    The impact of family environment on self-esteem and symptoms in early psychosis

    Expressed emotion (EE) and self-esteem (SE) have been implicated in the onset and development of paranoia and positive symptoms of psychosis. However, the impact of EE on patients' SE and ultimately on symptoms in the early stages of psychosis is still not fully understood. The main objectives of this study were to examine whether: (1) patients' SE mediated the effect of relatives' EE on patients' positive symptoms and paranoia; (2) patients' perceived EE mediated the effect of relatives' EE on patients' SE; (3) patients' SE mediated between patients' perceived EE and patients' symptomatology; and (4) patients' perceived EE and patients' SE serially mediated the effect of relatives' EE on patients' positive symptoms and paranoia. Incipient psychosis patients (at-risk mental states and first-episode of psychosis) and their respective relatives completed measures of EE, SE, and symptoms. Findings indicated that: (1) patients' perceived EE mediated the link between relatives' EE and patients' negative, but not positive, SE; (2) patients' negative SE mediated the effect of patients' perceived EE on positive symptoms and paranoia; (3) the association of relatives' EE with positive symptoms and paranoia was serially mediated by an increased level of patients' perceived EE leading to increases in negative SE; (4) high levels of patients' distress moderated the effect of relatives' EE on symptoms through patients' perceived EE and negative SE. Findings emphasize that patients' SE is relevant for understanding how microsocial environmental factors impact formation and expression of positive symptoms and paranoia in early psychosis. They suggest that broader interventions for patients and their relatives aiming at improving family dynamics might also improve patients' negative SE and symptoms

    Ecological validity of expressed emotion in early psychosis

    Expressed emotion (EE) is an aspect of the family environment that influences the course of multiple forms of psychopathology. However, there is limited research about how EE dimensions [i.e., criticism and emotional over-involvement (EOI)] are expressed in real-world settings. The present study used experience sampling methodology to investigate: 1) the criterion and construct validity of daily-life, momentary measures of criticism and EOI, and 2) the construct and ecological validity of psychometric EE-dimensions as assessed with the self-report Family Questionnaire (FQ). A total sample of 55 relatives (34 relatives of at-risk mental state patients and 21 of first-episode psychosis patients) were prompted randomly six times daily for 1-week to assess their current emotional experiences and cognitive appraisals. Relatives also completed the FQ. Momentary criticism and EOI were significantly associated with the two FQ-EE dimensions respectively, supporting the criterion validity of real-world assessed EE dimensions. As hypothesized, momentary and FQ-EE dimensions were associated with decreased positive affect, as well as with appraisals of less effective coping in daily life. Only momentary EE dimensions were associated with increased momentary negative affect. Partly in contrast with our hypotheses, momentary criticism and FQ-criticism were more consistently related to situational stress and burden than momentary EOI and FQ-EOI. Finally, neither momentary nor FQ-EE dimensions showed distinct patterns of associations with illness attributions. Findings partly support the construct validity of momentary criticism and EOI as well as the construct and ecological validity of the FQ as a sensitive measure of EE dimensions

    A Human iPSC-derived 3D platform using primary brain cancer cells to study drug development and personalized medicine

    Abstract A high throughput histology (microTMA) platform was applied for testing drugs against tumors in a novel 3D heterotypic glioblastoma brain sphere (gBS) model consisting of glioblastoma tumor cells, iPSC-derived neurons, glial cells and astrocytes grown in a spheroid. The differential responses of gBS tumors and normal neuronal cells to sustained treatments with anti-cancer drugs temozolomide (TMZ) and doxorubicin (DOX) were investigated. gBS were exposed to TMZ or DOX over a 7-day period. Untreated gBS tumors increased in size over a 4-week culture period, however, there was no increase in the number of normal neuronal cells. TMZ (100 uM) and DOX (0.3 uM) treatments caused ~30% (P~0.07) and ~80% (P < 0.001) decreases in the size of the tumors, respectively. Neither treatment altered the number of normal neuronal cells in the model. The anti-tumor effects of TMZ and DOX were mediated in part by selective induction of apoptosis. This platform provides a novel approach for screening new anti-glioblastoma agents and evaluating different treatment options for a given patient

    Relatives' attachment anxiety mediates the association between perceived loss and expressed emotion in early psychosis

    A common reaction experienced by family members of patients with psychosis is grief for the loss of their healthy relative. Importantly, high levels of perceived loss have been related to the manifestation of high expressed emotion (EE), which includes the negative attitudes expressed by relatives toward an ill family member. However, the mechanisms underlying the relationship between relatives' perceived loss and EE attitudes in the early stages of psychosis are still not fully understood. In this regard, attachment theory has been suggested as a useful framework for understanding this link. The current study aimed to examine: (1) whether relatives' perceived loss was associated with relatives' EE dimensions (i.e., criticism and emotional over-involvement (EOI)), and (2) whether such associations were mediated by relatives' attachment dimensions (i.e., anxiety and avoidance). Seventy-eight relatives of patients with early psychosis completed the Mental Illness Version of the Texas Inventory of Grief for the assessment of loss reactions. Attachment dimensions and EE attitudes were assessed by the Psychosis Attachment Measure and the Family Questionnaire, respectively. Findings indicated that relatives' perceived loss was associated with EE dimensions. Relatives' attachment anxiety, but not avoidance, mediated the relationship of perceived loss with both criticism and EOI. Findings highlight the importance of examining the role of relatives' attachment characteristics for understanding how perceptions of loss might impact the manifestation of EE attitudes in the early stages of psychosis. Family interventions aimed at assisting relatives to improve their management of negative emotional reactions to loss are fundamental to prevent impairing loss reactions and the entrenchment of high-EE attitudes

    Estimación del balance hídrico en una cuenca semiárida, El Mogor, Baja California, México

    El trabajo de investigación se efectuó en la subcuenca El Mogor en el Valle de Guadalupe (VG) en Baja California, México. Se realizó una caracterización integral, con el objetivo de hacer una cuantificación del aporte hidrogeológico de la subcuenca hacia el valle en el año hidrológico 2010-2011. Esta región presenta un clima semiárido y se realizan actividades agrícolas de gran importancia, donde sobresale la viticultura. Como parte de la caracterización, se estableció la conductividad hidráulica no saturada utilizando la ecuación de Kozeny-Carman (KC). Se llevó a cabo el cálculo de la evapotranspiración potencial (ETP) utilizando el método de Thornthwaite (Thw), el cual se comparó con la evaporación potencial (EP) estimada con la charola de Penman (CHP) y con la ETP calculada por la estación del CICESE (CIC), cuyo programa utiliza la ecuación de Penman-Monteith para su cálculo. Se estimó un balance hídrico (Almorox, 2003) utilizando el resultado obtenido de la ETP de Thw y los datos de precipitación (P), con los que se obtuvo una infiltración potencial (I) generalizada en la subcuenca de ~974 000 m3 y un escurrimiento (Es) de ~86 000 m3 . Asimismo, se calculó de forma directa el escurrimiento superficial mediante el uso de un vertedor triangular de pared delgada con el que se estimó un volumen de ~60 000 m3 . Finalmente, mediante fórmulas empíricas, se calculó una recarga (vertical, de sistema montañoso, horizontal y por percolación por agua de riego) de ~466 000 m3 , que fue equivalente al 7% de la precipitación infiltrable en el año hidrológico en estudio

    Predictors of criticism and emotional over-involvement in relatives of early psychosis patients

    Mechanisms underlying the manifestation of relatives' expressed emotion (EE) in the early stages of psychosis are still not properly understood. The present study aimed to examine whether relatives' psychological distress and subjective appraisals of the illness predicted EE dimensions over-and-above patients' poor clinical and functional status. Baseline patient-related variables and relatives attributes comprising criticism, emotional over-involvement (EOI), psychological distress, and illness attributions were assessed in 91 early psychosis patients and their respective relatives. Relatives were reassessed regarding EE dimensions at a 6-month follow-up. Relatives' psychological distress and illness attributions predicted criticism and EOI over-and-above patients' illness characteristics at both time points. Relatives' increased levels of anxiety, attributions of blame toward the patients, an emotional negative representation about the disorder, and decreased levels of self-blame attributions predicted EE-criticism at baseline. Relatives' anxiety and negative emotional representation of the disorder were the only significant predictors of EE-criticism at follow-up, whereas anxiety, attributions of control by the relative and an emotional negative representation about the disorder predicted EE-EOI both at baseline and follow-up assessments. Understanding the components that comprise and maintain EE attitudes should guide early psychosis caregivers in family interventions, enhancing proper management of psychological distress and reduction of negative appraisals about the illness. The prevention of high-EE attitudes over time in a sensitive period such as early psychosis might be critical in shaping the health of caregivers and the outcome of the affected relatives

    Magnetically Responsive Materials based on Polymeric Ionic Liquids and Graphene Oxide for Water Clean-up

    Hypothesis Owing to attractive interactions between negatively charged graphene oxide (GO) and a paramagnetic cationic polyelectrolyte (polyallydimethylammonium chloride with a FeCl4− counterion (Fe-polyDADMAC) it should be possible to generate magnetic materials. The benefit of using charge-based adsorption is that the need to form covalently linked magnetic materials is offset, which is expected to significantly reduce the time, energy and cost to make such responsive materials. These systems could have a wide use and application in water treatment. Experiments Non-covalent magnetic materials were formed through the mixing of Fe-pDADMAC and GO. A systematic study was conducted by varying polymer concentration at a fixed GO concentration. UV–Vis was used to confirm and quantify polymer adsorption onto GO sheets. The potential uses of the systems for water purification were demonstrated. Findings Fe-polyDADMAC adsorbs to the surface of GO and induces flocculation. Low concentrations of the polymer (20 mmol/L) induce restabilization. Difficult-to-recover gold nanoparticles can be separated from suspensions as well as the pollutant antibiotic tetracycline. Both harmful materials can be magnetically recovered from the dispersions. This system therefore has economical and practical applications in decontamination and water treatment
