Polarization-dependence of anomalous scattering in brominated DNA and RNA molecules, and importance of crystal orientation in single- and multiple-wavelength anomalous diffraction phasing
In this paper the anisotropy of anomalous scattering at the Br K-absorption edge in brominated nucleotides is investigated, and it is shown that this effect can give rise to a marked directional dependence of the anomalous signal strength in X-ray diffraction data. This implies that choosing the correct orientation for crystals of such molecules can be a crucial determinant of success or failure when using single- and multiple-wavelength anomalous diffraction (SAD or MAD) methods to solve their structure. In particular, polarized absorption spectra on an oriented crystal of a brominated DNA molecule were measured, and were used to determine the orientation that yields a maximum anomalous signal in the diffraction data. Out of several SAD data sets, only those collected at or near that optimal orientation allowed interpretable electron density maps to be obtained. The findings of this study have implications for instrumental choices in experimental stations at synchrotron beamlines, as well as for the development of data collection strategy programs
Structural disorder versus chiral magnetism in CrNbS
The crystal structure of a disordered form of CrNbS has been
characterized using diffraction and inelastic scattering of synchrotron
radiation. In contrast to the previously reported symmetry (P622), the
crystal can be described by a regular twinning of an average P6 structure
with three disordered positions of the Cr ions. Short-range correlations of the
occupational disorder result in a quite intense and structured diffuse
scattering; a static nature of the disorder was unambiguously attributed by the
inelastic x-ray scattering. The diffuse scattering has been modeled using a
reverse Monte-Carlo algorithm assuming a disorder of the Cr sub-lattice only.
The observed correlated disorder of the Cr sub-lattice reduces the temperature
of the magnetic ordering from 130 K to 88 K and drastically modifies the field
dependence of the magnetization as it is evidenced by the SQUID magnetometery.
We conclude, that in contrast to the helicoidal spin structure assumed for
P622 form, the compound under study is ferromagnetically ordered with a
pronounced in-plane anisotropy
Protocol for a mixed-methods exploratory investigation of care following intensive care discharge: the REFLECT study

It is unclear how many of these deaths are preventable. Ward-based management following discharge from ICU is an area that patients and healthcare staff are concerned about. The primary aim of REFLECT (Recovery Following Intensive Care Treatment) is to develop an intervention plan to reduce in-hospital mortality rates in patients who have been discharged from ICU. METHODS AND ANALYSIS: REFLECT is a multicentre mixed-methods exploratory study examining ward care delivery to adult patients discharged from ICU. The study will be made up of four substudies. Medical notes of patients who were discharged from ICU and subsequently died will be examined using a retrospective case records review (RCRR) technique. Patients and their relatives will be interviewed about their post-ICU care, including relatives of patients who died in hospital following ICU discharge. Staff involved in the care of patients post-ICU discharge will be interviewed about the care of this patient group. The medical records of patients who survived their post-ICU stay will also be reviewed using the RCRR technique. The analyses of the substudies will be both descriptive and use a modified grounded theory approach to identify emerging themes. The evidence generated in these four substudies will form the basis of the intervention development, which will take place through stakeholder and clinical expert meetings. ETHICS AND DISSEMINATION: Ethical approval has been obtained through the Wales Research and Ethics Committee 4 (17/WA/0107). We aim to disseminate the findings through international conferences, international peer-reviewed journals and social media. From Physics to Fixtures to Food: Current and Potential LED Efficacy
Light-emitting diodes (LEDs) have enabled a historic increase in the conversion of electric energy to photons, but this is approaching a physical limit. The theoretical maximum efficiency occurs when all input energy is converted to energy in photosynthetic photons. Blue LEDs can be 93% efficient, phosphor-converted “whites” 76% efficient, and red LEDs 81% efficient. These improvements open new opportunities for horticultural lighting. Here we review (1) fundamental physics and efficiency of LEDs, (2) the current efficacy of LEDs, (3) the effect of spectral quality on crop yield, and (4) the potential efficacy of horticultural fixtures. Advances in the conversion of photons to yield can be achieved by optimization of spectral effects on plant morphology, which vary among species. Conversely, spectral effects on photosynthesis are remarkably similar across species, but the conventional definition of photosynthetic photons (400–700 nm) may need to be modified. The upper limit of LED fixture efficacy is determined by the LED package efficacy multiplied by four factors inherent to all fixtures: current droop, thermal droop, driver (power supply) inefficiencies, and optical losses. With current LED technology, the calculations indicate efficacy limits of 3.4 µmol J−1 for white + red fixtures, and 4.1 µmol J−1 for blue + red fixtures. GUCY2C maintains intestinal LGR5+ stem cells by opposing ER stress
Long-lived multipotent stem cells (ISCs) at the base of intestinal crypts adjust their phenotypes to accommodate normal maintenance and post-injury regeneration of the epithelium. Their long life, lineage plasticity, and proliferative potential underlie the necessity for tight homeostatic regulation of the ISC compartment. In that context, the guanylate cyclase C (GUCY2C) receptor and its paracrine ligands regulate intestinal epithelial homeostasis, including proliferation, lineage commitment, and DNA damage repair. However, a role for this axis in maintaining ISCs remains unknown. Transgenic mice enabling analysis of ISCs (Lgr5-GFP) in the context of GUCY2C elimination (Gucy2c-/-) were combined with immunodetection techniques and pharmacological treatments to define the role of the GUCY2C signaling axis in supporting ISCs. ISCs were reduced in Gucy2c-/- mice, associated with loss of active Lgr5+ cells but a reciprocal increase in reserve Bmi1+ cells. GUCY2C was expressed in crypt base Lgr5+ cells in which it mediates canonical cyclic (c) GMPdependent signaling. Endoplasmic reticulum (ER) stress, typically absent from ISCs, was elevated throughout the crypt base in Gucy2c-/- mice. The chemical chaperone tauroursodeoxycholic acid resolved this ER stress and restored the balance of ISCs, an effect mimicked by the GUCY2C effector 8Br-cGMP. Reduced ISCs in Gucy2c-/-mice was associated with greater epithelial injury and impaired regeneration following sub-lethal doses of irradiation. These observations suggest that GUCY2C provides homeostatic signals that modulate ER stress and cell vulnerability as part of the machinery contributing to the integrity of ISCs. © Kraft et al
Cooperative Jahn-Teller Distortion in PrO2
We report neutron diffraction data on single crystal PrO2 which reveal a
cooperative Jahn-Teller distortion at TD = 120 +/- 2 K. Below this temperature
an internal distortion of the oxygen sublattice causes the unit cell of the
crystallographic structure to become doubled along one crystal axis. We discuss
several possible models for this structure. The antiferromagnetic structure
below TN = 13.5 K is found to consist of two components, one of which shares
the same doubled unit cell as the distorted crystallographic structure. We also
present measurements of the magnetic susceptibility, the specific heat capacity
and the electrical conductivity of PrO2. The susceptibility data show an
anomaly at a temperature close to TD. From the specific heat capacity data we
deduce that the ground state is doubly degenerate, consistent with a distortion
of the cubic local symmetry. We discuss possible mechanisms for this. The
conductivity shows an activated behaviour with an activation energy Ea = 0.262

Developing an intervention to equip nurses for acute life threatening events (ALTEs) in hospital:a phenomenological approach to healthcare research
Objectives: To understand staff's experiences of acute life threatening events (ALTEs) in a pediatric hospital setting. These data will inform an intervention to equip nurses with clinical and emotional skills for dealing with ALTEs. Method: A mixed design was used in the broader research program; this paper focuses on phenomenon-focused interviews analyzed using interpretative phenomenological analysis (IPA). Results: Emerging themes included staff's relationships with patients and the impact of personhood on their ability to perform competently in an emergency. More experienced nurses described "automatic" competence generated through increased exposure to ALTEs and were able to recognize "fumbling and shaking" as a normal stress response. Designating a role was significant to staff experience of effectiveness. Key to nurses' learning experience was reflection and identifying experiences as "teachable moments." Findings were considered alongside existing theories of self-efficacy, reflective thought, and advocacy inquiry to create an experiential learning intervention involving a series of clinical and role-related scenarios. Conclusion: The phenomenological work facilitated an in-depth reading of experience. It accentuated the importance of exposure to ALTEs giving nurses experiential knowledge to prepare them for the impact of these events. Challenges included bracketing the personhood of child patients, shifting focus to clinical tasks during the pressured demands of managing an ALTE, normalizing the physiological stress response, and the need for a forum and structure for reflection and learning. An intervention will be designed to provide experiential learning and encourage nurses to realize and benefit from their embodied knowledge
Autosomal recessive primary microcephaly: an analysis of locus heterogeneity and phenotypic variation
BACKGROUND AND OBJECTIVES: Locus heterogeneity is well established in autosomal recessive primary
microcephaly (MCPH) and to date five loci have been mapped. However, the relative contributions of
these loci have not been assessed and genotype-phenotype correlations have not been investigated.
DESIGN: A study population of 56 consanguineous families resident in or originating from northern
Pakistan was ascertained and assessed by the authors. A panel of microsatellite markers spanning
each of the MCPH loci was designed, against which the families were genotyped.
RESULTS: The head circumference of the 131 affected subjects ranged from 4 to 14 SD below the mean,
but there was little intrafamilial variation among affecteds (± 1 SD). MCPH5 was the most prevalent,
with 24/56 families consistent with linkage; 2/56 families were compatible with linkage to MCPH1,
10/56 to MCPH2, 2/56 to MCPH3, none to MCPH4, and 18/56 did not segregate with any of the
loci.
CONCLUSIONS: MCPH5 is the most common locus in this population. On clinical grounds alone, the
phenotype of families linked to each MCPH locus could not be distinguished. We have also shown that
further MCPH loci await discovery with a number of families as yet unlinked
Can changes in soil properties in organic banana production suppress fusarium wilt?
Organic agriculture promotes disease suppression through healthy soils by increasing biological activity and diversity through the application of organic fertilizers and increasing organic inputs. Fusarium wilt of bananas (Fusarium oxysporum f. sp. cubense) (Foc), also known as Panama disease, has been a devastating disease throughout the world. So far, no fungicides or cultural measures have been found that control Foc sufficiently. The aim of this research was to assess whether organic-based farming systems were more resilient than inorganic farming systems to soil borne diseases, in particular Fusarium wilts. A survey was conducted comparing five organic and five conventional banana plantations at paired sites in north Queensland, Australia. Soil samples were collected and analysed for chemical, physical and biological soil health indicators. Disease development of F. oxysporum f. sp. lycopersici in tomatoes and Foc in bananas were studied in pot trials to pursue clues for identifying Fusarium suppressive soil traits. Organic soils from the survey showed higher microbial activity and lower disease symptom expression (both with tomatoes and bananas) than conventional soils. In the survey, nematode diversity and soil sulphate content were recurring indicators in all experiments showing close correlations to pathogen growth, disease expression and plant health. Organic soils were lower in plant-parasitic nematodes and sulphate sulphur levels and higher in nematode diversity, labile soil C and microbial indicators. Soil conduciveness or suppression of Foc appeared to be largely governed by competition for carbon. Measurement of soil microbial enzyme activity, nematode community structure and diversity and possibly sulphate sulphur seem to provide a relatively reliable indicator for general disease suppression.
