    Coexistence of antiferrodistortive and ferroelectric distortions at the PbTiO3_3 (001) surface

    The c(2×\times2) reconstruction of (001) PbTiO3_3 surfaces is studied by means of first principles calculations for paraelectric (non-polar) and ferroelectric ([001] polarized) films. Analysis of the atomic displacements in the near-surface region shows how the surface modifies the antiferrodistortive (AFD) instability and its interaction with ferroelectric (FE) distortions. The effect of the surface is found to be termination dependent. The AFD instability is suppressed at the TiO2_2 termination while it is strongly enhanced, relative to the bulk, at the PbO termination resulting in a c(2x2) surface reconstruction which is in excellent agreement with experiments. We find that, in contrast to bulk PbTiO3_3, in-plane ferroelectricity at the PbO termination does not suppress the AFD instability. The AFD and the in-plane FE distortions are instead concurrently enhanced at the PbO termination. This leads to a novel surface phase with coexisting FE and AFD distortions which is not found in PbTiO3_3 bulk

    Multilayer studies and applications in template bi-epitaxial DC SQUIDS

    Multilayer deposition for the creation of a well-defined grain boundary, based on different in-plane orientations of c-axis oriented thin YBa2O2Cu3O7-δ layers on a single substrate, has been performed on three different kinds of substrates: (1102)-oriented Al2O3 , (100) SrTiO3, and (100) MgO. The multilayers consist of combinations of SrTiO3, MgO, CeO2, and YBa2O2Cu3O7-δ. The YBa 2O2Cu3O7-δ top layers on (1102) Al2O3 and (100) SrTiO3 were polycrystalline. Josephson junctions and DC superconducting quantum interference devices (SQUIDs) have been structured in the layers on MgO. Shapiro steps were observed. The Jcρn-product of the junctions at 4.2 K is on the order of 1 mV. The critical current decreases in good approximation linearly with increasing temperature, whereas the normal state resistance is nearly temperature independent. Voltage modulation was observed at temperatures up to 77 K

    Detecting time-fragmented cache attacks against AES using Performance Monitoring Counters

    Cache timing attacks use shared caches in multi-core processors as side channels to extract information from victim processes. These attacks are particularly dangerous in cloud infrastructures, in which the deployed countermeasures cause collateral effects in terms of performance loss and increase in energy consumption. We propose to monitor the victim process using an independent monitoring (detector) process, that continuously measures selected Performance Monitoring Counters (PMC) to detect the presence of an attack. Ad-hoc countermeasures can be applied only when such a risky situation arises. In our case, the victim process is the AES encryption algorithm and the attack is performed by means of random encryption requests. We demonstrate that PMCs are a feasible tool to detect the attack and that sampling PMCs at high frequencies is worse than sampling at lower frequencies in terms of detection capabilities, particularly when the attack is fragmented in time to try to be hidden from detection

    Mifepristone increases mRNA translation rate, triggers the unfolded protein response, increases autophagic flux, and kills ovarian cancer cells in combination with proteasome or lysosome inhibitors

    The synthetic steroid mifepristone blocks the growth of ovarian cancer cells, yet the mechanism driving such effect is not entirely understood. Unbiased genomic and proteomic screenings using ovarian cancer cell lines of different genetic backgrounds and sensitivities to platinum led to the identification of two key genes upregulated by mifepristone and involved in the unfolded protein response (UPR): the master chaperone of the endoplasmic reticulum (ER), glucose regulated protein (GRP) of 78 kDa, and the CCAAT/enhancer binding protein homologous transcription factor (CHOP). GRP78 and CHOP were upregulated by mifepristone in ovarian cancer cells regardless of p53 status and platinum sensitivity. Further studies revealed that the three UPR-associated pathways, PERK, IRE1α, and ATF6, were activated by mifepristone. Also, the synthetic steroid acutely increased mRNA translation rate, which, if prevented, abrogated the splicing of XBP1 mRNA, a non-translatable readout of IRE1α activation. Moreover, mifepristone increased LC3-II levels due to increased autophagic flux. When the autophagic–lysosomal pathway was inhibited with chloroquine, mifepristone was lethal to the cells. Lastly, doses of proteasome inhibitors that are inadequate to block the activity of the proteasomes, caused cell death when combined with mifepristone; this phenotype was accompanied by accumulation of poly-ubiquitinated proteins denoting proteasome inhibition. The stimulation by mifepristone of ER stress and autophagic flux offers a therapeutic opportunity for utilizing this compound to sensitize ovarian cancer cells to proteasome or lysosome inhibitors.Fil: Zhang, Lei.     Molecular and Cellular Biology Animations: Development and Impact on Student Learning

    Educators often struggle when teaching cellular and molecular processes because typically they have only two-dimensional tools to teach something that plays out in four dimensions. Learning research has demonstrated that visualizing processes in three dimensions aids learning, and animations are effective visualization tools for novice learners and aid with long-term memory retention. The World Wide Web Instructional Committee at North Dakota State University has used these research results as an inspiration to develop a suite of high-quality animations of molecular and cellular processes. Currently, these animations represent transcription, translation, bacterial gene expression, messenger RNA (mRNA) processing, mRNA splicing, protein transport into an organelle, the electron transport chain, and the use of a biological gradient to drive adenosine triphosphate synthesis. These animations are integrated with an educational module that consists of First Look and Advanced Look components that feature captioned stills from the animation representing the key steps in the processes at varying levels of complexity. These animation-based educational modules are available via the World Wide Web at http://vcell.ndsu.edu/animations. An in-class research experiment demonstrated that student retention of content material was significantly better when students received a lecture coupled with the animations and then used the animation as an individual study activity

    Near‐millimeter‐wave response of high Tc ramp‐type Josephson junctions

    We have studied the response of a YBCO/PBCO/YBCO ramp‐type junction to coherent radiation at 176 and 270 GHz. The I‐V characteristic of the junction closely resembles the prediction of the RSJ model. The I cR n product of the junction is 0.25 mV at 5 K. The millimeter‐wave radiation is coupled to the junction via a quasioptical structure that focuses the radiation onto the junction through a yttrium‐stabilized ZrO2 substrate. At 176 GHz, we have observed as many as six Shapiro steps at the maximum power level of our Gunn oscillator‐pumped frequency doubler. Shapiro steps are still clearly seen up to 65 K. The amplitudes of the zeroth, first, and second Shapiro steps, as functions of the square root of the radiation power, agree remarkably well with a Bessel function fit, indicating the junction is voltage‐biased at the radiation frequency (rf). At 270 GHz, due to a combination of the heavy rf loss in the ZrO2 substrate and the lack of radiation power, we have observed only the first Shapiro step

    Genetical Genomics Reveals Large Scale Genotype-By-Environment Interactions in Arabidopsis thaliana

    One of the major goals of quantitative genetics is to unravel the complex interactions between molecular genetic factors and the environment. The effects of these genotype-by-environment interactions also affect and cause variation in gene expression. The regulatory loci responsible for this variation can be found by genetical genomics that involves the mapping of quantitative trait loci (QTLs) for gene expression traits also called expression-QTL (eQTLs). Most genetical genomics experiments published so far, are performed in a single environment and hence do not allow investigation of the role of genotype-by-environment interactions. Furthermore, most studies have been done in a steady state environment leading to acclimated expression patterns. However a response to the environment or change therein can be highly plastic and possibly lead to more and larger differences between genotypes. Here we present a genetical genomics study on 120 Arabidopsis thaliana, Landsberg erecta × Cape Verde Islands, recombinant inbred lines (RILs) in active response to the environment by treating them with 3 h of shade. The results of this experiment are compared to a previous study on seedlings of the same RILs from a steady state environment. The combination of two highly different conditions but exactly the same RILs with a fixed genetic variation showed the large role of genotype-by-environment interactions on gene expression levels. We found environment-dependent hotspots of transcript regulation. The major hotspot was confirmed by the expression profile of a near isogenic line. Our combined analysis leads us to propose CSN5A, a COP9 signalosome component, as a candidate regulator for the gene expression response to shade

    The family of hsp70 (70 kilodalton heat shock protein) molecular chaperones plays an essential and diverse role in cellular physiology, Hsp70 proteins appear to elicit their effects by interacting with polypeptides that present domains which exhibit non-native conformations at distinct stages during their life in the cell. In this paper we review work pertaining to the functions of hsp70 proteins in chaperoning mitochondrial protein biogenesis. Hsp70 proteins function in protein synthesis, protein translocation across mitochondrial membranes, protein folding and finally the delivery of misfolded proteins to proteolytic enzymes in the mitochondrial matrix

    Mind the implementation gap?:Police reform and local policing in the Netherlands and Scotland

    In 2013 the governments of the Netherlands and Scotland established national police forces, replacing a tradition of largely autonomous regional police organizations. In both jurisdictions, these radical reforms have raised concerns about the consequences of these national police structures for local policing and for relationships with local communities and local government. Drawing on documentary sources and interview material from each jurisdiction and informed by insights from the policy implementation literature, the key question addressed in this article is how has the legislation that created the new national police forces been put into effect at a local level? Focusing on the impact on the governance, organization and delivery of local policing, the article reveals how the implementation in both jurisdictions involves interpretation and discretion by multiple actors so that gaps are emerging between the national ‘policy promises’ set out in the legislation and the ‘policy products’ experienced in local contexts
