    Influence of Maternal Gestational Treatment with Mycobacterial Antigens on Postnatal Immunity in an Experimental Murine Model

    BACKGROUND: It has been proposed that the immune system could be primed as early as during the fetal life and this might have an impact on postnatal vaccination. Therefore, we addressed in murine models whether gestational treatment with mycobacterial antigens could induce better immune responses in the postnatal life. METHODS/FINDINGS: BALB/c mice were treated subcutaneously (s.c.) at the second week of gestation with antigen (Ag)85A or heparin-binding hemagglutinin (HBHA) in the absence of adjuvant. Following birth, offspring mice were immunized intranasally (i.n.) with the same antigens formulated with the adjuvant cholera toxin (CT) at week 1 and week 4. One week after the last immunization, we assessed antigen-specific recall interferon gamma (IFN-gamma) responses by in vitro restimulation of lung-derived lymphocytes. Protection against infection was assessed by challenge with high dose Mycobacterium bovis Bacille Calmette-Guérin (BCG) given i.n. We found that recall IFN-gamma responses were higher in the offspring born to the treated mother compared to the untreated-mother. More importantly, we observed that the offspring born to the treated mother controlled infection better than the offspring born to the untreated mother. Since the gestational treatment was done in absence of adjuvant, essentially there was no antibody production observed in the pregnant mice and therefore no influence of maternal antibodies was expected. We hypothesized that the effect of maternal treatment with antigen on the offspring occurred due to antigen transportation through placenta. To trace the antigens, we conjugated fluorescent nanocrystals with Ag85A (Qdot-ITK-Ag85A). After inoculation in the pregnant mice, Qdot-ITK-Ag85A conjugates were detected in the liver, spleen of pregnant females and in all the fetuses and placentas examined.     Multi Objective PSO with Passive Congregation for Load Balancing Problem

    High-level architecture (HLA) and Distributed Interactive Simulation (DIS) are commonly used for the distributed system. However, HLA suffers from a resource allocation problem and to solve this issue, optimization of load balancing is required. Efficient load balancing can minimize the simulation time of HLA and this optimization can be done using the multi-objective evolutionary algorithms (MOEA). Multi-Objective Particle Swarm Optimization (MOPSO) based on crowding distance (CD) is a popular MOEA method used to balance HLA load. In this research, the efficiency of MOPSO-CD is further improved by introducing the passive congregation (PC) method. Several simulation tests are done on this improved MOPSO-CD-PC method and the results showed that in terms of Coverage, Spacing, Non-dominated solutions and Inverted generational distance metrics, the MOPSO-CD-PC performed better than the previous MOPSO-CD algorithm. Hence, it can be a useful tool to optimize the load balancing problem in HLA

    Salt Stress Tolerance in Rice: Emerging Role of Exogenous Phytoprotectants

    Excess salinity in soil is one of the major environmental factors that limit plant growth and yield of a wide variety of crops including rice. On the basis of tolerance ability toward salinity, rice is considered as salt-sensitive crop, and growth and yield of rice are greatly affected by salinity. In general, rice can tolerate a small amount of saltwater without compromising the growth and yield. However, it greatly depends on the types and species of rice and their growth stage. Salinity-induced ionic and osmotic stresses reduce rate of photosynthesis and consequently cause oxidative stress, which is also responsible for growth reduction. The negative effects of salt stress that mentioned ultimately reduced yield of most crops including rice, except some halophytes. In recent decades, researchers have developed various approaches toward making salt-tolerant rice varieties. Using phytoprotectants is found to be effective in conferring salt tolerance to rice plants.     Reflections on the multi-sectoral response to COVID-19 in Bangladesh's Rohingya refugee camps

    While the COVID-19 pandemic continues to impact people globally, refugees comprise a vulnerable population, particularly those living in densely populated areas. In Bangladesh, Cox’s Bazar is currently home to almost a million Rohingya refugees. Because of the lack of healthcare, sanitation and water, as well as overcrowding, refugees were at high risk of becoming ill during the early phases of the COVID-19 pandemic in 2020. Moreover, superstitions and lack of trust in the healthcare system threaten to put the community at further risk. To prevent tragic consequences, national and international attention and action are required to strengthen the health system for Rohingya refugees. The community will require surveillance and testing, infection prevention and control measures, adequate food supplies, and access to improved healthcare services. This paper calls for a multi-sectoral approach to developing an action plan and implementation strategy to minimize the impact of COVID-19 on this vulnerable population

    Investigation of Drug Distribution Process and Pharmacy Practices on Model Pharmacy in Dhaka City, Bangladesh

    Objectives: In Bangladesh, Drug distribution process is very important cause of our increasing economic and development. In the recent days Model pharmacy has become an important theme of study for the student in pharmacy. In Bangladesh the introduction of the model pharmacy practice likes other skilled and semi-skilled health professionals will certainly improve primary health care system. The aim of the study was to develop a strategy to improve the overall services of model pharmacy in Bangladesh. Methodology: A total of 20 model pharmacies were the study subject. The study cover the model pharmacy located in Dhaka city, where each pharmacy was accounted through a questioner on random basis. The form includes their facilities, management system, drug distribution process and counseling system. Results: Study showed that, out of 20 Pharmacists, 95% of them used Software tools for their drug dispensing system. There was a helpline system for individual, online order within a short time of delivery system, discount system and membership card. The most important significant factors are counseling by “A grade” pharmacists, patient database system, affordable price and quality of medicine. Conclusions: If we improve those model pharmacies and if the aim is successful, then it will be spread across Bangladesh and model pharmacy will be established gradually with digitalization.     Loss of Zbtb32 in NOD mice does not significantly alter T cell responses. [version 2; referees: 2 approved]

    Background: We previously identified the transcriptional regulator Zbtb32 as a factor that can promote T cell tolerance in the Non-Obese Diabetic (NOD) mouse, a model of Type 1 diabetes. Antigen targeted to DCIR2+ dendritic cells (DCs) in vivo inhibited both diabetes and effector T cell expansion in NOD mice. Furthermore, Zbtb32 was preferentially induced in autoreactive CD4 T cells stimulated by these tolerogenic DCIR2+ DCs, and overexpression of Zbtb32 in islet-specific T cells inhibited the diabetes development by limiting T cell proliferation and cytokine production. Methods: To further understand the role of Zbtb32 in T cell tolerance induction, we have now used CRISPR to target the Zbtb32 gene for deletion directly in NOD mice and characterized the mutant mice. We hypothesized that the systemic loss of Zbtb32 in NOD mice would lead to increased T cell activation and increased diabetes pathogenesis. Results: Although NOD.Zbtb32-/- male NOD mice showed a trend towards increased diabetes incidence compared to littermate controls, the difference was not significant. Furthermore, no significant alteration in lymphocyte number or function was observed. Importantly, in vitro stimulation of lymphocytes from NOD.Zbtb32-/- mice did not produce the expected hypersensitive phenotype observed in other genetic strains, potentially due to compensation by homologous genes. Conclusions: The loss of Zbtb32 in the NOD background does not result in the expected T cell activation phenotype

    Mental Health Condition among University Students of Bangladesh during the Critical COVID-19 Period

    Bangladesh’s education sector has been in a state of flux since COVID-19. During the pandemic, all university campuses were closed. There was a mental health issue among the students. This study aims to examine the mental health condition and the determinants that contribute to adverse mental health conditions among university students of Bangladesh. A survey was performed online among university students in Bangladesh, in mid-June 2020 when averaging 3345 affected cases of the population daily. The convenience sampling technique was used and the survey gathered data from 365 university students. The relationship between general information and Depression, Anxiety, and Stress Scale 21 (DASS-21) subscales of university students was determined. The questionnaire was administered to respondents during the pandemic, which ensured fast replies. Linear regression models were used for statistical analysis. University students indicated normal levels of depression (30.41%), anxiety (43.29%), and stress (47.40%). However, a disproportionate number of extremely depressed, anxious, and stressed university students suggested a mental health status of concern. There were significant connections between the individual’s opinion of social satisfaction, mental health concerns, and the present location’s safety with an undesirable mental health condition. Female students were shown to be much more anxious and stressed than male students. Capital Dhaka city students were more depressed and anxious than students outside of Dhaka. Financial and psychological support for students may help mitigate the psychological impact. Authorities should make effective efforts to reduce mental health problems among these students. This research may aid organizations, health care providers, and social workers in their attempts to prepare for and respond to pandemics
