Impact of genotype, age of tree and environmental temperature on androgenesis induction of Aesculus hippocastanum L.
Influence of ten different genotypes, age of trees and environmental temperature on induction of androgenesis and apperance albino horse chestnut embryos were studied. Efficiency of in vitro androgenesis via anther and microspore culture had been investigated. Microspores and anthers were used from the same closed flower bud. Androgenic response of different genotypes was measured and compared. Anther induction rates were from 5 to 37.6%, depending on genotype. The number ofembryos per isolated anther varied between 0.5 to 5.0 embryos in anther culture, while in microspore culture varied between 3.0 to 27 embryos, depending on genotype. A microspore culture was 5 - 6 times efficient than anther culture for same genotype. Age of the trees had no influence on androgenesis induction. Temperature of about 4 - 5°C was optimal for androgenic embryo induction. Albino horse chestnut embryos phenomenon depended on genotype. The number of albino appearing in anther washigher than in microspore culture. The same correlation of appearing albino was observed between short and long day. Flow cytogenetic analysis of androgenic embryos originating from anther and microspore culture was done after a first generation of regenerants. All androgenic embryos, the first generation from microspore culture were haploid, while 50% of the regenerants originating from anther culture were haploid, and the other half diploid
Contrasting and explaining purposeful and legitimizing uses of performance information: a mayorâs perspective
This study looks at purposeful and legitimizing types of performance information use in local governments. Drawing on a survey of Austrian mayors who are at the politico-administrative apex of local government, the paper shows that purposeful and legitimizing uses of performance information coexist, but they appear to be negatively associated. In exploring the contextual and organizational conditions under which legitimizing uses prevail over purposeful ones, the analysis shows that oversight (coercive) and political (normative) pressures, hierarchical culture, and low-performance information availability foster the dominance of the legitimizing use type over the purposeful one
Effect of activated charcoal, abscisic acid and polyethylene glycol on maturation, germination and conversion of Aesculus hippocastanum androgenic embryos
The influence of activated charcoal (AC), abscisic acid (ABA) and polyethylene glycol (PEG) on the maturation and conversion of horse chestnut (Aesculus hippocastanum L.) androgenic embryos weretested. Androgenic embryos originating from microspores and anther culture were maturated over 90 days. Androgenic embryos on media containing PEG (50 g l-1), in combination with AC (1 g l-1) showed arapid development of embryos in the cotyledonary stage and lowered percentage of abnormal structures. The best results of androgenic microspore embryo germination were observed on media supplemented with AC alone (99%) and in combination with PEG (100%). Also, the greatest number of androgenic microspore plants (18%) and androgenic anther plants (12%) were formed on media enriched with 1 % AC. Lowest germination percentages of 37 and 39% in microspore culture and 33 and38% in anther culture were obtained on maturation media with ABA 20 mg l-1 alone and in combination with AC 1g l-1. Flow cytometric analysis showed that most of the androgenic embryos were haploid, corresponding to their microspore origin, while half of these became diploid after maturation for 90 days. All regenerants originating from microspore culture were haploid immediately after germination, but only 10% embryos re ained haploidity after 3 years subculturing, while 10.5% were diploid, 73.5% tetraploid and 6% octaploid on hormone-free medium. Unlike those from anther culture, after 3 years of subculturing on hormone-free medium, there were no haploid regenerant from anther culture, while8.5% were diploid, 81% tetraploid and 10.5% octaploid

How do local governments respond to shocks? The role of anticipatory capacities and financial vulnerability
The increased uncertainty, volatility and complexity under which governments operate have put great emphasis on how governments respond to shocks and crises. Extant research has mostly studied the causes of and reactions to recent crises, some focusing on local government, with a relative paucity of research on how accounting is implicated in crisis responses at the organizational level. More research is needed to understand how accounting informs responses to crises and shocks influencing perceptions of organizational financial vulnerability and, more generally, affecting organizational capacity to anticipate shocks.
This paper builds on previous research on governmental financial resilience to understand how local governmentsâ responses to shocks are shaped by organizational perceptions of financial conditions and the presence of anticipatory capacities. The concept of financial resilience has been shown to uncover internal capacities that act as shaping forces during crises, contributing to and informing âbouncing backâ or âbouncing forwardâ response strategies. Based on a survey of over 600 local governments in Germany, Italy, and the UK, this paper looks at the role that anticipatory capacities, and associated vulnerabilities, play in determining organizational response strategies (bouncing back vs. bouncing forward) at times of crisis

Local government financial resilience: Germany, Italy and UK compared: with a practical tool-kit for local governments
The increased uncertainty, volatility and complexity under which local governments operate, coupled with recent shocks, starting with the 2008 financial crisis, but also including Brexit and the increasing influx of refugee migrants, have put great emphasis on governmental financial resilience, i.e., how governments cope with shocks affecting their finances.
This report increases our understanding of local government financial resilience by presenting the results of a survey of local governments across Italy, the UK and Germany. Analysing the combination of internal and external resilience dimensions against the background of recent crises and across countries, the project not only sheds light on different performance enabling capacities but also helps to achieve a greater understanding of how local governments maintain or build resilience.
The capacities, which also assist in keeping particular vulnerabilities in control, are specified in practical guidance linked to this report, a tool-kit for local governments to assess and develop their capacities in order to be better equipped to cope with disruptive events
EPS-SJ Exopolisaccharide Produced by the Strain Lactobacillus paracasei subsp paracasei BGSJ2-8 Is Involved in Adhesion to Epithelial Intestinal Cells and Decrease on E-coil Association to Caco-2 Cells
The aim of this study was to determine the role of an exopolysaccharide produced by natural dairy isolate Lactobacillus paracasel subsp. paracasel BGSJ2-8, in the adhesion to intestinal epithelial cells and a decrease in Escherichia coil's association with Caco-2 cells. Annotation of the BGSJ2-8 genome showed the presence of a gene cluster, epsSJ, which encodes the biosynthesis of the strain-specific exopolysaccharide EPS-SJ, detected as two fractions (P1 and P2) by size exclusion chromatography (SEC) coupled with multi angle laser light scattering (MALLS) detection. SEC MALLS analysis revealed that an EPS-SJ(-) mutant (EPS7, obtained by insertion mutagenesis of the glps_2198 gene encoding primary glycosyltransferase) does not produce the P2 fraction of EPS-SJ. Transmission electron microscopy showed that EPS7 mutant has a thinner cell wall compared to the EPS-SJ(+) strain BGSJ2-83 (a plasmid free derivative of BGSJ2-8). Interestingly, strain BGSJ2-83 showed higher adhesion to Caco-2 epithelial intestinal cell line than the EPS7 mutant. Accordingly, BGSJ2-83 effectively reduced E. coil ATCC25922's association with Caco-2 cells, while EPS7 did not show statistically significant differences. In addition, the effect of EPS-SJ on the proliferation of lymphocytes in gastrointestinal associated lymphoid tissue (GALT) was tested and the results showed that the reduction of GALT lymphocyte proliferation was higher by BGSJ2-83 than by the mutant. To the best of our knowledge this is the first report indicating that the presence of EPS (EPS-SJ) on the surface of lactobacilli can improve communication between bacteria and intestinal epithelium, implying its possible role in gut colonization
Governmental financial resilience under austerity in Austria, England and Italy: how do local governments cope with financial shocks?
The recent economic and fiscal crises provide an opportunity for learning lessons of general and practical relevance into how governments face shocks affecting their financial conditions. This article draws on the resilience concept to investigate the organizational capacities that are deployed and/or built by local governments (LGs) to respond to such shocks, looking at their combinations and interactions with environmental conditions. The paper presents the results of a multiple-case analysis of 12 European LGs across Austria, Italy and England. The analysis allows to highlight and operationalize different patterns of financial resilience, i.e. self-regulation, constrained or reactive adaptation, contented or powerless fatalism, that are the result of the interaction and development over time of different internal and external dimensions
Binding of SGTA to Rpn13 selectively modulates protein quality control
YesRpn13 is an intrinsic ubiquitin receptor of the 26S proteasome regulatory subunit that facilitates substrate capture prior to degradation. Here we show that the C-terminal region of Rpn13 binds to the tetratricopeptide repeat (TPR) domain of SGTA, a cytosolic factor implicated in the quality control of mislocalised membrane proteins (MLPs). The overexpression of SGTA results in a substantial increase in steady-state MLP levels, consistent with an effect on proteasomal degradation. However, this effect is strongly dependent upon the interaction of SGTA with the proteasomal component Rpn13. Hence, overexpression of the SGTA-binding region of Rpn13 or point mutations within the SGTA TPR domain both inhibit SGTA binding to the proteasome and substantially reduce MLP levels. These findings suggest that SGTA can regulate the access of MLPs to the proteolytic core of the proteasome, implying that a protein quality control cycle that involves SGTA and the BAG6 complex can operate at the 19S regulatory particle. Governance tools for board members : adapting strategy maps and balanced scorecards for directorial action
The accountability of members of the board of directors of publicly traded companies has increased over years. Corresponding to these developments, there has been an inadequate advancement of tools and frameworks to help directorial functioning. This paper provides an argument for design of the Balanced Scorecard and Strategy Maps made available to the directors as a means of influencing, monitoring, controlling and assisting managerial action. This paper examines how the Balanced Scorecard and Strategy Maps could be modified and used for this purpose. The paper suggests incorporating Balanced Scorecards in the Internal Process perspective, âinternalâ implying here not just âinternal to the firmâ, but also âinternal to the inter-organizational systemâ. We recommend that other such factors be introduced separately under a new âperspectiveâ depending upon what the board wants to emphasize without creating any unwieldy proliferation of measures. Tracking the Strategy Map over time by the board of directors is a way for the board to take responsibility for the firmâs performance. The paper makes a distinction between action variables and monitoring variables. Monitoring variables are further divided on the basis of two considerations: a) whether results have been met or not and b) whether causative factors have met the expected levels of performance or not. Based on directorial responsibilities and accountability, we take another look at how the variables could be specified more completely and accurately with directorial recommendations for executives
