    Influence of smoking and obesity on alveolar-arterial gas pressure differences and dead space ventilation at rest and peak exercise in healthy men and women

    SummaryBackground and aimsBesides exercise intolerance, the assessment of ventilatory and perfusion adequacy allows additional insights in the disease pathophysiology in many cardiovascular or pulmonary diseases. Valid measurements of dead space/tidal volume ratios (VD/VT), arterial (a′) – end-tidal (et) carbon dioxide (CO2) and oxygen (O2) pressure differences (p(a′-et)CO2) and (p(et-a′)O2), and alveolar (A)–a′ O2 pressure differences (p(A-a′)O2) require using blood samples in addition to gas exchange analyses on a breath-by-breath-basis. Smoking and nutritional status are also important factors in defining disorders. Using a large healthy population we considered the impact of these factors to develop useful prediction equations.Methods and resultsIncremental cycle exercise protocols were applied to apparently healthy volunteer adults who did not have structural heart disease or echocardiographic or lung function pathologies. Age, height, weight, and smoking were analysed for their influence on the target parameters in each gender. Reference values were determined by regression analyses. The final study sample consisted of 476 volunteers (190 female), aged 25–85 years. Smoking significantly influences p(A-a′)O2 and p(a′-et)CO2 at rest and peak exercise, and VD/VT during exercise. Obesity influences upper limits of VD/VT, p(a′-et)CO2 and p(et-a′)O2 at rest as well as p(A-a′)O2 and p(et-a′)O2 at exercise. Reference equations for never-smokers as well as for apparently healthy smokers considering influencing factors are given.ConclusionGender, age, height, weight, and smoking significantly influence gas exchange. Considering all of these factors this study provides a comprehensive set of reference equations derived from a large number of participants of a population-based study

    The Synthetic Biology Open Language (SBOL) Version 3:Simplified Data Exchange for Bioengineering

    The Synthetic Biology Open Language (SBOL) is a community-developed data standard that allows knowledge about biological designs to be captured using a machine-tractable, ontology-backed representation that is built using Semantic Web technologies. While early versions of SBOL focused only on the description of DNA-based components and their sub-components, SBOL can now be used to represent knowledge across multiple scales and throughout the entire synthetic biology workflow, from the specification of a single molecule or DNA fragment through to multicellular systems containing multiple interacting genetic circuits. The third major iteration of the SBOL standard, SBOL3, is an effort to streamline and simplify the underlying data model with a focus on real-world applications, based on experience from the deployment of SBOL in a variety of scientific and industrial settings. Here, we introduce the SBOL3 specification both in comparison to previous versions of SBOL and through practical examples of its use

    Synthetic biology builds upon genetics, molecular biology, and metabolic engineering by applying engineering principles to the design of biological systems. When designing a synthetic system, synthetic biologists need to exchange information about multiple types of molecules, the intended behavior of the system, and actual experimental measurements. The Synthetic Biology Open Language (SBOL) has been developed as a standard to support the specification and exchange of biological design information in synthetic biology, following an open community process involving both wet bench scientists and dry scientific modelers and software developers, across academia, industry, and other institutions. This document describes SBOL 3.0.0, which condenses and simplifies previous versions of SBOL based on experiences in deployment across a variety of scientific and industrial settings. In particular, SBOL 3.0.0, (1) separates sequence features from part/sub-part relationships, (2) renames Component Definition/Component to Component/Sub-Component, (3) merges Component and Module classes, (4) ensures consistency between data model and ontology terms, (5) extends the means to define and reference Sub-Components, (6) refines requirements on object URIs, (7) enables graph-based serialization, (8) moves Systems Biology Ontology (SBO) for Component types, (9) makes all sequence associations explicit, (10) makes interfaces explicit, (11) generalizes Sequence Constraints into a general structural Constraint class, and (12) expands the set of allowed constraints

    A Biobrick Library for Cloning Custom Eukaryotic Plasmids

    Researchers often require customised variations of plasmids that are not commercially available. Here we demonstrate the applicability and versatility of standard synthetic biological parts (biobricks) to build custom plasmids. For this purpose we have built a collection of 52 parts that include multiple cloning sites (MCS) and common protein tags, protein reporters and selection markers, amongst others. Importantly, most of the parts are designed in a format to allow fusions that maintain the reading frame. We illustrate the collection by building several model contructs, including concatemers of protein binding-site motifs, and a variety of plasmids for eukaryotic stable cloning and chromosomal insertion. For example, in 3 biobrick iterations, we make a cerulean-reporter plasmid for cloning fluorescent protein fusions. Furthermore, we use the collection to implement a recombinase-mediated DNA insertion (RMDI), allowing chromosomal site-directed exchange of genes. By making one recipient stable cell line, many standardised cell lines can subsequently be generated, by fluorescent fusion-gene exchange. We propose that this biobrick collection may be distributed peer-to-peer as a stand-alone library, in addition to its distribution through the Registry of Standard Biological Parts (http://partsregistry.org/)

    Gendered Risk Perceptions Associated with Human-Wildlife Conflict: Implications for Participatory Conservation

    This research aims to foster discourse about the extent to which gender is important to consider within the context of participatory approaches for biological conservation. Our objectives are to: (1) gender-disaggregate data about stakeholders' risk perceptions associated with human-wildlife conflict (HWC) in a participatory conservation context, and (2) highlight insights from characterizing gendered similarities and differences in the way people think about HWC-related risks. Two communal conservancies in Caprivi, Namibia served as case study sites. We analyzed data from focus groups (n = 2) to create gendered concept maps about risks to wildlife and livelihoods and any associations of those risks with HWC, and semi-structured interviews (n = 76; men = 38, women = 38) to measure explicit risk attitudes associated with HWC. Concept maps indicated some divergent perceptions in how groups characterized risks to wildlife and livelihoods; however, not only were identified risks to wildlife (e.g., pollution, hunting) dissimilar in some instances, descriptions of risks varied as well. Study groups reported similar risk perceptions associated with HWC with the exception of worry associated with HWC effects on local livelihoods. Gendered differences in risk perceptions may signal different priorities or incentives to participate in efforts to resolve HWC-related risks. Thus, although shared goals and interests may seem to be an obvious reason for cooperative wildlife management, it is not always obvious that management goals are shared. Opportunity exists to move beyond thinking about gender as an explanatory variable for understanding how different groups think about participating in conservation activities

    Paradigmatic or Critical? Resilience as a New Turn in EU Governance for the Neighbourhood

    Rising from the margins of EU aid documents, resilience became a centrepiece of the 2016 EU Global Security Strategy, especially in relation to the neighbourhood. While new resilience-thinking may signify another paradigmatic shift in EU modus operandi, the question that emerges is whether it is critical enough to render EU governance a new turn, to make it sustainable? This article argues that in order for resilience-framed governance to become more effective, the EU needs not just engage with ‘the local’ by way of externally enabling their communal capacity. More crucially, the EU needs to understand resilience for what it is – a self-governing project – to allow ‘the local’ an opportunity to grow their own critical infrastructures and collective agency, in their pursuit of ‘good life’. Is the EU ready for this new thinking, and not just rhetorically or even methodologically when creating new instruments and subjectivities? The bigger question is whether the EU is prepared to critically turn the corner of its neoliberal agenda to accommodate emergent collective rationalities of self-governance as a key to make its peace-building project more successful

This essay focuses on Azerbaijan’s relations with the European Union (EU). It will identify the policy strategies used by Azerbaijan to engage with the EU in two areas where their interests and objectives do not align, namely democracy and human rights promotion policies, and agenda-setting of the Nagorno-Karabakh conflict. The essay highlights, first, the relevance of national independence, ‘balancing’ and regime legitimacy in explaining Azerbaijan’s foreign policy strategies, and second, the mixed track record of Azerbaijan’s assertiveness, with different outcomes depending on whether the policy strategies are reactive or proactive
