On particle acceleration and trapping by Poynting flux dominated flows
Using particle-in-cell (PIC) simulations, we study the evolution of a
strongly magnetized plasma slab propagating into a finite density ambient
medium. Like previous work, we find that the slab breaks into discrete magnetic
pulses. The subsequent evolution is consistent with diamagnetic relativistic
pulse acceleration of \cite{liangetal2003}. Unlike previous work, we use the
actual electron to proton mass ratio and focus on understanding trapping vs.
transmission of the ambient plasma by the pulses and on the particle
acceleration spectra. We find that the accelerated electron distribution
internal to the slab develops a double-power law. We predict that emission from
reflected/trapped external electrons will peak after that of the internal
electrons. We also find that the thin discrete pulses trap ambient electrons
but allow protons to pass through, resulting in less drag on the pulse than in
the case of trapping of both species. Poynting flux dominated scenarios have
been proposed as the driver of relativistic outflows and particle acceleration

Temporal Correlation of Hard X-rays and Meter/Decimeter Radio Structures in Solar Flares
We investigate the relative timing between hard X-ray (HXR) peaks and
structures in metric and decimetric radio emissions of solar flares using data
from the RHESSI and Phoenix-2 instruments. The radio events under consideration
are predominantly classified as type III bursts, decimetric pulsations and
patches. The RHESSI data are demodulated using special techniques appropriate
for a Phoenix-2 temporal resolution of 0.1s. The absolute timing accuracy of
the two instruments is found to be about 170 ms, and much better on the
average. It is found that type III radio groups often coincide with enhanced
HXR emission, but only a relatively small fraction ( 20%) of the groups
show close correlation on time scales 1s. If structures correlate, the HXRs
precede the type III emissions in a majority of cases, and by 0.690.19 s
on the average. Reversed drift type III bursts are also delayed, but
high-frequency and harmonic emission is retarded less. The decimetric
pulsations and patches (DCIM) have a larger scatter of delays, but do not have
a statistically significant sign or an average different from zero. The time
delay does not show a center-to-limb variation excluding simple propagation
effects. The delay by scattering near the source region is suggested to be the
most efficient process on the average for delaying type III radio emission
Spatial analysis of solar type III events associated with narrow band spikes at metric wavelengths
The spatial association of narrow band metric radio spikes with type III
bursts is analyzed. The analysis addresses the question of a possible causal
relation between the spike emission and the acceleration of the energetic
electrons causing the type III burst. The spikes are identified by the
Phoenix-2 spectrometer (ETH Zurich) from survey solar observations in the
frequency range from 220 MHz to 530 MHz. Simultaneous spatial information was
provided by the Nancay Radioheliograph (NRH) at several frequencies. Five
events were selected showing spikes at one or two and type III bursts at two or
more Nancay frequencies. The 3-dimensional geometry of the single events has
been reconstructed by applying different coronal density models. As a working
hypothesis it is assumed that emission at the plasma frequency or its harmonic
is the responsible radiation process for the spikes as well as for the type III
bursts. It has been found that the spike source location is consistent with the
backward extrapolation of the trajectory of the type III bursts, tracing a
magnetic field line. In one of the analyzed events, type III bursts with two
different trajectories originating from the same spike source could be
identified. These findings support the hypothesis that narrow band metric

In vivo biofunctional evaluation of hydrogels for disc regeneration
Purpose Regenerative strategies aim to restore the original
biofunctionality of the intervertebral disc. Different
biomaterials are available, which might support disc
regeneration. In the present study, the prospects of success
of two hydrogels functionalized with anti-angiogenic peptides
and seeded with bone marrow derived mononuclear
cells (BMC), respectively, were investigated in an ovine
nucleotomy model.
Methods In a one-step procedure iliac crest aspirates
were harvested and, subsequently, separated BMC were
seeded on hydrogels and implanted into the ovine disc. For
the cell-seeded approach a hyaluronic acid-based hydrogel
was used. The anti-angiogenic potential of newly developed
VEGF-blockers was investigated on ionically crosslinked
metacrylated gellan gum hydrogels. Untreated discs
served as nucleotomy controls. 24 adult merino sheep were
used. After 6 weeks histological, after 12 weeks histological
and biomechanical analyses were conducted.
Results Biomechanical tests revealed no differences
between any of the implanted and nucleotomized discs. All
implanted discs significantly degenerated compared to
intact discs. In contrast, there was no marked difference
between implanted and nucleotomized discs. In tendency,
albeit not significant, degeneration score and disc height
index deteriorated for all but not for the cell-seeded
hydrogels from 6 to 12 weeks. Cell-seeded hydrogels
slightly decelerated degeneration.
Conclusions None of the hydrogel configurations was
able to regenerate biofunctionality of the intervertebral
disc. This might presumably be caused by hydrogel
extrusion. Great importance should be given to the development
of annulus sealants, which effectively exploit the
potential of (cell-seeded) hydrogels for biological disc
regeneration and restoration of intervertebral disc

Effects of mannoprotein E1 in liquid diet on inflammatory response and TLR5 expression in the gut of rats infected by Salmonella typhimurium
<p>Abstract</p> <p>Background</p> <p>Mannoproteins are yeast cell wall componend, and rich in mannose. The use of foods rich in mannose as carbohydrate, could have a bioprotective effect against entrobacteria intestinal infection. Nothing is known about mannoproteins' activity in inflammatory bowel processes induced by entrobacteria.</p> <p>This study investigates the effects of mannoprotein administration via a liquid diet on inflammatory response and TLR5 expression during intestinal tissue injury in a rat model of infection with <it>Salmonella typhimurium</it>.</p> <p>Methods</p> <p>Adult Wistar male rats were divided into three groups: control, and mannoprotein E<sub>1 </sub>at 10 or 15%. Animals were fed with a liquid diet supplemented or not with mannoprotein E<sub>1</sub>. Groups were infected by intragastrical administration of <it>S. typhimurium</it>. 24 h post-inoculation samples of spleen, ileum and liver were collected for microbiological studies. Gut samples were processed to determine levels of proinflammatory cytokines (mRNA) and TLR5 (mRNA and protein) by quantitative PCR and Western-blot, and the number of proliferative and apoptotic cells determined by immunohistochemistry.</p> <p>Results</p> <p>Ininfected levels of proinflammatory cytokines and TLR5 were higher in untreated controls than in the animals receiving mannoprotein. Proliferation was similar in both groups, whereas apoptosis was higher in controls. Curiosly, the mannoprotein effect was dose dependent.</p> <p>Conclusions</p> <p>Mannoprotein administration in a liquid diet seems to protect intestinal tissue against <it>S. typhimurium </it>infection. This protection seems to expressed as a lower pro-inflammatory response and TLR5 downregulation in gut epithelium, as well as by an inhibition of apoptosis. Nevertheless, the molecular mechanism by which mannoprotein is able to regulate these responses remain unclear. These results could open up new avenues in the use of mannoproteins as prebiotics in the therapeutic strategy for treatment of inflammatory gut processes induced by microbia.</p
Histological analysis of surgical lumbar intervertebral disc tissue provides evidence for an association between disc degeneration and increased body mass index
<p>Abstract</p> <p>Background</p> <p>Although histopathological grading systems for disc degeneration are frequently used in research, they are not yet integrated into daily care routine pathology of surgical samples. Therefore, data on histopathological changes in surgically excised disc material and their correlation to clinical parameters such as age, gender or body mass index (BMI) is limited to date. The current study was designed to correlate major physico-clinical parameters from a population of orthopaedic spine center patients (gender, age and BMI) with a quantitative histologic degeneration score (HDS).</p> <p>Methods</p> <p>Excised lumbar disc material from 854 patients (529 men/325 women/mean age 56 (15-96) yrs.) was graded based on a previously validated histologic degeneration score (HDS) in a cohort of surgical disc samples that had been obtained for the treatment of either disc herniation or discogenic back pain. Cases with obvious inflammation, tumor formation or congenital disc pathology were excluded. The degree of histological changes was correlated with sex, age and BMI.</p> <p>Results</p> <p>The HDS (0-15 points) showed significantly higher values in the nucleus pulposus (NP) than in the annulus fibrosus (AF) (Mean: NP 11.45/AF 7.87), with a significantly higher frequency of histomorphological alterations in men in comparison to women. Furthermore, the HDS revealed a positive significant correlation between the BMI and the extent of histological changes. No statistical age relation of the degenerative lesions was seen.</p> <p>Conclusions</p> <p>This study demonstrated that histological disc alterations in surgical specimens can be graded in a reliable manner based on a quantitative histologic degeneration score (HDS). Increased BMI was identified as a positive risk factor for the development of symptomatic, clinically significant disc degeneration.</p
The NlpD Lipoprotein Is a Novel Yersinia pestis Virulence Factor Essential for the Development of Plague
Yersinia pestis is the causative agent of plague. Previously we have isolated an attenuated Y. pestis transposon insertion mutant in which the pcm gene was disrupted. In the present study, we investigated the expression and the role of pcm locus genes in Y. pestis pathogenesis using a set of isogenic surE, pcm, nlpD and rpoS mutants of the fully virulent Kimberley53 strain. We show that in Y. pestis, nlpD expression is controlled from elements residing within the upstream genes surE and pcm. The NlpD lipoprotein is the only factor encoded from the pcm locus that is essential for Y. pestis virulence. A chromosomal deletion of the nlpD gene sequence resulted in a drastic reduction in virulence to an LD50 of at least 107 cfu for subcutaneous and airway routes of infection. The mutant was unable to colonize mouse organs following infection. The filamented morphology of the nlpD mutant indicates that NlpD is involved in cell separation; however, deletion of nlpD did not affect in vitro growth rate. Trans-complementation experiments with the Y. pestis nlpD gene restored virulence and all other phenotypic defects. Finally, we demonstrated that subcutaneous administration of the nlpD mutant could protect animals against bubonic and primary pneumonic plague. Taken together, these results demonstrate that Y. pestis NlpD is a novel virulence factor essential for the development of bubonic and pneumonic plague. Further, the nlpD mutant is superior to the EV76 prototype live vaccine strain in immunogenicity and in conferring effective protective immunity. Thus it could serve as a basis for a very potent live vaccine against bubonic and pneumonic plague
Repeat Composition of CenH3-chromatin and H3K9me2-marked heterochromatin in Sugar Beet (Beta vulgaris)

Sugar beet (Beta vulgaris) is an important crop of temperate climate zones, which provides nearly 30 % of the world’s annual sugar needs. From the total genome size of 758 Mb, only 567 Mb were incorporated in the recently published genome sequence, due to the fact that regions with high repetitive DNA contents (e.g. satellite DNAs) are only partially included. Therefore, to fill these gaps and to gain information about the repeat composition of centromeres and heterochromatic regions, we performed chromatin immunoprecipitation followed by sequencing (ChIP-Seq) using antibodies against the centromere-specific histone H3 variant of sugar beet (CenH3) and the heterochromatic mark of dimethylated lysine 9 of histone H3 (H3K9me2).
Results
ChIP-Seq analysis revealed that active centromeres containing CenH3 consist of the satellite pBV and the Ty3-gypsy retrotransposon Beetle7, while heterochromatin marked by H3K9me2 exhibits heterogeneity in repeat composition. H3K9me2 was mainly associated with the satellite family pEV, the Ty1-copia retrotransposon family Cotzilla and the DNA transposon superfamily of the En/Spm type. In members of the section Beta within the genus Beta, immunostaining using the CenH3 antibody was successful, indicating that orthologous CenH3 proteins are present in closely related
species within this section.
Conclusions
The identification of repetitive genome portions by ChIP-Seq experiments complemented the sugar beet reference sequence by providing insights into the repeat composition of poorly characterized CenH3-chromatin and H3K9me2-heterochromatin. Therefore, our work provides the basis for future research and application concerning the sugar beet centromere and repeat rich heterochromatic regions characterized by the presence of H3K9me2

Penelitian tugas akhir ini mengkaji tentang Profil Wisatawan di
Museum Radya Pustaka Surakarta. Tujuan dari penelitian ini adalah untuk
mengetahui dari daerah mana saja wisatawan yang berkunjung ke Museum
Radya Pustaka, bagaimana ciri-ciri wisatawan yang berkunjung ke Museum
Radya Pustaka serta harapan-harapan yang diinginkan wisatawan terhadap
Museum Radya Pustaka.
Penelitian dilakukan dengan metode kualitatif. Pengumpulan data
dilakukan melalui wawancara dengan narasumber wisatawan yang berkujung
di Museum Radya Pustaka Surakarta tempat penulis melakukan penelitian,
serta studi pustaka dan studi dokumen guna menambah sumber data.
Hasil penelitian menunjukkan bahwa (1) Sebagian besar wisatawan
yang datang berasal dari Semarang sebesar 32%. (2) Mayoritas wisatawan
yang berkunjung ke Museum Radya Pustaka berusia antara 17-25 tahun dan
kebanyakan dari mereka adalah pelajar atau mahasiswa dengan prosentase
52%. (3) Sebagian besar wisatawan yang datang ke Museum Radya Pustaka
adalah bertujuan untuk melakukan penelitian yaitu sebesar 34%. (4) Harapan
wisatawan yang berkunjung terhadap kelangsungan Museum Radya Pustaka
sebagian besar adalah agar ditingkatkan lagi pengelolaan dan keamanan
museum, agar kejadian hilangnya benda-benda koleksi museum tidak terulang
lagi dikemudian hari.
Kesimpulan dari hasil penelitian ini bahwa wisatawan yang berkujung
ke Museum Radya Pustaka Surakarta mayoritas berasal dari Semarang,
mayoritas berusia 17-25 tahun dan kebanyakan dari mereka adalah berprofesi
sebagai pelajar dan mahasiswa. Kebanyakan wisatawan yang datang bertujuan
untuk melakukan penelitian, serta harapan wisatawan terhadap Museum
Radya Pustaka adalah supaya lebih ditingkatkan lagi pengelolaan dan
keamanan museum
