Nanotechnology in ligature-induced periodontitis: protective effect of a doxycycline gel with nanoparticules
OBJECTIVES: The aim of this study was to test the efficacy of a locally applied 8.5% nanostructured doxycycline (DOX) gel in preventing alveolar bone loss in experimental periodontal disease (EPD) in rats by using the tapping mode atomic force microscopy (AFM). MATERIAL AND METHODS: EPD was induced in 24 Wistar rats. Animals were treated with the doxycycline gel topically, immediately after EPD induction, and 3 times a day during 11 days. Four groups (n=6) were formed as follows: Naïve group (animals not subjected to EPD nor treated); non-treated (NT) group (animals subjected to EPD, but not treated); vehicle gel (VG) group (animals subjected to EPD and treated with topical gel vehicle); and DOX group (test group): animals subjected to EPD and treated with the 8.5% DOX gel. In order to investigate topographical changes in histological sections, a novel simple method was used for sample preparation, by etching sections from paraffin-embedded specimens with xylol. Nanostructured transdermal hormone replacement therapy for relieving menopausal symptoms: a confocal Raman spectroscopy study
OBJECTIVE: To determine the safety and efficacy of a transdermal nanostructured formulation of progesterone (10%) combined with estriol (0.1%) + estradiol (0.25%) for relieving postmenopausal symptoms. METHODS: A total of 66 postmenopausal Brazilian women with climacteric symptoms of natural menopause received transdermal nanostructured formulations of progesterone and estrogens in the forearm daily for 60 months to mimic the normal ovarian secretory pattern. Confocal Raman spectroscopy of hormones in skin layers was performed. Clinical parameters, serum concentrations of estradiol and follicle-stimulating hormone, blood pressure, BI-RADS classification from bilateral mammography, and symptomatic relief were compared between baseline and 60 months post-treatment. Nanotechnology in hormone replacement therapy: Safe and efficacy of transdermal estriol and estradiol nanoparticles after 5 Years Follow-Up Study
This study aimed to evaluate the safety and efficacy of a novel protocol of transdermal Hormone Replacement Therapy (HRT) based on a nanostructured formulation of Estriol (0.1 %) + Estradiol (0.25 %) restoring serum levels and relieving menopausal symptoms. We evaluated 122 women with mean age of 56.88 (± 6.27) as part a longitudinal prospective study on post-menopausal women with natural menopause, received in the right forearm a transdermal formulation of (EE) daily for 60 months. Clinical parameters including the degree of satisfaction with symptomatic relief, serum concentrations of estradiol, weight, blood pressure, and bilateral mammography BI-RADS were compared between the baseline and five years after treatment. New evidence regarding this HRT protocol was assessed. The transdermal nanoformulation estradiol improved clinical parameters. Satisfaction with treatment was 92 %. Serum concentrations of estradiol changed significantly after treatment (p 0.05) over the years. No vaginal bleeding was observed. Bilateral mammography assessment of the breasts following 60 months of HRT with bioidentical estradiol treatment found normal results in all women. This paper shows for the first time the effectiveness of a nanostructured transdermal formulation enhancer on the delivery of estradiol and estriol measured in vivo using Raman Confocal Spectroscopy. The Nanostructured formulation is safe and effective in reestablishing estradiol serum levels and relieving menopausal symptoms. Anti-inflammatory effect of diclofenac diethylammonium gel on acute phase of ligature induced periodontitis in rats
This study aimed to evaluate the effect of a diclofenac diethylammonium gel 10 mg/g (DD) on acute phase of ligature induced periodontitis model in rats. Experimental Periodontitis Disease (EPD) was induced in 30 Wistar rats subjected to ligature placement on left molars. Animals were treated with (DD), immediately after (EPD) induction. Saline-based gel (SG) was utilized as negative control and DD gel 10 mg/g was the tested substance. Animals were randomly assigned into the groups. The periodontium and the surrounding gingiva were examined at histopathology, as well as the neutrophil influx into the gingiva was assayed using myeloperoxidase activity levels by ELISA method. DD treatment reduced tissue lesion at histopathology coupled to decreased myeloperoxidase activity production in gingival tissue when compared to the saline gel control group (p < 0.05). The DD gel was able to provide a significant myeloperoxidase decreasing in gingiva tissue confirming to be effective in reducing gingival inflammation in this model.Colegio de Farmacéuticos de la Provincia de Buenos Aire
Biolipídio B2: desenvolvimento de um veículo estável para reposição hormonal transdérmica nanoestruturada
A administração de hormônios pela via oral se configura como um grave problema, uma vez
que diversos efeitos colaterais estão relacionados a tal procedimento. O objetivo do presente
estudo foi avaliar a segurança e a eficácia de um novo produto destinado a Terapia de
Reposição Hormonal transdérmica nanoestruturada (TRHN), com base em uma formulação
patenteada sob No. US 2013/0123220A1. Tal formulação foi capaz de restabelecer os níveis
séricos de Estradiol (0,1%) + Estradiol (0,25%), em 122 mulheres menopausadas com idade
média de 56,88 (± 6,27). A avaliação faz parte de um estudo prospectivo longitudinal.
Parâmetros clínicos, incluindo o grau de satisfação com alívio sintomático, concentrações
séricas de estradiol, peso, pressão arterial, foram comparados entre a o início e o final do
tratamento. Os achados mostram que o BIOLIPÍDEO/B2®
foi eficaz e seguro em restabelecer
os níveis séricos hormonais sem efeitos colaterais. A satisfação com tratamento foi de 92%.
As concentrações no soro de estradiol foram significativamente maiores após tratamento
(p<0,05). Peso e pressão arterial sistólica e diastólica não apresentaram diferenças
significativas (p>0,05) ao longo do tratamento. Não Foi observada hemorragia vaginal.
Avaliação de mamografia bilateral das mamas o tratamento encontraram resultados normais
em todas as mulheres. Este estudo mostra pela primeira vez a eficácia de uma formulação
transdérmica nanoestruturada na entrega transdérmica de estradiol e o estriol medido in vivo
utilizando Espectroscopia Raman confocal. A formulação do BIOLIPÍDEO/B2®
é segura e
eficaz em restabelecer os níveis séricos de estradiol e de aliviar os sintomas da menopausa. A
formulação pode servir como uma boa escolha para a terapia de reposição hormonal para

Testosterone Nanoemulsion Prevents Prostate Cancer: PC-3 and LNCaP Cell Viability In Vitro
For many years, it has been speculated that elevated testosterone levels may be critically involved in the genesis and proliferation of prostate cancer. Methods: The effect of testosterone on the metabolic activity of hormone-independent [PC-3] and hormone-dependent [LNCAP] cancer cells was investigated in vitro. Additionally, the impact of testosterone nanoemulsion [nanocare®] on cell viability was accessed by flow cytometry. Results: Despite the dependency of the normal prostate and of most prostatic cancers upon androgens, the obtained results indicate that, contrary to prevailing opinion, the supplementation of testosterone with higher doses in nanoemulsion was able to lower the metabolic activity and viability of prostate cancer cells. Conclusions: We conclude that the growth of hormone-independent and hormone-dependent prostate cancer cells was reduced by the exposure of a nanoemulsion of bioidentical testostosterone in vitro. To the best of our knowledge, this is the first time that the potential effect of a testosterone nanoemulsion on the metabolic activity of prostate cancer cells has been shown. Such tests suggest that the growth of hormone-independent and hormone-dependent prostate cancer cells was reduced by the administration of bioidentical testostosterone, and this might be an interesting strategy for prostate cancer treatment in diagnosed patients
Vaginal probiotics on the outcomes of premature rupture of membranes: a pilot randomised clinical trial
The published literature shows that a balanced vaginal microbiota can have a favourable impact on decreasing the prevalence of premature rupture of membranes (PROM). We studied the effects of vaginal probiotics (Lactobacillus rhamnosus and L. gasseri) on the outcomes of PROM in pregnant women between 24 and 36 weeks of gestation. We performed a randomised, triple-blind, placebo-controlled study. A total of 27 participants were divided into two groups—group 1 (probiotics; n = 16) and group 2 (placebo; n = 11). Although most outcomes did not show much variation, the latency period and gestational age at delivery were higher in group 1 than in group 2. Thus, vaginal use of L. rhamnosus and L. gasseri along with standard treatment appears to increase the latency period and gestational age at delivery in women with PROM.IMPACT STATEMENT What is already known on this subject? Delaying delivery increases the risk of infections, but the prolongation of pregnancy allows further foetal maturation, thus reducing the risk of complications associated with premature birth. Moreover, a more extended latency period in pregnancies of <37 weeks of gestation is associated with a better neonatal prognosis. What do the results of this study add? The use of probiotics (L. rhamnosus and L. gasseri) prolongs the latency period in pregnant women with premature rupture of membranes. What are the implications of these findings for clinical practice and/or further research? Educação escolar em comunidades quilombolas do território de identidade do Velho Chico-BA
Este artigo visa apresentar algumas reflexões a partir de uma pesquisa de doutorado realizada em três escolas, situadas em comunidades quilombolas do município de Bom Jesus da Lapa-BA, no Território de Identidade do Velho Chico. Buscamos analisar de que modo a educação escolar dialoga com as especificidades étnicoculturais das comunidades quilombolas. Para a coleta de dados, utilizamos entrevistas semiestruturadas com professores, gestores, coordenadora pedagógica, pais e lideranças quilombolas, grupos focais com os alunos, análises dos projetos político-pedagógicos das escolas, observações de eventos, registros fotográficos e anotações em diário de campo. Os resultados demonstram que as escolas tratam as especificidades étnico-culturais das comunidades quilombolas de maneira pontual, descontextualizada e superficial, em datas comemorativas, no componente curricular de história, gincanas e outros eventos. Todavia, constatamos algumas experiências pedagógicas significativas que vêm transgredindo os currículos escolares para dialogarcom o universo sociocultural dos aluno
