    Impact of annual praziquantel treatment on urogenital schistosomiasis in a seasonal transmission focus in central Senegal

    In Sub-Saharan Africa, urogenital schistosomiasis remains a significant public health problem, causing 150.000 deaths/year with approximately 112 million cases diagnosed. The Niakhar district is a disease hotspot in central Senegal where transmission occurs seasonally with high prevalences. The aim of this study was to determine the effect of annual treatment over 3 years on the seasonal transmission dynamics of S. haematobium in 9 villages in the Niakhar district. Adults and children aged between 5 and 60 years were surveyed from 2011 to 2014. Urine samples were collected door-to-door and examined for S. haematobium eggs at baseline in June 2011, and all participants were treated in August 2011 with PZQ (40 mg/kg). After this initial examination, evaluations were conducted at 3 successive time points from September 2011 to March 2014, to measure the efficacy of the annual treatments and the rates of reinfection. Each year, during the transmission period, from July to November-December, malacological surveys were also carried out in the fresh water bodies of each village to evaluate the infestation of the snail intermediate hosts. At baseline, the overall prevalence of S. haematobium infection was 57.7%, and the proportion of heavy infection was 45.3%, but one month after the first treatment high cure rates (92.9%) were obtained. The overall infection prevalence and proportion of heavy infection intensities were drastically reduced to 4.2% and 2.3%, respectively. The level of the first reinfection in February-March 2012 was 9.5%. At follow-up time points, prevalence levels varied slightly between reinfection and treatment from 9.5% in June 2012 to 0.3% in March 2013, 11.2 in June 2013, and 10.1% April 2014. At the end of the study, overall prevalence was significantly reduced from 57.7% to 10.1%. The overall rate of infested Bulinid snails was reduced after repeated treatment from 0.8% in 2012 to 0.5% in 2013. Repeated annual treatments are suggested to have a considerable impact on the transmission dynamics of S. haematobium in Niakhar, due to the nature of the epidemiological system with seasonal transmission. Thus, to maintain this benefit and continue to reduce the morbidity of urogenital schistosomiasis, other approaches should be integrated into the strategy plans of the National program to achieve the goal of urogenital schistosomiasis elimination in seasonal foci in Senegal

    A comprehensive analysis of drug resistance molecular markers and Plasmodium falciparum genetic diversity in two malaria endemic sites in Mali.

    BACKGROUND: Drug resistance is one of the greatest challenges of malaria control programme in Mali. Recent advances in next-generation sequencing (NGS) technologies provide new and effective ways of tracking drug-resistant malaria parasites in Africa. The diversity and the prevalence of Plasmodium falciparum drug-resistance molecular markers were assessed in Dangassa and Nioro-du-Sahel in Mali, two sites with distinct malaria transmission patterns. Dangassa has an intense seasonal malaria transmission, whereas Nioro-du-Sahel has an unstable and short seasonal malaria transmission. METHODS: Up to 270 dried blood spot samples (214 in Dangassa and 56 in Nioro-du-Sahel) were collected from P. falciparum positive patients in 2016. Samples were analysed on the Agena MassARRAY® iPLEX platform. Specific codons were targeted in Pfcrt, Pfmdr1, Pfdhfr, and Pfdhps, Pfarps10, Pfferredoxin, Pfexonuclease and Pfmdr2 genes. The Sanger's 101-SNPs-barcode method was used to assess the genetic diversity of P. falciparum and to determine the parasite species. RESULTS: The Pfcrt_76T chloroquine-resistance genotype was found at a rate of 64.4% in Dangassa and 45.2% in Nioro-du-Sahel (p = 0.025). The Pfdhfr_51I-59R-108N pyrimethamine-resistance genotype was 14.1% and 19.6%, respectively in Dangassa and Nioro-du-Sahel. Mutations in the Pfdhps_S436-A437-K540-A581-613A sulfadoxine-resistance gene was significantly more prevalent in Dangassa as compared to Nioro-du-Sahel (p = 0.035). Up to 17.8% of the isolates from Dangassa vs 7% from Nioro-du-Sahel harboured at least two codon substitutions in this haplotype. The amodiaquine-resistance Pfmdr1_N86Y mutation was identified in only three samples (two in Dangassa and one in Nioro-du-Sahel). The lumefantrine-reduced susceptibility Pfmdr1_Y184F mutation was found in 39.9% and 48.2% of samples in Dangassa and Nioro-du-Sahel, respectively. One piperaquine-resistance Exo_E415G mutation was found in Dangassa, while no artemisinin resistance genetic-background were identified. A high P. falciparum diversity was observed, but no clear genetic aggregation was found at either study sites. Higher multiplicity of infection was observed in Dangassa with both COIL (p = 0.04) and Real McCOIL (p = 0.02) methods relative to Nioro-du-Sahel. CONCLUSIONS: This study reveals high prevalence of chloroquine and pyrimethamine-resistance markers as well as high codon substitution rate in the sulfadoxine-resistance gene. High genetic diversity of P. falciparum was observed. These observations suggest that the use of artemisinins is relevant in both Dangassa and Nioro-du-Sahel

    Africa alive corridors : transdisciplinary research based on African footprints

It is AEON contribution No. 208 and Iphakade publication 281.DATA AVAILABILITY : The data that support the maps and findings are available on request from the corresponding author.The idea of Africa Alive Corridors (AAC) evolved from Gondwana geological mapping to a comprehensive, more inclusive and dynamic approach to transdisciplinary research known as Earth Stewardship Science. Twenty designated corridors explore the geo-biological and cultural heritage of different regions of Africa over various periods, from deep time to the Anthropocene. Each corridor reveals a specific lens through which to investigate some of the rich scientific narratives embedded within it. The concept also facilitates learning and knowledge exchange across numerous disciplines: archeology, geology, geophysics, oceanography, glaciology, biology, botany, ecology, agriculture, engineering, spatial statistics, social sciences, and the humanities. This contribution analyses ten selected corridors in southern and western Africa, the Congo Basin, East Africa, and Madagascar. The various research themes explored include Earth impact hazard, origins of humankind, Snowball Earth, coastal food systems and conservation, the biogeography of lemurs, human settlement dynamics in Cameroon, tectonically linked earthquake occurrences in Algeria and Morocco, modelling land-use changes in the Western Rift Valley, trades and civilizations of the Mali Empire, Mbira music, and contemporary art. The ongoing work on these—and ten other—corridors has considerable potential to host new international collaborations to develop the links between society and natural sciences in Africa.     Développement de l’orpaillage et Mutations dans les Villages Aurifères du Sud-est du Sénégal

    Dans le sud-est du Sénégal, en l’occurrence la région de Kédougou, le développement croissant de l’orpaillage à partir des années 2000 a entraîné de nombreuses mutations dans les villages aurifères et les zones d’orpaillage. Celles-ci sont à la fois d’ordre social, démographique, politique, économique et environnemental. Cet article est centré sur l’analyse de ces mutations ainsi que de leurs multiples implications sur la vie des individus, des groupes et des communautés, sur l’environnement, sur les micro-économies et sur l’organisation sociopolitique des villages et des zones concernés. Il montre en définitive que, jusqu’ici, l’orpaillage a plutôt donné lieu à un développement problématique conduisant vers un phénomène de « phagédénisme multidimensionnel », c’est-à-dire la tendance à l’extension, au développement et à l’aggravation des problèmes sur le plan social, économique, environnemental, etc.AbstractIn south-eastern Senegal, namely the Kedougou region, the increasing development of gold panning from the 2000s has led to many changes in auriferous villages and gold panning areas. These changes are of social, demographic, political, economic and environmental nature. This paper focuses on the analysis of changes and their multiple implications on the lives of individuals, groups and communities, as well as the environment, micro-economies and the socio-political organization of villages and areas concerned. It ultimately shows that so far gold panning has rather resulted in a problematic development leading to the “multidimensional phagedena” phenomenon, that is to say the tendency to spreading, deepening and worsening problems at social, economic, environmental levels, etc

    Efficient enrichment cloning of TAL effector genes from Xanthomonas

    Many plant-pathogenic xanthomonads use a type III secretion system to translocate Transcription Activator-Like (TAL) effectors into eukaryotic host cells where they act as transcription factors. Target genes are induced upon binding of a TAL effector to double-stranded DNA in a sequence-specific manner. DNA binding is governed by a highly repetitive protein domain, which consists of an array of nearly identical repeats of ca. 102 base pairs. Many species and pathovars of Xanthomonas, including pathogens of rice, cereals, cassava, citrus and cotton, encode multiple TAL effectors in their genomes. Some of the TAL effectors have been shown to act as key pathogenicity factors, which induce the expression of susceptibility genes to the benefit of the pathogen. However, due to the repetitive character and the presence of multiple gene copies, high-throughput cloning of TAL effector genes remains a challenge. In order to isolate complete TAL effector gene repertoires, we developed an enrichment cloning strategy based on • genome-informed in silico optimization of restriction digestions, • selective restriction digestion of genomic DNA, and • size fractionation of DNA fragments.Our rapid, cheap and powerful method allows efficient cloning of TAL effector genes from xanthomonads, as demonstrated for two rice-pathogenic strains of Xanthomonas oryzae from Africa.
