Can Hatred Speak? On the Linguistic Dimensions of Hate Crime
Starting from the pressing issue of hate crime and hate speech, as addressed by several EU Framework Decisions and recently by the International Criminal Tribunal for the former Yugoslavia (ICTY), the article focuses on the complex and often contested relation of language and violence. Whereas recent accounts are usually concerned with the violent dimension of speech and language, the article approaches the question from a different methodological angle and asks in which way hatred and violence might be understood as a form of speaking and address. This approach is based on the more general thesis that it is possible neither to gain an adequate understanding of speech and language without considering their violent force, nor to gain an adequate account of interpersonal violence without considering its linguistic dimension. In order to support this view, it will be argued that it is precisely the symbolic-linguistic character of hate crime that is responsible for its particular injurious force as well as for its dehumanizing effects. Moreover, hate crimes are not only linguistically in character, they are also directed against the possibility of language and speech itself, insofar as they aim at making us speechless, depriving us of the possibility of speaking out and being heard. This leads to an account of hate crime as a form of multi-addressed violence that not only calls for a strong concept of responsibility, but also requires a differentiated response by all social institutions and authorities
Isolierte Neutronensterne und ihre sub-stellaren Begleiter
Neben den Radiopulsaren, die die Mehrheit der gegenwärtig beobachteten Neutronensterne ausmachen, gibt es radioleise junge Neutronensterne mit rein thermischer Röntgenstrahlung, die einen direkten Blick auf ihre Oberfläche erlauben. In dieser Arbeit wird untersucht, wie gut die unter Anwendung verschiedener Annahmen simulierte Anzahl von solchen Neutronensternen mit den Beobachtungen übereinstimmt und wieviel mehr Neutronensterne man unter den detektierten, schwachen Röntgenquellen aufgrund dieser Simulationen erwarten kann. Wichtige Fragen sind der Einfluss der Verteilung der Vorgängersterne und der Absorption durch das Interstellare Medium auf die vorhergesagten Neutronensternzahlen. Des weiteren wird nach neuen Kondidaten solcher Neutronensterne gesucht, und zwar in Röntgendaten von ROSAT (RASS, HRI, PSPC pointings) und XMM-Newton, sowie mit Hilfe des SLOAN Quellenkatalogs im sichtbaren Licht. Ein weiteres Projekt ist die Suche nach substellaren Begleitern, also Planeten oder Braunen Zwergen, um junge und nahe Neutronensterne. Zwei Neutronensterne haben eine ausreichend hohe Eigenbewegung, so dass bereits jeweils eine Wiederholungsbeobachtung analysiert werden konnte. Für alle untersuchten 10 Neutronensterne sind obere Grenzen der Helligkeiten im Nahen Infrarot bestimmt worden.Most known neutron stars are radio pulsars, but there are also radio-quiet neutron stars with pure thermal X-ray emission. These radio-quiet neutron stars are of special interest because their thermal X-ray spectra are caused by photospheric emission - allowing a direct view onto the stars' surfaces. This work investigates first the performance of a population synthesis model of neutron stars with thermal X-ray emission. This model is based on several assumptions. Important questions in this work concern the influence of the distribution of the progenitor stars on the observable neutron star number and the role of an inhomogeneous distribution of the X-ray-absorbing interstellar medium. More neutron stars with strong thermal X-ray emission are exprected. This work reports also about a search for new candidates of such neutron stars using data from ROSAT (RASS, PSPC, HRI), XMM-Newton and the sloan Digital Sky Survey. Furthermore we search for substellar companions around young, nearby neutron stars. Two objects are moving fast enough to obtain and analyze the second imaging epoch. For all investigated 10 neutron stars the limits at Near Infrared wavelegths are given

AbstractZiel der vorliegenden Arbeit war die Synthese und biologische Testung
potentiell antitumorwirksamer Eisen(diarylsalen)-Komplexe, deren zytotoxische
Wirkung auf einer Bindung an die Phosphatbrücken der DNA und nachfolgender
DNA-Spaltung durch Reaktive Sauerstoffspezies beruhen könnte. Um den
Einfluss der Ligandenstruktur auf die zytotoxische Wirksamkeit der
synthetisierten Eisen(II)- und Eisen(III)-Komplexe mit der allgemeinen
Grundstruktur
[1,6-Bis(2-hydroxyphenyl)-3,4-diaryl-2,5-diazahexa-1,5-dien]eisen(II) bzw.
[1,6-Bis(2-hydroxyphenyl)-3,4-diaryl-2,5-diazahexa-1,5-dien]eisen(III)-chlorid
näher untersuchen zu können, wurden Liganden mit unterschiedlichen
Substitutionsmustern und Variationen am Azomethingrundgerüst dargestellt. Mit
Hilfe von in vitro Zytotoxizitäts-Assays wurde anschließend die
Antitumorwirkung der dargestellten Verbindungen an sechs verschiedenen
Tumorzelllinien (MCF7, MDA-MB-231, HeLa, LNCaP/FGC, LAMA84 und K562)
überprüft, wobei eindeutige Struktur-Wirkungs-Beziehungen abgeleitet werden
konnten. Dabei zeigte sich u.a., dass die Oxidationsstufe des
Eisenzentralatoms der Komplexe einen bedeutenden Einfluss auf die zytotoxische
Wirkung der Verbindungen hatte. Eisen(III)(diarylsalen)-Komplexe besaßen an
allen Zelllinien wesentlich stärkere zytotoxische Wirkungen als ihre
Eisen(II)-Analoga. Weiterhin konnte eine Abhängigkeit der antiproliferativen
Wirkung der getesteten Verbindungen von der Konfiguration der 1,2-Diarylethan-
Partialstruktur und von der Art und der Position der Substituenten der
Brücken - bzw. Basis -Aromaten gefunden werden. Besonders auffällig war die
herausragende zytotoxische Wirksamkeit der Eisen(diarylsalene) an der MDA-
MB-231-Zelllinie. Die synthetisierten Eisen(diarylsalen)-Komplexe zeigten
bereits ab einer Konzentration von 0.1 bzw. 0.5 Mikromol stark zytozide
Effekte. Derart hohe zelltoxische Wirkungen von Metallkomplexen an dieser
Brustkrebszelllinie sind bisher noch nicht beschrieben worden. Durch
Zellaufnahmestudien, die unter Verwendung atomabsorptionsspektroskopischer
Methoden durchgeführt wurden, konnte ein Zusammenhang zwischen der
Bioverfügbarkeit der Eisen(diarylsalen)-Komplexe und der antiproliferativen
Wirkung der Verbindungen herausgearbeitet werden. Diese Untersuchungen ließen
dabei vermuten, dass die unterschiedlichen antiproliferativen Effekte der
Eisen(diarylsalen)-Komplexe nicht allein durch eine selektive Aufnahme bedingt
sind, sondern vielmehr spezifischere, strukturabhängige Mechanismen
nachgeschaltet sein könnten. Mittels der DAPI-Färbung und dem Nachweis von
ssDNA konnten die apoptoseauslösenden Eigenschaften der Eisenkomplexe belegt
werden, wobei wiederum eine deutliche Abhängigkeit der apoptoseinduzierenden
Wirkung von der Struktur der Komplexe gefunden wurde. Die zytotoxische
Aktivität und die apoptoseauslösenden Eigenschaften der Komplexe waren
Ausgangspunkt für die Untersuchungen der Wechselwirkungen der Verbindungen mit
der DNA. Durch die Aufnahme von DNA-Schmelzpunktkurven konnte gezeigt werden,
dass die Eisenkomplexe an die DNA binden und diese dadurch gegen
denaturierende Einflüsse stabilisieren. Mittels gelelektrophoretischer
Untersuchungen konnte die DNA-Spaltungsaktivität der Verbindungen nachgewiesen
werden. Für Eisen(diarylsalen)-Komplexe mit chinoider Teilstruktur wurde dabei
eine selbstaktivierende Nukleasenwirkung gefunden. Damit stellen die im
Rahmen dieser Arbeit synthetisierten und pharmakologisch, biochemisch näher
charakterisierten Eisen(diarylsalen)-Komplexe einen viel versprechenden Ansatz
auf dem Weg zu effizienteren Zytostatika dar.The target of this investigation was the synthesis and biological testing of
iron(diarylsalen) complexes with potential anti-tumor activity. It is believed
that the cytostatic effect of these compounds is caused by binding to the
phosphate backbone and subsequent cleavage of DNA through the generation of
reactive oxygen species. The synthesized iron(II) and iron(III) complexes had
the basic structure of
[1,6-bis(2-hydroxyphenyl)-3,4-diaryl-2,5-diazahexa-1,5-diene]iron(II) and
[1,6-bis(2-hydroxyphenyl)-3,4-diaryl-2,5-diazahexa-1,5-diene]iron(III)-chlorid
respectively. Ligands with varying substitution patterns and variations of the
azomethine backbones were prepared in order to investigate the influence of
the ligand structure onto their cytotoxic activity. The anti-tumor potential
of these compounds was tested in cytotoxicity assays with different cell lines
(MCF7, MDA-MB-231, HeLa, LNCaP/FGC, LAMA84 und K562) indicating a close
structure-activity relationship. The anti-proliferation activity of all tested
compounds was depended on the configuration of the 1,2-diarylethane-
substructure as well as on the nature and position of the substituents of the
basis - or bridge -aromatics. Especially, the effect of iron(diarylsalen)
complexes on MDA-MB-231-cells was extraordinary strong. The synthesized iron
complexes had highest cytotoxic activity so far documented in literature for
breast cancer cell lines, starting at concentrations as low as 0.1 and 0.5
micro mol, respectively. In addition, it was also demonstrated that the
oxidation state of the central iron atom significantly influences the
cytotoxicity. Iron(III)(diarylsalen) complexes showed an enhanced activity
compared to their iron(II)-analogs in all tested cell lines. Cell uptake
assays, analyzed by atomic absorption spectroscopy, showed a correlation
between bioavailability and anti-proliferating activity. They also suggest
that the differences in anti-proliferating activities were caused not only by
high uptake rates of these compounds, but also through other structure
dependent and highly selective mechanisms. The capability of these compounds
to induce apoptosis in micro molar concentrations was demonstrated by
observation of nuclear morphology changes with DAPI staining and by detection
of ssDNA in treated cells. The results also showed a dependency of the
efficiency of apoptosis induction with the chemical structure. Further, DNA
melting point studies indicated a direct binding of the compounds to DNA and
gel electrophoresis of circular plasmid DNA demonstrated in vitro DNA cleavage
activity, with a self induction effect of iron salen complexes with
hydroquinone substructure. These findings point at DNA damage as the major
cause of apoptosis. Overall, in this work the synthesized, pharmacological and
biochemical characterized iron(diarylsalen) complexes present a promising
approach towards cytostatic agents with highly enhanced anti-tumor activity
