Fretting of CoCrMo and Ti6Al4V Alloys in Modular Prostheses
Implantation of a total hip replacements (THR) is an effective intervention in the management of arthritis. Modularity at the taper junction of THR was introduced in order to improve the ease with which the surgeon could modify the length of the taper section and the overall length of the replacement. Cobalt chromium (Co–28Cr–6Mo) and titanium (Ti–6Al–4V) alloys are the most commonly used materials for the device. This study investigates the fretting behaviour of both CoCr–CoCr and CoCr–Ti couplings and analyses their damage mechanisms. A reciprocating tribometer ball on plate fretting contact was instrumented with in situ electrochemistry to characterise the damage inflicted by tribocorrosion on the two couplings. Fretting displacements amplitudes of 10, 25 and 50 mm at an initial contact pressure of 1 GPa were assessed. The results reveal larger metallic volume loss from the CoCr–CoCr alloy compared to the CoCr–Ti alloy, and the open circuit potential indicates a depassivation of the protective oxide layer at displacement amplitudes .25 mm. In conclusion, the damage mechanisms of CoCr–CoCr and CoCr–Ti fretting contacts were identified to be wear and fatigue dominated mechanisms respectively
Public Attitudes Towards Moral Enhancement. Evidence that Means Matter Morally
To gain insight into the reasons that the public may have for endorsing or eschewing pharmacological moral enhancement for themselves or for others, we used empirical tools to explore public attitudes towards these issues. Participants (N = 293) from the United States were recruited via Amazon’s Mechanical Turk and were randomly assigned to read one of several contrastive vignettes in which a 13-year-old child is described as bullying another student in school and then is offered an empathy-enhancing program. The empathy-enhancing program is described as either involving taking a pill or playing a video game on a daily basis for four weeks. In addition, participants were asked to imagine either their own child bullying another student at school, or their own child being bullied by another student. This resulted in a 2 × 2 between-subjects design. In an escalating series of morally challenging questions, we asked participants to rate their overall support for the program; whether they would support requiring participation; whether they would support requiring participation of children who are at higher risk to become bullies in the future; whether they would support requiring participation of all children or even the entire population; and whether they would be willing to participate in the program themselves. We found that people were significantly more troubled by pharmacological as opposed to non-pharmacological moral enhancement interventions. The results indicate that members of the public for the greater part oppose pharmacological moral bioenhancement, yet are open to non-biomedical means to attain moral enhancement. [248 words]
Mortality from lung cancer in workers exposed to sulfur dioxide in the pulp and paper industry.
Our objective in this study was to evaluate the mortality of workers exposed to sulfur dioxide in the pulp and paper industry. The cohort included 57,613 workers employed for at least 1 year in the pulp and paper industry in 12 countries. We assessed exposure to SO(2) at the level of mill and department, using industrial hygiene measurement data and information from company questionnaires; 40,704 workers were classified as exposed to SO(2). We conducted a standardized mortality ratio (SMR) analysis based on age-specific and calendar period-specific national mortality rates. We also conducted a Poisson regression analysis to determine the dose-response relations between SO(2) exposure and cancer mortality risks and to explore the effect of potential confounding factors. The SMR analysis showed a moderate deficit of all causes of death [SMR = 0.89; 95% confidence interval (CI), 0.87-0.96] among exposed workers. Lung cancer mortality was marginally increased among exposed workers (SMR = 1.08; 95% CI, 0.98-1.18). After adjustment for occupational coexposures, the lung cancer risk was increased compared with unexposed workers (rate ratio = 1.49; 95% CI, 1.14-1.96). There was a suggestion of a positive relationship between weighted cumulative SO(2) exposure and lung cancer mortality (p-value of test for linear trend = 0.009 among all exposed workers; p = 0.3 among workers with high exposure). Neither duration of exposure nor time since first exposure was associated with lung cancer mortality. Mortality from non-Hodgkin lymphoma and from leukemia was increased among workers with high SO(2) exposure; a dose-response relationship with cumulative SO(2) exposure was suggested for non-Hodgkin lymphoma. For the other causes of death, there was no evidence of increased mortality associated with exposure to SO(2). Although residual confounding may have occurred, our results suggest that occupational exposure to SO(2) in the pulp and paper industry may be associated with an increased risk of lung cancer
The use of a P. falciparum specific coiled-coil domain to construct a self-assembling protein nanoparticle vaccine to prevent malaria.
The parasitic disease malaria remains a major global public health concern and no truly effective vaccine exists. One approach to the development of a malaria vaccine is to target the asexual blood stage that results in clinical symptoms. Most attempts have failed. New antigens such as P27A and P27 have emerged as potential new vaccine candidates. Multiple studies have demonstrated that antigens are more immunogenic and are better correlated with protection when presented on particulate delivery systems. One such particulate delivery system is the self-assembling protein nanoparticle (SAPN) that relies on coiled-coil domains of proteins to form stable nanoparticles. In the past we have used de novo designed amino acid domains to drive the formation of the coiled-coil scaffolds which present the antigenic epitopes on the particle surface.
Here we use naturally occurring domains found in the tex1 protein to form the coiled-coil scaffolding of the nanoparticle. Thus, by engineering P27A and a new extended form of the coiled-coil domain P27 onto the N and C terminus of the SAPN protein monomer we have developed a particulate delivery system that effectively displays both antigens on a single particle that uses malaria tex1 sequences to form the nanoparticle scaffold. These particles are immunogenic in a murine model and induce immune responses similar to the ones observed in seropositive individuals in malaria endemic regions.
We demonstrate that our P27/P27A-SAPNs induce an immune response akin to the one in seropositive individuals in Burkina Faso. Since P27 is highly conserved among different Plasmodium species, these novel SAPNs may even provide cross-protection between Plasmodium falciparum and Plasmodium vivax the two major human malaria pathogens. As the SAPNs are also easy to manufacture and store they can be delivered to the population in need without complication thus providing a low cost malaria vaccine
Sleep and recovery in physicians on night call: a longitudinal field study
<p>Abstract</p> <p>Background</p> <p>It is well known that physicians' night-call duty may cause impaired performance and adverse effects on subjective health, but there is limited knowledge about effects on sleep duration and recovery time. In recent years occupational stress and impaired well-being among anaesthesiologists have been frequently reported for in the scientific literature. Given their main focus on handling patients with life-threatening conditions, when on call, one might expect sleep and recovery to be negatively affected by work, especially in this specialist group. The aim of the present study was to examine whether a 16-hour night-call schedule allowed for sufficient recovery in anaesthesiologists compared with other physician specialists handling less life-threatening conditions, when on call.</p> <p>Methods</p> <p>Sleep, monitored by actigraphy and Karolinska Sleep Diary/Sleepiness Scale on one night after daytime work, one night call, the following first and second nights post-call, and a Saturday night, was compared between 15 anaesthesiologists and 17 paediatricians and ear, nose, and throat surgeons.</p> <p>Results</p> <p>Recovery patterns over the days after night call did not differ between groups, but between days. Mean night sleep for all physicians was 3 hours when on call, 7 h both nights post-call and Saturday, and 6 h after daytime work (p < 0.001). Scores for mental fatigue and feeling well rested were poorer post-call, but returned to Sunday morning levels after two nights' sleep.</p> <p>Conclusions</p> <p>Despite considerable sleep loss during work on night call, and unexpectedly short sleep after ordinary day work, the physicians' self-reports indicate full recovery after two nights' sleep. We conclude that these 16-hour night duties were compatible with a short-term recovery in both physician groups, but the limited sleep duration in general still implies a long-term health concern. These results may contribute to the establishment of safe working hours for night-call duty in physicians and other health-care workers.</p
Reproducibility of exhaled nitric oxide in smokers and non-smokers: relevance for longitudinal studies
<p>Abstract</p> <p>Background</p> <p>Currently, there is much interest in measuring fractional exhaled nitric oxide (<b>FE<sub>NO</sub></b>) in populations. We evaluated the reproducibility of <b>FE<sub>NO </sub></b>in healthy subjects and determined the number of subjects necessary to carry out a longitudinal survey of <b>FE<sub>NO </sub></b>in a population containing smokers and non-smokers, based on the assessed reproducibility.</p> <p>Methods</p> <p>The reproducibility of <b>FE<sub>NO </sub></b>was examined in 18 healthy smokers and 21 non-smokers. <b>FE<sub>NO </sub></b>was assessed once at 9 AM on five consecutive days; in the last day this measurement was repeated at 2 PM. Respiratory symptoms and medical history were assessed by questionnaire. The within- and between-session repeatability of <b>FE<sub>NO </sub></b>and log-transformed <b>FE<sub>NO </sub></b>was described. The power of a longitudinal study based on a relative increase in <b>FE<sub>NO </sub></b>was estimated using a bilateral t-test of the log-transformed <b>FE<sub>NO </sub></b>using the between-session variance of the assay.</p> <p>Results</p> <p><b>FE<sub>NO </sub></b>measurements were highly reproducible throughout the study. <b>FE<sub>NO </sub></b>was significantly higher in males than females regardless of smoking status. <b>FE<sub>NO </sub></b>was positively associated with height (p < 0.001), gender (p < 0.034), smoking (p < 0.0001) and percent FEV<sub>1</sub>/FVC (p < 0.001) but not with age (p = 0.987). The between-session standard deviation was roughly constant on the log scale. Assuming the between-session standard deviation is equal to its longitudinal equivalent, either 111 or 29 subjects would be necessary to achieve an 80% power in detecting a 3% or a 10% increase in <b>FE<sub>NO </sub></b>respectively.</p> <p>Conclusion</p> <p>The good reproducibility of <b>FE<sub>NO </sub></b>is not influenced by gender or smoking habits. In a well controlled, longitudinal study it should allow detecting even small increases in <b>FE<sub>NO </sub></b>with a reasonable population size.</p
Aβ Peptide Fibrillar Architectures Controlled by Conformational Constraints of the Monomer
Anomalous self-assembly of the Aβ peptide into fibrillar amyloid deposits is strongly correlated with the development of Alzheimer's disease. Aβ fibril extension follows a template guided “dock and lock” mechanism where polymerisation is catalysed by the fibrillar ends. Using surface plasmon resonance (SPR) and quenched hydrogen-deuterium exchange NMR (H/D-exchange NMR), we have analysed the fibrillar structure and polymerisation properties of both the highly aggregation prone Aβ1–40 Glu22Gly (Aβ40Arc) and wild type Aβ1–40 (Aβ40WT). The solvent protection patterns from H/D exchange experiments suggest very similar structures of the fibrillar forms. However, through cross-seeding experiments monitored by SPR, we found that the monomeric form of Aβ40WT is significantly impaired to acquire the fibrillar architecture of Aβ40Arc. A detailed characterisation demonstrated that Aβ40WT has a restricted ability to dock and isomerise with high binding affinity onto Aβ40Arc fibrils. These results have general implications for the process of fibril assembly, where the rate of polymerisation, and consequently the architecture of the formed fibrils, is restricted by conformational constraints of the monomers. Interestingly, we also found that the kinetic rate of fibril formation rather than the thermodynamically lowest energy state determines the overall fibrillar structure
Existence of a Tribo-Modified Surface Layer on SBR Elastomers: Balance Between Formation and Wear of the Modified Layer
In most of the tribological contacts, the composition and tribological properties of the original interface will change during use. The tribo-films, with modified properties compared to the bulk, are dynamic structures that play a significant role in friction. The existence of a tribo-modified surface layer and its importance on the overall friction of elastomers has been shown both theoretically and experimentally before. The characteristics of the modified surface layer deserve specific attention since the tribological properties of elastomers in contact with a rough counter-surface are determined by these modified surfaces together with the properties of bulk of the material. Both the formation of the modified layer and the break down (wear) of it are of importance in determining the existence and thickness of the tribo-modified layer. In this study, the importance of the wear is emphasized by comparing two styrene butadiene rubber-based elastomers in contact with a granite sphere. A current status of perception of the removal and the stability of the modified surface layers on rubbers is introduced as well as experimental work related to this matter and discussion within literature. Pin-on-disk friction tests are performed on two SBR-based samples in contact with a granite sphere under controlled environmental conditions to form the modified surface layer. Although the hysteresis part of the friction force which has a minor contribution in the overall friction is not markedly different, the total measured friction coefficient differs significantly. Mechanical changes both inside and outside the wear track are determined by atomic force microscope nano-indentations at different timescales to examine the modified surface layer on the test samples. The specific wear rates of the two tribo-systems are compared, and the existence of the modified surface layer, the different measured friction coefficient and the running-in distances toward steady-state friction are explained considering different wear rates. A conceptual model is presented, correlating the energy input into the tribo-system and the existence of a modified surface layer
Varicella susceptibility and transmission dynamics in Slovenia
<p>Abstract</p> <p>Background</p> <p>A cross-sectional, age-stratified study was conducted to determine varicella-zoster seroprevalence and force of infection in Slovenia.</p> <p>Methods</p> <p>3689 serum samples were tested for VZV IgG antibodies with an enzyme immunoassay. Semiparametric and parametric modelling were used to estimate the force of infection.</p> <p>Results</p> <p>Overall, 85.6% of serum samples were seropositive. Age-specific prevalence rose rapidly in preschool children and over 90% of 8 years old tested positive for VZV. However, 2.8% of serum samples among women of childbearing age were seronegative. Semiparametric modelling yielded force of infection estimates of 0.182 (95% CI 0.158-0.206), 0.367 (95% CI 0.285-0.448) and 0.008 (95% CI 0.0-0.032) for age groups 0.5- < 6, 6-11 and ≥12 years, respectively, and 0.175 (95% CI 0.147-0.202), 0.391 (95% CI 0.303-0.480) and 0.025 (95% CI 0.003-0.046) for age groups 0.5- < 5, 5-9 and ≥10 years, respectively.</p> <p>Conclusions</p> <p>Regardless of the age grouping used, the highest transmission occurred in children in their first years of school.</p
Root trenching: a useful tool to estimate autotrophic soil respiration? A case study in an Austrian mountain forest
We conducted a trenching experiment in a mountain forest in order to assess the contribution of theautotrophic respiration to total soil respiration and evaluate trenching as a technique to achieve it. We hypothesised that the trenching experiment would alter both microbial biomass and microbial community structure and that Wne roots (less than 2 mm diameter) would be decomposed within one growing season. Soil CO2 eZux was measured roughlybiweekly over two growing seasons. Root presence and morphology parameters, as well as the soil microbial community were measured prior to trenching, 5 and 15 months after trenching. The trenched plots emitted about 20 and 30% less CO2 than the control plots in the Wrst and secondgrowing season, respectively. Roots died in trenched plots, but root decay was slow. After 5 and 15 months, Wne root biomass was decreased by 9% (not statistically diferent)and 30%, (statistically diVerent) respectively. When wecorrected for the additional trenched-plot CO2 eZux due to Wne root decomposition, the autotrophic soil respiration rose to »26% of the total soil respiration for the Wrst growing season, and to »44% for the second growing season.Soil microbial biomass and community structure was not altered by the end of the second growing season.
