    Few crucial links assure checkpoint efficiency in the yeast cell-cycle network

    Motivation: The ability of cells to complete mitosis with high fidelity relies on elaborate checkpoint mechanisms. We study S- and M-phase checkpoint responses in silico in the budding yeast with a stochastic dynamical model for the cell-cycle. We aim to provide an unbiased functional classification of network interactions that reflect the contribution of each link to checkpoint efficiency in the presence of cellular fluctuations. Results: We developed an algorithm BNetDyn to compute stochastic dynamical trajectories for an input gene network and its structural perturbations. User specified output measures like the mutual information between trigger and output nodes are then evaluated on the stationary state of the Markov process. Systematic perturbations of the yeast cell-cycle model by Li et al. classify each link according to its effect on checkpoint efficiencies and stabilities of the main cell-cycle phases. This points to the crosstalk in the cascades downstream of the SBF/MBF transcription activator complexes as determinant for checkpoint optimality; a finding that consistently reflects recent experiments. Finally our stochastic analysis emphasizes how dynamical stability in the yeast cell-cycle network crucially relies on backward inhibitory circuits next to forward induction.     Rotational Reconstruction of Sapphire (0001)

    The structure of the (31×31)R±9∘(\sqrt{31}\times \sqrt{31})R\pm9^\circ reconstructed phase on sapphire (0001) surface is investigated by means of a simulation based on the energy minimization. The interaction between Al adatoms is described with the semi-empirical many-body Sutton-Chen potential, corrected for the charge transfer between the metallic overlayer and the substrate. The interactions between the Al adatoms and sapphire substrate are described with a simple three-dimensional potential field which has the hexagonal periodicity of sapphire surface. Our energy analysis gave evidence that the structure which is observed at room temperature is in fact a frozen high-temperature structure. In accordance with the X-ray scattering, a hexagonal domain pattern separated by domain walls has been found. The Al adatoms, distributed in two monolayers, are ordered and isomorphic to metallic Al(111) in the domains and disordered in the domain walls. The main reason for the rotational reconstruction is the lattice misfit between the metallic Al and sapphire.Comment: 15 pages with 4 eps figures in text.     Modelling ballast behaviour under dynamic loading. Part 1: A 2D polygonal discrete element method approach

    International audienceDiscrete element simulation provides some insight into the alteration of railway ballast after repeated train passings. The present Part 1 is devoted to a 2D model of this granular layer interposed between the deformable ground and the rail sleeper, to which a large number of loading cycles is applied. Ballast grains are modelled as indeformable polygonal solids. A detailed account of the application to this frictional dynamical problem of the Non-Smooth Contact Dyna- mics numerical method is given. Validation is obtained through comparison with physical experiments performed on assemblies of prismatic mineral grains. Numerical results on the settlement of a track submitted to 20,000 loading cycles or more are presented

    Vibration damping in polygonal plates using the acoustic black hole effect : model based on the image source method

    A method for damping flexural vibrations in thin polygonal plates using the acoustic black hole effect is proposed. Acoustic black holes in thin plates consist of a zone of decreasing thickness in which flexural waves slow down, acting as wave sinks. An acoustic black hole of axisymmetric thickness profile is here tested on different polygonal plates. A parabolic edge is added in order to focus waves into the black hole area. The experimental results show significant reduction of the vibration level. A model based on the image source method provides qualitative agreement with the experiments

    A 802.11g and UMTS Simultaneous Reception Front-End Architecture using a double IQ structure

    International audienceIn this paper, we address the architecture of multistandard simultaneous reception receivers and we aim to reduce the complexity of the analog front-end. To this end, we propose an architecture using the double orthogonal translation technique in order to multiplex two signals received on different frequency bands. A study case concerning the simultaneous reception of 802.11g and UMTS signals is developed in this paper. Theoretical and simulation results show that this type of multiplexing does not significantly influence the evolution of the signal to noise ratio of the signals

    The Relationships Between Skeletal Muscle Index and Bone Variables in a Group of Young Adults

    International audienceThe purpose of this study was to investigate the relationships between skeletal muscle index (SMI) and bone variables in a group of young adults. Three hundred and thirty-five young adults (129 men and 206 women) whose ages ranged from 18 to 35 yr voluntarily participated in this study. Weight and height were measured, and body mass index (BMI) was calculated. Body composition, bone mineral content (BMC), bone mineral density (BMD), geometric indices of hip bone strength and trabecular bone score (TBS) were determined for each individual by Dual-energy X-ray absorptiometry (DXA). Appendicular skeletal mass (ASM, in kg) was calculated by summing the muscle masses of the 4 limbs, assuming that all nonfat and nonebone mass is skeletal muscle. Skeletal muscle index (SMI) was defined as ASM/height². In young men, SMI was positively correlated to WB BMC (r = 0.63; p < 0.001), WB BMD (r = 0.53; p < 0.001), L1-L4 BMC (r = 0.33; p < 0.001), L1-L4 BMD (r = 0.30; p < 0.001), L1-L4 TBS (r = 0.26; p < 0.01), TH BMC (r = 0.61; p < 0.001), TH BMD (r = 0.46; p < 0.001), FN BMC (r = 0.51; p < 0.001), FN BMD (r = 0.46; p < 0.001), FN cross-sectional area (CSA) (r = 0.56; p < 0.001), FN cross-sectional moment of inertia (CSMI) (r = 0.52; p < 0.001) and FN section modulus (Z) (r = 0.54; p < 0.001) but negatively correlated to FN strength index (SI) (r = -0.24; p < 0.01). In young women, SMI was positively correlated to WB BMC (r = 0.61; p < 0.001), WB BMD (r = 0.60; p < 0.001), L1-L4 BMC (r = 0.35; p < 0.001), L1-L4 BMD (r = 0.33; p < 0.001), L1-L4 TBS (r = 0.29; p < 0.001), TH BMC (r = 0.61; p < 0.001), TH BMD (r = 0.53; p < 0.001), FN BMC (r = 0.45; p < 0.001), FN BMD (r = 0.49; p < 0.001), FN CSA (r = 0.60; p < 0.001), FN CSMI (r = 0.52; p < 0.001), and FN Z (r = 0.40; p < 0.001) but negatively correlated to FN SI (r = -0.20; p < 0.01). The current study suggests that SMI is a positive determinant of bone mineral density and geometric indices of hip bone strength in young adults

    Osteoprotegerin in relation to insulin resistance and blood lipids in sub-Saharan African women with and without abdominal obesity

    BackgroundOsteoprotegerin (OPG), a soluble member of the tumor necrosis factor receptor superfamily that inhibits bone resorption, has been suggested as a potential marker of cardiovascular risk. This study aimed to assess the relationship between insulin resistance, lipid profile and OPG levels in obese and non-obese sub-Saharan African women.MethodsSixty obese (44) and non-obese (16) volunteer women aged 18 to 40years were recruited in this cross-sectional study. Their clinical (age, height, weight, waist circumference, systolic and diastolic blood pressures) and biochemical parameters (fasting blood glucose, total cholesterol, high density lipoprotein-cholesterol (HDL-C)) were measured using standard methods. Insulin levels were measured using an electrochemiluminescence immunoassay, while OPG levels were measured using the ELISA technique. Low density lipoprotein-cholesterol (LDL-C), body mass index (BMI) and Homeostasis Model Assessment of Insulin Resistance (HOMA-IR) were calculated using standard methods. Abdominal obesity was defined as a waist circumference ≥ 80cm.ResultsOPG levels were higher in obese than in normal subjects, though the difference was not significant (p = 0.9). BMI, waist circumference, percent body fat and systolic blood pressure were significantly higher in obese than in non-obese subjects (p < 0.05). In these subjects, only age significantly correlated with OPG levels (r = 0.831, p = 0.003), while none of the anthropometric nor metabolic parameter did, even after adjustment for age. In obese subjects, OPG levels fairly correlated with HDL-C (r = 0.298, p = 0.058), and significantly correlated with HOMA-IR (r = −0.438, p = 0.018). After adjustment for age, OPG levels remained negatively correlated to HOMA-IR (r = −0.516, p = 0.020) and LDL-C (r = −0.535, p = 0.015) and positively correlated to HDL-C (r = 0.615, p = 0.004). In multiple linear regression analysis, age was a main determinant of OPG levels in non-obese (β = 0.647, p = 0.006) and obese (β = 0.356, p = 0.044) women. HDL-C was also associated to OPG levels in obese women (β = 0.535, p = 0.009).ConclusionThe positive correlation of OPG with HDL-C and HOMA-IR, and its negative correlation with LDL-C suggest that it may be a marker of insulin sensitivity/resistance and atherogenic risk in obese African women
