Limnology of Two Contrasting Hydroelectric Reservoirs (Storage and Run-of-River) in Southeast Brazil
Infecção pelo Sugarcane yellow leaf virus causa alterações na eficiência fotossintética e acúmulo de carboidratos nas folhas de cana-de-açúcar
Infection by Sugarcane yellow leaf virus (ScYLV) causes severe leaf symptoms in sugarcane (Saccharum spp.) hybrids, which indicate alterations in its photosynthetic apparatus. To gain an overview of the physiological status of infected plants, we evaluated chlorophyll a fluorescence and gas exchange assays, correlating the results with leaf metabolic surveys, i.e., photosynthetic pigments and carbohydrate contents. When compared to healthy plants, infected plants showed a reduction in potential quantum efficiency for photochemistry of photosystem (PSII) and alterations in the filling up of the plastoquinone (PQ) pool. They also showed reduction in the CO2 net exchange rates, probably as a consequence of impaired quantum yield. In addition, reductions were found in the contents of photosynthetic leaf pigments and in the ratio chlorophyll a/chlorophyll b (chla/chlb). Carbohydrate content in the leaves was increased as a secondary effect of the ScYLV infection. This article discusses the relation of virus replication and host defense responses with general alterations in the photosynthetic apparatus and in the metabolism of infected plants.O vírus do amarelecimento foliar da cana-de-açúcar (Sugarcane yellow leaf virus, ScYLV) causa sintomas foliares severos e típicos de infecção por luteovirus, em cana-de-açúcar (Saccharum spp.). Uma vez que alterações no sistema fotossintético da planta seriam esperadas, avaliaram-se a análise da emissão da fluorescência da clorofila a e as trocas gasosas durante a fotossíntese, relacionando esses dados com análises metabólicas, ou seja, conteúdos de pigmentos fotossintéticos e carboidratos presentes nas folhas. As plantas infetadas apresentaram redução na eficiência quântica fotoquímica potencial do fotossistema II (PSII) e alterações no preenchimento do pool de plastoquinona (PQ). Essas plantas apresentaram, também, redução nas taxas de troca líquida de CO2, provavelmente em conseqüência da redução na eficiência quântica. Paralelamente, reduções nos conteúdos de pigmentos fotossintéticos foliares e na razão clorofila a/clorofila b (chla/chlb) foram verificadas. Adicionalmente, o conteúdo de açúcares nas folhas foi aumentado, provavelmente como um efeito secundário da infecção viral. A relação entre a replicação viral e as respostas de defesa da hospedeira com as alterações encontradas no aparelho fotossintético e no metabolismo das plantas infetadas é discutida.101
Software Tomato Analyzer para a determinação do tamanho do embrião em sementes radiografadas
A primary interest of image analysis of X-rayed seeds is to identify whether the extent of fill in the embryo cavity is associated with to seed physiological quality. The objective of this research was to verify the accuracy of the freely available Tomato Analyzer (TA) software developed at The Ohio State University to determine the ratio of embryo size over total seed area. Seeds of pumpkin, watermelon, cucumber and cotton were X-rayed and analyzed by the software which defines seed and embryo boundaries and automatically generates numerical values to quantify that ratio. Results showed that the TA has the sensitivity to evaluate the extent of embryo growth within the cucurbits and cotton seeds and is a promising alternative for this assessment in other seed species.Uma das prioridades atuais da pesquisa sobre análise de imagens de sementes radiografadas é determinar a proporção entre a área ocupada pelo embrião e o espaço disponível na cavidade interna da semente e associar esse parâmetro ao potencial fisiológico. Este trabalho procurou avaliar a eficiência do software Tomato Analyzer para quantificar o grau de desenvolvimento do embrião. Sementes de abóbora, melancia, pepino e algodão foram radiogafadas e as imagens processadas foram submetidas à análise pelo software, que define o perímetro da semente e o do embrião e gera valores utilizados para expressar a relação percentual entre a área do embrião e a da cavidade interna da semente. Os resultados indicaram que o softwareTomato Analyzer permite determinar o grau de desenvolvimento do embrião de sementes de cucurbitáceas e de algodão, indicado pela área embrionária ou do espaço livre existente entre o embrião e o tegumento; essa alternativa pode ser viável para avaliar sementes de outras espécies.FAPESPCNP
Quantitative And Qualitative Analysis Of Microorganisms In Root-filled Teeth With Persistent Infection: Monitoring Of The Endodontic Retreatment.
The aim of this study was to investigate in vivo microorganisms detected in root-filled teeth with post-treatment apical periodontitis and quantify colony-forming units (CFU) during endodontic retreatment. Fifteen root-filled teeth had their previous gutta-percha removed and were randomly instrumented before being divided into three groups and medicated with either [Ca(OH)2 + 2% CHX gel], [Ca(OH)2 + 0.9% NaCl] or 2% CHX gel. Samples were taken after removal of gutta-percha (S1), after chemomechanical preparation using 2% CHX gel (S2), and after inter-appointment dressing (S3) for 7 or 14 days later. Cultivable bacteria recovered from infected root canals at the three stages were counted and identified by means of culture and PCR assay (16S rDNA). Quantitative data were statistically analyzed by using Mann-Whitney test in which pairs of groups were compared (P < 0.05). CFU counts decreased significantly from S1 to S2 (P < 0.05). No significant difference was found between S2 and S3 (P = 0.3093) for all three experimental groups. Chemomechanical preparation and intra-canal dressing promoted significant median reductions of 99.61% and 99.57%, respectively, in the number of bacteria compared to S1 samples. A total of 110 cultivable isolates were recovered by culture technique from 32 different species and 7 different genera. Out of the 13 target species-specific primer of bacteria analyzed, 11 were detected during endodontic retreatment. The great majority of taxa found in post-treatment samples were Gram-positive bacteria, although Gram-negative bacteria were found by molecular methods. Moreover, our results showed that gutta-percha removal and chemomechanical preparation are effective for root canal disinfection, whereas additional intra-canal dressing did not improve disinfection.7302-
Deficiência de boro no metabolismo de pontas de raízes de tomateiro
Variations in growth, total soluble sugars, mono and polyphenols, proteins and ribonucleic acids were studied in root tips of tomato (Lycopersicon esculentum Mill. cv. Campbell 33) submitted to boron deficiency for 0, 2, 4, and 6 days. From the second day onwards, boron deficiency reduced growth of the plants and also the concentration of total soluble sugars (a fact probably associated with the increased synthesis of structural material) and monophenols (a phenomenon associated to the increased oxidation of these compounds). Polyphenols, proteins and RNA were increased in the boron-deficient root tips. Boron may act more directly on the ribonucleic acid metabolism than on protein metabolism.Estudaram-se as variações de crescimento, açúcares solúveis totais, mono e polifenóis, proteínas e ácidos ribonucléicos em pontas de raízes de tomateiro (Lycopersicon esculentum Mill. cv. Campbell 33), submetido à deficiência de boro por 0, 2, 4 e 6 dias. A partir do segundo dia, a deficiência de boro reduziu o crescimento das plantas, os teores de açúcares solúveis (fato provavelmente associado à maior síntese de material estrutural) e de monofenóis (associado à maior oxidação desses compostos). Polifenóis, proteínas e ARN sofreram aumentos nas raízes deficientes. Possivelmente o boro tem ação mais direta sobre o metabolismo de ácidos ribonucléicos que de proteínas

Este trabalho procurou verificar a eficiência do SVIS® (Seed Vigor Imaging System) para determinar o vigor de sementes de abóbora colhidas durante diferentes estádios de maturação. Utilizou-se a cultivar híbrida Jabras, proveniente do cruzamento entre Cucurbita maxima (linhagem feminina) e Cucurbita moschata (linhagem masculina)
The extracisternal approach in vestibular schwannoma surgery and facial nerve preservation
The classical surgical technique for the resection of vestibular schwannomas (VS) has emphasized the microsurgical anatomy of cranial nerves. We believe that the focus on preservation of the arachnoid membrane may serve as a safe guide for tumor removal. Method: the extracisternal approach is described in detail. We reviewed charts from 120 patients treated with this technique between 2006 and 2012. Surgical results were evaluated based on the extension of resection, tumor relapse, and facial nerve function. Results: Overall gross total resection was achieved in 81% of the patients. the overall postoperative facial nerve function House-Brackmann grades I-II at one year was 93%. There was no recurrence in 4.2 years mean follow up. Immunophenotyping of peripheral blood, lymph node, and bone marrow T lymphocytes during canine leishmaniosis and the impact of antileishmanial chemotherapy
Dogs are a major reservoir of Leishmania infantum, etiological agent of canine leishmaniosis (CanL) a zoonotic visceral disease of worldwide concern. Therapeutic protocols based on antileishmanial drugs are commonly used to treat sick dogs and improve their clinical condition. To better understand the impact of Leishmania infection and antileishmanial drugs on the dog's immune response, this study investigates the profile of CD4+ and CD8+ T cell subsets in peripheral blood, lymph node, and bone marrow of sick dogs and after two different CanL treatments. Two CanL groups of six dogs each were treated with either miltefosine or meglumine antimoniate combined with allopurinol. Another group of 10 clinically healthy dogs was used as control. Upon diagnosis and during the following 3 months of treatment, peripheral blood, popliteal lymph node, and bone marrow mononuclear cells were collected, labeled for surface markers CD45, CD3, CD4, CD8, CD25, and intracellular nuclear factor FoxP3, and T lymphocyte subpopulations were immunophenotyped by flow cytometry. CanL dogs presented an overall increased frequency of CD8+ and CD4+CD8+ double-positive T cells in all tissues and a decreased frequency of CD4+ T cells in the blood. Furthermore, there was a higher frequency of CD8+ T cells expressing CD25+FoxP3+ in the blood and bone marrow. During treatment, these subsets recovered to levels similar to those of healthy dogs. Nevertheless, antileishmanial therapy caused an increase of CD4+CD25+FoxP3+ T cells in all tissues, associated with the decrease of CD8+CD25−FoxP3− T cell percentages. These findings may support previous studies that indicate that L. infantum manipulates the dog's immune system to avoid the development of a protective response, ensuring the parasite's survival and the conditions that allow the completion of Leishmania life cycle. Both treatments used appear to have an effect on the dog's immune response, proving to be effective in promoting the normalization of T cell subsets.publishersversionpublishe
