Lysine degradation by ruminal Fusobacterium necrophorum
Three experiments were conducted to characterize lysine fermentation by Fusobacterium necrophorum, a ruminal bacterium that is known to degrade amino acids. In Experiment 1, 7 strains of Fusobacterium necrophorum were inoculated into media containing lysine (50 mM), lactate (50 mM), or lysine plus lactate (50 mM each) as the major energy substrate to evaluate growth and ammonia production. All strains grew with lysine, lactate, or lactate plus lysine as the primary substrate. When grown with lysine, all strains produced ammonia as an end product, even if lactate was also present. Smaller concentrations of ammonia for medium containing lactate plus lysine when compared with lysine alone indicate that the Fusobacterium strains used lactate as a growth substrate that stimulated utilization of ammonia. In Experiment 2, the 2 strains tested were able to degrade extensively both lysine and glutamic acid. Some evidence was detected for partial utilization for growth of histidine, methionine, and tryptophan by strain A21. In Experiment 3, the minimum inhibitory concentration (MIC) of the antibiotic tylosin was 25 μg/mL when Fusobacterium necrophorum strains A21 and B35 were grown in either lysine or lactate-enriched medium. The MIC of monensin was 6.25 and 3.9 μg/mL for strains A21 and B35, respectively, when grown in lysine-enriched medium, but \u3e 50 and 10.9 μg/mL when the strains were grown in lactate-enriched medium. Bioavailability of lysine from hydroxymethyl lysine
Twelve mature sheep were used as a ruminant model to estimate the bioavailability of lysine in hydroxymethyl lysine (HML) compared with a commercial product of rumen-protected lysine (RPL; LysiPEARL, Kemin Industries, Inc.) with known availability. The sheep were fed a diet with a forage to concentrate ratio similar to that of dairy diets. Following a control period in which plasma lysine was measured when sheep received no supplemental lysine, the sheep were provided 2 of 4 treatments during periods 2 and 3; treatments included RPL to provide 3 or 6 g/day of available lysine (actual amounts of product provided were based on the manufacturer\u27s data related to ruminal escape and intestinal availability) and 3 or 6 g/day of lysine provided as HML. Blood samples were collected at the end of each feeding period at 3 hours after feeding. Both HML and RPL significantly increased plasma lysine concentrations. By comparison with plasma lysine concentrations when known amounts of bioavailable lysine were provided as RPL, the bioavailability of lysine in HML was estimated to be 94%. 36Cl measurements in groundwaters of the deep confined aquifer of the Lake Chad Basin (LCB) were performed in order to constrain groundwater geochemical ages and residence times. Twenty-seven wells were sampled in Nigeria, Niger and Chad in the southern parts of the large (700 000 km2) multilayered aquifer of the LCB. 36Cl/Cl values range between 11±1.10-15 to 148±8.10-15 at/at. The highest ratios are observed near the recharge zone of the Nigerian part of the Continental Terminal aquifer, while the lowest ones are found in wells located near the southern fringe of the present-day lake Chad. Chloride concentrations are low (below 100 mg/l) and not correlated to the 36Cl/Cl values, indicating negligible dissolution of evaporites in most samples. Reliable 36Cl ages can be calculated along the different flow paths investigated, suggesting residence times of the deep groundwaters larger than 300 000 years. These results are consistent with new AMS-14C data below the detection limit but are in contradiction with previous 14C data obtained in the area
Chemical compositions and biological activities of commiphora gileadensis: A review
Background: A significant medicinal plant with several medicinal applications is Commiphora gileadensis. Due to the C. gileadensis medicinal properties, such as anti-cancer, antibacterial, antioxidant, cytotoxic, and antiviral activities, secondary phytoconstituents of the plant are crucial for medicine. Objective: This review aims to identify the biological activities and chemical compositions of C. gileadensis. Method: Data for the study were gathered from book chapters, research articles, review articles, and conference papers as part of the study's review approach, which was based on a thorough search of the literature. Results: Based on the results obtained, it may be inferred that the chemical composition and lipid constituents of C. gileadensis extracts provide substantiation for their utilisation in traditional medicinal practises. The primary chemical constituents seen in the sample were β-pinene (7.0% ± 0.1%), cymene (13.6 ± 0.1%), α-pinene (14.4%), terpinen-4-ol (18.7 ± 0.1%), and sabinene (22.7 ± 0.1%). Additionally, the sample exhibited the presence of various lipids, including phosphatidylethanolamine (PE) at a concentration of 7.6%, hexosylceramide (Hex1Cer) at 18%, and ceramide (Cer) at 69%. Conclusion: As a result, this review presented an overview of C. gileadensis' biological activities and chemical compositions. The review suggests that more attention be paid to the leaves barks. It stems from C. gileadensis, as well as the type of extraction techniques and advanced characterization because extending research materials could lead to the discovery of novel bioactive components
The global burden of cancer attributable to risk factors, 2010–19: a systematic analysis for the Global Burden of Disease Study 2019
BACKGROUND: Understanding the magnitude of cancer burden attributable to potentially modifiable risk factors is crucial for development of effective prevention and mitigation strategies. We analysed results from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2019 to inform cancer control planning efforts globally. METHODS: The GBD 2019 comparative risk assessment framework was used to estimate cancer burden attributable to behavioural, environmental and occupational, and metabolic risk factors. A total of 82 risk–outcome pairs were included on the basis of the World Cancer Research Fund criteria. Estimated cancer deaths and disability-adjusted life-years (DALYs) in 2019 and change in these measures between 2010 and 2019 are presented. FINDINGS: Globally, in 2019, the risk factors included in this analysis accounted for 4·45 million (95% uncertainty interval 4·01–4·94) deaths and 105 million (95·0–116) DALYs for both sexes combined, representing 44·4% (41·3–48·4) of all cancer deaths and 42·0% (39·1–45·6) of all DALYs. There were 2·88 million (2·60–3·18) risk-attributable cancer deaths in males (50·6% [47·8–54·1] of all male cancer deaths) and 1·58 million (1·36–1·84) risk-attributable cancer deaths in females (36·3% [32·5–41·3] of all female cancer deaths). The leading risk factors at the most detailed level globally for risk-attributable cancer deaths and DALYs in 2019 for both sexes combined were smoking, followed by alcohol use and high BMI. Risk-attributable cancer burden varied by world region and Socio-demographic Index (SDI), with smoking, unsafe sex, and alcohol use being the three leading risk factors for risk-attributable cancer DALYs in low SDI locations in 2019, whereas DALYs in high SDI locations mirrored the top three global risk factor rankings. From 2010 to 2019, global risk-attributable cancer deaths increased by 20·4% (12·6–28·4) and DALYs by 16·8% (8·8–25·0), with the greatest percentage increase in metabolic risks (34·7% [27·9–42·8] and 33·3% [25·8–42·0]). INTERPRETATION: The leading risk factors contributing to global cancer burden in 2019 were behavioural, whereas metabolic risk factors saw the largest increases between 2010 and 2019. Reducing exposure to these modifiable risk factors would decrease cancer mortality and DALY rates worldwide, and policies should be tailored appropriately to local cancer risk factor burden
“Theory Becoming Alive”: The Learning Transition Process of Newly Graduated Nurses in Canada
Background
Newly graduated nurses often encounter a gap between theory and practice in clinical settings. Although this has been the focus of considerable research, little is known about the learning transition process.
Purpose
The purpose of this study was to explore the experiences of newly graduated nurses in acute healthcare settings within Canada. This study was conducted to gain a greater understanding of the experiences and challenges faced by graduates.
Methods
Grounded theory method was utilized with a sample of 14 registered nurses who were employed in acute-care settings. Data were collected using in-depth interviews. Constant comparative analysis was used to analyze data.
Results
Findings revealed a core category, “Theory Becoming Alive,” and four supporting categories: Entry into Practice, Immersion, Committing, and Evolving. Theory Becoming Alive described the process of new graduate nurses’ clinical learning experiences as well as the challenges that they encountered in clinical settings after graduating.
Conclusions
This research provides a greater understanding of learning process of new graduate nurses in Canada. It highlights the importance of providing supportive environments to assist new graduate nurses to develop confidence as independent registered nurses in clinical areas. Future research directions as well as supportive educational strategies are described
Prevention of Recurrent Laryngeal Nerve Injury in Thyroid Surgery: Are Neuromonitoring Techniques Needed?
Background: Identification of the recurrent laryngeal nerve is the most
essential in thyroid surgery. Positive identification of the nerve is
essential for preservation of nerve integrity and function. Some
authors recommend using of nerve monitoring techniques. Methods: 359
thyroid surgeries with obligatory exposure of the RLN were reviewed.
Meticulous dissection and exploration of the nerve was the main
technique in our study. Results: The nerve was identified in 97% of
cases, Temporary RLN palsy was found in 6 patients and permanent palsy
was found in 1 case. No neuromonitoring techniques were used.
Discussion: Our results and the literature review showed that the nerve
can be clearly identified and protected in thyroid surgery without
using any neuromonitoring techniques. In cases without identification,
capsular dissection is the safest way to protect the nerve.

substitute to meticulous dissection and were never with better results
than conventional meticulous dissection
