    Combined effects of salinity changes and salicylic acid exposure in Mytilus galloprovincialis

    Pharmaceuticals and Personal care products (PPCPs) are frequently released into several marine matrices, representing significant environmental and ecotoxicological risks. Among the widest spread PPCPs in aquatic systems is Salicylic acid (SA), with known negative effects on marine and freshwater species. Nevertheless, the toxicity resulting from these emerging pollutants, including SA, together with climate change has still received little attention up to date. Among climate change related factors salinity is one that most affects aquatic organisms. To better understand the combined impacts of SA and salinity, the present study evaluated the biochemical alterations induced in Mytilus galloprovincialis mussels exposed to SA and different salinity levels, acting individually and in combination. The effects observed clearly highlighted that cellular damages were mainly observed at higher salinity (35), with no additive or synergistic effects derived from the combined presence of SA. Higher antioxidant capacity of mussels in the presence of SA may prevent increased LPO levels in comparison to uncontaminated mussels. Nevertheless, in the presence of SA mussels revealed loss of redox balance, regardless of the salinity level. Furthermore, mussels exposed to SA at control salinity showed increased metabolic capacity which decreased when exposed to salinities 25 and 35. These findings may indicate the protective capacity of mussels towards higher stressful conditions, with lower energy reserves expenditure when in the presence of SA and salinities out of their optimal range. Although limited cellular damages were observed, changes on mussel's redox balance, antioxidant mechanisms and metabolism derived from the combined exposure to SA and salinity changes may compromise mussel's growth and reproduction. Overall, the present study highlights the need to investigate the impacts induced by pollutants under present and future climate change scenarios, towards a more realistic environmental risk assessment.publishe

    Toxic impacts induced by Sodium lauryl sulfate in Mytilus galloprovincialis

    Pharmaceuticals and personal care products (PPCPs) are continuously dispersed into the environment, as a result of human and veterinary use, reaching aquatic coastal systems and inhabiting organisms. However, information regarding to toxic effects of these compounds towards marine invertebrates is still scarce, especially in what regards to metabolic capacity and oxidative status alterations induced in bivalves after chronic exposure. In the present study, the toxic impacts of Sodium lauryl sulfate (SLS), an anionic surfactant widely used as an emulsifying cleaning agent in household and cosmetics, were evaluated in the mussel Mytilus galloprovincialis, after exposure for 28 days to different concentrations (0.0; 0.5; 1.0; 2.0 and 4.0 mg/L). For this, effects on mussels respitation rate, metabolic capacity and oxidative status were evaluated. The obtained results indicate a significant decrease on mussel's respiration rate after exposure to different SLS concentrations, an alteration that was accompanied by a decrease of bioconcentration factor along the increasing exposure gradient, especially at the highest exposure concentration. Nonetheless, the amount of SLS accumulated in organisms originated alterations in mussel's metabolic performance, with higher metabolic capacity up to 2.0 mg/L followed by a decrease at the highest tested concentration (4.0 mg/L). Mussels exposed to SLS revealed limited antioxidant defense mecanhisms but cellular damage was only observed at the highest exposure concentration (4.0 mg/L). In fact, up to 2.0 mg/L of SLS limited toxic impacts were observed, namely in terms of oxidative stress and redox balance. However, since mussel's respiration rate was greatly affected by the presence of SLS, the present study may highlight the potential threat of SLS towards marine bivalves, limiting their filtration capacity and, thus, affecting their global physiological development (including growth and reproduction) and ultimely their biochemical performance (afecting their defense capacity towards stressful conditons).publishe

    The Potential Health Benefits of Polyphenol-Rich Extracts from Cichorium intybus

    Phytochemicals can exert their bioactivity without reaching the systemic circulation; scarcely absorbed antioxidants might reach the large bowel contributing to protection from oxidative damage-induced gastrointestinal diseases. In the present work, we aimed to study the relationship between potential activity of polyphenol-rich extracts from Cichorium intybus L. and changes in morphological characteristics on Caco-2 cells. Phytochemicals content (carotenoids and flavonoids) and total antioxidant activity of Red Chicory of Treviso and Variegated Chicory of Castelfranco were evaluated. The bioactivity of polyphenol-rich extracts from chicories was studied in in vitro Caco-2 cell monolayers model. Morphological characteristics changes to test the antioxidant and/or prooxidant effect were verified by histological analysis and observed by Electronic Scansion Microscopy (SEM). On Caco-2 cell model, the polyphenols fractions from chicories have indicated a moderate antioxidant behavior until 17 μM concentration, while 70 μM and 34 μM exert cytotoxic effects for Treviso’s and Castelfranco’s Chicory, respectively, highlighted by TEER decreasing, increased permeability, and alteration of epithelium. Our findings support the beneficial effects of these products in counteracting the oxidative stress and cellular damage, induced in vitro on Caco-2 cell model, through interaction with the mucopolysaccharide complexes in the glycocalyx, maintaining in vivo a healthy and effective intestinal barrier

    A Consensus Proposal for Nutritional Indicators to Assess the Sustainability of a Healthy Diet: The Mediterranean Diet as a Case Study

    BACKGROUND: There is increasing evidence of the multiple effects of diets on public health nutrition, society, and environment. Sustainability and food security are closely interrelated. The traditional Mediterranean Diet (MD) is recognized as a healthier dietary pattern with a lower environmental impact. As a case study, the MD may guide innovative inter-sectorial efforts to counteract the degradation of ecosystems, loss of biodiversity, and homogeneity of diets due to globalization through the improvement of sustainable healthy dietary patterns. This consensus position paper defines a suite of the most appropriate nutrition and health indicators for assessing the sustainability of diets based on the MD. METHODS: In 2011, an informal International Working Group from different national and international institutions was convened. Through online and face-to-face brainstorming meetings over 4 years, a set of nutrition and health indicators for sustainability was identified and refined. RESULTS: Thirteen nutrition indicators of sustainability relating were identified in five areas. Biochemical characteristics of food (A1. Vegetable/animal protein consumption ratios; A2. Average dietary energy adequacy; A3. Dietary Energy Density Score; A4. Nutrient density of diet), Food Quality (A5. Fruit and vegetable consumption/intakes; A6. Dietary Diversity Score), Environment (A7. Food biodiversity composition and consumption; A8. Rate of Local/regional foods and seasonality; A9. Rate of eco-friendly food production and/or consumption), Lifestyle (A10. Physical activity/physical inactivity prevalence; A11. Adherence to the Mediterranean dietary pattern), Clinical Aspects (A12. Diet-related morbidity/mortality statistics; A13. Nutritional Anthropometry). A standardized set of information was provided for each indicator: definition, methodology, background, data sources, limitations of the indicator, and references. CONCLUSION: The selection and analysis of these indicators has been performed (where possible) with specific reference to the MD. Sustainability of food systems is an urgent priority for governments and international organizations to address the serious socioeconomic and environmental implications of short-sighted and short-term practices for agricultural land and rural communities. These proposed nutrition indicators will be a useful methodological framework for designing health, education, and agricultural policies in order, not only to conserve the traditional diets of the Mediterranean area as a common cultural heritage and lifestyle but also to enhance the sustainability of diets in general

    The objective of the present research was to evaluate if and to what extent the encounter between different cultures in a context of globalization may have influenced the adherence to the Mediterranean diet (MD) in adolescents. The study comprised a structured questionnaire packet that inquired about anthropometric data, demographic information and both dietary and lifestyle habits. The KIDMED questionnaire was used to evaluate the adherence to the MD. A total of 103 respondents, aged 10–24 years (39.8% born in Italy or first-generation immigrants from Mediterranean countries) were included in the study. The results showed the highest percentage of adolescents having a medium adherence to the MD (44.7%), without differences according to geographical area of origin. Furthermore, the study proved that a higher adherence to the MD was significantly associated with daily breakfast (p = 0.008) and the consumption of fruit and vegetables (p = 0.002 and p = 0.000, respectively). Volunteers representing some of the different cultures and geographical proveniences currently present in Italy were perfectly integrated regarding diet and lifestyle, aspects considered in this study. Our investigation suggests the existence of incorrect dietary habits. It is certainly necessary to implement public health policies targeting adolescents to promote healthier lifestyle choices; the nutritional patterns of the MD should be among these choices

    Migration as Cultural Phenomenon in a Globalized World: A Pilot Study on Lifestyle and Eating Behaviours of Adolescents Living in Rome

    The aim of this research was to assess, through an observational study, lifestyle and eating behaviours of adolescents (native, and first- and second-generation immigrants), in order to understand if the migration process may have influenced these aspects. The study was carried out by a structured questionnaire packet that investigated anthropometric data, eating habits, smoking, alcohol consumption, physical activity, and sedentary behaviours. A total of 105 respondents, aged 10–24 years (51.4% first-generation immigrants, 19.1% second-generation immigrants, and 29.5% natives) were included in the study. The results showed statistical differences in some social aspects by migration status, such as place of residence, living arrangement, parental educational level, and eating differently from family members. Despite these differences, volunteers were perfectly integrated regarding most eating habits and lifestyle behaviour, underlying a process of acculturation. Moreover, our study indicates the existence of inadequate dietary habits, such as skipping breakfast. It is important to implement effective nutrition interventions for adolescents to promote healthier lifestyle choices, considering that they should also include cultural components of dietary habits

    Molecularly imprinted polymers as effective capturing receptors in a pseudo-ELISA immunoassay for procalcitonin detection in veterinary species

    In this study, a new sandwich-type immunoenzymatic assay, based on a molecularly imprinted polymer (MIP) as an artificial antibody (pseudo-ELISA), was developed for the determination of procalcitonin (PCT) in veterinary species. The quantification of PCT in human medicine represents the state of the art for the diagnosis of sepsis; instead the clinical studies on the relevance of PCT as a sepsis predictor in veterinary patients are few, likely due to the total absence of validated assays. MIPs have been widely used as antibody mimics for important applications, and MIP-based sandwich assays have emerged as promising analytical tools for the detection of disease biomarkers. Herein, a polynorepinephrine (PNE)-based imprinted film was directly synthesized on the well surface of a 96-well plate. Subsequently, based on a commercial ELISA kit, the PCT quantification was accomplished via a colorimetric sandwich assay by replacing the capture antibody of the kit with the PNE-based MIP. This method was performed to detect canine and equine PCT in buffer and in plasma samples. Under optimal conditions, the results obtained in plasma samples showed a limit of detection (LOD) of 5.87 ng mL-1 and a reproducibility (CVav%) of 10.0% for canine samples, while a LOD = 4.46 ng mL-1 and CVav% = 7.61% were obtained for equine samples
