Toxicity effects of ethanol extract of Simarouba versicolor on reproductive parameters in female Wistar rats
Simarouba versicolor is popularly known as pau-paraiba. It belongs to the Simaroubaceae family and is found in the Northeast of São Paulo and in some parts of the states of Pará and Mato Grosso do Sul. It is known that this and other species from this genus have several activities: insecticides, antiinflammatory, antitumor, anthelmintic, among others. The effects of ethanolic extract of the bark of S. versicolor (Sv-EtOH) on the reproductive system of rats were investigated; also the influence of estrogenic and/or antiestrogenic activity, estrous cycle, pregnancy, lactation and offspring development was observed. In the experimental protocols with ethanolic extract of S. versicolor, the doses were 10, 20 and 40 mg/kg. To evaluate estrogenic and anti-estrogenic activity, uterotrophic assay and histopathology of the uterus were performed. In the estrous cycle tests, animals were treated daily, for 30 days, to count number of cycles and record the biochemical profile. During pre-implantation and post-implantation, the effect of the extract on pregnancy, lactation and development of the progeny was observed. Uterotrophic assay exhibited anti-estrogenic activity at extract doses of 10, 20 and 40 mg/kg. The animals exposed to all doses had no changes in their estrous cycle, pregnancy, lactation and biochemical profile. All results were within normal range; however, creatinine concentration increased significantly at all doses as compared to the control. The ethanol extract of S. versicolor presented antiestrogen activity, low systemic toxicity and had no reproductive toxicity at investigated doses.Key words: Simarouba versicolor, reproduction, rats
Covalent functionalization of multi-walled carbon nanotubes with a gadolinium chelate for efficient T1-weighted magnetic resonance imaging
Given the promise of carbon nanotubes (CNTs) for photothermal therapy, drug delivery, tissue engineering, and gene therapy, there is a need for non-invasive imaging methods to monitor CNT distribution and fate in the body. In this study, non-ionizing whole-body high field magnetic resonance imaging (MRI) is used to follow the distribution of water-dispersible non-toxic functionalized CNTs administrated intravenously to mice. Oxidized CNTs are endowed with positive MRI contrast properties by covalent functionalization with the chelating ligand diethylenetriaminepentaacetic dianhydride (DTPA), followed by chelation to Gd. The structural and magnetic properties, MR relaxivities, cellular uptake, and application for MRI cell imaging of Gd-CNTs in comparison to the precursor oxidized CNTs are evaluated. Despite the intrinsic T contrast of oxidized CNTs internalized in macrophages, the anchoring of paramagnetic gadolinium onto the nanotube sidewall allows efficient T contrast and MR signal enhancement, which is preserved after CNT internalization by cells. Hence, due to their high dispersibility, Gd-CNTs have the potential to produce positive contrast in vivo following injection into the bloodstream. The uptake of Gd-CNTs in the liver and spleen is assessed using MRI, while rapid renal clearance of extracellular Gd-CNTs is observed, confirming the evidences of other studies using different imaging modalities
HIV Aspartic Peptidase Inhibitors Modulate Surface Molecules and Enzyme Activities Involved with Physiopathological Events in Fonsecaea pedrosoi
Fonsecaea pedrosoi is the main etiological agent of chromoblastomycosis, a recalcitrant disease that is extremely difficult to treat. Therefore, new chemotherapeutics to combat this fungal infection are urgently needed. Although aspartic peptidase inhibitors (PIs) currently used in the treatment of human immunodeficiency virus (HIV) have shown anti-F. pedrosoi activity their exact mechanisms of action have not been elucidated. In the present study, we have investigated the effects of four HIV-PIs on crucial virulence attributes expressed by F. pedrosoi conidial cells, including surface molecules and secreted enzymes, both of which are directly involved in the disease development. In all the experiments, conidia were treated with indinavir, nelfinavir, ritonavir and saquinavir (100 μM) for 24 h, and then fungal cells were used to evaluate the effects of HIV-PIs on different virulence attributes expressed by F. pedrosoi. In comparison to untreated controls, exposure of F. pedrosoi cells to HIV-PIs caused (i) reduction on the conidial granularity; (ii) irreversible surface ultrastructural alterations, such as shedding of electron dense and amorphous material from the cell wall, undulations/invaginations of the plasma membrane with and withdrawal of this membrane from the cell wall; (iii) a decrease in both mannose-rich glycoconjugates and melanin molecules and an increase in glucosylceramides on the conidial surface; (iv) inhibition of ergosterol and lanosterol production; (v) reduction in the secretion of aspartic peptidase, esterase and phospholipase; (vi) significant reduction in the viability of non-pigmented conidia compared to pigmented ones. In summary, HIV-PIs are efficient drugs with an ability to block crucial biological processes of F. pedrosoi and can be seriously considered as potential compounds for the development of new chromoblastomycosis chemotherapeutics
Hadroproduction of the Chi1 and Chi2 States of Charmonium in 800 GeV/c Proton-Silicon Interactions
The cross sections for the hadroproduction of the Chi1 and Chi2 states of
charmonium in proton-silicon collisions at sqrt{s}=38.8 GeV have been measured
in Fermilab fixed target Experiment 771. The Chi states were observed via their
radiative decay to J/psi+gamma, where the photon converted to e+e- in the
material of the spectrometer. The measured values for the Chi1 and Chi2 cross
sections for x_F>0 are 263+-69(stat)+-32(syst) and 498+-143(stat)+-67(syst) nb
per nucleon respectively. The resulting sigma(Chi1}/sigma(Chi2) ratio of
0.53+-0.20(stat)+-0.07(syst), although somewhat larger than most theoretical
expectations, can be accomodated by the latest theoretical estimates.Comment: 4 pages, 4 figure
Measurement of the p\bar{p}\sqrt{s}$ = 1.8 TeV
We update the measurement of the top production cross section using the CDF
detector at the Fermilab Tevatron. This measurement uses decays to
the final states +jets and +jets. We search for quarks from
decays via secondary-vertex identification or the identification of
semileptonic decays of the and cascade quarks. The background to the
production is determined primarily through a Monte Carlo simulation.
However, we calibrate the simulation and evaluate its uncertainty using several
independent data samples. For a top mass of 175 , we measure
pb and pb using
the secondary vertex and the lepton tagging algorithms, respectively. Finally,
we combine these results with those from other decay channels and

Search for Narrow Diphoton Resonances and for gamma-gamma+W/Z Signatures in p\bar p Collisions at sqrt(s)=1.8 TeV
We present results of searches for diphoton resonances produced both
inclusively and also in association with a vector boson (W or Z) using 100
pb^{-1} of p\bar p collisions using the CDF detector. We set upper limits on
the product of cross section times branching ratio for both p\bar
p\to\gamma\gamma + X and p\bar p\to\gamma\gamma + W/Z. Comparing the inclusive
production to the expectations from heavy sgoldstinos we derive limits on the
supersymmetry-breaking scale sqrt{F} in the TeV range, depending on the
sgoldstino mass and the choice of other parameters. Also, using a NLO
prediction for the associated production of a Higgs boson with a W or Z boson,
we set an upper limit on the branching ratio for H\to\gamma\gamma. Finally, we
set a lower limit on the mass of a `bosophilic' Higgs boson (e.g. one which
couples only to \gamma, W, and Z$ bosons with standard model couplings) of 82
GeV/c^2 at 95% confidence level.Comment: 30 pages, 11 figure
Search for Chargino-Neutralino Associated Production at the Fermilab Tevatron Collider
We have searched in collisions at = 1.8 TeV for events
with three charged leptons and missing transverse energy. In the Minimal
Supersymmetric Standard Model, we expect trilepton events from
chargino-neutralino (\chione \chitwo) pair production, with subsequent decay
into leptons. We observe no candidate , ,
or events in 106 pb integrated
luminosity. We present limits on the sum of the branching ratios times cross
section for the four channels: \sigma_{\chione\chitwo}\cdot
BR(\chione\chitwo\to 3\ell+X) 81.5 \mgev\sp and
M_\chitwo > 82.2 \mgev\sp for , ~\mgev\sp and
M_\squark= M_\gluino.Comment: 9 pages and 3 figure
Search for a Fourth-Generation Quark More Massive than the Z0 Boson in ppbar Collisions at sqrt(s) = 1.8 TeV
We present the results of a search for pair production of a fourth-generation
charge -1/3 quark (b') in sqrt(s)=1.8 TeV ppbar collisions using 88 pb^(-1) of
data obtained with the Collider Detector at Fermilab. We assume that both
quarks decay via the flavor-changing neutral current process b' -> bZ and that
the b' mass is greater than m_Z + m_b. We studied the decay mode b'b'bar -> ZZ
b bbar where one Z0 decays into e^+e^- or mu^+ mu^- and the other decays
hadronically, giving a signature of two leptons plus jets. An upper limit on
the cross section of ppbar -> b'b'bar times [BR (b' -> bZ)]^2 is established as
a function of the b' mass. We exclude at 95% confidence level a b' quark with
mass between 100 and 199 GeV/c^2 for BR(b' -> bZ) = 100%.Comment: 12 pages, 2 figures, submitted to Phys. Rev. Letters on 9/12/9
Measurement of the Helicity of W Bosons in Top Quark Decays
We use the transverse momentum spectrum of leptons in the decay chain t-->bW
with W-->l nu to measure the helicity of the W bosons in the top quark rest
frame. Our measurement uses a ttbar sample isolated in 106 +/- 4 inverse
picobarns of data collected in ppbar collisions at sqrt(s)=1.8 TeV with the CDF
detector at the Fermilab Tevatron. Assuming a standard V--A weak decay, we find
that the fraction of W's with zero helicity in the top rest frame is F_0 = 0.91
+/- 0.37 (stat) +/- 0.13 (syst), consistent with the standard model prediction
of F_0=0.70 for a top mass of 175 GeV/c**2.Comment: Submitted to PRL. 8 pages, 2 figure
