    Limited Activity Of Miltefosine In Murine Models Of Cryptococcal Meningoencephalitis And Disseminated Cryptococcosis

    Miltefosine is an alkyl phosphocholine with good oral bioavailability and in vitro activity against Cryptococcus species that has gained interest as an additional agent for cryptococcal infections. Our objective was to further evaluate the in vivo efficacy of miltefosine in experimental in vivo models of cryptococcal meningoencephalitis and disseminated cryptococcosis. Mice were infected intracranially or intravenously with either C. neoformans USC1597 or H99. Miltefosine treatment (1.8 to 45 mg/kg of body weight orally once daily) began at either 1 h or 1 day postinoculation. Fluconazole (10 mg/kg orally twice daily) or amphotericin B deoxycholate (3 mg/kg intraperitoneally once daily) served as positive controls. In our standard models, miltefosine did not result in significant improvements in survival or reductions in fungal burden against either C. neoformans isolate. There was a trend toward improved survival with miltefosine at 7.2 mg/kg against disseminated cryptococcosis with the H99 strain but only at a low infecting inoculum. In contrast, both fluconazole and amphotericin B significantly improved survival in mice with cryptococcal meningoencephalitis and disseminated cryptococcosis due to USC1597. Amphotericin B also improved survival against both cryptococcal infections caused by H99. Combination therapy with miltefosine demonstrated neither synergy nor antagonism in both models. These results demonstrate limited efficacy of miltefosine and suggest caution with the potential use of this agent for the treatment of C. neoformans infections.Pharmac

    Structural Insights into Dissimilatory Sulfite Reductases: Structure of Desulforubidin from Desulfomicrobium Norvegicum

    Dissimilatory sulfite reductases (dSiRs) are crucial enzymes in bacterial sulfur-based energy metabolism, which are likely to have been present in some of the earliest life forms on Earth. Several classes of dSiRs have been proposed on the basis of different biochemical and spectroscopic properties, but it is not clear whether this corresponds to actual physiological or structural differences. Here, we describe the first structure of a dSiR from the desulforubidin class isolated from Desulfomicrobium norvegicum. The desulforubidin (Drub) structure is assembled as α2β2γ2, in which two DsrC proteins are bound to the core [DsrA]2[DsrB]2 unit, as reported for the desulfoviridin (Dvir) structure from Desulfovibrio vulgaris. Unlike Dvir, four sirohemes and eight [4Fe–4S] clusters are present in Drub. However, the structure indicates that only two of the Drub coupled siroheme-[4Fe–4S] cofactors are catalytically active. Mass spectrometry studies of purified Drub and Dvir show that both proteins present different oligomeric complex forms that bind two, one, or no DsrC proteins, providing an explanation for conflicting spectroscopic and biochemical results in the literature, and further indicating that DsrC is not a subunit of dSiR, but rather a protein with which it interacts

    Major practicum as a learning site for exercise science professionals: A pilot study

    Exercise science is now an integral part of the allied health framework in Australia and graduates from accredited programmes are equipped with skills recognised as being important in the prevention and management of lifestyle-related diseases. This pilot study sought to determine the experiences of 11 final-year exercise science students in their major practicum and identify skills learned and developed while on placement. Analysis of the interview data established that the students worked with clients from a broad range of sociocultural and socioeconomic backgrounds, both within and between practicum sites; the students' experiences and their preparedness to engage with clients from different backgrounds varied as a result. Although the students generally reported being technically skilled for their major placement, many reported being underprepared to deal with people from different backgrounds. However, all participants held that their interpersonal skills greatly improved in response to their placement and several remarked that they developed their problem-solving skills through watching and assisting their supervisors work with clients. The present study confirms the practicum as a critical learning site for improving communication and problem-solving skills with exercise science and exercise physiology students

    The role played by cell-substrate interactions in the pathogenesis of osteoclast-mediated peri-implant osteolysis

    Prosthetic wear debris-induced peri-implant osteolysis is a major cause of aseptic loosening after total joint replacement. In this condition, wear particles released from the implant components induce a granulomatous inflammatory reaction at the interface between implant and adjacent bone, leading to progressive bone resorption and loss of fixation. The present study was undertaken to characterize definitively the phenotype of osteoclast-like cells associated with regions of peri-implant focal bone resorption and to compare the phenotypic features of these cells with those of mononucleated and multinucleated cells associated with polyethylene wear particles. Peri-implant tissues were obtained from patients undergoing hip revision surgery for aseptic loosening after total joint replacement. Cells were examined for the expression of several markers associated with the osteoclast phenotype using immunohistochemistry, histochemistry, and/or in situ hybridization. CD68 protein, a marker expressed by multiple macrophage lineage cell types, was detected in mononucleated and multinucleated cells associated with polyethylene particles and the bone surface. Cathepsin K and tartrate-resistant acid phosphatase were expressed highly in both mononucleated and multinucleated cells associated with the bone surface. Levels of expression were much lower in cells associated with polyethylene particles. High levels of β(3 )integrin protein were detected in cells in contact with bone. Multinucleated cells associated with polyethylene particles exhibited faint positive staining. Calcitonin receptor mRNA expression was detected solely in multinucleated cells present in resorption lacunae on the bone surface and was absent in cells associated with polyethylene particles. Our findings provide further evidence that cells expressing the full repertoire of osteoclast phenotypic markers are involved in the pathogenesis of peri-implant osteolysis after total joint replacement. They also demonstrate that foreign body giant cells, although believed to be phenotypically and functionally distinct from osteoclasts, express many osteoclast-associated genes and gene products. However, the levels and patterns of expression of these genes in the two cell types differ. We speculate that, in addition to the role of cytokines and growth factors, the substrate with which these cells interact plays a critical role in their differential phenotypic and functional properties

    A New Rodent Model to Assess Blood Stage Immunity to the Plasmodium falciparum Antigen Merozoite Surface Protein 119 Reveals a Protective Role for Invasion Inhibitory Antibodies

    Antibodies capable of inhibiting the invasion of Plasmodium merozoites into erythrocytes are present in individuals that are clinically immune to the malaria parasite. Those targeting the 19-kD COOH-terminal domain of the major merozoite surface protein (MSP)-119 are a major component of this inhibitory activity. However, it has been difficult to assess the overall relevance of such antibodies to antiparasite immunity. Here we use an allelic replacement approach to generate a rodent malaria parasite (Plasmodium berghei) that expresses a human malaria (Plasmodium falciparum) form of MSP-119. We show that mice made semi-immune to this parasite line generate high levels of merozoite inhibitory antibodies that are specific for P. falciparum MSP-119. Importantly, protection from homologous blood stage challenge in these mice correlated with levels of P. falciparum MSP-119–specific inhibitory antibodies, but not with titres of total MSP-119–specific immunoglobulins. We conclude that merozoite inhibitory antibodies generated in response to infection can play a significant role in suppressing parasitemia in vivo. This study provides a strong impetus for the development of blood stage vaccines designed to generate invasion inhibitory antibodies and offers a new animal model to trial P. falciparum MSP-119 vaccines

    Mercury Concentrations in Fishes of the Madeira River: Results and Recomendations for Human Consumption

    Samples of eleven fish species were collected during September and October of 1991 in the gold mining area of Cachoeira de Teotônio and in the control area Guajará-Mirim, along the Madeira River, (Rondônia, Brazil) and anaiized for total mercury by cold vapor atomic absorption spectrofotometry. Almost all fish predators from the gold mining area had mercury concentrations above the maximal level of 0.5 ug.g"1, permitted for human consumption by Brazilian Secretary of Health and the World Health Organization and much greater than those found in predatory fish species in the control area were all below this critical value (0.5 pg.g\'X demonstrating the influence of anthropogenic contamination in the gold mining area. In both areas, non-predators fish species had lower mercury concentrations than predators fish species, indicating the effects of biomagnification of mercury in the food chain. Safety limits for fish consumption by the human populations in this gold mining area were estabilished considering the mercury levels encontred in fish and the critical intake rate associated with the signs of mercury poisoning. Pacu, could be consumed without restrictions, matrinchã, curimatã, mandi and tucunaré, could be consumed with moderation and large predatory fish species (mostly Siluriformes), could be consumed only sporadically. Some factors that could influence mercury contaminations in "ribeirinhos" are discussed.A concentração de mercúrio total foi determinada em onze espécies de peixes coletados de setembro a outubro de 1991, na área de garimpo de ouro da Cachoeira de Tcotônio e da área considerada controle em Guajará-Mirim, ambas no rio Madeira, Estado de Rondônia. Utilizou-se, para a análise, a técnica de espectrofotometria de absorção atômica com gerador de vapor frio de Hg. Quase todos os peixes predadores da área de garimpo tiveram concentrações de mercúrio acima do nível critico de 0,5 pg.g"1 permitido para consumo humano pelo Ministério da Saúde do Brasil e Organização Mundial de Saúde, sendo bem maiores que as concentrações de mercúrio nas espécies predadoras da área controle, evidenciando a influência do garimpo de ouro na contaminação dos peixes por mercúrio. As espécies não-predadoras tiveram concentrações de mercúrio abaixo daquelas das espécies predadoras para as duas áreas, indicando o efeito da biomagnificação do mercúrio na cadeia alimentar. Procurou-se estabelecer limites para o consumo de peixes pelas populações humanas da área de garimpo estudada, calculando-se a taxa de ingestão necessária para se desenvolver os primeiros sintomas de contaminação mercurial. Concluiu-se que espécies como o pacu podem ser consumidas sem restrição, já os peixes como matrinchã, curimatã, mandi e tucunaré, deveriam ser consumidos com moderação e que a maioria dos peixes predadores (em geral Siluriformes), deveriam ser consumidos apenas esporadicamente. Discutem-se alguns fatores que poderiam estar interagindo no processo de contaminação por mercúrio dos ribeirinhos dessa área de garimpo

    Sqrt{shat}_{min} resurrected

    We discuss the use of the variable sqrt{shat}_{min}, which has been proposed in order to measure the hard scale of a multi parton final state event using inclusive quantities only, on a SUSY data sample for a 14 TeV LHC. In its original version, where this variable was proposed on calorimeter level, the direct correlation to the hard scattering scale does not survive when effects from soft physics are taken into account. We here show that when using reconstructed objects instead of calorimeter energy and momenta as input, we manage to actually recover this correlation for the parameter point considered here. We furthermore discuss the effect of including W + jets and t tbar+jets background in our analysis and the use of sqrt{shat}_{min} for the suppression of SM induced background in new physics searches.Comment: 23 pages, 9 figures; v2: 1 figure, several subsections and references as well as new author affiliation added.
