis, a introdução do hidrogénio e estratégias de otimização da rede, permitem
uma visão geral e um pouco mais perto da realidade, de como os vários setores – como a produção
de eletricidade, a cogeração, o setor industrial, os transportes, entre outros – se influenciam entre
si e como por consequência, influenciam os sistemas exteriores.
Os sistemas de energia atuais, nomeadamente os sistemas de Portugal e de Espanha têm uma
percentagem relevante de geração de energia renovável quando olhamos para o panorama europeu
e mundial. No entanto, observa-se um aumento muito significativo na quantidade de energia
renovável produzida a cada ano, prevendo-se penetrações de geração renovável variável muito
perto dos 100%, com o objetivo de atingir esse valor absoluto num futuro que podemos considerar
bastante próximo. Os sistemas de energia com estas percentagens de produção renovável,
apresentam desafios muito diferentes dos sistemas atuais. Por esta razão, este estudo dá ênfase a
alguns fatores que achamos relevantes nos sistemas de energia de Portugal e de Espanha neste
novo paradigma energético. O grande desafio passa pelo balanço de consumo e produção de energia, quando temos grandes
percentagens de produção variável devido aos recursos renováveis como o solar e eólico,
apresentarem um pequeno grau de previsibilidade de produção.
A complementaridade entre várias tecnologias de produção renovável que utilizam diferentes
recursos renováveis, como o conhecido exemplo entre a complementaridade da produção solar
com a produção eólica, contribuem para uma menor flutuação das curvas de produção de um
sistema.
Uma estratégia que se tornará mais relevante no futuro será a própria complementaridade entre
sistemas. Diferentes sistemas de energia apresentam diferentes curvas de consumo, tal como
diferentes curvas de produção de energia. A diferente disponibilidade de recursos endógenos
consequentemente resulta em diferentes perfis de produção. A linha de transmissão aqui ganha
um papel na complementaridade entre sistemas de energia criando um balanço entre os sistemas
por meio de exportações e importações, onde o consumo de um sistema é suprimido pelo excesso
de produção renovável de outro sistema. Neste futuro, a gestão, aumento e até criação da
capacidade da linha de transmissão que liga os vários sistemas energéticos da Europa ganha uma
maior importância.
Neste trabalho também se estudou a influência do hidrogénio como vetor energético, de forma a
perceber como influência os sistemas de energia, produzido através de eletrólise de água acoplado
a centrais solares fotovoltaicos e eólicas. O efeito da introdução do hidrogénio nos sistemas fazse sentir essencialmente no aumento da produção das centrais a gás natural pela realocação de
energia solar e eólica para o efeito, e na redução da necessidade de bombagem de água por ser o
hidrogénio um vetor energético que proporciona flexibilidade ao sistema.
De acordo com a capacidade instalada para 2030 segundo os respetivos planos para Portugal e
Espanha, os modelos indicam maiores percentagens de renováveis na eletricidade e na energia
primária superiores que nos dados estimados.
A conclusão central deste estudo está relacionada com o papel da capacidade da linha de
transmissão entre Portugal e Espanha e entre Espanha e França, na prespetiva dos sistemas de
energia de 2030. O aumento de produção renovável em ambos os sistemas resultam em maior
potencial de exportação, e consequentemente, é necessário um aumento da capacidade da linha
de transmissão para transportar todo o excesso de energia.
Os modelos criados neste trabalho indicam que a capacidade da linha de transmissão estimada
segundo os planos para 2030 no sentido de Portugal para Espanha, está de acordo com os valores
obtidos, mas que a capacidade no sentido de Espanha para os sistemas externos (França), está
subestimada. O foco é então no reforço acima das expectativas da interligação entre Espanha e
França, permitindo o fluxo de exportações em horas de produção solar, típicas do perfil de
produção renovável da Península Ibérica.
No entanto, devido ao consumo e produção de Portugal e Espanha serem muito semelhantes por
se encontrarem em locais geograficamente semelhantes, influenciados pelo mesmo ambiente e
condições, estes sistemas apresentam muito pouca complementaridade. Os resultados obtidos
remetem também à Peninsula Ibérica com um perfil de consumo e produção próprio relativamente
aos sistemas externos (Europa), e à relevância do aumento da interligação no sentido da Europa
(França). Portanto, o valor do excedente de eletricidade do ponto de vista interno entre Portugal e Espanha
tem pouco valor, mas numa perspetiva de complementaridade de regiões da Europa, vai de
encontro à atual conjuntura em que o reforço de interligações é considerado crucial para manter
a resiliência dos sistemas energéticos, tornando-os mais independentes de combustíveis fósseis
por meio de trocas de energia renovável.
Neste trabalho não foram tidos em conta fatores económicos sendo que o foco foi uma análise
técnica, mas não desprezando que a visão económica influência diretamente decisões de operação
que não estão necessariamente ligadas à opção tecnicamente mais eficiente de produção de
energia. Este facto determina diretamente as trocas de energia entre sistemas (importações e
exportações) tal como o armazenamento. Por vezes as importações e exportações ocorrem devido
a valores de custo de energia superiores ou inferiores aos custos de produção.This dissertation focuses on the study of the national energy and climate plans for 2030 in Portugal
and Spain, through a tool for modeling energy systems.
These plans define achievable medium-term goals that contribute to the reduction of greenhouse
gas (GHG) emissions, through decarbonization strategies in the energy sector that essentially
involve increasing energy production from renewable sources that come to replace energy
generation technologies based on fossil fuels. The objective of this work is to simulate and analyse
the operation of electricity systems of both countries based on the objectives defined in the
mentioned plans.
The results obtained in this study show whether the measures of the 2030 plans contribute to
achieving the ambitious goals defined for 2050, what are the biggest challenges to achieve these
goals and which may have to be different.
The main conclusions drawn from this study focus on the importance of increasing the capacity
of the transmission line in relation to what is estimated in the plans defined for 2030 mainly in
Spain. The simulations indicate that for the characteristics of the systems according to estimates
made for 2030, the capacity of the transmission line from Portugal to Spain is adequately
dimensioned for the energy flows obtained, however, in the direction of Spain for the outside
systems (France), it was concluded that the capacity of the line needs to be about double that
estimated.
This study allowed an analysis not only of the systems in Portugal and Spain individually, but
also from the prespective of Iberian Peninsula with a own consumption and production profile in
relation to external systems (Europe), emphasizing the importance of increasing interconnection
towards Europe (France)
Antifungal Activity of Brazilian Medicinal Plants against Candida Species
Due to the resistance of Candida sp. to the usual antifungal, the demand for active principles found in the plants has been the target of diverse studies around the world. There are few in vivo and human studies on the antifungal activity of medicinal plants in the mouth. Native and imported medicinal plants, used by the Brazilian population for traditional medicine use, are the subject of study in this chapter. Thirty-eight Brazilian plants were related to information on species, family, name, used part, and medical indication of popular use. All the species mentioned had their extracts tested in vitro against C. albicans, C. tropicalis, C. krusei, C. parapsilosis, among other species that occur more frequently in the mouth. In the articles consulted, there is a great variation in Candida species tested and in minimum inhibitory concentration. The in vitro studies serve as information for the continuity of studies on the best performing plants, validate the popular belief about the use, and provide subsidies for the development of new products that are effective in the control of oral and systemic candidiasis and that are cheap and accessible for the population
