    IGF-I receptor phosphorylation is impaired in cathepsin X-deficient prostate cancer cells

    The cysteine-type peptidase cathepsin X is highly upregulated in several cancers and presumably promotes tumor invasion through bypassing cellular senescence. Here, we present first evidence that the underlying mechanism may involve the regulation of the insulin-like growth factor (IGF) system, a well-known activator of proliferating tumor cells. Cathepsin X deficiency leads to a reduced phosphorylation of the IGF-I receptor in response to IGF-I stimulation. In addition, downstream signaling through focal adhesion kinase was also affected. Taken together, our results indicate that cathepsin X is able to assist in IGF signaling, which may be an important progress toward understanding cathepsin X-dependent tumorigenesis

    What fecal analyses reveal about Manniophyton fulvum consumption in LuiKotale bonobos (Pan paniscus): A medicinal plant revisited

    Observations of animals in the wild can result in the discovery of plants for human medicinal purposes. In this context, our closest relatives, the great apes, are of particular interest. The Euphorbiaceae Manniophyton fulvum possesses both phytochemical and biomechanical properties. Its use in the genus Pan (P. troglodytes; P. paniscus) is thought to be based on its mechanical properties promoting the egestion of intestinal parasites, but additional observations from different habitats where the behavior is performed may shed more light on its true purpose. To improve our understanding of what triggers this behavior, we investigated M. fulvum consumption in wild bonobos at LuiKotale, Democratic Republic of the Congo between December 2018 and July 2020. Specifically, we tested the hypothesis that M. fulvum ingestion is related to gastro-intestinal parasite expulsion. Of 649 focal follows of 37 individuals from two habituated communities, consumption of M. fulvum was observed on 111 days (N = 507), independent of seasons, environmental factors and the plant's availability. A total of 588 fecal samples were assessed for the presence/absence of gastro-intestinal parasites. We found strongyle eggs in 2.89% of samples and their presence was not associated with the ingestion of M. fulvum or environmental conditions. We discuss the importance of seasonality in the life cycle of strongyle species that may influence the pattern of M. fulvum consumption observed at LuiKotale. Our data open additional perspectives concerning behavioral parameters such as the existence of a cultural component when comparing ingestion behavior between communities

    Derivative based global sensitivity measures

    The method of derivative based global sensitivity measures (DGSM) has recently become popular among practitioners. It has a strong link with the Morris screening method and Sobol' sensitivity indices and has several advantages over them. DGSM are very easy to implement and evaluate numerically. The computational time required for numerical evaluation of DGSM is generally much lower than that for estimation of Sobol' sensitivity indices. This paper presents a survey of recent advances in DGSM concerning lower and upper bounds on the values of Sobol' total sensitivity indices S_itotS\_{i}^{tot}. Using these bounds it is possible in most cases to get a good practical estimation of the values of S_itotS\_{i}^{tot} . Several examples are used to illustrate an application of DGSM

    Salmonella Heidelberg isolates has become an emerging pathogen during the 80s in the United States (Martin et al., 1989). Approximately 60% of human cases reported to the CDC in 1995 were caused by only four serovars, including S. Enteridis (24,7%), S. Typhimurium (23,5%), S. Newport (6,2%) and S. Heidelberg (5,1%), (CDC, Salmonella surveillance) and were frequently isolated from chicken and pork (Sawari et al., 2001)

    Discovery of novel herpes simplexviruses in wild gorillas, bonobos, and chimpanzees supports zoonotic origin of HSV-2

    Viruses closely related to human pathogens can reveal the origins of human infectious diseases. Human herpes simplexvirus type 1 (HSV-1) and type 2 (HSV-2) are hypothesized to have arisen via host-virus codivergence and cross-species transmission. We report the discovery of novel herpes simplexviruses during a large-scale screening of fecal samples from wild gorillas, bonobos, and chimpanzees. Phylogenetic analysis indicates that, contrary to expectation, simplexviruses from these African apes are all more closely related to HSV-2 than to HSV-1. Molecular clock-based hypothesis testing suggests the divergence between HSV-1 and the African great ape simplexviruses likely represents a codivergence event between humans and gorillas. The simplexviruses infecting African great apes subsequently experienced multiple cross-species transmission events over the past 3 My, the most recent of which occurred between humans and bonobos around 1 Ma. These findings revise our understanding of the origins of human herpes simplexviruses and suggest that HSV-2 is one of the earliest zoonotic pathogens

    Identification of Radiopure Titanium for the LZ Dark Matter Experiment and Future Rare Event Searches

    The LUX-ZEPLIN (LZ) experiment will search for dark matter particle interactions with a detector containing a total of 10 tonnes of liquid xenon within a double-vessel cryostat. The large mass and proximity of the cryostat to the active detector volume demand the use of material with extremely low intrinsic radioactivity. We report on the radioassay campaign conducted to identify suitable metals, the determination of factors limiting radiopure production, and the selection of titanium for construction of the LZ cryostat and other detector components. This titanium has been measured with activities of 238^{238}Ue_{e}~<<1.6~mBq/kg, 238^{238}Ul_{l}~<<0.09~mBq/kg, 232^{232}The_{e}~=0.28±0.03=0.28\pm 0.03~mBq/kg, 232^{232}Thl_{l}~=0.25±0.02=0.25\pm 0.02~mBq/kg, 40^{40}K~<<0.54~mBq/kg, and 60^{60}Co~<<0.02~mBq/kg (68\% CL). Such low intrinsic activities, which are some of the lowest ever reported for titanium, enable its use for future dark matter and other rare event searches. Monte Carlo simulations have been performed to assess the expected background contribution from the LZ cryostat with this radioactivity. In 1,000 days of WIMP search exposure of a 5.6-tonne fiducial mass, the cryostat will contribute only a mean background of 0.160±0.0010.160\pm0.001(stat)±0.030\pm0.030(sys) counts.Comment: 13 pages, 3 figures, accepted for publication in Astroparticle Physic

    Assessing Host-Virus Codivergence for Close Relatives of Merkel Cell Polyomavirus Infecting African Great Apes

    It has long been hypothesized that polyomaviruses (PyV; family Polyomaviridae) codiverged with their animal hosts. In contrast, recent analyses suggested that codivergence may only marginally influence the evolution of PyV. We reassess this question by focusing on a single lineage of PyV infecting hominine hosts, the Merkel cell polyomavirus (MCPyV) lineage. By characterizing the genetic diversity of these viruses in seven African great ape taxa, we show that they exhibit very strong host specificity. Reconciliation analyses identify more codivergence than noncodivergence events. In addition, we find that a number of host and PyV divergence events are synchronous. Collectively, our results support codivergence as the dominant process at play during the evolution of the MCPyV lineage. More generally, our results add to the growing body of evidence suggesting an ancient and stable association of PyV and their animal hosts

    Simulations of events for the LUX-ZEPLIN (LZ) dark matter experiment

    The LUX-ZEPLIN dark matter search aims to achieve a sensitivity to the WIMP-nucleon spin-independent cross-section down to (1–2)×10−12 pb at a WIMP mass of 40 GeV/c2. This paper describes the simulations framework that, along with radioactivity measurements, was used to support this projection, and also to provide mock data for validating reconstruction and analysis software. Of particular note are the event generators, which allow us to model the background radiation, and the detector response physics used in the production of raw signals, which can be converted into digitized waveforms similar to data from the operational detector. Inclusion of the detector response allows us to process simulated data using the same analysis routines as developed to process the experimental data
