    Motown [RE]Vival: The Use of Culinary and Urban Farming to Revitalize the Dying City of Detroit

    Motown: Detroit, Michigan, once known as the automotive capital of the world, received its claim to fame through the birth of the assembly line by Henry Ford in 1905. His innovation steered the big boom of the automotive industry in the early 20th century, and later the mass movement to the city. Sadly, that all came crashing down with the fall of the industrial period. The loss of nearly all its auto plants, postwar white flight and suburbanization caused the city to suffer economic turmoil. By 1980 the population had decreased 35% since its peak in 1950. Today? The city endures the loss of over 60% of its population. With lack of people and a scarcity of business, Detroit is also fighting food desertion with 69% of households being food insecure. The once bustling factories and auto plants now sit dormant with nearly 900 structures unused. What if we could change that and restore Motown’s former glory? Industrialized cities can reclaim their purpose through innovation that engages the public and provides a new purposeful identity. My thesis aims to breathe life back into the dying city of Detroit, through adaptive reuse strategies accompanied by a new program, food. Turning a portion of Detroit into a culinary hub to include demo kitchens, restaurants, and small business supports. While also incorporating an urban farming sector to educate, grow, and sell produce straight from their metaphorical backyard. Not only will this bring a fresh focus, but it will also build revenue and encourage population growth. My proposal seeks to educate the locals on farm to table, food research learning and distribution, by incorporating a culinary research institution. This new institute will touch on the historical significance of the surrounding industrial buildings while informing the possibilities of a bright future as a culinary core. The education center will offer a variety of learning through field study and stress the importance of food technology. The questions are What do the current and future people of Detroit need to maintain a healthy social, economic, and communal relationship? How can my proposed design aid in the fight for urban regeneration and food security? Can these tactics work as an exemplary model for other fallen industrialized cities to follow. Many other architects have proposed ideas. Specifically, they have focused on housing, office, and multi-use. None have introduced the idea of a cuisine city center that could aid in the current food desertion epidemic. With 19 neighborhoods without adequate food access, my plan will bring an opportunity to revitalize these communities. And in whole, pull the dying city of Detroit out of neglect

    The purpose of this study is to analyze the determination of the rate structure of the electric utilities. It consists of three chapters. The first chapter deals with the determination of the rate basis of electric utilities. The calculation of the rate base is explained, and also the problems of the price level changes. The second chapter shows the calculation of the rate of return and the measures used for testing the fair rate of return. The third chapter is an analysis showing the effect of the different cost and demand factors in determining the rate structure of electric utilities

    Female-limited X chromosome evolution reveals that lifespan is mainly modulated by interlocus rather than intralocus sexual conflict

    Sexual dimorphism in somatic investment may be shaped by two distinct forms of sexual conflict; under intralocus sexual conflict (IASC), males and females have different optimal levels of somatic investment but are constrained from reaching their respective optima by their shared genome, while under interlocus sexual conflict (IRSC), males and females have different optimal sexual strategies, which could have direct or indirect effects on levels of somatic investment. We investigated effects of IASC and IRSC on two aspects of somatic investment, immune defence strategies and longevity, using previously established female-limited experimental evolution lines in Drosophila melanogaster. We found little evidence for any effect of either type of sexual conflict on investment in the immune defence resistance or tolerance. Nor did we find convincing evidence that longevity is subject to IASC in this species. However, we did find evidence that increased female control over mating rate had important and opposite effects on longevity between the sexes. Specifically, females that had adapted to high levels of female control over mating had a longer lifespan when kept in mixed-sex groups, while males had shorter longevity, perhaps due to increased investment in post-copulatory sexual selection. These novel results show that female control over mating rates may have important and unexpected effects on patterns of somatic investment

    Alternative splicing of TGF-betas and their high-affinity receptors TβRI, TβRII and TβRIII (betaglycan) reveal new variants in human prostatic cells

    <p>Abstract</p> <p>Background</p> <p>The transforming growth factors (TGF)-β, TGF-β1, TGF-β2 and TGF-β3, and their receptors [TβRI, TβRII, TβRIII (betaglycan)] elicit pleiotropic functions in the prostate. Although expression of the ligands and receptors have been investigated, the splice variants have never been analyzed. We therefore have analyzed all ligands, the receptors and the splice variants TβRIB, TβRIIB and TGF-β2B in human prostatic cells.</p> <p>Results</p> <p>Interestingly, a novel human receptor transcript TβRIIC was identified, encoding additional 36 amino acids in the extracellular domain, that is expressed in the prostatic cancer cells PC-3, stromal hPCPs, and other human tissues. Furthermore, the receptor variant TβRIB with four additional amino acids was identified also in human. Expression of the variant TβRIIB was found in all prostate cell lines studied with a preferential localization in epithelial cells in some human prostatic glands. Similarly, we observed localization of TβRIIC and TGF-β2B mainly in the epithelial cells with a preferential localization of TGF-β2B in the apical cell compartment. Whereas in the androgen-independent hPCPs and PC-3 cells all TGF-β ligands and receptors are expressed, the androgen-dependent LNCaP cells failed to express all ligands. Additionally, stimulation of PC-3 cells with TGF-β2 resulted in a significant and strong increase in secretion of plasminogen activator inhibitor-1 (PAI-1) with a major participation of TβRII.</p> <p>Conclusion</p> <p>In general, expression of the splice variants was more heterogeneous in contrast to the well-known isoforms. The identification of the splice variants TβRIB and the novel isoform TβRIIC in man clearly contributes to the growing complexity of the TGF-β family.</p

    Populations of Radial Glial Cells Respond Differently to Reelin and Neuregulin1 in a Ferret Model of Cortical Dysplasia

    Radial glial cells play an essential role during corticogenesis through their function as neural precursors and guides of neuronal migration. Both reelin and neuregulin1 (NRG1) maintain the radial glial scaffold; they also induce expression of Brain Lipid Binding Protein (BLBP), a well known marker of radial glia. Although radial glia in normal ferrets express both vimentin and BLBP, this coexpression diverges at P3; vimentin is expressed in the radial glial processes, while BLBP appears in cells detached from the ventricular zone. Our lab developed a model of cortical dysplasia in the ferret, resulting in impaired migration of neurons into the cortical plate and disordered radial glia. This occurs after exposure to the antimitotic methylazoxymethanol (MAM) on the 24th day of development (E24). Ferrets treated with MAM on E24 result in an overall decrease of BLBP expression; radial glia that continue to express BLBP, however, show only mild disruption compared with the strongly disrupted vimentin expressing radial glia. When E24 MAM-treated organotypic slices are exposed to reelin or NRG1, the severely disrupted vimentin+ radial glial processes are repaired but the slightly disordered BLBP+ processes are not. The realignment of vimentin+ processes was linked with an increase of their BLBP expression. BLBP expressing radial glia are distinguished by being both less affected by MAM treatment and by attempts at repair. We further investigated the effects induced by reelin and found that signaling was mediated via VLDLR/Dab1/Pi3K activation while NRG1 signaling was mediated via erbB3/erbB4/Pi3K. We then tested whether radial glial repair correlated with improved neuronal migration. Repairing the radial glial scaffold is not sufficient to restore neuronal migration; although reelin improves migration of neurons toward the cortical plate signaling through ApoER2/Dab1/PI3K activation, NRG1 does not

    Diethylstilbestrol (DES), an estrogen agonist, increases prolactin (PRL) cells through transdifferentiation of follicle-stimulating hormone (FSH) and luteinizing hormone (LH) cells to PRL cells as well as proliferation of PRL cells in adult male mouse pituitary. Since hyperacetylation of histone H3 is implicated in the regulation of activation of various genes, we examined the effect of DES on the state of histone H3 acetylation. DES significantly reduced the immunohistochemical signal for acetylated histone H3 at lysine 9 (H3K9ac) in PRL, LH and FSH cells, but not for H3K18ac or H3K23ac. DES-treated mice were injected intraperitoneally with HDAC inhibitors (HDACi), sodium phenylbutyrate (NaPB) or valproic acid (VPA), to mimic the acetylation level of histone H3. As expected, HDACi treatment restored the level of H3K9ac expression in these cells, and also inhibited DES-induced increase in PRL cells. Furthermore, NaPB and VPA also abrogated the effects of DES on the population density of both LH and FSH cells. Similarly, the numbers of proliferating and apoptotic cells in the pituitary in NaPB- or VPA-treated mice were comparable to those of the control mice.     The Intersectionality of Race, Leadership, and Mental Health Industries: What Contributes to the Underrepresentation?

    The research study explores the experiences of African American leaders who work within a specific mental health agency. The study focuses on the factors that contribute to their attempt reaching, retaining, and being recruited for hierarchical positions while considering the intersectionality of race, leadership, and mental health industries. Fourteen participants agreed to participate in the study, which consisted of a one-on-one meeting in private settings. The qualitative research design for the study was phenomenology, because it focused on the lived experiences of each participant as it related to the overall research. Underrepresentation, intersectionality, cultural stigma, and race-based oppression were each contributing factors found most common throughout the study. The findings for the research suggest each of the aforementioned factors directly contribute to the experiences of African American leaders seeking, retaining, and being recruited for hierarchical positions within mental health industries
