    Early life-history studies of nearshore rockfishes and lingcod off Central California, 1987-92

    This study focused on the physical and biological processes that influence the distribution, abundance, growth, and survival of young-of-the-year (YOY) rockfishes and lingcod along the central California coast. The annual somatic and reproductive condition of adult female blue rockfish corresponded to annual upwelling. Resulting larval production may correspond to the reproductive potential of adults; however, ultimate recruitment success of YOY is also effected by oceanographic conditions during their planktonic stage. Within a year, each species of settled YOY was observed concurrently and in relatively similar abundances at all study dive sites along the central coast. Most species of YOY exhibited similar growth patterns among stations and years. We found a high degree of interannual variability in the condition of adults and relative abundances of YOY. We believe a large part of this variability is due to annual oceanographic conditions, specifically upwelling. Marine reserves, which would protect populations of reproductively mature rockfishes and lingcod and insure 1arval production, have been suggested as an alternative to present management strategies for these species. However, a crucial question is whether or not larvae from adult fish in reserves would significantly contribute to replenishing stocks in other areas.     SUPPLY CHAIN COMPETENCY: RECIPE FOR CEREAL AND LIVESTOCK MARKETING IN ALBERTA?

    This study examines the nature of Supply Chain Management (SCM) in the Canadian barley industry, economic theories related to SCM, identifies SCM drivers and reviews the Canadian barley marketing system. Two surveys were conducted; one on the feed barley segment of the market; another on the malt barley segment of the market. These surveys provide an outline of the attributes sought by buyers of feed barley in Alberta and by buyers of malt barley in Canada and the United States. A further goal of these surveys was to assess the extent of motivations for SCM in the barley supply chain. Study methods include scaling, factor analysis and stated preference techniques to analyze purchasers' preferences for specific product attributes, business relationships and product source. The major attributes of feed barley sought by Alberta feed manufacturers appear to be physical characteristics such as moisture level, absence of foreign material, high bushel weight and uniform appearance of kernels. Features identified as of moderate importance included levels of certain key amino acids, starch level in the barley sample, as well as such seller characteristics as whether the seller was personally known to the buyer, and willingness of the seller to enter into a long-term supply contract. At the level of the Alberta feed mill industry, results therefore indicate that physical, readily identifiable attributes dominate in the selection of feed barley. As a result, the study identified that SCM is not yet a part of the awareness of barley buyers at feed mills. Among buyers of malt barley, physical or easily assessed attributes such as size of kernel, germination percentage, variety and location where produced ranked highly in a factor analysis as important to malt barley buyers. While results from the sample of Canadian and US buyers did not indicate strong potential for SCM in the malt barley sector, the study found there to be differences in attributes desired by US versus Canadian malt purchasers. Main differences were the concern of US buyers with the region where the barley was grown, and the apparently much higher willingness of US buyers to obtain their malt barley from more than one source. These differences may suggest a potential for SCM in malt barley focused on procuring supplies from regions identified as preferred locations for barley used in malt production.Industrial Organization,

    ELISA analysis of β-secretase cleavage of the Swedish amyloid precursor protein in the secretory and endocytic pathways

    Limiting beta amyloid (Aβ) production could become an important therapeutic target in Alzheimer's disease (AD). Aβ is derived by the sequential cleavage of amyloid precursor protein (APP) by β- and γ-secretases. A double missense mutation in APP found in a Swedish pedigree (APPsw) elevates Aβ40 and Aβ42 production. Aβ production and, therefore, β-secretase cleavage of APPsw reportedly occur in the endoplasmic reticulum (ER), Golgi and endocytic compartments. However, the relative contribution of β-secretase cleavage occurring in each compartment has not been determined. Experiments described here use a novel ELISA to measure the β-cleaved product, APPswβ. Using this ELISA, we provide new information regarding the relative amount of β-secretase cleavage of APPsw that occurs in secretory and endocytic pathways. Using a dilysine retrieval motif to retain APPsw in the ER, we discovered that less than 15% of the β-secretase cleavage was still detected. Experiments utilizing endocytosis-impaired mutants of APPsw revealed that little or no β-secretase cleavage of APPsw appears to take place through endocytosis. Surprisingly, deletion of the entire cytoplasmic tail increased both APPswβ and Aβ secretion, suggesting that protein interactions with this region normally impede β-secretase cleavage.     Clinicopathological determinants of an elevated systemic inflammatory response following elective potentially curative resection for colorectal cancer

    Introduction: The postoperative systemic inflammatory response (SIR) is related to both long- and short-term outcomes following surgery for colorectal cancer. However, it is not clear which clinicopathological factors are associated with the magnitude of the postoperative SIR. The present study was designed to determine the clinicopathological determinants of the postoperative systemic inflammatory response following colorectal cancer resection. Methods: Patients with a histologically proven diagnosis of colorectal cancer who underwent elective, potentially curative resection during a period from 1999 to 2013 were included in the study (n = 752). Clinicopathological data and the postoperative SIR, as evidenced by postoperative Glasgow Prognostic Score (poGPS), were recorded in a prospectively maintained database. Results: The majority of patients were aged 65 years or older, male, were overweight or obese, and had an open resection. After adjustment for year of operation, a high day 3 poGPS was independently associated with American Society of Anesthesiologists (ASA) grade (hazard ratio [HR] 1.96; confidence interval [CI] 1.25–3.09; p = 0.003), body mass index (BMI) (HR 1.60; CI 1.07–2.38; p = 0.001), mGPS (HR 2.03; CI 1.35–3.03; p = 0.001), and tumour site (HR 2.99; CI 1.56–5.71; p < 0.001). After adjustment for year of operation, a high day 4 poGPS was independently associated with ASA grade (HR 1.65; CI 1.06–2.57; p = 0.028), mGPS (HR 1.81; CI 1.22–2.68; p = 0.003), NLR (HR 0.50; CI 0.26–0.95; p = 0.034), and tumour site (HR 2.90; CI 1.49–5.65; p = 0.002). Conclusions: ASA grade, BMI, mGPS, and tumour site were consistently associated with the magnitude of the postoperative systemic inflammatory response, evidenced by a high poGPS on days 3 and 4, in patients undergoing elective potentially curative resection for colorectal cancer

    Durability of Intercalated Graphite Epoxy Composites in Low Earth Orbit

    The electrical conductivity of graphite epoxy composites can be substantially increased by intercalating (inserting guest atoms or molecules between the graphene planes) the graphite fibers before composite formation. The resulting high strength, low density, electrically conducting composites have been proposed for EMI shielding in spacecraft. Questions have been raised, however, about their durability in the space environment, especially with respect to outgassing of the intercalates, which are corrosive species such as bromine. To answer those concerns, six samples of bromine intercalated graphite epoxy composites were included in the third Evaluation of Oxygen Interaction with Materials (EOIM-3) experiment flown on the Space Shuttle Discovery (STS-46). Changes in electrical conductivity, optical reflectance, surface texture, and mass loss for SiO2 protected and unprotected samples were measured after being exposed to the LEO environment for 42 hours. SiO2 protected samples showed no degradation, verifying conventional protection strategies are applicable to bromine intercalated composites. The unprotected samples showed that bromine intercalation does not alter the degradation of graphite-epoxy composites. No bromine was detected to have been released by the fibers allaying fears that outgassing could be disruptive to the sensitive electronics the EMI shield is meant to protect

    Modelling the dynamics of Plasmodium falciparum histidine-rich protein 2 in human malaria to better understand malaria rapid diagnostic test performance

    Background: Effective diagnosis of malaria is a major component of case management. Rapid diagnostic tests (RDTs) based on Plasmodium falciparumhistidine-rich protein 2 (PfHRP2) are popular for diagnosis of this most virulent malaria infection. However, concerns have been raised about the longevity of the PfHRP2 antigenaemia following curative treatment in endemic regions. Methods. A model of PfHRP2 production and decay was developed to mimic the kinetics of PfHRP2 antigenaemia during infections. Data from two human infection studies was used to fit the model, and to investigate PfHRP2 kinetics. Four malaria RDTs were assessed in the laboratory to determine the minimum detectable concentration of PfHRP2. Results: Fitting of the PfHRP2 dynamics model indicated that in malaria nave hosts, P. falciparum parasites of the 3D7 strain produce 1.4 × 10 g of PfHRP2 per parasite per replication cycle. The four RDTs had minimum detection thresholds between 6.9 and 27.8 ng/mL. Combining these detection thresholds with the kinetics of PfHRP2, it is predicted that as few as 8 parasites/L may be required to maintain a positive RDT in a chronic infection. Conclusions: The results of the model indicate that good quality PfHRP2-based RDTs should be able to detect parasites on the first day of symptoms, and that the persistence of the antigen will cause the tests to remain positive for at least seven days after treatment. The duration of a positive test result following curative treatment is dependent on the duration and density of parasitaemia prior to treatment and the presence and affinity of anti-PfHRP2 antibodies

    Pharmaceutical companies’ documented and online privacy practices: Development of an index measure and initial test

    Consumers have begun to take a more proactive approach to their healthcare by accessing pharmaceutical companies Websites to obtain health and drug information. In exchange for these benefits, companies require consumers to voluntarily disclose information. However, research has shown that consumers continue to be concerned about how their information is managed, used, and distributed by companies, especially if accessed via the Web. To date, there has been limited empirical research to examine the actual online practices of companies when it comes to privacy, especially those of pharmaceutical companies. Using the Delphi expert panel process, we identified the components of a hierarchical benchmarking index to examine the documented and actual online practices of 100 Website registrations with pharmaceutical companies. In this paper, we outline the development of an index to measure the personal information privacy violations of pharmaceutical companies using hierarchical linear technique. Second, we provided empirical evidence regarding the magnitude of voluntary adherence to the Fair Information Practices (FIPs) by pharmaceutical companies based upon the personal information privacy violations. Our results revealed that companies with headquarters in Europe had fewer personal information privacy violations than those in the US. Moreover, our results indicate that fewer personal information privacy violations occur for chronic conditions than for non-chronic conditions, as well as fewer violations occur with Website registrations for updates than for discounts. Finally, both Europe and UK demonstrated more overall adherence to the FIPs than the US and Asia. This suggests that self-regulation may not be sufficien
