    Biological reference points for Atlantic surfclam (Spisula solidissima) in warming seas

    Atlantic surfclam (Spisula solidissima) are a large, commercially important shellfish in the United States faced with several important management challenges. Compared to many harvested fish and shellfish, their life history is relatively unknown. They are undergoing contraction in the southern and inshore parts of their range, as well as expansion into deeper water. Atlantic surfclam are thermally sensitive, and the changes in their distribution track changes in maximum bottom temperature. Sessile species cannot emigrate and are limited to recruitment and mortality as mechanisms for redistribution in response to changing climate. Management of Atlantic surf clam should account for these challenges. We describe a simulation designed to calculate biological reference points that will work well for Atlantic surfclam relative to biological and fishery goals, over a range of life history parameters, assessment uncertainties, and increases in temperature. Simulations of the trade-off between somatic growth and mortality under increased temperature led to target fishing mortality rates higher than the status quo, but also to increased variability in yield. Results suggest that increasing temperature may adversely affect the Atlantic surfclam industry, which prefers stable catches to short term increases in yield, due to market limitations. The results of this analysis are specific to Atlantic surfclam, but the methods described here could be used to enhance management for other harvested species facing similar challenges

    Polygenic risk scores for schizophrenia and major depression are associated with socio-economic indicators of adversity in two British community samples

    Schizophrenia (SCZ) and major depressive disorder (MDD) are complex psychiatric disorders which contribute substantially to the global burden of disease. Both psychopathologies are heritable with some genetic overlap between them. Importantly, SCZ and MDD have also been found to be associated with environmental risk factors. However, rather than being independent of genetic influences, exposure to environmental risk factors may be under genetic control, known as gene-environment correlation (rGE). In this study we investigated rGE in relation to polygenic risk scores for SCZ and MDD in adults, derived from large genome-wide association studies, across two different British community samples: Understanding Society (USoc) and the National Child Development Study (NCDS). We tested whether established environmental risk factors for SCZ and/or MDD are correlated with polygenic scores in adults and whether these associations differ between the two disorders and cohorts. Findings partially overlapped between disorders and cohorts. In NCDS, we identified a significant correlation between the genetic risk for MDD and an indicator of low socio-economic status, but no significant findings emerged for SCZ. In USoc, we replicated associations between indicators of low socio-economic status and the genetic propensity for MDD. In addition, we identified associations between the genetic susceptibility for SCZ and being single or divorced. Results across both studies provide further evidence that the genetic risk for SCZ and MDD were associated with common environmental risk factors, specifically MDD’s association with lower socio-economic status

    Fishing and bottom water temperature as drivers of change in maximum shell length in Atlantic surfclams (Spisula solidissima)

    Maximum shell length of Atlantic surfclams (Spisula solidissima) on the Middle Atlantic Bight (MAB) continental shelf, obtained from federal fishery survey data from 1982-present, has decreased by 15-20 mm. Two potential causes of this decreasing trend, fishery removal of large animals and stress due to warming bottom temperatures, were investigated using an individual-based model for post-settlement surfclams and a fifty-year hindcast of bottom water temperatures on the MAB. Simulations showed that fishing and/or warming bottom water temperature can cause decreases in maximum surfclam shell length (body size) equivalent to those observed in the fished stock. Independently, either localized fishing rates of 20% or sustained bottom temperatures that are 2 degrees C warmer than average conditions generate the observed decrease in maximum shell length. However, these independent conditions represent extremes and are not sustained in the MAB. The combined effects of fishing and warmer temperatures can generate simulated length decreases that are similar to observed decreases. Interannual variability in bottom water temperatures can also generate fluctuations in simulated shell length of up to 20 mm over a period of 10-15 years. If the change in maximum size is not genotypic, simulations also suggest that shell size composition of surfclam populations can recover if conditions change; however, that recovery could take a decade to become evident

    An Overview Of Factors Affecting Distribution Of The Atlantic Surfclam (Spisula solidissima), A Continental Shelf Biomass Dominant, During A Period Of Climate Change

    The Atlantic surfclam (Spisula solidissitna) is a dominant member of the biological community of the Middle Atlantic Bight continental shelf and a commercially harvested species. Climate warming is affecting the biology and distribution of this species, which provides an opportunity to investigate the processes and conditions that are restructuring this fishery and the implications for ecological and socioeconomic systems. A Management Strategy Evaluation (MSE) developed for the surfclam fishery provides a mechanistic description of the surfclam\u27s response to climate change and understanding of the cascade of effects initiated by changes in oceanographic conditions that ultimately appear as social and economic effects. This understanding in turn informs development of management policies for the resource. This overview considers the components of the surfclam MSE, relevant results, and implications for management and policy. The lessons learned from the surfclam MSE provide a basis for applying similar approaches to other ecologically important species that are also commercially exploitable resources

    The Exposed Surface Area To Volume Ratio: Is Shell More Efficient Than Limestone In Promoting Oyster Recruitment?

    Planting oyster cultch is a common management approach used to enhance recruitment. The two most popular cultch materials are shell and limestone. Both are sold by volume or weight; however, once deposited on oyster grounds, only a small portion of the total surface area of each particle is available for recruitment. Shell and limestone have different surface area to volume properties, and thus provide differential settlement opportunities. Exposed surface area to volume (expSA/V) ratios of oyster shell and limestone fragments were compared, as an indicator of their recruitment potential and cost-effectiveness for cultch planting. Samples were collected from the Primary Public Oyster Seed Grounds in Louisiana by vibracore, and from the Pass Christian Tonging Grounds in Mississippi by dredge. Shell (including whole shell and fragments) and limestone particles greater than or equal to 8 mm by geometric shape were classified and their expSA/V was calculated. Mean expSA/V ratios of shell were approximately three to nine times higher than limestone. For limestone of similar particle size to provide an equivalent recruitment benefit for the same cost would require that the cost of purchase, transport, and planting be three to nine times lower than shell. Thus, shell is likely to be a more efficient material than limestone for recruitment enhancement. Nevertheless, the higher variability in expSA/V of shell and other factors such as the expected lifetime and the relative performance of small and large particles of materials should also be considered. Analysis of a Louisiana limestone plant and associated oyster cultch showed that the proportion of small and large limestone particles and the relative proportion of whole shells and fragments can greatly alter expSA/V. In this case, the a priori expectation that oyster shell would outperform limestone did not materialize because of the quantity of small limestone particles of favorable shapes in the deployed material. Even so, as yet unknown is the possible reduction in performance in situ of smaller particles that might occur if they increase the one-dimensionality of the plant

    Too close for comfort: spatial patterns in acorn barnacle populations

    Spatial patterns in aggregations form as a result of the interplay between costs and benefits experienced by individuals. Such self-organisation of aggregations can be explained using a zonal model in which a short-range zone of repulsion and longer-range zone of attraction surrounding individuals leads to emergent pattern properties. The signal of these processes can be detected using spatial pattern analyses. Furthermore, in sessile organisms, post-settlement mortality reveals the relative costs and benefits of positions within the aggregation. Acorn barnacles are known to require contact with conspecifics for reproduction and are therefore believed to aggregate for this purpose; isolated individuals may also be more susceptible to abiotic stress and predation. At short distances, however, competition for space and resources is likely to occur. In this study spatial patterns of barnacles (Semibalanus balanoides L.) were analysed using pair-correlation functions. Individuals were dispersed at distances below 0.30 cm, but peak relative density occurred at a distance of 0.36 cm from conspecifics. This is much closer than required for reproductive access, implying a strong aggregative drive, up to the point of physical contact with neighbours. Nevertheless, analysis of dead barnacles illustrated that such proximity carries a cost as barnacles with many neighbours were more likely to have died. The inferences obtained from these patterns are that barnacles aggregate as closely as they can, and that local neighbourhood competition is a powerful determinant of mortality. These processes give rise to the observed pattern properties

    High Distribution of CD40 and TRAF2 in Th40 T Cell Rafts Leads to Preferential Survival of this Auto-Aggressive Population in Autoimmunity

    CD40-CD154 interactions have proven critical in autoimmunity, with the identification of CD4(lo)CD40(+) T cells (Th40 cells) as harboring an autoaggressive T cell population shedding new insights into those disease processes. Th40 cells are present at contained levels in non-autoimmune individuals but are significantly expanded in autoimmunity. Th40 cells are necessary and sufficient in transferring type 1 diabetes in mouse models. However, little is known about CD40 signaling in T cells and whether there are differences in that signaling and subsequent outcome depending on disease conditions. When CD40 is engaged, CD40 and TNF-receptor associated factors, TRAFs, become associated with lipid raft microdomains. Dysregulation of T cell homeostasis is emerging as a major contributor to autoimmune disease and thwarted apoptosis is key in breaking homeostasis.Cells were sorted into CD4(hi) and CD4(lo) (Th40 cells) then treated and assayed either as whole or fractionated cell lysates. Protein expression was assayed by western blot and Nf-kappaB DNA-binding activity by electrophoretic mobility shifts. We demonstrate here that autoimmune NOD Th40 cells have drastically exaggerated expression of CD40 on a per-cell-basis compared to non-autoimmune BALB/c. Immediately ex-vivo, untreated Th40 cells from NOD mice have high levels of CD40 and TRAF2 associated with the raft microdomain while Th40 cells from NOR and BALB/c mice do not. CD40 engagement of Th40 cells induces Nf-kappaB DNA-binding activity and anti-apoptotic Bcl-X(L) expression in all three mouse strains. However, only in NOD Th40 cells is anti-apoptotic cFLIP(p43) induced which leads to preferential survival and proliferation. Importantly, CD40 engagement rescues NOD Th40 cells from Fas-induced death.CD40 may act as a switch between life and death promoting signals and NOD Th40 cells are poised for survival via this switch. This may explain how they expand in autoimmunity to thwart T cell homeostasis

    Galectin-9 Controls CD40 Signaling through a Tim-3 Independent Mechanism and Redirects the Cytokine Profile of Pathogenic T Cells in Autoimmunity

    While it has long been understood that CD40 plays a critical role in the etiology of autoimmunity, glycobiology is emerging as an important contributor. CD40 signaling is also gaining further interest in transplantation and cancer therapies. Work on CD40 signaling has focused on signaling outcomes and blocking of its ligand, CD154, while little is known about the actual receptor itself and its control. We demonstrated that CD40 is in fact several receptors occurring as constellations of differentially glycosylated forms of the protein that can sometimes form hybrid receptors with other proteins. An enticing area of autoimmunity is differential glycosylation of immune molecules leading to altered signaling. Galectins interact with carbohydrates on proteins to effect such signaling alterations. Studying autoimmune prone NOD and non-autoimmune BALB/c mice, here we reveal that in-vivo CD40 signals alter the glycosylation status of non-autoimmune derived CD4 T cells to resemble that of autoimmune derived CD4 T cells. Galectin-9 interacts with CD40 and, at higher concentrations, prevents CD40 induced proliferative responses of CD4loCD40+ effector T cells and induces cell death through a Tim-3 independent mechanism. Interestingly, galectin-9, at lower concentrations, alters the surface expression of CD3, CD4, and TCR, regulating access to those molecules and thereby redirects the inflammatory cytokine phenotype and CD3 induced proliferation of autoimmune CD4loCD40+ T cells. Understanding the dynamics of the CD40 receptor(s) and the impact of glycosylation status in immunity will gain insight into how to maintain useful CD40 signals while shutting down detrimental ones
