    Radio Band Observations of Blazar Variability

    The properties of blazar variability in the radio band are studied using the unique combination of temporal resolution from single dish monitoring and spatial resolution from VLBA imaging; such measurements, now available in all four Stokes parameters, together with theoretical simulations, identify the origin of radio band variability and probe the characteristics of the radio jet where the broadband blazar emission originates. Outbursts in total flux density and linear polarization in the optical-to-radio bands are attributed to shocks propagating within the jet spine, in part based on limited modeling invoking transverse shocks; new radiative transfer simulations allowing for shocks at arbitrary angle to the flow direction confirm this picture by reproducing the observed centimeter-band variations observed more generally, and are of current interest since these shocks may play a role in the gamma-ray flaring detected by Fermi. Recent UMRAO multifrequency Stokes V studies of bright blazars identify the spectral variability properties of circular polarization for the first time and demonstrate that polarity flips are relatively common. All-Stokes data are consistent with the production of circular polarization by linear-to-circular mode conversion in a region that is at least partially self-absorbed. Detailed analysis of single-epoch, multifrequency, all-Stokes VLBA observations of 3C 279 support this physical picture and are best explained by emission from an electron-proton plasma.Comment: 6 pages, 5 figures, uses, jaa.sty.     Blickling Hall Lake is a relatively large (10.1 ha.) shallow (average depth 95 cm) estate lake in the grounds of Blickling Hall, near Aylsham, North Norfolk. It was formed in the early 1700s through the damming of a small tributary of the River Bure, presently the major inflow to the lake. Water levels in the lake are controlled by a sluice and there is one outflow which ultimately feeds into the River Bure. The site has considerable current conservation interest in the form of an extensive fringing reedswamp, which provides good habitat for birds and dragonflies in particular. Currently, however the lake itself is of minimal conservation interest

    Prognostic microRNAs in high-grade glioma reveal a link to oligodendrocyte precursor differentiation.

    MicroRNA expression can be exploited to define tumor prognosis and stratification for precision medicine. It remains unclear whether prognostic microRNA signatures are exclusively tumor grade and/or molecular subtype-specific, or whether common signatures of aggressive clinical behavior can be identified. Here, we defined microRNAs that are associated with good and poor prognosis in grade III and IV gliomas using data from The Cancer Genome Atlas. Pathway analysis of microRNA targets that are differentially expressed in good and poor prognosis glioma identified a link to oligodendrocyte development. Notably, a microRNA expression profile that is characteristic of a specific oligodendrocyte precursor cell type (OP1) correlates with microRNA expression from 597 of these tumors and is consistently associated with poor patient outcome in grade III and IV gliomas. Our study reveals grade-independent and subtype-independent prognostic molecular signatures in high-grade glioma and provides a framework for investigating the mechanisms of brain tumor aggressiveness

    MicroRNA-222 reprogrammed cancer-associated fibroblasts enhance growth and metastasis of breast cancer

    BACKGROUND: Cancer-associated fibroblasts (CAFs) are known to impact on tumour behaviour, but the mechanisms controlling this are poorly understood. METHODS: Breast normal fibroblasts (NFs) or CAFs were isolated from cancers by laser microdissection or were cultured. Fibroblasts were transfected to manipulate miR-222 or Lamin B receptor (LBR). The fibroblast-conditioned medium was collected and used to treat epithelial BC lines MDA-MB-231 and MDA-MB-157. Migration, invasion, proliferation or senescence was assessed using transwell, MTT or X-gal assays, respectively. RESULTS: MiR-222 was upregulated in CAFs as compared with NFs. Ectopic miR-222 expression in NFs induced CAF-like expression profiles, while miR-222 knockdown in CAFs inhibited CAF phenotypes. LBR was identified as a direct miR-222 target, and was functionally relevant since LBR knockdown phenocopied miR-222 overexpression and LBR overexpression phenocopied miR-222 knockdown. MiR-222 overexpression, or LBR knockdown, was sufficient to induce NFs to show the CAF characteristics of enhanced migration, invasion and senescence, and furthermore, the conditioned medium from these fibroblasts induced increased BC cell migration and invasion. The reverse manipulations in CAFs inhibited these behaviours in fibroblasts, and inhibited paracrine influences on BC cells. CONCLUSION: MiR-222/LBR have key roles in controlling pro-progression influences of CAFs in BC. This pathway may present therapeutic opportunities to inhibit CAF-induced cancer progression

    Spatially Resolved Magnetic Field Structure in the Disk of a T Tauri Star

    Magnetic fields in accretion disks play a dominant role during the star formation process but have hitherto been observationally poorly constrained. Field strengths have been inferred on T Tauri stars themselves and possibly in the innermost part of the accretion disk, but the strength and morphology of the field in the bulk of the disk have not been observed. Unresolved measurements of polarized emission (arising from elongated dust grains aligned perpendicular to the field) imply average fields aligned with the disks. Theoretically, the fields are expected to be largely toroidal, poloidal, or a mixture of the two, which imply different mechanisms for transporting angular momentum in the disks of actively accreting young stars such as HL Tau. Here we report resolved measurements of the polarized 1.25 mm continuum emission from HL Tau's disk. The magnetic field on a scale of 80 AU is coincident with the major axis (~210 AU diameter) of the disk. From this we conclude that the magnetic field inside the disk at this scale cannot be dominated by a vertical component, though a purely toroidal field does not fit the data well either. The unexpected morphology suggests that the magnetic field's role for the accretion of a T Tauri star is more complex than the current theoretical understanding.Comment: Accepted for publication in Natur
