    INOVASI PELAYANAN PUBLIK MELALUI UP3SK DI DINAS KEPENDUDUKAN DAN PENCATATAN SIPIL KABUPATEN PINRANG

    Penelitian ini bertujuan untuk menganalisis inovasi pelayanan publik melalui UP3SK di Dinas Kependudukan dan Pencatatan Sipil Kabupaten Pinrang. Penelitian ini merupakan penelitian kualitatif, dalam penelitian yang dilakukan bersifat deskriptif yaitu untuk menggambarkan kenyataan dari kejadian yang diteliti. Metode penelitian ini dianggap mampu menjelaskan penelitian ini secara mendalam. Fokus penelitian ini pada teori atribut inovasi oleh Rogers yaitu: Relative Advantage (Keuntungan Relatif), Compatibility (kesesuaian), Complexity (Kerumitan), Triability (Kemungkinan Dicoba), Observability (Kemudahan Diamati). Hasil penelitian menunjukkan bahwa inovasi pelayanan publik melalui UP3SK di Dinas Kependudukan dan Pencatatan Sipil Kabupaten Pinrang dapat dilihat berdasarkan atribut inovasi menurut Rogers. Pada atribut relative advantage diketahui bahwa keuntungan dari adanya inovasi pelayanan UP3SK adalah meminimalisir praktek percaloan dan lebih memudahkan masyarakat. Pada atribut compatibility menjelaskan bahwa inovasi pelayanan UP3SK sesuai dengan keinginan masyarakat dalam proses pengurusan dokumen yang lebih mudah. Atribut complexity menjelaskan bahwa tidak ada kerumitan dalam pengurusan dan juga dalam proses pelayanan UP3SK. Namun ada beberapa kendala yaitu belum lengkapnya sarana dan prasarana dalam proses pelayanan UP3SK, serta tidak jelasnya aturan petunjuk pelaksanaan dan petunjuk teknis dalam pelaksanaan pelayanan UP3SK. Atribut triability juga menjelaskan bahwa tidak adanya fase uji coba kepada masyarakat dalam pelayanan UP3SK. Hanya ada percobaan teknis di Kantor Dinas Kependudukan dan Pencatatan Sipil Kabupaten Pinrang. Pada atribut observability menunjukkan bahwa selama pelayanan UP3SK berjalan, telah sesuai dengan keinginan Dinas Kependudukan dan Pencatatan Sipil Kabupaten Pinrang yakni untuk memudahkan masyarakat dalam pengurusan dokumen penting dan meminimalisir percaloan atau pungutan liar

    Airport Cargo Logistics and Economic Outcome of Supply Chain: An Empirical Analysis

    Abstract Purpose – The United Arab Emirates (UAE) will have the third largest international air cargo market in the world by 2018 according to a new report by the International Air Transport Association (IATA). Handling airport cargo in and along with the integrated supply chain has become increasingly challenging in a competitive environment, particularly in Abu Dhabi. In this perspective, the purpose of the study is to investigate the influence of five factors-price sensitivity, cost leadership, airport charges, provision of unique product and services, development of airport facilities on competitive advantage. Design/Methodology/approach – An online questionnaire link sent to 168 airport logistics executives, out of which 102 respondents submitted. A convenience sampling method was used and the statistical software PLS was employed to measure the suitability of the conceptual framework. Findings – All the five factors or independent variables are positively related to the dependent variable, which confirms the support to the conceptual framework and hypothesis through various statistical analyses. Managerial Implications – Managers need to understand the relationship of the five factors, which are positively related to the dependent variable. Designing, developing and delivering on all the constructs as promised to the stakeholders are vital to achieve competitive advantage. Originality/Value – Due to lack of empirical research work in airport logistics, particularly in UAE, this study will be value added research work and fills the research gap as well.     Role of the redox responsive transcription factor, NRF2 in immune cell function

    Dendritic cells (DCs) are potent innate antigen presenting cells which are able to sense and engulf pathogens from sites of infection, which are then processed and presented to adaptive T lymphocytes in secondary lymphoid organs. Therefore, they are critical for the initiation and modulation of primary antigen-specific adaptive immune responses. They also play a critical role in the maintenance of T cell tolerance. T cells play vital roles in mediating both cellular and humoral-specific adaptive immune responses. There are various types of T cells present within the immune system including the cytotoxic CD8 T cells and T helper CD4 cells. CD4 T cells can be further subdivided into various subtypes examples of which include T helper 1 cells (Th1), Th2, Th17 and T regulatory cells. Each subset has its own distinctive function, transcriptional regulation and effector cytokine profile. It is established that appropriate DC and T cell immune function is highly dependent on their intracellular redox status. Cellular redox homeostasis is maintained through a balance between oxidising agents e.g. reactive oxygen species (ROS) and anti-oxidant or reducing agents e.g. glutathione (GSH). Excessive ROS production resulting in oxidative stress is extremely deleterious to the cell and if left unimpeded can result in cell necrosis, tissue damage and the onset of disease. As a result, mammalian cells have evolved an inducible adaptive defence system which provides protection against such oxidative or chemical insult. The functionality of this cellular defence system is principally governed by the activity of the redox-sensitive transcription factor Nrf2. In response to oxidative stress, Nrf2 induces the transcription of a battery of cytoprotective and antioxidant genes involved in GSH synthesis, detoxification of xenobiotics and their reactive metabolites and the maintenance of cellular redox homeostasis. It is now emerging that Nrf2 plays a pivotal role in immunity. However, its precise role in DC and T cell function is unclear. Using immature bone marrow-derived DCs (iDCs) from Nrf2+/+ and Nrf2-/- mice, the work presented in this thesis demonstrates that Nrf2 deficiency in iDCs resulted in lowered GSH levels, enhanced iDCs co-stimulatory receptor expression, impaired endocytic and phagocytic capacity, and increased iDC-mediated antigen-specific CD8 T cell stimulatory capacity in response to both an antigenic and self-peptide. Furthermore, artificially lowering GSH levels in the iDCs did not recapitulate the Nrf2 deficient iDC phenotype. Moreover, Nrf2-/- DCs exhibited an enhanced capacity to present cell-associated peptide antigens to antigen-specific CD8 T cells, resulting in increased CD8 T cell effector function. Loss of Nrf2 in LPS-stimulated DCs results in a lowered Th1 cytokine profile. These results have implications for Nrf2 in DC-mediated CD8 T-cell immunity, peripheral CD8 T cell tolerance and CD4 effector differentiation. The role of Nrf2 in T cell function is poorly understood. Using thymocytes, splenocytes and lymph-node derived T cells from Nrf2+/+ and Nrf2-/- mice, we demonstrate that loss of Nrf2 did not affect the development of CD4+CD25+ natural occurring Treg, mature CD4 and CD8 T cell populations within the thymus. Furthermore, Nrf2 deficiency did not alter the composition of CD4 and CD8 T cell populations within secondary lymphoid organs. It was observed that splenic Nrf2-/- naïve T cells exhibited enhanced ROS generation, accompanied by low level increases in T cell activation markers. However, the marginal augmentation of Nrf2-/- naïve T cell activation status did not result in increased T cell receptor (TCR)-triggered T cell proliferation. In contrast, Nrf2 deficient effector T cells exhibited enhanced TCR/CD3-triggered proliferation, associated with increased Th1 and decreased Th2 effector function. Importantly, we demonstrated that Nrf2-/- effector T cells secreted increased levels of IL-17A and IL-22, a signature cytokine profile indicative of the more recently identified Th17 cell lineage. This was also observed under Th17 polarising conditions, further suggesting that loss of Nrf2 predisposes effector Th17 development. The implications of the latter findings are significant given the pivotal role that Th17 cells play in the pathogenesis of a variety of autoimmune diseases including multiple sclerosis (MS) and Systemic lupus erythematosus (SLE). Nrf2 plays a critical role in the detoxification of xenobiotics in the liver, which as the primary drug-metabolising organ, is subjected to an array of xenobiotics and their respective metabolites. Individuals vary in their responses to xenobiotic exposure from adaptation to severe adverse drug reactions. However, it is unknown whether this human disparity in drug response is a consequence of inter-individual variation in the Nrf2 adaptive defence system to xenobiotic stress. In light of this, we aimed to firstly investigate whether variation in the Nrf2 adaptive system was present within individuals in response to a chemical inducer of Nrf2, CDDO-Me. To address this issue, basal and induced Nrf2 protein levels and downstream NQO1 expression were measured in activated human T cell blasts, in response to increasing concentrations of CDDO-Me. Examination of various donor-derived T cells, demonstrated that humans vary in their Nrf2 response to CDDO-Me, with respect to nuclear Nrf2 and NQO1 mRNA expression. Therefore we concluded that inter-individual variation does exist in the human’s Nrf2 adaptive system in response to a known Nrf2 probe. Overall, the experimental results obtained in this PhD programme have provided detail on the role of Nrf2 in immune cells and highlights the potential for therapeutically targeting Nrf2 in immune-mediated disease

    O presente trabalho teve como objetivo divulgar a utilização do esporte como ferramenta para promover nos jovens de comunidades vulneráveis o desenvolvimento local tendo como instrumento de estímulo a expectativa gerada pela mídia nos alunos de Artes Marciais Mistas (MMA) do Instituto Nogueira. Foi aplicada a técnica do questionário com 18 alunos, com a faixa etária entre 12 e 17 anos, sendo 13 meninos e cinco meninas, todos matriculados no instituto localizado na comunidade Manguinhos, na zona da Leopoldina da cidade do Rio de Janeiro. Os resultados demonstram que ocorreu estímulo positivo nos jovens, por meio dos mecanismos de persuasão realizados pela mídia. A maioria percebeu no MMA uma chance de ascensão social, além de conquistar um corpo ideal semelhante ao dos lutadores apresentados pela mídia

    Case Report Multifocal Buruli Ulcer Associated with Secondary Infection in HIV Positive Patient

    Buruli ulcer is a chronic and infectious skin disease, caused by Mycobacterium ulcerans. It leads to large skin ulceration and sometimes bone infection which is responsible for deformities. Here, we report a case of multifocal form of Buruli ulcer associated with secondary infection in a 46-year-old human immunodeficiency virus (HIV) positive woman. The antimycobacterial drugs combined to surgery allowed curing this multifocal case and rose up two relevant issues: the susceptibility of immune reconstitution inflammatory syndrome (IRIS) occurrence and Mycobacterium dissemination. The deep immune depression, the underline biological, and clinical disorders of the patient might contribute to IRIS occurrence and Buruli ulcer dissemination. Future investigations have to be conducted on the mechanism of IRIS on set and on Mycobacterium ulcerans dissemination after ARV drugs initiation and the patient related underline clinical or biological disorders

    Histological evidence for a supraspinous ligament in sauropod dinosaurs

    Supraspinous ossified rods have been reported in the sacra of some derived sauropod dinosaurs. Although different hypotheses have been proposed to explain the origin ofthis structure, histological evidence has never been provided to support or reject any of them. In order to establish its origin, we analyse and characterize the microstructure of thesupraspinous rod of two sauropod dinosaurs from the Upper Cretaceous of Argentina. The supraspinous ossified rod is almost entirely formed by dense Haversian bone. Remains ofprimary bone consist entirely of an avascular tissue composed of two types of fibre-like structures, which are coarse and longitudinally (parallel to the main axis of the element) oriented. These structures are differentiated on the basis of their optical properties under polarized light. Very thin fibrous strands are also observed in some regions. These small fibres are all oriented parallel to one another but perpendicular to the element main axis. Histological features of the primary bone tissue indicate that the sacral supraspinous rod corresponds to an ossified supraspinous ligament. The formation of this structure appears to have been a non-pathological metaplastic ossification, possibly induced by the continuous tensile forces applied to the element.Fil: Cerda, Ignacio Alejandro.
