Spatial, High-Accuracy, Positioning-Encoding Sensor (SHAPES) for large space system control applications
The Spatial, High-Accuracy, Position-Encoding Sensor is a controls sensor suitable for the determination of the static shape and vibrational motion of large space structures and similar systems and for the determination of position and velocity in rendezvous and docking. It uses a combination of electro-optical techniques to measure the three-dimensional coordinates distributed over the structure at reading rates high compared to the rates at which the coordinates are changing. The technical approach is that of measuring the distance to and the direction of points on the structure from a single sensor head. Many points can be measured simultaneously from a single head without significantly increasing the complexity of the system
Components required for in vitro cleavage and polyadenylation of eukaryotic mRNA
We have studied in vitro cleavage/polyadenylation of precursor RNA containing herpes simplex virus type 2 poly A site sequences and have analysed four RNA/protein complexes which form during in vitro reactions. Two complexes, A and B, form extremely rapidly and are then progressively replaced by a third complex, C which is produced following cleavage and polyadenylation of precursor RNA. Substitution of ATP with cordycepin triphosphate prevents polyadenylation and the formation of complex C however a fourth complex, D, results which contains cleaved RNA. A precursor RNA lacking GU-rich downstream sequences required for efficient cleavage/ polyadenylation fails to form complex B and produces a markedly reduced amount of complex A. As these GU-rich sequences are required for efficient cleavage, this establishes a relationship between complex B formation and cleavage/polyadenylation of precursor RNA in vitro. The components required for in vitro RNA processing have been separated by fractionation of the nuclear extract on Q-Sepharose and Biorex 70 columns. A Q-Sepharose fraction forms complex B but does not process RNA. Addition of a Biorex 70 fraction restores cleavage activity at the poly A site but this fraction does not appear to contribute to complex formation. Moreover, in the absence of polyethylene glycol, precursor RNA is not cleaved and polyadenylated, however, complexes A and B readily form. Thus, while complex B is necessary for in vitro cleavage and polyadenylation, it may not contain all the components required for this processing
Optical fiber coupling method and apparatus
Systems are described for coupling a pair of optical fibers to pass light between them, which enables a coupler to be easily made, and with simple equipment, while closely controlling the characteristics of the coupler. One method includes mounting a pair of optical fibers on a block having a large hole therein, so the fibers extend across the hole while lying adjacent and parallel to one another. The fibers are immersed in an etchant to reduce the thickness of cladding around the fiber core. The fibers are joined together by applying a liquid polymer so the polymer-air interface moves along the length of the fibers to bring the fibers together in a zipper-like manner, and to progressively lay a thin coating of the polymer on the fibers
Ranging system which compares an object reflected component of a light beam to a reference component of the light beam
A system is described for measuring the distance to an object by comparing a first component of a light pulse that is reflected off the object with a second component of the light pulse that passes along a reference path of known length, which provides great accuracy with a relatively simple and rugged design. The reference path can be changed in precise steps so that it has an equivalent length approximately equal to the path length of the light pulse component that is reflected from the object. The resulting small difference in path lengths can be precisely determined by directing the light pulse components into opposite ends of a detector formed of a material that emits a second harmonic light output at the locations where the opposite going pulses past simultaneously across one another
Acute phase protein levels in dogs with mast cell tumours and sarcomas
<p><b>Context:</b> The acute phase protein response is part of a non-specific and complex host response to inflammation. It occurs shortly after tissue injury and may be induced by a range of different causes, including infectious, inflammatory, neoplastic, traumatic or immunological disease. Although it was conventionally believed that APPs were exclusively hepatocyte derived, there is increasing evidence to support extra-hepatic generation in neoplastic and other disease states. In people, C-reactive protein (CRP) has been shown to be of value in identifying metastatic disease from primary renal tumours as well as showing promise for monitoring rejection of renal transplants. Serum CRP correlates with survival in colorectal cancer and oesophageal squamous cell carcinoma while serum amyloid A (SAA) concentrations correlate with cancer activity, stage and prognosis in gastric tumours. Recent immunohistochemical studies in people with oesophageal carcinoma suggest that tumour tissue may itself elaborate APP with a poorer survival and outcome associated with tumours elaborating higher levels of CRP. A similar association has been seen between alpha-1 acid glycoprotein (AGP) and colorectal tumours and ovarian carcinoma.</p>
<p>As yet, studies regarding APP values in neoplastic conditions in dogs are limited, and many are non-specific. In veterinary patients, elevated levels of AGP have been identified in dogs with a range of tumours with localisation to liver and splenic tissue in one study. Another study found higher levels of AGP in dogs with non-specific tumours of grade III-IV based on the WHO Tumour Node Metastasis (TNM) scale and elevated serum AGP has been documented in non-specific tumour-bearing cats. Elevated CRP levels have been documented in both dogs and cats with lymphoma and serum CRP may be used as an indicator of complete remission status in dogs with multicentric lymphoma. Elevated levels of CRP, Haptoglobin (Hp) and SAA have been identified in dogs with mammary tumours, with significant increases over normal in the presence of metastatic disease, primary tumours greater than 5cm in diameter and those with ulceration.</p>
<p>In this study we evaluated an APP profile using four APPs (CRP, Hp, SAA and AGP), in dogs with mast cell tumours (MCTs) and sarcomas to assess whether the APP profile would change in reflection of tumour presence; whether the extent of any change would correlate with tumour grade; and whether the changes would differ with tumour type.</p>
<p><b>Approach:</b> Patients with naturally occurring MCTs and sarcomas presenting for staging and treatment were included if they met the study criteria. Criteria for inclusion were that the patient was not currently being treated with steroids, did not have a recent history of infectious or inflammatory disease other than the tumour, a definitive histological diagnosis was available and a full staging procedure was completed prior to surgery using standard oncological protocols to identify metastatic disease where present. Following surgical resection each tumour was submitted for full histological evaluation and grading to include assessment of the margins of excision. Cases were only enrolled in the study if blood sampling formed part of the clinical investigation and/or treatment, and where residual blood was available after diagnostic sampling which would otherwise have been disposed of as clinical waste. In brief, the CRP levels were determined by immunoturbidometric assay and Hp by means of haemoglobin binding capacity assay. SAA was measured with a commercial canine ELISA kit (TriDelta Development, Dublin, Ireland) and AGP was measured with a commercial radial immunodiffusion assay (J-Path Inc, Tokyo, Japan).</p>
<p><b>Results:</b> All comparisons using continuous data were checked for normality and equality of variances and appropriate statistical tests were employed (student’s t test operationalised as a two-sample Welch’s test for samples of unequal sizes and variances, Mann-Whitney, Chi-square and Fishers exact tests as appropriate). In MCTs, the CRP and AGP were elevated above reference ranges, Hp showed no significant change and SAA dropped relative to the reference range. In sarcoma patients CRP, Hp and AGP were all elevated above reference ranges. None of the tumour grade differences were significant apart from SAA in sarcoma patients where values in grade 2 sarcoma were significantly higher than those in grade 1.</p>
<p><b>Interpretation and notes of caution:</b> The numbers in our groups were small which compromises the validity of statistical evaluation so our results must be interpreted with caution. However some interesting relationships have emerged from the initial evaluation which suggests that APP profiles may have potential for screening in patients with neoplastic disease. For patients with MCTs, CRP and AGP levels would be expected to increase, with a concurrent drop in SAA levels. In sarcoma patients CRP, AGP and Hp can all be expected to increase. These initial results need to be evaluated in larger numbers of cases with naturally occurring disease to validate the findings, to assess whether the presence and extent of metastatic disease has a significant effect, and also to confirm whether the values alter after surgical resection of the primary tumour.</p>
<p><b>Significance of findings:</b> If there are consistent and specific changes in APP profiles associated with different tumour types in dogs, as is the case with a wide range of cancers in humans, then there may be potential for APP profiles on routine blood samples to be used as indicators of disease, or where monitoring for recurrence. Whether they could also have potential for assessment of the presence of metastatic disease and prognosis as in people is unknown as yet.</p>
Reversing the trend: interventions to treat intracranial haemorrhage associated with anticoagulation
Elevated interferon-stimulated gene transcription in peripheral blood mononuclear cells occurs in patients infected with genotype 1 but not genotype 3 hepatitis C virus
Hepatitis C virus (HCV) can be classified into seven distinct genotypes that are associated with differing pathologies and respond differently to antiviral therapy. In the UK, genotype 1 and 3 are present in approximately equal proportions. Chronic infection with HCV genotype 3 is associated with increased liver steatosis and reduced peripheral total cholesterol levels, which potentially influences peripheral immune responses. To understand these differences, we investigated host gene transcription in peripheral blood mononuclear cells by microarray and quantitative PCR in patients with genotype 1 (n = 22) or genotype 3 infection (n = 22) and matched healthy controls (n = 15). Enrichment of genes involved in immune response and inflammatory pathways were present in patients infected with HCV genotype 1; however, no differences in genes involved in lipid or cholesterol metabolism were detected. This genotype-specific induction of genes is unrelated to IL28B genotype or previous treatment failure. Our data support the hypothesis that genotype 1 infection drives a skewed Type I interferon response and provides a foundation for future investigations into the host–pathogen interactions that underlie the genotype-specific clinical outcomes of chronic HCV infection
Influences of forested and grassland vegetation on late Quaternary ecosystem development as recorded in lacustrine sediments
Geosphere-biosphere interactions are ubiquitous features of the Earth surface, yet the development of interactions between newly exposed lithologic surfaces and colonizing plants during primary succession after glaciation are lacking temporal detail. To assess the nature, rate, and magnitude of vegetation influence on parent material and sediment delivery, we analyzed ecosystem and geochemical proxies from lacustrine sediment cores at a grassland site and a forested site in the northern United States. Over time, terrigenous inputs declined at both sites, with increasing amounts of organic inputs toward present. The similarities between sites were striking given that the grassland sequence began in the Early Holocene, and the forested sequence began after the last glacial maximum. Multiple mechanisms of chemical weathering, hydrologic transport, and changes in source material potentially contribute to this pattern. Although there were strong links between vegetation composition and nitrogen cycling at each site, it appears that changes in forest type, or from oak woodland to grassland, did not exert a large influence on elemental (K, Ti, Si, Ca, Fe, Mn, and S) abundance in the sedimentary sequences. Rather, other factors in the catchment-lake system determined the temporal sequence of elemental abundance

Effects of past climate variability on fire and vegetation in the cerrãdo savanna of the Huanchaca Mesetta, NE Bolivia
Cerrãdo savannas have the greatest fire activity
of all major global land-cover types and play a significant
role in the global carbon cycle. During the 21st century,
temperatures are projected to increase by ∼ 3
â—¦C coupled
with a precipitation decrease of ∼ 20 %. Although these conditions
could potentially intensify drought stress, it is unknown
how that might alter vegetation composition and fire
regimes. To assess how Neotropical savannas responded to
past climate changes, a 14 500-year, high-resolution, sedimentary
record from Huanchaca Mesetta, a palm swamp located
in the cerrãdo savanna in northeastern Bolivia, was analyzed
with phytoliths, stable isotopes, and charcoal. A nonanalogue,
cold-adapted vegetation community dominated the
Lateglacial–early Holocene period (14 500–9000 cal yr BP,
which included trees and C3 Pooideae and C4 Panicoideae
grasses. The Lateglacial vegetation was fire-sensitive and fire
activity during this period was low, likely responding to fuel
availability and limitation. Although similar vegetation characterized
the early Holocene, the warming conditions associated
with the onset of the Holocene led to an initial increase
in fire activity. Huanchaca Mesetta became increasingly firedependent
during the middle Holocene with the expansion
of C4 fire-adapted grasses. However, as warm, dry conditions,
characterized by increased length and severity of the
dry season, continued, fuel availability decreased. The establishment
of the modern palm swamp vegetation occurred at
5000 cal yr BP. Edaphic factors are the first-order control on
vegetation on the rocky quartzite mesetta. Where soils are
sufficiently thick, climate is the second-order control of vegetation
on the mesetta. The presence of the modern palm
swamp is attributed to two factors: (1) increased precipitation
that increased water table levels and (2) decreased frequency
and duration of surazos (cold wind incursions from
Patagonia) leading to increased temperature minima. Natural
(soil, climate, fire) drivers rather than anthropogenic
drivers control the vegetation and fire activity at Huanchaca
Mesetta. Thus the cerrãdo savanna ecosystem of the Huanchaca
Plateau has exhibited ecosystem resilience to major
climatic changes in both temperature and precipitation since
the Lateglacial period
