    Alat ekstraksi madu yang diputar secara manual masih sering terjadi kesalahan dalam putaran pada alat ekstraksi madu. Pengontrolan kecepatan pada alat ektraksi madu menggunakan DC menyempurnakan alat ekstraksi yang masih diputar secara manual. Hal tersebut memiliki proses tingkat efisiensi yang lebih baik dari segi waktu dan tenaga yang dibutuhkan. Digunakan Kontrol Logika Fuzzy untuk mengurangi kesalahan putaran pada alat ekstraksi madu, sehingga putaran motor dapat sesuai dengan kecepatan yang diinginkan. Hasil pengujian menggunakan kontroler logika fuzzy melalui beberapa tahap, yaitu fuzzifikasi, kaidah atur (Rule-Base) dan inferensi, dan defuzzifikasi. Metode fuzzifikasi yang digunakan adalah metode min-max sedangkan pada defuzzifikasi menggunakan metode Weighted Average dengan keluaran sistem menggunakan metode Tsukamoto. Presentase error yang dihasilkan atas respon secara keseluruhan sistem adalah 0.4129%

    This study aims to analyze the effect of shielding gas flow velocity on GMAW Aluminum 6061 welding on tensile strength, hardness, and Scanning Electron Microscopy (SEM) in bevel groove joints. The tensile test results from the reasoning for variations in the flow velocity of the protective gas 5 liters/minute have an average value of 4.56 kN, 10 liters/minute have an average value of 5.86 kN, 15 liters/minute have an average value of 6.73 kN. From the results of the micro vikers test which has decreased, for the hardness of the 5 liter/minute gas shield speed variation in the welding section is 71.53 HVN, the HAZ 1 section is 75.83 HVN, the HAZ 2 section is 76.27 HVN, 10 liters/minute in the weld section is 67.70 HVN, the HAZ 1 section is 65.56 HVN, the HAZ 2 section is 69.50 HVN, 15 liters/minute in the weld section is 57.83 HVN, the HAZ 1 section is 59.23 HVN, the HAZ 2 is 67.73 HVN.     Pengecoran adalah proses menghasilkan logam menjadi bentuk yang ditentukan dengan cara melebur logam padat menjadi bentuk cair, menuangkannya ke dalam cetakan dan membiarkannya mengeras sesuai bentuk yang diinginkan. Pada penelitian menggunakan proses sand casting dan simulasi menggunakan software SolidCast dengan material FC25. Hasil uji kekerasan pada bagian atas sebesar 123,826 HV, bagian tengah sebesar 168,847 HV, dan bagian bawah sebesar 132,6 HV. Temperatur tertinggi pada pengecoran memiliki nilai masing-masing yaitu, T1 sebesar 1023,75 ºC, T2 sebesar 1023,75 ºC, T3 sebesar 1023,75 ºC, dan hasil simulasi Ts sebesar 1023 ºC. Proses penurunan temperatur dihitung hingga 80 menit dengan temperatur masing-masing yaitu, T1 sebesar 370,75 ºC tetap menjadi penurunan suhu yang tertinggi, T2 sebesar 362,75 ºC ,T3 sebesar 330,25 ºC dan hasil simulasi Ts sebesar 330 ºC

    Aluminum alloy is one type of metal that is used as a construction material on ships. Determination of welding parameters is very important in the welding process, especially the strength of the welding current and the shape of the seam on the mechanical properties of the weld. This study was conducted to determine how much influence on the mechanical properties if the current used in welding is varied with the current strength values of 120 A, 140 A, and 160 A. The material used is Aluminum Series 6063 with a thickness of 5 mm with a single V joint connection of 60°. The welding method applied in this research is GMAW welding with ER 5356 filler with a diameter of 1 mm as the filler metal. By conducting tensile, hardness, microstructure, and elemental tests, the highest tensile strength was achieved at a current of 120 A with a stress value of 235 Mpa, strain 0.029, and modulus of elasticity 8102 Mpa. The lowest tensile strength occurs at a current of 160 A with a stress value of 167.6 Mpa, a strain of 0.032, and a modulus of elasticity of 5252 Mpa. The results of the SEM-EDX test are influenced by the low and high variations in current and heat input so that there are differences in the content of compounds in it, this indicates that the hardness value in the weld area is always greater than in the HAZ area and the parent metal for each variation of current strength. At a current of 120 A, the hardness test obtained the highest strength of 63.4 HVN, the lowest hardness at a current of 160 A with a value of 57.23 HVN

    DRAM-3 modulates autophagy and promotes cell survival in the absence of glucose

    Macroautophagy is a membrane-trafficking process that delivers cytoplasmic constituents to lysosomes for degradation. The process operates under basal conditions as a mechanism to turnover damaged or misfolded proteins and organelles. As a result, it has a major role in preserving cellular integrity and viability. In addition to this basal function, macroautophagy can also be modulated in response to various forms of cellular stress, and the rate and cargoes of macroautophagy can be tailored to facilitate appropriate cellular responses in particular situations. The macroautophagy machinery is regulated by a group of evolutionarily conserved autophagy-related (ATG) proteins and by several other autophagy regulators, which either have tissue-restricted expression or operate in specific contexts. We report here the characterization of a novel autophagy regulator that we have termed DRAM-3 due to its significant homology to damage-regulated autophagy modulator (DRAM-1). DRAM-3 is expressed in a broad spectrum of normal tissues and tumor cells, but different from DRAM-1, DRAM-3 is not induced by p53 or DNA-damaging agents. Immunofluorescence studies revealed that DRAM-3 localizes to lysosomes/autolysosomes, endosomes and the plasma membrane, but not the endoplasmic reticulum, phagophores, autophagosomes or Golgi, indicating significant overlap with DRAM-1 localization and with organelles associated with macroautophagy. In this regard, we further proceed to show that DRAM-3 expression causes accumulation of autophagosomes under basal conditions and enhances autophagic flux. Reciprocally, CRISPR/Cas9-mediated disruption of DRAM-3 impairs autophagic flux confirming that DRAM-3 is a modulator of macroautophagy. As macroautophagy can be cytoprotective under starvation conditions, we also tested whether DRAM-3 could promote survival on nutrient deprivation. This revealed that DRAM-3 can repress cell death and promote long-term clonogenic survival of cells grown in the absence of glucose. Interestingly, however, this effect is macroautophagy-independent. In summary, these findings constitute the primary characterization of DRAM-3 as a modulator of both macroautophagy and cell survival under starvation conditions

Bus station of Pilangsari is located in Pilangsari village, Ngrampal sub-district, Sragen district, Central Java province. The low public interest in using the terminal, the remote location of the terminal, and less than optimal terminal services have caused this terminal to not operate optimally. Efforts need to be made to improve the operation of the Bus station of Pilangsari, namely by relocating the terminal location from a remote place to a strategic place and then adding public open spaces and train stations in it. The existence of public open space aims to invite the public into the bus station area, so that the bus station area can live and be able to provide an attraction for the bus station itself. While the existence of the train station aims to improve services for the community in using the bus station. The application of this concept is expected to be able to improve the operation of the bus station of Pilangsari
