Distribution of the invasive ctenophore <i>Mnemiopsis leidyi</i> in the Belgian part of the North Sea
The invasive ctenophore Mnemiopsis leidyi A. Agassiz, 1865 was recorded for the first time in Northern Europe in summer 2005, while the first records in the North Sea date back to summer 2006. The first sightings in the Belgian part of the North Sea were made in August 2007 in the port of Zeebrugge, but most probably M. leidyi had already been present for a longer period in this area. The high densities in the port of Zeebrugge suggest that M. leidyi entered the Belgian marine waters via ballast water transport, comparable with the invasion in the Black and Caspian Seas and the Dutch part of the North Sea. In the period 2009ā2011, M. leidyi was found in all ports and all along the Belgian coastline, up to 27 km offshore. Further offshore, no M. leidyi were found in zooplankton samples and small meshed otter trawl samples. Sightings of adult individuals in the coldest winter months imply that the species can survive Belgian winters. Highest densities (17 ind.m-3) were found in the Sluice dock in the port of Oostende. Along the coastline, average densities of 0.4 ind.m 3 were recorded. As M. leidyi might previously have been misidentified on the basis of morphological features alone, we also determined the species with genetic identification tools. Taking into account the notorious impact of this species in its native and in other invaded waters, it is recommended to continue the monitoring of M. leidyi populations in the Belgian part of the North Sea
Weight-loss induced changes in physical activity and activity energy expenditure in overweight and obese subjects before and after energy restriction
Activity energy expenditure (AEE) is the component of daily energy expenditure that is mainly influenced by the amount of physical activity (PA) and by the weight of the body displaced. This study aimed at analyzing the effect of weight loss on PA and AEE. The body weight and PA of 66 overweight and obese subjects were measured at baseline and after 12 weeks of 67% energy restriction. PA was measured using a tri-axial accelerometer for movement registration (Tracmor) and quantified in activity counts. Tracmor recordings were also processed using a classification algorithm to recognize 6 common activity types engaged in during the day. A doubly-labeled water validated equation based on Tracmor output was used to estimate AEE. After weight loss, body weight decreased by 13Ā±4%, daily activity counts augmented by 9% (95% CI: +2%, +15%), and this increase was weakly associated with the decrease in body weight (R(2)ā=ā7%; P<0.05). After weight loss subjects were significantly (P<0.05) less sedentary (-26 min/d), and increased the time spent walking (+11 min/d) and bicycling (+4 min/d). However, AEE decreased by 0.6Ā±0.4 MJ/d after weight loss. On average, a 2-hour/day reduction of sedentary time by increasing ambulatory and generic activities was required to restore baseline levels of AEE. In conclusion, after weight loss PA increased but the related metabolic demand did not offset the reduction in AEE due to the lower body weight. Artificial MiRNA Knockdown of Platelet Glycoprotein lbĪ±: A Tool for Platelet Gene Silencing.
In recent years, candidate genes and proteins implicated in platelet function have been identified by various genomic approaches. To elucidate their exact role, we aimed to develop a method to apply miRNA interference in platelet progenitor cells by using GPIbĪ± as a proof-of-concept target protein. After in silico and in vitro screening of siRNAs targeting GPIbĪ± (siGPIBAs), we developed artificial miRNAs (miGPIBAs), which were tested in CHO cells stably expressing GPIb-IX complex and megakaryoblastic DAMI cells. Introduction of siGPIBAs in CHO GPIb-IX cells resulted in 44 to 75% and up to 80% knockdown of GPIbĪ± expression using single or combined siRNAs, respectively. Conversion of siGPIBAs to miGPIBAs resulted in reduced silencing efficiency, which could however be circumvented by tandem integration of two hairpins targeting different regions of GPIBA mRNA where 72% GPIbĪ± knockdown was achieved. CHO GPIb-IX cells transfected with the miGPIBA construct displayed a significant decrease in their ability to aggregate characterized by lower aggregate numbers and size compared to control CHO GPIb-IX cells. More importantly, we successfully silenced GPIbĪ± in differentiating megakaryoblastic DAMI cells that exhibited morphological changes associated with actin organization. In conclusion, we here report the successful use of miRNA technology to silence a platelet protein in megakaryoblastic cells and demonstrate its usefulness in functional assays. Hence, we believe that artificial miRNAs are suitable tools to unravel the role of a protein of interest in stem cells, megakaryocytes and platelets, thereby expanding their application to novel fields of basic and translational research
A cross-sectional study of nutrient intake and health status among older adults in Yogyakarta Indonesia
Many communities around the world, particularly developing countries including Indonesia, are experiencing population ageing. There is little knowledge regarding the impact of malnutrition, or its prevalence within rural compared to urban areas, on the nutritional, functional and mental status of community-living older residents in these countries. Hence, a cross-sectional study was conducted to determine socio-demographic and anthropometric characteristics, nutritional, mental and functional status, and energy and nutrient intake of community-dwelling Indonesians from both rural and urban areas of Yogyakarta. Older individuals were included in the study if they had been living in Yogyakarta for the last year and were aged ā„65 years (n = 527; mean Ā± SD age of 74 Ā± 7 years). Rural compared with urban participants had a lower level of education and income, more hospital admissions, less dietary protein intake, lower cognitive function, poorer nutritional status and grip strength, but faster gait speed while being more dependent on assistance to perform daily activities (all p < 0.05). Cognitive function was more strongly associated than nutritional status with physical function. Rural older Indonesians living in Yogyakarta were more likely than urban older people to be malnourished and cognitively impaired, and to have associated reductions in functional capacity and independence. Food services using energy- and protein-fortified meals to assist vulnerable community-residing older adults meet their dietary requirements and maintain good health and quality of life: findings from a pilot study
The effects of "standard (STD)" vs. "protein- and energy-enriched (HEHP)" food-service meals on the nutrient intake, nutritional status, functional capacity, and wellbeing of older adults was investigated using a 12 week, double-blinded, parallel group design. All participants received dietetics counseling and either an STD (2.3 MJ and 30 g protein per meal) or a HEHP (4.6 MJ and 60 g protein) hot lunchtime meal for at least 3 days/week; those who did not want food-service meals were included in the control group (CON). Twenty-nine participants completed the study (STD = 7; HEHP = 12; CON = 10). From baseline to week 12, the HEHP subjects increased their mean daily energy intake from 6151 Ā± 376 kJ to 8228 Ā± 642 kJ (p = 0.002 for effect of time) and protein intake from 67 Ā± 4 g to 86 Ā± 8 g (p = 0.014 for effect of time). The MNA (Mini Nutritional Assessment) score was increased significantly in HEHP by 4.0 Ā± 1.1 points (p = 0.001), but not in the STD and CON groups (2.8 Ā± 2.1 points and 1.8 Ā± 1.1 points, p > 0.05). No difference was found for other clinical outcomes between the groups. Effects of substitution, and adding of carbohydrate and fat to whey-protein on energy intake, appetite, gastric emptying, glucose, insulin, ghrelin, cck and glp-1 in healthy older men - a randomized controlled trial
Protein-rich supplements are used widely for the management of malnutrition in the elderly. We reported previously that the suppression of energy intake by whey protein is less in older than younger adults. The aim was to determine the effects of substitution, and adding of carbohydrate and fat to whey protein, on ad libitum energy intake from a buffet meal (180-210 min), gastric emptying (3D-ultrasonography), plasma gut hormone concentrations (0-180 min) and appetite (visual analogue scales), in healthy older men. In a randomized, double-blind order, 13 older men (75 Ā± 2 years) ingested drinks (~450 mL) containing: (i) 70 g whey protein (280 kcal; 'Pāāā'); (ii) 14 g protein, 28 g carbohydrate, 12.4 g fat (280 kcal; 'Māāā'); (iii) 70 g protein, 28 g carbohydrate, 12.4 g fat (504 kcal; 'Mā
āā'); or (iv) control (~2 kcal). The caloric drinks, compared to a control, did not suppress appetite or energy intake; there was an increase in total energy intake (drink + meal, p < 0.05), which was increased most by the Mā
āā-drink. Pāāā- and Mā
āā-drink ingestion were associated with slower a gastric-emptying time (n = 9), lower ghrelin, and higher cholecystokinin (CCK) and glucagon-like peptide-1 (GLP-1) than Māāā (p < 0.05). Glucose and insulin were increased most by the mixed-macronutrient drinks (p < 0.05). Peptide-siRNA nanocomplexes targeting NF-ĪŗB subunit p65 suppress nascent experimental arthritis
The NF-ĪŗB signaling pathway is implicated in various inflammatory diseases, including rheumatoid arthritis (RA); therefore, inhibition of this pathway has the potential to ameliorate an array of inflammatory diseases. Given that NF-ĪŗB signaling is critical for many immune cell functions, systemic blockade of this pathway may lead to detrimental side effects. siRNAs coupled with a safe and effective delivery nanoplatform may afford the specificity lacking in systemic administration of small-molecule inhibitors. Here we demonstrated that a melittin-derived cationic amphipathic peptide combined with siRNA targeting the p65 subunit of NF-ĪŗB (p5RHH-p65) noncovalently self-assemble into stable nanocomplexes that home to the inflamed joints in a murine model of RA. Specifically, administration of p5RHH-p65 siRNA nanocomplexes abrogated inflammatory cytokine expression and cellular influx into the joints, protected against bone erosions, and preserved cartilage integrity. The p5RHH-p65 siRNA nanocomplexes potently suppressed early inflammatory arthritis without affecting p65 expression in off-target organs or eliciting a humoral response after serial injections. These data suggest that this self-assembling, largely nontoxic platform may have broad utility for the specific delivery of siRNA to target and limit inflammatory processes for the treatment of a variety of diseases
Mapping children's presence in the neighbourhood
Within the work of Gert Biesta, public spaces are considered as the main fields where processes of civic learning can take place. Learning is always āin place and timeā. Place matters, not only as a spatial background or set of conditions , but as a pedagogical process in itself. So in order to facilitate processes of civic learning we need to understand how spaces function as a co-educator in its own right.. In my PhD I studied how the neighbourhood can be understood as a co-educator. The processes through which children are socialised into a given order are not universal, nor neutral. One might state that children grow up into very different orders. The neighbourhood is an important factor into these diversified socialisation processes, but the neighbourhood is also made by its residents and users themselves, including children. This emphasises that socialisation is a relational process and not a functional oneway introduction of children into a prescribed social ordeIn this contribution, a methodological framework will be developed for understanding and studying the neighbourhood of children as a co-educator. The basis of this framework is formed by the synergy between spatial, social and personal dimensions of the neighbourhood
