    S-palmitoylation (S-acylation) is an emerging dynamic post-translational modification of cysteine residues within proteins.Current assays for protein S-palmitoylation involve either in vivo labelling or chemical cleavage of S-palmitoyl groupsto reveal a free cysteine sulfhydryl that can be subsequently labelled with an affinity handle (acyl-exchange). Assays for protein S-palmitoylation using acylexchange chemistry therefore require blocking of non-S-palmitoylated cysteines, typically using Nethylmaleimide, to prevent non-specific detection. This in turn necessitates multiple precipitation based clean-up steps to remove reagents between stages, often leading to variable sample loss, reduced signal or protein aggregation. These combine to reduce the sensitivity, reliability and accuracy of these assays and also requires a substantial amount of time to perform. By substituting these precipitation steps with chemical scavenging of N-ethylmaleimide by 2,3-dimethyl-1,3-butadiene in an aqueous Diels-Alder 4+2 cyclo-addition reaction it is possible to greatly improve sensitivity and accuracy while reducing hands-on and overall time required for assays

    National audienceL'application de l'Ingénierie Dirigée par les Modèles (IDM) est souvent réservée aux processus de génie logiciel (par exemple, spécification, génération de code, maintenance, rétro-ingénierie, évolution). Cependant, elle peut aussi être bénéfique pour des initiatives davantage orientées métier ou liées à la prise de décisions stratégiques, telles que l'Architecture d'Entreprise (AE). L'AE est le processus de traduction de la vision métier/stratégie d'une entreprise en un changement effectif, via la description de son état présent et futur (par exemple, concernant son système d'information). Même si différentes approches ont déjà proposé plusieurs sortes de méthodes et d'outillages pour l'AE dans une entreprise, il n'existe pas véritablement à l'heure actuelle de framework IDM intégré combinant fédération de données d'AE, adaptabilité du standard de représentation et support pour des points de vue multiples. Le présent article rapporte notre expérience en cours de construction du framework IDM TEAP (basé sur le standard TOGAF et l'outillage SmartEA) visant notamment à traiter ces trois défis dans un contexte industriel d'AE

    In-vitro and in-vivo evidence for uncoupling of BCR internalization and signaling in chronic lymphocytic leukemia

    B-cell receptor activation, occurring within lymph nodes, plays a key role in the pathogenesis of chronic lymphocytic leukemia and is linked to prognosis. As well as activation of downstream signaling, receptor ligation triggers internalization, transit to acidified endosomes and degradation of ligand-receptor complexes. In the present study we investigated the relationship between these two processes in normal and leukemic B-cells. We found that leukemic B-cells, particularly anergic cases lacking the capacity to initiate downstream signaling, internalize and accumulate ligand in acidified endosomes more efficiently than normal B-cells. Furthermore, ligation of either surface CD79B, a Bcell receptor component required for downstream signaling, or surface IgM by cognate agonistic antibody, showed that the two molecules internalize independently of each other in leukemic but not normal B-cells. Since association with surface CD79B is required for surface retention of IgM, this suggests that uncoupling of B-cell receptor internalization from signaling may be due to dissociation of these two molecules in leukemic cells. Comparison of lymph node with peripheral blood cells from chronic lymphocytic leukemia patients showed that, despite recent B-cell receptor activation, lymph node B-cells expressed higher levels of surface IgM. This surprising finding suggests that the B-cell receptors of lymph node and peripheral blood derived leukemic cells might be functionally distinct. Finally, long-term therapy with the Bruton’s tyrosine kinase inhibitors ibrutinib or acalabrutinib resulted in a switch to an anergic pattern of B-cell receptor function with reduced signaling capacity, surface IgM expression and more efficient internalization

    International audienceBuilding on previous experiments of reverse engineering with the Squeak language, we have designed a new prototype on the DotNet platform that should be able to exploit the full capabilities of the Visual Basic 9.0 language, including the facilities offered for dynamic typing. The objective of this work is to prove that we are able to implement an automatic model mining facility when the system consists of a legacy with good reflectivity capabilities. The initial work presented in this paper comforts the previous results obtained for Squeak and constitute the basis of a reverse engineering development toolkit that is intended to be extended in the coming months

    Genetic algorithm with logistic regression for prediction of progression to Alzheimer\u27s disease

    Assessment of risk and early diagnosis of Alzheimer\u27s disease (AD) is a key to its prevention or slowing the progression of the disease. Previous research on risk factors for AD typically utilizes statistical comparison tests or stepwise selection with regression models. Outcomes of these methods tend to emphasize single risk factors rather than a combination of risk factors. However, a combination of factors, rather than any one alone, is likely to affect disease development. Genetic algorithms (GA) can be useful and efficient for searching a combination of variables for the best achievement (eg. accuracy of diagnosis), especially when the search space is large, complex or poorly understood, as in the case in prediction of AD development. This study showed the potential of GA application in the neural science area. It demonstrated that the combination of a small set of variables is superior in performance than the use of all the single significant variables in the model for prediction of progression of disease. Variables more frequently selected by GA might be more important as part of the algorithm for prediction of disease development
