A008 Presence of tissue factor and other components of atherosclerosis in human aortic valve stenosis
BackgroundIt is now generally accepted that calcific aortic valve disease is an atherosclerotic-like process. Recent studies in an experimental model of aortic valve sclerosis demonstrated the presence of tissue factor (TF), the main contributor to atherosclerotic plaque thrombogenicity, in diseased valve leaflets. We assessed the hypothesis that human aortic valve disease is an atherosclerotic-like process in which TF plays an important role and evaluated the valvular expression and localization of TF and other components of atherosclerosis.MethodsCalcified aortic valves (n=52) were obtained from patients undergoing aortic valve replacement. Leaflet structure, cellular and lipid infiltration and expression of TF, its inhibitors, VEGF and other components of atherosclerosis were evaluated by histological and immunohistochemical staining. TF, TFPI, osteopontin, MMP- 9, TIMP-1 and VEGF antigen were measured by ELISA and TF and alkaline phosphatase activity were determined using chromogenic assays. Finally, we performed semi-quantification of TF transcripts by RT- PCR and further analyzed protein expression by Western blot.ResultsHistological and immunohistochemical staining of the valve leaflets revealed neovascularisation at the centre of the lesions, overall macrophage and myofibroblast infiltration and the abundant presence of MMP-9. On the other hand, TF and TFPI were associated with calcification and extracellular lipid deposits in the fibrosa and the subendothelial layer of the aortic side of the leaflets. Correspondingly, TF antigen and activity were found to be higher in calcified regions of the valve leaflets (733.29±70.49pg/mgvs 429.40±73.17pg/mg and 144.75±14.65pg/mgvs 40.15±6.19pg/mg respectively (p<0.0001)). Similar results were found for osteopontin, MMP-9, TIMP-1 and VEGF. In contrast, TFPI antigen was found to be much lower in these calcified regions (722.54±153.92pg/mgvs 2459.28±285.36pg/mg (p<0.0001)).ConclusionThese results demonstrate that aortic valve lesions display several characteristics of atherosclerosis, including TF expression. In addition, we showed that TF is colocalized with calcification and lipid deposition. Further studies are now set up to evaluate the role of TF in aortic valve disease and its association with other components of the atherosclerotic process
A model intercomparison project to study the role of plant functional diversity in the response of tropical forests to drought
Uncertainty in how the land carbon (C) sink will change over time contributes to uncertainty in Earth system model (ESM) projections of climate change. Much of the land sink is thought to reside in old-growth tropical forests, but recent analyses suggest a diminishing C sink in these forests due to rising temperatures and drought. Thus, there is an urgent need to better understand tropical forest responses to drought and to incorporate this understanding into ESMs. Previous work with vegetation demographic models (VDMs) – which represent the dynamics of individuals or cohorts, along with hydrology and biogeochemistry − suggest that functional diversity can enhance tropical forest resilience to climate change. However, there is little understanding of how different approaches to representing trait diversity and demography affect model outcomes. To explore the potential for trait diversity to moderate tropical forest responses to drought, we explored the behavior of nine VDMs, ranging from models with detailed site-level parameterizations to more generalized land models designed as ESM components. The behavior of each model was studied using soil and meteorological data collected at each of two tropical forest sites: Paracou Research Station, French Guiana, and Tapajos National Forest, Brazil. Low and high trait-diversity scenarios were simulated for each model using historical meteorology, as well as reduced rainfall scenarios.
Few models showed strong effects of trait diversity on drought resistance (short-term response of forest biomass to rainfall reduction), but most models showed positive effects of diversity on resilience (long-term recovery of forest biomass following the initial biomass loss due to rainfall reduction). Long-term recovery was always associated with shifts in community composition towards greater drought-tolerance. However, there were large differences among models in the degree and time-scale of recovery. These differences were unrelated to the goodness-of-fit of model predictions to observations of biomass, productivity, and soil moisture, suggesting that site-level calibration of model parameters is unlikely to strongly affect biodiversity-ecosystem functioning relationships in VDMs. Rather, the degree to which diversity moderated drought responses depended on which axes of trait variation were represented in the model, as well as model assumptions that affect the time-scale over which community composition shifts in response to environmental change. Our study suggests that incorporating trait diversity and demography into ESMs would likely lead to altered climate projections, but additional empirical and modeling work is needed to provide the ESM community with clear guidance on model development
Les haies fourragères dans les hauts de l'Ouest à la Réunion : l'intégration du Calliandra calothyrsus pour une protection productive
A la Réunion, les andosols constituent plus de la moitié des sols cultivés. Du fait de leur constitution et de leur histoire culturale, ils présentent des propriétés spécifiques qui leur confèrent une sensibilité particulière à l'érosion pluviale. La conservation des sols ne peut satisfaire les paysans que si elle n'entrave pas la production et s'ils y voient une valorisation possible. Dans les "Hauts de l'Ouest", l'agroforesterie à vocation fourragère apparaît comme un compromis satisfaisant pour l'agriculteur. Dans ce contexte, nous nous sommes particulièrement attachés à évaluer les potentialités protectives et productives d'une légumineuse, le Calliandra calothyrsus. Notre étude nous a permis de dégager des perspectives prometteuses pour cet arbuste. Il semble contribuer efficacement à la restauration de fertilité, et à la lutte anti-érosive en améliorant l'état structural du sol. Produisant un fourrage de qualité en quantité, il ne paraît pas par ailleurs avoir d'incidence sur les rendements culturau

Beyond variance: simple random distributions are not a good proxy for intraspecific variability in systems with environmental structure
The role of intraspecific variability (IV) in shaping community dynamics and species coexistence has been intensively discussed over the past decade and modelling studies have played an important role in that respect. However, these studies often implicitly assume that IV can be represented by independent random draws around species-specific mean parameters. This major assumption has largely remained undiscussed, although a great part of observed IV is structured in space or time, in particular when environmental dimensions that influence individual performance are imperfectly characterised or unobserved in the field. To test the impact of this strong assumption on the outcome of community dynamics models, we designed a simulation experiment where we varied the level of knowledge of the environment in virtual communities, resulting in different relative importance of explained vs unexplained spatial individual variation in performance. We used a community dynamics simulator to generate communities where the unexplained individual variation is, or is not, added as an unstructured random noise. Communities simulated with unstructured IV never reached the community diversity and composition of those where all the variation was explained and structured (perfect knowledge model). This highlights that incorporating unstructured IV (i.e. a random noise) to account for unexplained (but structured) variation can lead to incorrect simulations of community dynamics. In addition, the effects of unstructured IV on community diversity and composition depended on the relative importance of structured vs unstructured IV, i.e. on the level of knowledge of the environment, which may partly explain the contrasting results of previous studies on the effect of IV on species coexistence. In particular, the effect of unstructured IV on community diversity was positive when the proportion of structured IV vs unstructured IV in the model was low, but negative when this proportion was high. This is because unstructured random noise can either limit the competitive exclusion of inferior competitors in low dimensions or destabilise tight niche partitioning in high dimension. Our study suggests that it is crucial to account for the sources and structure of observed IV in real communities to better understand its effect on community assembly and properly include it in community dynamics models
