    Characteristics of local pulmonary response following intranasal application of Aspergillus fumigatus conidia

    In this study, histopathology of local pulmonary response following intranasal inoculation of different doses of Aspergillus fumigatus conidia in laboratory rats was evaluated. Development of response was evaluated on days seven and twenty one, following conidia inoculation by cell infiltration and by presence of A. fumigatus conidia in homogenates of lung tissue. Total and differential peripheral blood leukocyte counts and state of leukocyte adhesion/aggregation were monitored to estimate the presence of systemic response in infected individuals. Mycological examination revealed the presence of conidia in lung tissue homogenates of infected animals, with high number of non-germinating spores on day twenty one, following the inoculation of lower conidia dose. Histopathological examination revealed the presence of lymphocytes perivascularly and in vascular lumen in the lungs. Presented data demonstrate pulmonary immune response following A. fumigatus conidia administration.Aspergillus fumigatus je oportunistička gljiva koja najčeŔće dovodi do pulmonarnih aspergiloza kod imunokompromitovanih organizama. U ovom radu su prikazani rezultati istraživanja izazivanja infekcije aspergilusom kod pacova. U cilju dobijanja podataka o prisustvu i karakteristikama lokalnog inflamatornog odgovora u plućima određen je diferencijalni sastav leukocita periferne krvi i testirana je njihova aktivnost, izvrÅ”eni su histopatoloÅ”ki pregled pluća i provera prisustva spora A. fumigatus u homogenatima pluća. NaÅ”i podaci su pokazali prisustvo lokalnog odgovora u plućima posle 7 dana dok podaci dobijeni posle 21 dan ne pokazuju značajne razlike u odnosu na kontrolnu grupu. Povećani broj neklijajućih spora u homogenatu pluća sedmog dana nakon inokulacije može da se objasni prisustvom limfocitnog infiltrata u plućima i aktivnoŔću limfocita in situ. Povećan broj klijajućih spora 21. dana nakon inokulacije može da bude posledica kapaciteta malog broja preostalih spora da klijaju.Projekat ministarstva br. 14303

    Influence of cadmium chloride on the frequency of micronuclei in da and AO rats

    Heavy metal cadmium (Cd), a well-known environmental hazard, exerts a number of toxic and genotoxic effects. Experimental animals, Albino Oxford (AO) and Dark August (DA) rat strains, were treated intraperitoneally with three different concentrations of cadmium chloride (CdCl2): 0.5, 1, and 2 mg CdCl2 per kg of body weight, while the control animals received equal volume of sterile phosphate buffered saline. In this investigation individuals of both sexes aged 3, 6 and 12 month were used. Frequency of micronuclei formation was evaluated in polychromatic erythrocytes (PCEs), 24h hours after treatment. The results showed that that cadmium-cloride (CdCl2) exibits the genotoxic effects causing an increase of the frequency of micronuclei depending on concentration, sex, age and strain

    Age structure of yellow-necked mouse (Apodemus flavicollis Melchior, 1834) in two samples obtained from live traps and owl pellets

    The age structure of yellow-necked mouse (Apodemus flavicollis Melchior 1834) has been analyzed in individuals obtained by two methods: Trapping with Sherman live traps and obtainment of skulls from long-eared owl (Asio otus Linnaeus, 1758) pellets (predator diet analysis). One hundred and forty-four mice were analyzed for the degree of wear of the surface of molar crowns, and an additional 74 measurements were performed on captive-born Apodemus flavicollis individuals. We used a refined model of comparison that included seven classes of mouse age, rather than four classes, as suggested by other authors.Uzrasna struktura Apodemus flavicollis (Melchior, 1834) analizirana je na uzorku pribavljenom primenom dve metode: izlovljavanjem Å ermanovim klopkama i prikupljanjem lobanja miÅ”eva iz gvalica sove Asio otus Linnaeus. Kod ukupno 144 jedinke studiran je stepen izrabljenosti molarnih krunica; izvrÅ”ena su i 74 dodatna merenja kod jedinki gajenih u laboratorijskim uslovima.     Toxigenic and pathogenic fungi in Norway rat (Rattus norvegicus Berk., 1769) from natural populations in semiagricultural habitats

    In this study, the presence of toxigenic and pathogenic fungi was detected in individuals of natural populations of Norway rats from semiagricultural habitats. The presence of fungi was noted in 19 out of 30 (63%) individuals examined. Six fungal species were isolated and identified, of which majority belonged to Hyphomycetes (Deuteromycotina) and Mucor racemosus from Zygomycotina. All of detected species are of public health importance and some of them might influence animals health.U ovom radu su prikazana istraživanja prisustva toksigenih i patogenih gljiva u jedinkama sivog pacova (Rattus norvegicus Berk., 1769) iz prirodnih populacija sa polupoljoprivrednih staniÅ”ta (Omoljica, Kovin, LeÅ”tane, Ovča). Detekcija prisustva gljiva u bioloÅ”kom materijalu (bris kože nosne Å”upljine i homogenat pluća) je vrÅ”ena na selektivnoj Sabouraud Maltose Agar (SMA) podlozi. Istraživanje je pokazalo prisustvo Å”est vrsta gljiva od kojih pet pripada klasi Hyphomyceta (Deuteromycotina), a Mucor racemosus klasi Zygomycotina. Vrste roda Aspergillus su detektovane u homogenatima pluća Å”est jedinki, a Penicillium kod pet jedinki. Paecilomyces varioti je izolovan iz tkiva Å”est jedinki sivog pacova. Brisevi kože nosne Å”upljine su dali pozitivne rezultate u 15 (50%) od 30 jedinki (četiri pozitivna za Aspergillus, četiri za Penicillium, Å”est za Paecilomyces varioti i jedna za Mucor racemosus). Međutim, u samo 9 slučajeva zabeleženo je poklapanje prisustva gljiva u koži nosne Å”upljine i pluća. Nalazi koji pokazuju prisustvo patogena, izazivača teÅ”kih gljivičnih oboljenja, ukazuju na sivog pacova kao na potencijalni rezervoar ovih agenasa. Dobijeni podaci predstavljaju, prema naÅ”em saznanju, prve podatke o prisustvu gljiva kod pacova iz prirodne sredine zabeležene kod nas.Projekat ministarstva br. 14303

    Sojne razlike u toksičnosti antagoniste vitamina K varfarina kod pacova

    Warfarin (3-(alpha-acetonylbenzy1)-4-hydroxy coumarin) is a vitamin K (VK) antagonist that inhibits vitamin K-dependent (VKD) processes, such as blood coagulation. It also exerts an influence on some non-VKD-related activities. In this study, the effect of sub-acute (30-day) oral warfarin (2 and 1 mg L-1) intake on hematological parameters was examined in two rat strains, Albino Oxford (AO) and Dark Agouti (DA), that differ in their sensitivity to certain chemicals. Greater susceptibility to the anticoagulant effect of 2 mg L-1 of warfarin was observed in AO rats and was associated with an increase in the relevant hematological parameters in this strain. Although both strains responded to 2 mg L-1 of warfarin with quantitative changes in the peripheral blood leukocytes, differential bone marrow and lung responses were observed. Strain-related differences in the pro-inflammatory activity of peripheral blood granulocytes and in mononuclear cell IFN-gamma production were observed. U ovom radu je ispitan efekat subakutnog (30 dana) oralnog unosa varfarina na hematoloÅ”ke parametre i aktivnost leukocita periferne krvi kod dva soja pacova Albino Oxford (AO) i Dark Agouti (DA) koji se raz- likuju u osetljivosti na iste hemijske agense. Kod jedinki AO soja zapažena je veća smrtnost nakon konzumiranja doze od 4 mg Lā€“1 kao i veća osetljivost na antikoagulantno dejstvo varfarina pri nižim dozama (2 mg Lā€“1) koje je praćeno povećanjem nekih hematoloÅ”kih parametara. Iako kod jedinki oba soja dolazi do povećanja broja neutrofilnih leukocita periferne krvi pri dozi od 2 mg Lā€“1, promene u osnovnim proinflamatornim aktivnostima ovih ćelija su zapažene samo kod jedinki DA soja. Promene u broju neutrofilnih leukocita u krvi DA jedinki su praćene povećanjem broja granulocitnih prekursora u koÅ”tanoj srži, dok prisustvo neutrofila u plućima AO jedinki ukazuje na razmenu ćelija između periferne krvi i plućnog intravaskularnog pula ćelija. Diferencijalne sojnoā€“zavisne promene u aktivnosti mononuklearnih ćelija periferne krvi su takođe zapažene. Razlike u efektu oralno unetog varfarina mogu da imaju implikacije za osobe na oralnoj varfarinskoj terapiji

    Rodenticide efficacy of sodium selenite baits in laboratory conditions

    We examined the acceptance and palatability of baits containing different contents of sodium selenite as a rodenticide, in Swiss mice under laboratory conditions. In a no-choice and choice feeding test, the animals were exposed to baits containing 0.1, 0.05, 0.025 and 0.0125% of sodium selenite. The total bait consumption by Swiss mice in the no-choice feeding test was highly negatively correlated, while total sodium selenite intake was medium-positively correlated to the sodium selenite content in the bait. In the same test, daily intakes significantly depended on the content of sodium selenite in the bait, while the exposure and associated interactions of contents of sodium selenite and exposure had no statistically significant impact. Baits with sodium selenite contents of 0.05 and 0.1% had the most lethal effects. The negative impact of the sodium selenite content on bait acceptance and palatability was confirmed in choice feeding tests. Baits containing 0.05 and 0.1% of sodium selenite displayed the biological potential to be used as a rodenticide. It is necessary to improve its insufficient acceptability and palatability by adding adequate additives to the bait. The results of this study should be verified in experiments with wild rodents
