    Contextualizing Teacher Education Emphases for Classroom Diversity

    Nominal attention has been dedicated to standards of best practice that local teachers should demonstrate in teaching culturally and linguistically diverse (CLD) students. The CREDE standards address the gap and emphasize five transnational, universals of best practice for CLD students/families. However, recent research indicates that teachers practices indicative of the most important of these, contextualization, are among the least robust of those observed. Necessarily, future research is needed to unpack these findings. In the interim, we argue that teachers’ critical reflection on their own socialization is essential to the fundamental understandings necessary for standards-based practices with these students and families

    Retention of First-Generation Mexican American Paraeducators in Teacher Education: The Juggling Act of Nontraditional Students

    This paper discusses the dynamics and challenges encountered by culturally and linguistically diverse (CLD) paraeducators who are participating in a 2+2, distance-delivered, teacher education program in the Midwest. The theoretical framework that serves as the basis of this case study is Thomas and Collier’s Prism Model (Collier, 19878: Collier & Thomas, 1989; Thomas & Collier, 1997), which focuses on the four essential dimensions of the student biography (linguistic, socio-cultural, academic, and cognitive). This case study should be understood as an account of the lived experiences of 30 CLD paraeducators in a unique recruitment and retention program designed to support all four dimensions of the student biography. To these ends, the researcher seeks to understand this pioneering project within a complex socio-political system and its implications for recruitment, retention and graduation of CLD students. Furthermore, the paper provides critical insights to inform the field concerning future program design

    Effect of Transnational Standards on U.S. Teacher Education

    The Standards for Effective Pedagogy and Learning (CREDE, 2014) specify five transnational universals of teaching that are especially effective for the rapidly growing population of English language learners in North America. CLASSIC is an evidence-based, CREDE-aligned model of teacher education for classroom educators of English language learners. CLASSIC has utilized with more than 10,000 teachers in 100 school districts, located in eight states, in collaboration with eight different universities. This study examined the impact of the transnational standards of CLASSIC curricula on teachers’ observed practices with English language learners as measured by the recently developed Inventory of Situationally and Culturally Responsive Teaching (ISCRT). Despite some variability, over 110 participating teachers in 37 U.S. schools demonstrated statistically significant improvements in their delivery of effective pedagogy, across a wide range (18 of 22) ISCRT indicators; teachers exhibited highest levels of growth in instructional conversations, joint productive activity, and challenging activities

    A Laminin G-EGF-Laminin G Module in Neurexin IV Is Essential for the Apico-Lateral Localization of Contactin and Organization of Septate Junctions

    Septate junctions (SJs) display a unique ultrastructural morphology with ladder-like electron densities that are conserved through evolution. Genetic and molecular analyses have identified a highly conserved core complex of SJ proteins consisting of three cell adhesion molecules Neurexin IV, Contactin, and Neuroglian, which interact with the cytoskeletal FERM domain protein Coracle. How these individual proteins interact to form the septal arrays that create the paracellular barrier is poorly understood. Here, we show that point mutations that map to specific domains of neurexin IV lead to formation of fewer septae and disorganization of SJs. Consistent with these observations, our in vivo domain deletion analyses identified the first Laminin G-EGF-Laminin G module in the extracellular region of Neurexin IV as necessary for the localization of and association with Contactin. Neurexin IV protein that is devoid of its cytoplasmic region is able to create septae, but fails to form a full complement of SJs. These data provide the first in vivo evidence that specific domains in Neurexin IV are required for protein-protein interactions and organization of SJs. Given the molecular conservation of SJ proteins across species, our studies may provide insights into how vertebrate axo-glial SJs are organized in myelinated axons

    The DESI Sky Continuum Monitor System

    The Dark Energy Spectroscopic Instrument (DESI) is an ongoing spectroscopic survey to measure the dark energy equation of state to unprecedented precision. We describe the DESI Sky Continuum Monitor System, which tracks the night sky brightness as part of a system that dynamically adjusts the spectroscopic exposure time to produce more uniform data quality and to maximize observing efficiency. The DESI dynamic exposure time calculator (ETC) will combine sky brightness measurements from the Sky Monitor with data from the guider system to calculate the exposure time to achieve uniform signal-to-noise ratio (SNR) in the spectra under various observing conditions. The DESI design includes 20 sky fibers, and these are split between two identical Sky Monitor units to provide redundancy. Each Sky Monitor unit uses an SBIG STXL-6303e CCD camera and supports an eight-position filter wheel. Both units have been completed and delivered to the Mayall Telescope at the Kitt Peak National Observatory.     MicroRNA-335-5p suppresses voltage-gated sodium channel expression and may be a target for seizure control

    There remains an urgent need for new therapies for treatment-resistant epilepsy. Sodium channel blockers are effective for seizure control in common forms of epilepsy, but loss of sodium channel function underlies some genetic forms of epilepsy. Approaches that provide bidirectional control of sodium channel expression are needed. MicroRNAs (miRNA) are small noncoding RNAs which negatively regulate gene expression. Here we show that genome-wide miRNA screening of hippocampal tissue from a rat epilepsy model, mice treated with the antiseizure medicine cannabidiol, and plasma from patients with treatment-resistant epilepsy, converge on a single target-miR-335-5p. Pathway analysis on predicted and validated miR-335-5p targets identified multiple voltage-gated sodium channels (VGSCs). Intracerebroventricular injection of antisense oligonucleotides against miR-335-5p resulted in upregulation of Scn1a, Scn2a, and Scn3a in the mouse brain and an increased action potential rising phase and greater excitability of hippocampal pyramidal neurons in brain slice recordings, consistent with VGSCs as functional targets of miR-335-5p. Blocking miR-335-5p also increased voltage-gated sodium currents and SCN1A, SCN2A, and SCN3A expression in human induced pluripotent stem cell-derived neurons. Inhibition of miR-335-5p increased susceptibility to tonic-clonic seizures in the pentylenetetrazol seizure model, whereas adeno-associated virus 9-mediated overexpression of miR-335-5p reduced seizure severity and improved survival. These studies suggest modulation of miR-335-5p may be a means to regulate VGSCs and affect neuronal excitability and seizures. Changes to miR-335-5p may reflect compensatory mechanisms to control excitability and could provide biomarker or therapeutic strategies for different types of treatment-resistant epilepsy

    The DESI One-Percent survey: constructing galaxy-halo connections for ELGs and LRGs using auto and cross correlations

    In the current Dark Energy Spectroscopic Instrument (DESI) survey, emission line galaxies (ELGs) and luminous red galaxies (LRGs) are essential for mapping the dark matter distribution at z1z \sim 1. We measure the auto and cross correlation functions of ELGs and LRGs at 0.8<z1.00.8<z\leq 1.0 from the DESI One-Percent survey. Following Gao et al. (2022), we construct the galaxy-halo connections for ELGs and LRGs simultaneously. With the stellar-halo mass relation (SHMR) for the whole galaxy population (i.e. normal galaxies), LRGs can be selected directly by stellar mass, while ELGs can also be selected randomly based on the observed number density of each stellar mass, once the probability PsatP_{\mathrm{sat}} of a satellite galaxy becoming an ELG is determined. We demonstrate that the observed small scale clustering prefers a halo mass-dependent PsatP_{\mathrm{sat}} model rather than a constant. With this model, we can well reproduce the auto correlations of LRGs and the cross correlations between LRGs and ELGs at rp>0.1r_{\mathrm{p}}>0.1 Mpch1\mathrm{Mpc}\,h^{-1}. We can also reproduce the auto correlations of ELGs at rp>0.3r_{\mathrm{p}}>0.3 Mpch1\mathrm{Mpc}\,h^{-1} (s>1s>1 Mpch1\mathrm{Mpc}\,h^{-1}) in real (redshift) space. Although our model has only seven parameters, we show that it can be extended to higher redshifts and reproduces the observed auto correlations of ELGs in the whole range of 0.8<z<1.60.8<z<1.6, which enables us to generate a lightcone ELG mock for DESI.     Low-dose carotid computed tomography angiography using pure iterative reconstruction

    The aim of this study was to assess if a low-dose carotid computed tomography angiography (CTA) performed with pure iterative reconstruction (IR) is comparable to a conventional dose CTA protocol. Methods: Twenty patients were included. Radiation dose was divided into a low-dose acquisition reconstructed with pure IR and a conventional dose acquisition reconstructed with 40% hybrid IR. Dose, image noise, contrast resolution, spatial resolution, and carotid artery stenosis were measured. Results: Mean effective dose was significantly lower for low-dose than conventional dose studies (1.84 versus 3.71 mSv; P < 0.001). Subjective image noise, contrast resolution, and spatial resolution were significantly higher for the low-dose studies. There was excellent agreement for stenosis grading accuracy between low- and conventional dose studies (Cohen κ = 0.806). Conclusions: A low-dose carotid CTA protocol reconstructed with pure IR is comparable to a conventional dose CTA protocol in terms of image quality and diagnostic accuracy while enabling a dose reduction of 49.6%
