    El individuo en situaciones extraordinarias puede verse sometido a prácticas que salen radicalmente de lo cotidiano. En estas condiciones aparecen espacios de experiencia inéditos para el individuo que desencadenan procesos rápidos de creación subjetiva, asimilaciones bruscas, resemantizaciones, re-jerarquización de elementos, rupturas subjetivas, emergencia de zonas fosilizadas o sumergidas. Se pueden producir estos cambios subjetivos bruscos porque las configuraciones cotidianas no son suficientes para dar cuenta de las nuevas experiencias. Este es el caso que nos toca analizar. La construcción de la subjetividad en torno a ciertas dimensiones que analizaremos no es homogénea ni lineal. Sin embargo podemos afirmar que en el caso de todos los sujetos partícipes del proceso de recuperación del hotel BAUEN se vislumbran cambios subjetivos dado que sus “configuraciones cotidianas no son suficientes para dar cuenta de las nuevas experiencias”.     Communicational guidance of educational institutions. Towards the institutionalization of the communicator/educator proffesionals practices in Province of Buenos Aires

    El campo de intervención de comunicadores/educadores ha sido profundamente desarrollado por profesionales a lo largo de los últimos 30 años. Sin embargo, si bien puede reconocerse la importancia académica de su accionar, en nuestro país no se han desarrollado políticas públicas que den respuesta a los emergentes comunicacionales de manera institucionalizada y sistematizada, que puedan posicionar al comunicador dentro del equipo de gestión de las instituciones educativas. Mediante el presente trabajo de investigación me propongo analizar el proceso de institucionalización de las intervenciones profesionales de comunicadores educadores en la gestión comunicacional de las instituciones educativas en la provincia de Buenos Aires a partir de la entrada en vigencia del Reglamento de Instituciones Educativas (2011), para la elaboración de lineamientos en materia de política educativa.The field of intervention of communicators/educators has been deeply developed by professionals over the last 30 years. However, although the academic importance of their actions can be recognized, in our country public policies have not been developed that respond to communicational emergencies in an institutionalized and systematized way, that can position the communicator within the management team of educational institutions.     Study and Optimization of Design Parameters in Water Loop Heat Pump Systems for Office Buildings in the Iberian Peninsula

    Water loop heat pump (WLHP) air conditioning systems use heat pumps connected to a common water circuit to fulfill the energy demands of different thermal zones in a building. In this study, the energy consumption was analyzed for the air conditioning of an office building in the typical climate of four important cities of the Iberian Peninsula. The energy consumption of one water loop heat pump system was compared with a conventional water system. Two design parameters, the range in the control temperatures and the water loop thermal storage size, were tested. Energy redistribution is an important advantage of the WLHP system, but significant savings came from high efficiency parameters in the heat pumps and minor air flow rates in the cooling tower. The low thermal level in the water loop makes this technology appropriate to combine with renewable sources. Using natural gas as the thermal energy source, a mean decrease in CO2 emissions of 8.1% was reached. Simulations showed that the installation of big thermal storage tanks generated small energy savings. Besides, the total annual consumption in buildings with high internal loads can be reduced by keeping the water loop as cool as possible

    El presente informe monográfico que lleva por título “Niveles de calidad en el servicio de orientadores turísticos en el destino Tarapoto, provincia y región San Martín, año 2017”, tiene como objetivo determinar el nivel de la calidad del servicio brindado por los orientadores turísticos del destino Tarapoto. Se realizó una amplia revisión bibliográfica acerca de calidad de servicio y la búsqueda de antecedentes de investigación sobre el tema. Asimismo, se realizó un sondeo a los turistas nacionales y extranjeros con el fin de relevar información sobre sus percepciones con respecto al servicio de los orientadores turísticos, y comparar con la bibliografía disponible. Se llegó a las siguientes conclusiones: baja demanda del servicio de guiado turístico, la calidad del servicio brindado por los orientadores turísticos es considerado bueno/excelente por los turistas que visitan Tarapoto, el aspecto más valorado por los turistas del servicio de los orientadores turísticos es la empatía.This monographic report titled as "Levels of quality in the service of tourist counselors in the destination Tarapoto, province and region San Martin, year 2017", aims to describe the level of quality of service provided by tourism counselors of the destination Tarapoto. An extensive bibliographic review was made about quality of service and the search of research background on the subject. Likewise, a survey was applied to national and foreign tourists in order to gather information about their perceptions regarding the service of tourist counselors, and compare them with the available bibliography. The following conclusions were reached: low demand of the tourist guide service, the quality of the service provided by the tourist counselors is considered good / excellent by the tourists that visit Tarapoto; the aspect most valued by the tourists of the tour guide service is the empathy.Trabajo de suficiencia profesionalAp

    Ca2+ Dynamics in the Secretory Vesicles of Neurosecretory PC12 and INS1 Cells

    Producción CientíficaWe have investigated the dynamics of the free [Ca2+] inside the secretory granules of neurosecretory PC12 and INS1 cells using a low-Ca2+-affinity aequorin chimera fused to synaptobrevin-2. The steady-state secretory granule [Ca2+] ([Ca2+]SG] was around 20–40 lM in both cell types, about half the values previously found in chromaffin cells. Inhibition of SERCA-type Ca2+ pumps with thapsigargin largely blocked Ca2+ uptake by the granules in Ca2+-depleted permeabilized cells, and the same effect was obtained when the perfusion medium lacked ATP. Consistently, the SERCA-type Ca2+ pump inhibitor benzohydroquinone induced a rapid release of Ca2+ from the granules both in intact and permeabilized cells, suggesting that the continuous activity of SERCA-type Ca2+ pumps is essential to maintain the steady-state [Ca2+]SG. Both inositol 1,4, 5-trisphosphate (InsP3) and caffeine produced a rapid Ca2+ release from the granules, suggesting the presence of InsP3 and ryanodine receptors in the granules. The response to high-K+ depolarization was different in both cell types, a decrease in [Ca2+]SG in PC12 cells and an increase in [Ca2+]SG in INS1 cells. The difference may rely on the heterogeneous response of different vesicle populations in each cell type. Finally, increasing the glucose concentration triggered a decrease in [Ca2+]SG in INS1 cells. In conclusion, our data show that the secretory granules of PC12 and INS1 cells take up Ca2+ through SERCA-type Ca2+ pumps and can release it through InsP3 and ryanodine receptors, supporting the hypothesis that secretory granule Ca2+ may be released during cell stimulation and contribute to secretion

    A confocal study on the visualization of chromaffin cell secretory vesicles with fluorescent targeted probes and acidic dyes

    Producción CientíficaSecretory vesicles have low pH and have been classically identified as those labelled by a series of acidic fluorescent dyes such as acridine orange or neutral red, which accumulate into the vesicles according to the pH gradient. More recently, several fusion proteins containing enhanced green fluorescent protein (EGFP) and targeted to the secretory vesicles have been engineered. Both targeted fluorescent proteins and acidic dyes have been used, separately or combined, to monitor the dynamics of secretory vesicle movements and their fusion with the plasma membrane. We have now investigated in detail the degree of colocalization of both types of probes using several fusion proteins targeted to the vesicles (synaptobrevin2- EGFP, Cromogranin A-EGFP and neuropeptide Y-EGFP) and several acidic dyes (acridine orange, neutral red and lysotracker red) in chromaffin cells, PC12 cells and GH3 cells. We find that all the acidic dyes labelled the same population of vesicles. However, that population was largely different from the one labelled by the targeted proteins, with very little colocalization among them, in all the cell types studied. Our data show that the vesicles containing the proteins more characteristic of the secretory vesicles are not labelled by the acidic dyes, and vice versa. Peptide glycyl-L-phenylalanine 2-naphthylamide (GPN) produced a rapid and selective disruption of the vesicles labelled by acidic dyes, suggesting that they could be mainly lysosomes. Therefore, these labelling techniques distinguish two clearly different sets of acidic vesicles in neuroendocrine cells. This finding should be taken into account whenever vesicle dynamics is studied using these techniques

    Prevalence of post-intensive care syndrome in mechanically ventilated patients with COVID-19

    Coronavirus disease 19 (COVID-19) patients usually require long periods of mechanical ventilation and sedation, which added to steroid therapy, favours a predisposition to the development of delirium and subsequent mental health disorders, as well as physical and respiratory sequelae. The aim of this study was to determine the prevalence of post-intensive care syndrome (PICS) at 3 months after hospital discharge, in a cohort of mechanically ventilated patients with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). An ambispective, observational study was conducted in three hospitals with intensive care unit (ICU) follow-up clinics. We studied adults who survived a critical illness due to SARS-CoV-2 infection requiring invasive mechanical ventilation. A physical (muscle strength and pulmonary function), functional [12-Item Short Form Health Survey (SF-12), and Barthel score], psychological [hospital anxiety and depression (HADS) and posttraumatic stress disorder symptom severity scales], and cognitive [Montreal cognitive assessment (MoCA) test] assessment were performed. A total of 186 patients were evaluated at 88 days (IQR 68–121) after hospital discharge. Mean age was 59 ± 12 years old, 126 (68%) patients were men, and median length of mechanical ventilation was 14 days (IQR 8–31). About 3 out of 4 patients (n = 139, 75%) met PICS criteria. Symptoms of cognitive and psychiatric disorders were found in 59 (32%) and 58 (31%) patients, respectively. Ninety-one (49%) patients had muscle weakness. Pulmonary function tests in patients with no respiratory comorbidities showed a normal pattern in 93 (50%) patients, and a restrictive disorder in 62 (33%) patients. Also, 69 patients (37%) were on sick leave, while 32 (17%) had resumed work at the time of assessment. In conclusion, survivors of critical illness due to SARS-CoV-2 infection requiring mechanical ventilation have a high prevalence of PICS. Physical domain is the most frequently damaged, followed by cognitive and psychiatric disorders.     Monitoring mitochondrial [Ca2+] dynamics with rhod-2, ratiometric pericam and aequorin

    Producción CientíficaThe dynamics of mitochondrial [Ca2+] ([Ca2+]M) plays a key role in a variety of cellular processes. The most important methods available to monitor [Ca2+]M are fluorescent dyes such as rhod-2 and specifically targeted proteins such as aequorin and pericam. However, significant discrepancies, both quantitative and qualitative, exist in the literature between the results obtained with different methods. We have made here a systematic comparison of the response of several fluorescent dyes, rhod-2 and rhod-FF, and two Ca2+-sensitive proteins, aequorin and pericam. Our results show that measurements obtained with aequorin and pericam are consistent in terms of dynamic Ca2+ changes. Instead, fluorescent dyes failed to follow Ca2+ changes adequately, especially during repetitive stimulation. In particular, measures obtained with rhod-2 or rhod-FF evidenced the previously reported Ca2+-dependent inhibition of mitochondrial Ca2+ uptake, but data obtained with aequorin or pericam under the same conditions did not. The reason for the loss of response of fluorescent dyes is unclear. Loading with these dyes produced changes in mitochondrial morphology and membrane potential, which were small and reversible at low concentrations (1–2 M), but produced large and prolonged damage at higher concentrations. In addition, cells loaded with low concentrations of rhod-2 suffered large changes in mitochondrial morphology after light excitation.
