    Development patterns of an isolated oligo-mesophotic carbonate buildup, early Miocene, Yadana field, offshore Myanmar

    The development history of an oligo-mesophotic, early Miocene, isolated carbonate system (>160 m in thickness), forming the uppermost part of the Oligo-Miocene Yadana buildup (northern Andaman Sea), has been evidenced from the integration of sedimentological core studies from 4 wells (cumulated core length: 343 m), well correlations, seismic interpretation and analysis of the ecological requirements of the main skeletal components. Three types of carbonate factory operated on the top of the platform, depending on water-depth, turbidity and nutrient level: (1) a scleractinian factory developing under mesophotic conditions during periods of high particulate organic matter supplies, (2) an echinodermal factory occupying dysphotic to aphotic area of the platform coevally with the scleractinian factory, (3) a large benthic foraminiferal-coralline algal factories prevailing under oligo-mesophotic and oligo-mesotrophic conditions. The limited lateral changes in facies between wells, together with the seismic expression of the Yadana buildup, suggest deposition on a flat-topped shelf. Carbonate production and accumulation on the Yadana platform was mainly controlled by light penetration, nutrient content and hydrodynamic conditions. Scleractinian-rich facies resulted from transport of coral pieces derived from mesophotic environments (mounds?) and deposited in deeper, low light, mud-rich environments in which lived abundant communities of suspension feeders such as ophiuroids. Changes in monsoonal intensity, terrestrial runoff from the Irrawaddy River, upwelling currents and internal waves activity during the early Miocene are likely responsible for significant variations in water turbidity and nutrient concentration in the Andaman Sea, thus promoting the development of an oligo-mesophotic, incipiently drowned platform

    AVHRR and VISSR satellite instrument calibration results for both Cirrus and marine stratocumulus IFO periods

    Accurate characterizations of some cloud parameters are dependent upon the absolute accuracy of satellite radiance measurements. Visible wavelength measurements from both the AVHRR and VISSR instruments are often used to study cloud characteristics. Both of these instruments were radiometrically calibrated prior to launch, but neither has an onboard device to monitor degradation after launch. During the FIRE/SRB cirrus Intensive Field Operation (IFO), a special effort was made to monitor calibration of these two instruments onboard the NOAA-9 and GOES-6 spacecraft. In addition, several research groups have combined their efforts to assess the long-term performance of both instruments. These results are presented, and a limited comparison is made with the ERBE calibration standard

    Transdisciplinary participatory-action-research from questions to actionable knowledge for sustainable viticulture development

    Viticulture negatively impacts the environment, biodiversity, and human health; however, despite the widely acknowledged challenges that this intensive agricultural activity poses to sustainable development, measures to reduce its invasiveness are constantly being deferred or rebuffed. Constraints to change are linked to vine cultivation methods, the impacts of climate change on vine resilience and disease sensitivity, and socio-economic models, as well as growing criticisms from society. Research and training have thus far failed to provide solutions or mobilise stakeholders on a large scale. Such resistance to sustainable practices development calls into question the effectiveness of knowledge production systems and relations between scientists, winegrowers, and society: Have scientific disciplines overly isolated themselves from each other and from the wider society to the point of losing the capacity to incorporate alternative forms of knowledge and reasoning and achieve collaborative action? Herein, we describe our findings from a participatory action research project that began in Westhalten, France, in 2013 and ultimately spread to Switzerland and Germany over the next 6 years. We show that participatory action research can mobilise long-term collaborations between winegrowers, NGOs, advisers, elected officials, members of civil society, and researchers, despite differing visions of viticulture and the environment. The epistemological framework of this research promotes consensus-building by valuing complexity and dissensus in knowledge and reasoning such that all actors are involved in experimentation and the production of results. From these findings, consensus statements were collectively elaborated in qualitative and quantitative registers. Once acknowledged by the scientific community, these consensus statements became shareable knowledge. We propose that this renewed interdisciplinarity associating the human and social sciences with agronomic and biological sciences in collaboration with stakeholders produces actionable knowledge that mobilises and engages winegrowers to conceive and implement sustainable viticulture on a transnational scale

    The serine protease domain of MASP-3: enzymatic properties and crystal structure in complex with ecotin.

    International audienceMannan-binding lectin (MBL), ficolins and collectin-11 are known to associate with three homologous modular proteases, the MBL-Associated Serine Proteases (MASPs). The crystal structures of the catalytic domains of MASP-1 and MASP-2 have been solved, but the structure of the corresponding domain of MASP-3 remains unknown. A link between mutations in the MASP1/3 gene and the rare autosomal recessive 3MC (Mingarelli, Malpuech, Michels and Carnevale,) syndrome, characterized by various developmental disorders, was discovered recently, revealing an unexpected important role of MASP-3 in early developmental processes. To gain a first insight into the enzymatic and structural properties of MASP-3, a recombinant form of its serine protease (SP) domain was produced and characterized. The amidolytic activity of this domain on fluorescent peptidyl-aminomethylcoumarin substrates was shown to be considerably lower than that of other members of the C1r/C1s/MASP family. The E. coli protease inhibitor ecotin bound to the SP domains of MASP-3 and MASP-2, whereas no significant interaction was detected with MASP-1, C1r and C1s. A tetrameric complex comprising an ecotin dimer and two MASP-3 SP domains was isolated and its crystal structure was solved and refined to 3.2 Ã…. Analysis of the ecotin/MASP-3 interfaces allows a better understanding of the differential reactivity of the C1r/C1s/MASP protease family members towards ecotin, and comparison of the MASP-3 SP domain structure with those of other trypsin-like proteases yields novel hypotheses accounting for its zymogen-like properties in vitro

    Epilepsy Caused by an Abnormal Alternative Splicing with Dosage Effect of the SV2A Gene in a Chicken Model

    Photosensitive reflex epilepsy is caused by the combination of an individual's enhanced sensitivity with relevant light stimuli, such as stroboscopic lights or video games. This is the most common reflex epilepsy in humans; it is characterized by the photoparoxysmal response, which is an abnormal electroencephalographic reaction, and seizures triggered by intermittent light stimulation. Here, by using genetic mapping, sequencing and functional analyses, we report that a mutation in the acceptor site of the second intron of SV2A (the gene encoding synaptic vesicle glycoprotein 2A) is causing photosensitive reflex epilepsy in a unique vertebrate model, the Fepi chicken strain, a spontaneous model where the neurological disorder is inherited as an autosomal recessive mutation. This mutation causes an aberrant splicing event and significantly reduces the level of SV2A mRNA in homozygous carriers. Levetiracetam, a second generation antiepileptic drug, is known to bind SV2A, and SV2A knock-out mice develop seizures soon after birth and usually die within three weeks. The Fepi chicken survives to adulthood and responds to levetiracetam, suggesting that the low-level expression of SV2A in these animals is sufficient to allow survival, but does not protect against seizures. Thus, the Fepi chicken model shows that the role of the SV2A pathway in the brain is conserved between birds and mammals, in spite of a large phylogenetic distance. The Fepi model appears particularly useful for further studies of physiopathology of reflex epilepsy, in comparison with induced models of epilepsy in rodents. Consequently, SV2A is a very attractive candidate gene for analysis in the context of both mono- and polygenic generalized epilepsies in humans
