    The physiological expression of inducible nitric oxide synthase (iNOS) in the human colon

    Inducible nitric oxide synthase (iNOS) is expressed in the colonic epithelium in both inflammatory bowel disease and colorectal cancer. Nitric oxide (NO), the product of this enzyme, has been implicated in the pathogenesis of both conditions. However, there are conflicting data on whether iNOS is expressed in the normal, uninflamed human colon. To evaluate the expression of iNOS in histologically normal, non-inflamed human colonic mucosa. Reverse transcription polymerase chain reaction (RT-PCR), immunoblotting, and immunohistochemistry were used to investigate the expression of iNOS in 17 histologically normal specimens obtained at colectomy performed for colorectal neoplasia. In addition, 16 endoscopic mucosal biopsies, taken from normal individuals, were also evaluated. Eleven surgical specimens and 16 endoscopic biopsies from patients with refractory ulcerative colitis were used as inflammatory controls. All types of specimens expressed iNOS mRNA. Immunoblotting revealed a protein of approximately 130 kDa consistent with iNOS in mucosal extracts of 77% of normal individuals, and 85% of diseased controls. Immunolabelling localised this protein to the surface epithelium in most of the normal specimens and also to the crypt epithelium and inflammatory cells in the diseased controls. These findings provide evidence that iNOS is often expressed in the surface epithelium of non-inflamed human colon, suggesting that it is induced by local luminal factors, such as bacterial lipopolysaccharide (endotoxin). The resultant NO produced at this site might act as an oxidative barrier, reducing bacterial translocation and providing a means of defence against pathogenic microorganisms

    The influence of ocean acidification on nitrogen regeneration and nitrous oxide production in the North-West European shelf sea

    The assimilation and regeneration of dissolved inorganic nitrogen, and the concentration of N2O, was investigated at stations located in the NW European shelf sea during June/July 2011. These observational measurements within the photic zone demonstrated the simultaneous regeneration and assimilation of NH4+, NO2− and NO3−. NH4+ was assimilated at 1.82–49.12 nmol N L−1 h−1 and regenerated at 3.46–14.60 nmol N L−1 h−1; NO2− was assimilated at 0–2.08 nmol N L−1 h−1 and regenerated at 0.01–1.85 nmol N L−1 h−1; NO3− was assimilated at 0.67–18.75 nmol N L−1 h−1 and regenerated at 0.05–28.97 nmol N L−1 h−1. Observations implied that these processes were closely coupled at the regional scale and nitrogen recycling played an important role in sustaining phytoplankton growth during the summer. The [N2O], measured in water column profiles, was 10.13 ± 1.11 nmol L−1 and did not strongly diverge from atmospheric equilibrium indicating that sampled marine regions where neither a strong source nor sink of N2O to the atmosphere. Multivariate analysis of data describing water column biogeochemistry and its links to N-cycling activity failed to explain the observed variance in rates of N-regeneration and N-assimilation, possibly due to the limited number of process rate observations. In the surface waters of 5 further stations, Ocean Acidification (OA) bioassay experiments were conducted to investigate the response of NH4+ oxidising and regenerating organisms to simulated OA conditions, including the implications for [N2O]. Multivariate analysis was undertaken which considered the complete bioassay dataset of measured variables describing changes in N-regeneration rate, [N2O] and the biogeochemical composition of seawater. While anticipating biogeochemical differences between locations, we aimed to test the hypothesis that the underlying mechanism through which pelagic N-regeneration responded to simulated OA conditions was independent of location and that a mechanistic understanding of how NH4+ oxidation, NH4+ regeneration and N2O production responded to OA could be developed. Results indicated that N-regeneration process responses to OA treatments were location specific; no mechanistic understanding of how N-regeneration processes respond to OA in the surface ocean of the NW European shelf sea could be developed

    A Novel Power-Band based Data Segmentation Method for Enhancing Meter Phase and Transformer-Meter Pairing Identification

    This paper presents a novel power-band-based data segmentation (PBDS) method to enhance the identification of meter phase and meter-transformer pairing. Meters that share the same transformer or are on the same phase typically exhibit strongly correlated voltage profiles. However, under high power consumption, there can be significant voltage drops along the line connecting a customer to the distribution transformer. These voltage drops significantly decrease the correlations among meters on the same phase or supplied by the same transformer, resulting in high misidentification rates. To address this issue, we propose using power bands to select highly correlated voltage segments for computing correlations, rather than relying solely on correlations computed from the entire voltage waveforms. The algorithm's performance is assessed by conducting tests using data gathered from 13 utility feeders. To ensure the credibility of the identification results, utility engineers conduct field verification for all 13 feeders.     The homotopy type of the loops on (n−1)(n-1)-connected (2n+1)(2n+1)-manifolds

    For n≥2n\geq 2 we compute the homotopy groups of (n−1)(n-1)-connected closed manifolds of dimension (2n+1)(2n+1). Away from the finite set of primes dividing the order of the torsion subgroup in homology, the pp-local homotopy groups of MM are determined by the rank of the free Abelian part of the homology. Moreover, we show that these pp-local homotopy groups can be expressed as a direct sum of pp-local homotopy groups of spheres.     Age and growth, reproduction and diet of a sublittoral population of the rock goby Gobius paganellus (Teleostei, Gobiidae)

Printed in the Netherlands.Basic biological information for a sublittoral population of the rock goby Gobius paganellus Linnaeus, 1758 is presented based on a 2-year study involving 1680 specimens. The length-weight relationship was given by TW = 0.0089 * TL^3,163 (where TW= total weight in g; TL= total length in mm). Age at length data were inferred by modal analysis of the monthly length-frequency distributions. The parameters of the fitted Von Bertalanffy growth equation (with seasonal component, birth date on the 1st of January) were L1 = 13.8 cm; K = 0.73 yr^-1; to = -0.22 yr; C = 0.95; W = 0.07. This growth rate is much higher than that described for northern Europe populations (where K is about 0.3 yr^-1) and is probably associated with a shorter life span. Macroscopic examination of the gonads, and analysis of the monthly values of the gonadosomatic index, indicated that reproduction occurs in winter and early spring, with a maximum in February and March, when water temperatures are lowest. Individuals become sexually mature around 6-7 cm TL, a size that can be reached in less than 1 year. By contrast, individuals of this goby in the British Isles mature in their second or third year.     Pole/Zero Design of Agonist/Antagonist Actuation

    Objective: This brief analyzes the open-loop dynamics of coupled (dedicated) and decoupled tendon tension control methods in an agonist/antagonist (AA) actuator pair, a fundamental control unit used in orthopedic testbeds known as joint motion simulators (JMSs). Methods: A linear mathematical model of an AA actuator pair is derived. Transfer functions from tendon tension control signal to tendon tension and joint position are derived. Sources of key dynamics are explained. Results: The system's dynamic model shows that both the dedicated and decoupled approaches have a low-frequency pole pair, a low-frequency zero pair, and a mid-range zero pair. A frequency domain identification of the flexion/extension axis of an existing elbow JMS validates the locations of these dynamics. The interaction between tendon tension control and joint position is shown to be controllable in decoupled control, but not in dedicated control. The bandwidth reduction due to the low-frequency pole pair and low-frequency zero pair are shown to be controllable in decoupled control, but not in dedicated control. Conclusion: Decoupled control is superior to dedicated control for AA actuator pairs in JMS designs because it reduces actuator interaction and has a larger tension control bandwidth. Significance: This analysis describes the sources of the dynamics seen in the open-loop frequency response of both methods and shows the superiority of the decoupled method in tension control bandwidth and in lack of interactions with position control

    An Iterative Bidirectional Gradient Boosting Algorithm for CVR Baseline Estimation

    This paper presents a novel iterative, bidirectional, gradient boosting (bidirectional-GB) algorithm for estimating the baseline of the Conservation Voltage Reduction (CVR) program. We define the CVR baseline as the load profile during the CVR period if the substation voltage is not lowered. The proposed algorithm consists of two key steps: selection of similar days and iterative bidirectional-GB training. In the first step, pre- and post-event temperature profiles of the targeted CVR day are used to select similar days from historical non-CVR days. In the second step, the pre-event and post-event similar days are used to train two GBMs iteratively: a forward-GBM and a backward-GBM. After each iteration, the two generated CVR baselines are reconciled and only the first and the last points on the reconciled baseline are kept. The iteration repeats until all CVR baseline points are generated. We tested two gradient boosting methods (i.e., GBM and LighGBM) with two data resolutions (i.e., 15- and 30-minute). The results demonstrate that both the accuracy and performance of the algorithm are satisfactory.Comment: 5 pages, 8 figures, 2 table
