    Removal of Cr(VI) from aqueous solutions by a bacterial biofilm supported on zeolite : optimisation of the operational conditions and scale-up of the bioreactor

    The aim of this study was to investigate the feasibility of a bioreactor system and its scale-up to remove Cr(VI) from solution. The bioreactor is based on an innovative process that combines bioreduction of Cr(VI) to Cr(III) by the bacterium Arthrobacter viscosus and Cr(III) sorption by a specific zeolite. Batch studies were conducted in a laboratory-scale bioreactor, taking into account different operating conditions. Several variables, such as biomass concentration, pH and zeolite pre-treatment, were evaluated to increase removal efficiency. The obtained results suggest that the Cr removal efficiency is improved when the initial biomass concentration is approximately 5 g L–1 and the pH in the system is maintained at an acidic level. Under the optimised conditions, approximately 100 % of the Cr(VI) was removed.     Inactivation of the monofunctional peptidoglycan glycosyltransferase SgtB allows Staphylococcus aureus to survive in the absence of lipoteichoic acid

    The cell wall of Staphylococcus aureus is composed of peptidoglycan and the anionic polymers lipoteichoic acid (LTA) and wall teichoic acid. LTA is required for growth and normal cell morphology in S. aureus. Strains lacking LTA are usually viable only when grown under osmotically stabilizing conditions or after the acquisition of compensatory mutations. LTA-negative suppressor strains with inactivating mutations in gdpP, which resulted in increased intracellular c-di-AMP levels, were described previously. Here, we sought to identify factors other than c-di-AMP that allow S. aureus to survive without LTA. LTA-negative strains able to grow in unsupplemented medium were obtained and found to contain mutations in sgtB, mazE, clpX, or vraT. The growth improvement through mutations in mazE and sgtB was confirmed by complementation analysis. We also showed that an S. aureus sgtB transposon mutant, with the monofunctional peptidoglycan glycosyltransferase SgtB inactivated, displayed a 4-fold increase in the MIC of oxacillin, suggesting that alterations in the peptidoglycan structure could help bacteria compensate for the lack of LTA. Muropeptide analysis of peptidoglycans isolated from a wild-type strain and sgtB mutant strain did not reveal any sizable alterations in the peptidoglycan structure. In contrast, the peptidoglycan isolated from an LTA-negative ltaS mutant strain showed a significant reduction in the fraction of highly cross-linked peptidoglycan, which was partially rescued in the sgtB ltaS double mutant suppressor strain. Taken together, these data point toward an important function of LTA in cell wall integrity through its necessity for proper peptidoglycan assembly

    Locational preferences and spatial arrangement in the barrow landscape of Serra do Barbanza (North-western Iberia)

    As anywhere else around the world, GIS is an essential tool in Galician Archaeology (NW Spain) when examining and analysing spatial data. This is also true for the study of mounds in that area, since spatial analysis and statistics have become increasingly used for contrasting hypotheses regarding the locational preferences of these monuments, usually inferred from observations made during fieldwork or taken from studies conducted elsewhere. Drawing on this basis, in this paper, we have analysed the locational patterns of the tumuli of the upper tracts of the Serra do Barbanza (Galicia, NW Spain). Using a site-predictive modelling approach, several environmental covariates were analysed in order to explore their potential role in explaining the distribution of prehistoric mounds in the area. Subsequently, we studied the clustering of these monuments via second-order modelling. Our results suggest that tumuli in the Serra do Barbanza tended to cluster on a very local scale, a trend that can only be explained by intended site spacing strategies that may have taken place over millennia. Finally, by using significance testing via Monte Carlo Simulation, we have modelled both the effect of possible preferences regarding the location of mounds and the potential impact of tradition, with pre-existent megaliths possibly fostering the construction of more monuments in the nearby areas

    A time-domain fourth-order-convergent numerical algorithm to integrate black hole perturbations in the extreme-mass-ratio limit

    We obtain a fourth order accurate numerical algorithm to integrate the Zerilli and Regge-Wheeler wave equations, describing perturbations of nonrotating black holes, with source terms due to an orbiting particle. Those source terms contain the Dirac's delta and its first derivative. We also re-derive the source of the Zerilli and Regge-Wheeler equations for more convenient definitions of the waveforms, that allow direct metric reconstruction (in the Regge-Wheeler gauge).Comment: 30 pages, 12 figure

    Improved outer boundary conditions for Einstein's field equations

    In a recent article, we constructed a hierarchy B_L of outer boundary conditions for Einstein's field equations with the property that, for a spherical outer boundary, it is perfectly absorbing for linearized gravitational radiation up to a given angular momentum number L. In this article, we generalize B_2 so that it can be applied to fairly general foliations of spacetime by space-like hypersurfaces and general outer boundary shapes and further, we improve B_2 in two steps: (i) we give a local boundary condition C_2 which is perfectly absorbing including first order contributions in 2M/R of curvature corrections for quadrupolar waves (where M is the mass of the spacetime and R is a typical radius of the outer boundary) and which significantly reduces spurious reflections due to backscatter, and (ii) we give a non-local boundary condition D_2 which is exact when first order corrections in 2M/R for both curvature and backscatter are considered, for quadrupolar radiation.Comment: accepted Class.     Development and operation of a pixel segmented liquid-filled linear array for radiotherapy quality assurance

    A liquid isooctane (C8_{8}H18_{18}) filled ionization linear array for radiotherapy quality assurance has been designed, built and tested. The detector consists of 128 pixels, each of them with an area of 1.7 mm ×\times 1.7 mm and a gap of 0.5 mm. The small pixel size makes the detector ideal for high gradient beam profiles like those present in Intensity Modulated Radiation Therapy (IMRT) and radiosurgery. As read-out electronics we use the X-Ray Data Acquisition System (XDAS) with the Xchip developed by the CCLRC. Studies concerning the collection efficiency dependence on the polarization voltage and on the dose rate have been made in order to optimize the device operation. In the first tests we have studied dose rate and energy dependences, and signal reproducibility. Dose rate dependence was found lower than 2.5 % up to 5 Gy min−1^{-1}, and energy dependence lower than 2.1 % up to 20 cm depth in solid water. Output factors and penumbras for several rectangular fields have been measured with the linear array and were compared with the results obtained with a 0.125 cm3^{3} air ionization chamber and radiographic film, respectively. Finally, we have acquired profiles for an IMRT field and for a virtual wedge. These profiles have also been compared with radiographic film measurements. All the comparisons show a good correspondence. Signal reproducibility was within a 2% during the test period (around three months). The device has proved its capability to verify on-line therapy beams with good spatial resolution and signal to noise ratio.Comment: 16 pages, 12 figures Submitted to Phys. Med. Bio

    Stability and reproducibility of 6013 deep inspiration breath-holds in left-sided breast cancer

    Purpose: Patients with left-sided breast cancer frequently receive deep inspiration breath-hold (DIBH) radiotherapy to reduce the risk of cardiac side effects. The aim of the present study was to analyze intra-breath-hold stability and inter-fraction breath-hold reproducibility in clinical practice. Material and methods: Overall, we analyzed 103 patients receiving left-sided breast cancer radiotherapy using a surface-guided DIBH technique. During each treatment session the vertical motion of the patient was continuously measured by a surface guided radiation therapy (SGRT) system and automated gating control (beam on/off) was performed using an audio-visual patient feedback system. Dose delivery was automatically triggered when the tracking point was within a predefined gating window. Intra-breath-hold stability and inter-fraction reproducibility across all fractions of the entire treatment course were analyzed per patient. Results: In the present series, 6013 breath-holds during beam-on time were analyzed. The mean amplitude of the gating window from the baseline breathing curve (maximum expiration during free breathing) was 15.8 mm (95%-confidence interval: [8.5–30.6] mm) and had a width of 3.5 mm (95%-CI: [2–4.3] mm). As a measure of intra-breath-hold stability, the median standard deviation of the breath-hold level during DIBH was 0.3 mm (95%-CI: [0.1–0.9] mm). Similarly, the median absolute intra-breath-hold linear amplitude deviation was 0.4 mm (95%-CI: [0.01–2.1] mm). Reproducibility testing showed good inter-fractional reliability, as the maximum difference in the breathing amplitudes in all patients and all fractions were 1.3 mm on average (95%-CI: [0.5–2.6] mm). Conclusion: The clinical integration of an optical surface scanner enables a stable and reliable DIBH treatment delivery during SGRT for left-sided breast cancer in clinical routine

    Removal of hexavalent chromium of contaminated soil by coupling electrokinetic remediation and permeable reactive biobarriers

    PURPOSE: In this study, a novel and ecological alternative have been developed to treat soils contaminated with hexavalent chromium coupling two well-known systems: electrokinetic remediation and permeable reactive biobarriers. The electric field promotes the electromigration of the hexavalent chromium oxyanions towards the anode. The biobarriers were placed before the anode electrode, in order to promote the reduction and retention of the chromium migrating in its direction. Thus, this technology provided a global treatment to soil removal without subsequent treatments of the contaminated effluents. METHODS: The electrokinetic system was coupled with two different permeable reactive biobarriers composed by Arthrobacter viscosus bacteria, supported either in activated carbon or zeolite. An electric field of 10 V was applied and two different treatment times of 9 and 18 days were tested. RESULTS: Removal values of 60% and 79% were obtained when electrokinetic treatment was coupled with zeolite and activated carbon biobarriers, respectively, for a test period of 18 day. The reduction of hexavalent chromium to trivalent chromium was around 45% for both systems. CONCLUSIONS: In this work, two types of biobarriers were efficiently coupled to electrokinetic treatment to decontaminate soil with Cr(VI). Furthermore, the viability of the new coupling technology developed (electrokinetic + biobarriers) to treat low-permeability polluted soils was demonstrated.This work was supported by the Spanish Ministry of Science and Innovation (CTQ2008-03059/PPQ), Xunta de Galicia (08MDS034314PR).
