    SPS-assisted Synthesis of SICp reinforced high entropy alloys: reactivity of SIC and effects of pre-mechanical alloying and post-annealing treatment

    In this work a traditional high entropy alloy (FeCoNiCrAl) was reinforced by uniformly distributed reactive silicon carbide (SiC) particles by a powder metallurgy synthetic route, using as precursors simply mixed powders or mechanically prealloyed ones. The reactive sintering produced a single isomorphic BCC structure. The sample microstructure resulted equiassic, more homogenous in samples based on prealloyed powders. The instability of SiC in the presence of metal precursors resulted in the formation of more stable carbides and silicides, as well as in carbon diffusion in the high entropy alloy matrix and partially unreacted SiC particles. The formation of these newly formed fine precipitates, as well as the presence of residual SiC were useful to increase the hardness of the alloy

    Gli acciai per lavorazioni a caldo sono caratterizzati da buona tenacità ed elevata durezza a caldo.La metallurgia delle polveri, grazie alla produzione di materiali con microstruttura più fine,consente di migliorare entrambe queste caratteristiche. In questo lavoro, una polvere di AISI H13atomizzata in gas è stata macinata meccanicamente in modo da ridurre sia la dimensione delleparticelle che la dimensione della grana cristallina. I risultati mostrano che dopo 1000 minuti dimacinazione in un mulino planetario a sfere la dimensione del grano può essere ridotta in modosignificativo fino a 15nm. L’elevato grado di deformazione introdotto nelle particelle attiva ilprocesso di sinterizzazione e di conseguenza la polvere può essere sinterizzata a temperature e pertempi inferiori. Per produrre campioni ad alta densità e per mantenere la dimensione della granafine (<1?m) la polvere è stata sinterizzata tramite Spark Plasma Sintering. La microstruttura e ladurezza dei campioni prodotti da polvere macinata sono state investigate e confrontate con quelle diun materiale prodotto con polvere atomizzata

    Impacts of orography on large-scale atmospheric circulation

    Some of the largest and most persistent circulation errors in global numerical weather prediction and climate models are attributable to the inadequate representation of the impacts of orography on the atmospheric flow. Existing parametrization approaches attempting to account for unresolved orographic processes, such as turbulent form drag, low-level flow blocking or mountain waves, have been successful to some extent. They capture the basic impacts of the unresolved orography on atmospheric circulation in a qualitatively correct way and have led to significant progress in both numerical weather prediction and climate modelling. These approaches, however, have apparent limitations and inadequacies due to poor observational evidence, insufficient fundamental knowledge and an ambiguous separation between resolved and unresolved orographic scales and between different orographic processes. Numerical weather prediction and climate modelling has advanced to a stage where these inadequacies have become critical and hamper progress by limiting predictive skill on a wide range of spatial and temporal scales. More physically-based approaches are needed to quantify the relative importance of apparently disparate orographic processes and to account for their combined effects in a rational and accurate way in numerical models. We argue that, thanks to recent advances, significant progress can be made by combining theoretical approaches with observations, inverse modelling techniques and high-resolution and idealized numerical simulations

    Uncertainty in the Representation of Orography in Weather and Climate Models and Implications for Parameterized Drag

    The representation of orographic drag remains a major source of uncertainty for numerical weather prediction (NWP) and climate models. Its accuracy depends on contributions from both the model grid‐scale orography (GSO) and the subgrid‐scale orography (SSO). Different models use different source orography datasets and different methodologies to derive these orography fields. This study presents the first comparison of orography fields across several operational global NWP models. It also investigates the sensitivity of an orographic drag parameterisation to the inter‐model spread in SSO fields and the resulting implications for representing the northern hemisphere winter circulation in a NWP model. The inter‐model spread in both the GSO and the SSO fields is found to be considerable. This is due to differences in the underlying source dataset employed and in the manner in which this dataset is processed (in particular how it is smoothed and interpolated) to generate the model fields. The sensitivity of parameterised orographic drag to the inter‐model variability in SSO fields is shown to be considerable and dominated by the influence of two SSO fields: the standard deviation and the mean gradient of the SSO. NWP model sensitivity experiments demonstrate that the inter‐model spread in these fields is of first‐order importance to the inter‐model spread in parameterised surface stress, and to current known systematic model biases. The revealed importance of the SSO fields supports careful reconsideration of how these fields are generated, guiding future development of orographic drag parameterisations and re‐evaluation of the resolved impacts of orography on the flow

    Molecular genetic analysis of severe coagulation factor XI deficiency in six Italian patients

    BACKGROUND AND OBJECTIVES: Factor XI (FXI) deficiency is a rare autosomal recessive coagulopathy which is, however, frequent among Ashkenazi Jews. Two mutations, type II (Glu117stop) and type III (Phe283Leu), account for the majority of abnormal alleles in this population. The aim of this study was to analyze the molecular basis of FXI deficiency in six unrelated Italian probands with severe deficiency, a population hitherto largely unexplored. DESIGN AND METHODS: All patients showed unmeasurable functional FXI levels in plasma. Mutational screening was performed by sequencing. Haplotype analysis was performed using intragenic polymorphisms. Expression studies were performed by transient transfection in COS-1 cells. RESULTS: Sequencing identified two novel mutations: a nonsense mutation (Cys118stop) in exon 5 in two probands, and a 6-bp deletion (643-648delATCGAC) in exon 7 in one proband. The Cys118stop is predicted to cause FXI deficiency by a secretion defect and/or by increased mRNA degradation. The 6-bp deletion causes the loss of residues Ile197 and Asp198. There was a remarkable secretion impairment of the deleted FXI protein. In four of the six probands, the type II mutation was found. Haplotype analysis in patients carrying the type II mutation revealed that they share a common haplotype, perhaps derived from a Jewish ancestor. INTERPRETATION AND CONCLUSIONS: The identification and characterization of two novel mutations widen the mutational spectrum of FXI deficiency. Haplotype analysis is compatible with a Jewish origin of the type II mutation. The high occurrence of the type II mutation among Italian patients will be helpful to direct future genetic screenings

    Genetic study of quantitative traits supports the use of Guzerá as dual-purpose cattle.

    Objective: The aim of this study was to estimate genetic parameters for 305-day cumulative milk yield and components, growth, and reproductive traits in Guzerá cattle. Methods: The evaluated traits were 305-day first-lactation cumulative yields (kg) of milk (MY305), fat (FY305), protein (PY305), lactose (LY305), and total solids (SY305); age at first calving (AFC) in days; adjusted scrotal perimeter (cm) at the ages of 365 (SP365) and 450 (SP450) days; and adjusted body weight (kg) at the ages of 210 (W210), 365 (W365) and 450 (W450) days. The (co)variance components were estimated using the restricted maximum likelihood method for single-trait, bi-trait and tri-trait analyses. Contemporary groups and additive genetic effects were included in the general mixed model. Maternal genetic and permanent environmental effects were also included for W210. Results: The direct heritability estimates ranged from 0.16 (W210) to 0.32 (MY305). The maternal heritability estimate for W210 was 0.03. Genetic correlation estimates among milk production traits and growth traits ranged from 0.92 to 0.99 and from 0.92 to 0.99, respectively. For milk production and growth traits, the genetic correlations ranged from 0.33 to 0.56. The genetic correlations among AFC and all other traits were negative ( 0.43 to 0.27). Scrotal perimeter traits and body weights showed genetic correlations ranging from 0.41 to 0.46, and scrotal perimeter and milk production traits showed genetic correlations ranging from 0.11 to 0.30. The phenotypic correlations were similar in direction (same sign) and lower than the corresponding genetic correlations. Conclusion: These results suggest the viability and potential of joint selection for dairy and beef traits in Guzerá cattle, taking into account reproductive traits.First online

    High-fat diet fuels prostate cancer progression by rewiring the metabolome and amplifying the MYC program

    Systemic metabolic alterations associated with increased consumption of saturated fat and obesity are linked with increased risk of prostate cancer progression and mortality, but the molecular underpinnings of this association are poorly understood. Here, we demonstrate in a murine prostate cancer model, that high-fat diet (HFD) enhances the MYC transcriptional program through metabolic alterations that favour histone H4K20 hypomethylation at the promoter regions of MYC regulated genes, leading to increased cellular proliferation and tumour burden. Saturated fat intake (SFI) is also associated with an enhanced MYC transcriptional signature in prostate cancer patients. The SFI-induced MYC signature independently predicts prostate cancer progression and death. Finally, switching from a high-fat to a low-fat diet, attenuates the MYC transcriptional program in mice. Our findings suggest that in primary prostate cancer, dietary SFI contributes to tumour progression by mimicking MYC over expression, setting the stage for therapeutic approaches involving changes to the diet

    Varespladib and cardiovascular events in patients with an acute coronary syndrome: the VISTA-16 randomized clinical trial

    IMPORTANCE: Secretory phospholipase A2(sPLA2) generates bioactive phospholipid products implicated in atherosclerosis. The sPLA2inhibitor varespladib has favorable effects on lipid and inflammatory markers; however, its effect on cardiovascular outcomes is unknown. OBJECTIVE: To determine the effects of sPLA2inhibition with varespladib on cardiovascular outcomes. DESIGN, SETTING, AND PARTICIPANTS: A double-blind, randomized, multicenter trial at 362 academic and community hospitals in Europe, Australia, New Zealand, India, and North America of 5145 patients randomized within 96 hours of presentation of an acute coronary syndrome (ACS) to either varespladib (n = 2572) or placebo (n = 2573) with enrollment between June 1, 2010, and March 7, 2012 (study termination on March 9, 2012). INTERVENTIONS: Participants were randomized to receive varespladib (500 mg) or placebo daily for 16 weeks, in addition to atorvastatin and other established therapies. MAIN OUTCOMES AND MEASURES: The primary efficacy measurewas a composite of cardiovascular mortality, nonfatal myocardial infarction (MI), nonfatal stroke, or unstable angina with evidence of ischemia requiring hospitalization at 16 weeks. Six-month survival status was also evaluated. RESULTS: At a prespecified interim analysis, including 212 primary end point events, the independent data and safety monitoring board recommended termination of the trial for futility and possible harm. The primary end point occurred in 136 patients (6.1%) treated with varespladib compared with 109 patients (5.1%) treated with placebo (hazard ratio [HR], 1.25; 95%CI, 0.97-1.61; log-rank P = .08). Varespladib was associated with a greater risk of MI (78 [3.4%] vs 47 [2.2%]; HR, 1.66; 95%CI, 1.16-2.39; log-rank P = .005). The composite secondary end point of cardiovascular mortality, MI, and stroke was observed in 107 patients (4.6%) in the varespladib group and 79 patients (3.8%) in the placebo group (HR, 1.36; 95% CI, 1.02-1.82; P = .04). CONCLUSIONS AND RELEVANCE: In patients with recent ACS, varespladib did not reduce the risk of recurrent cardiovascular events and significantly increased the risk of MI. The sPLA2inhibition with varespladib may be harmful and is not a useful strategy to reduce adverse cardiovascular outcomes after ACS. TRIAL REGISTRATION: clinicaltrials.gov Identifier: NCT01130246.
