Increasing genetic variability in black oats using gamma irradiation

    Get PDF
The black oat (Avena strigosa Schreb) is commonly used for forage, soil cover, and green manure. Despite its importance, little improvement has been made to this species, leading to high levels of genotypic disuniformity within commercial cultivars. The objective of this study was to evaluate the efficiency of different doses of gamma rays [ ^60 Co] applied to black oat seeds on the increase of genetic variability of agronomic traits. We applied doses of 0, 10, 50, 100, and 200 Gy to the genotype ALPHA 94087 through exposure to [ ^60 Co]. Two experiments were conducted in the winter of 2008. The first aimed to test forage trait measurements such as plant height, dry matter yield, number of surviving tillers, and seedling stand. The second test assessed seed traits, such as yield and dormancy levels. Gamma irradiation seems not to increase seed yield in black oats, but it was effective in generating variability for the other traits. Tiller number and plant height are important selection traits to increase dry matter yield. Selection in advanced generations of mutant populations can increase the probability of identifying superior genotypes

Cross-cultural validity of the Animated Activity Questionnaire for patients with hip and knee osteoarthritis: a comparison between the Netherlands and Brazil

    Get PDF
Background: The Animated Activity Questionnaire (AAQ) was developed in the Netherlands to assess activity limitations in individuals with hip/knee osteoarthritis (HKOA). The AAQ is easy to implement and minimizes the disadvantages of questionnaires and performance-based tests by closely mimicking real-life situations. The AAQ has already been cross-culturally validated in six other countries. Objective: To assess the cross-cultural validity, the construct validity, the reliability of the AAQ in a Brazilian sample of individuals with HKOA, and the influence of formal education on the construct validity of the AAQ. Methods: The Brazilian sample (N = 200), mean age 64.4 years, completed the AAQ and the Western Ontario and McMaster Universities Index (WOMAC). A subgroup of participants performed physical function tests and completed the AAQ twice with a one-week interval. The Dutch sample (N = 279) was included to examine Differential Item Functioning (DIF) between the scores obtained in the Netherlands and Brazil. For this purpose, ordinal regression analyses were used to evaluate whether individuals with the same level of activity limitations from the two countries (the Dutch as the reference group) scored similarly in each AAQ item. To evaluate the construct validity, correlation coefficients were calculated between the AAQ, the WOMAC domains, and the performance-based tests. To evaluate reliability, the Cronbach's alpha coeffi-cient, the intraclass correlation coefficient, and the standard error of measurement (SEM) were calculated. Results: The AAQ showed significant correlations with all the WOMAC domains and performance -based tests (rho=0.46-0.77). The AAQ showed high internal consistency (Cronbach's alpha=0.94), excellent test-retest reliability (ICC2,1 = 0.98), and small SEM (2.25). Comparing to the scores from the Netherlands, the AAQ showed DIF in two items, however, they did not impact on the total AAQ score (rho=0.99). Conclusion: Overall, the AAQ showed adequate cross-cultural validity, construct validity, and reliability, which enables its use in Brazil and international/multicenter studies. Relationship between professional motivations and the expectation of staying at the same workplace: a cross-sectional descriptive study with physicians in Portugal

    Get PDF
There is a global trend related to the migration of health professionals, particularly physicians, to the most densely populated areas. In Portugal, the unequal distribution of medical professionals is justified by the fact that in the most favoured areas, there are more career opportunities, better infrastructures and equipment, better working conditions and supervision, more and better social facilities and better salary. The aim of this work was to identify professional reasons associated with the expectation of staying at the same workplace. It was selected a simple random sample of 594 physicians out of a total of 18,711 registered physicians in Northern Regional Section of the Order of Physicians (NRSOP), Portugal Most physicians would like to stay at the same workplace due to: the prospect of training and working at the current location, the organizational level of the unit, the level of differentiation of the hospital/health centre, the availability to use the state-of-the-art medical and surgical technology, better rewards, the level of work differentiation in the unit/service, the possibility of reconciling public and private practice, good references of the current unit/working group, avoid unit/workgroup with bad references and the prospect of career progression. This study revealed that, for physicians, professional motivations/purposes have an influence on the expectations of staying or not at the same workplace. Therefore, the study recommends that policy makers should prioritize professional reasons when formulating and implementing measures to promote the fixing of medical professionals in order to achieve greater equity in access to health care. This work is supported by: the European Structural and Investment Funds in the FEDER component, through the Operational Competitiveness and Internationalization Programme (COMPETE 2020) [Project No. 006971 (UID/SOC/04011)]; and national funds, through the FCT – Portuguese Foundation for Science and Technology under the project UID/SOC/04011/2013.info:eu-repo/semantics/publishedVersio

Field theory description of vacuum replicas

    Get PDF
In this paper we develop a systematic quantum field theory based approach to the vacuum replica recently found to exist in effective low energy models in hadronic physics. A local operator creating the replica state is constructed explicitly. We show that a new effective quark-quark force arises in result of replica existence. Phenomenological implications of such a force are also briefly discussed.Comment: RevTeX4, 23 pages, 4 Postscript figures, uses epsfig.sty, to appear in Phys.Rev.

SPLUNC1 degradation by the cystic fibrosis mucosal environment drives airway surface liquid dehydration

    Get PDF
The multi-organ disease cystic fibrosis (CF) is caused by mutations in the cystic fibrosis transmembrane regulator gene (CFTR) that lead to diminished transepithelial anion transport. CF lungs are characterised by airway surface liquid (ASL) dehydration, chronic infection/inflammation and neutrophilia. Dysfunctional CFTR may upregulate the epithelial Na+ channel (ENaC), further exacerbating dehydration. We previously demonstrated that short palate lung and nasal epithelial clone 1 (SPLUNC1) negatively regulates ENaC in normal airway epithelia. Here, we used pulmonary tissue samples, sputum and human bronchial epithelial cells (HBECs) to determine whether SPLUNC1 could regulate ENaC in a CF-like environment. We found reduced endogenous SPLUNC1 in CF secretions, and rapid degradation of recombinant SPLUNC1 (rSPLUNC1) by CF secretions. Normal sputum, containing SPLUNC1 and SPLUNC1-derived peptides, inhibited ENaC in both normal and CF HBECs. Conversely, CF sputum activated ENaC, and rSPLUNC1 could not reverse this phenomenon. Additionally, we observed upregulation of ENaC protein levels in human CF bronchi. Unlike SPLUNC1, the novel SPLUNC1-derived peptide SPX-101 resisted protease degradation, bound apically to HBECs, inhibited ENaC and prevented ASL dehydration following extended pre-incubation with CF sputum. Our data indicate that CF mucosal secretions drive ASL hyperabsorption and that protease-resistant peptides, e.g. SPX-101, can reverse this effect to rehydrate CF ASL
