    Oxidative Stress and Mitochondrial Dysfunction in Down's Syndrome: Relevance to Aging and Dementia

    Genome-wide gene deregulation and oxidative stress appear to be critical factors determining the high variability of phenotypes in Down's syndrome (DS). Even though individuals with trisomy 21 exhibit a higher survival rate compared to other aneuploidies, most of them die in utero or early during postnatal life. While the survivors are currently predicted to live past 60 years, they suffer higher incidence of age-related conditions including Alzheimer's disease (AD). This paper is centered on the mechanisms by which mitochondrial factors and oxidative stress may orchestrate an adaptive response directed to maintain basic cellular functions and survival in DS. In this context, the timing of therapeutic interventions should be carefully considered for the successful treatment of chronic disorders in the DS population

    SCHOOL PRINCIPALS' TRANSFORMATIONAL LEADERSHIP BEHAVIORS AS A PREDICTOR OF TEACHERS' PERCEPTIONS OF ORGANIZATIONAL COMMITMENT

    The aim of this research was to investigate the effect of principlas’ transformational leader behaviors on teachers’ organizational commitment. In the research, causal-comparative and relarional survey methods were used. The study group, defined according to convenient sampling method, was composed of 260 public school teachers working in five provinces in the Central Anatolia Region in Turkey. The data was gathered with “Transformational Leadership Scale” and “Teachers’ Organizational Commitment Scale”, in the spring semester of the 2021-2022 academic year. Descriptive statistical analysis, independent groups t-test, test of variance (ANOVA), Pearsons’ corelational analysis and regresion analysis were employed in the analysis phase. The findings showed that both principals’ transformational leadership levels and teachers’ organizational commitment levels were moderate. It was observed that teachers’ perceptions about their principals’ transformational leadership behaviours and organizational commitment levels didn’t show a significant difference among the groups of gender, age and work experience. It was found that there was a significant relationship between principals’ transformational leadership behaviours and teachers’ organizational commitment, and the former was a significant predictor of the latter. It was concluded that principals’ transformational leadership behaviors might be effective in enhancing teachers’ organizational commitment. In this frame practical, theoretical and research recommendations were developed

    Utility investigation of automated techniques in hematopoietic progenitor cell count and viability assessment in the Good Manufacturing Practice (GMP) settingg

    Aim: To compare our parameters as regards: i) cell count via two different automated cell count techniques, and ii) viability via automated trypan blue exclusion and 7-aminoactinomycin D (7-AAD) staining. Method: We used the trypan blue exclusion technique and an automated cell counter and for viability testing, and the trypan blue exclusion technique and the 7-AAD evaluation by flow cytometry. The trypan blue exclusion and the radio frequency techniques were used for automated cell counting. Flow cytometric analysis was performed by evaluating the yielded cellular products for 7-AAD uptake during the cell count of CD34+ cells. Results: The mean values for cell count were estimated as 3.44±1.22x106/ml (range, 2.48-5.71x106/ml) and 4.14±1.94x106/ml (range, 1.77-7.43x106/ml) for the trypan blue exclusion and radio frequency techniques, respectively. Additionally, the mean values for viability analyses via the automated trypan blue exclusion and 7-AAD were 93.38±6.09% (range, 79.00-98.00%) and 99.49±0.60% (range, 98.40-100.00%), respectively. Conclusions: Our study has responded to two fundamental questions: whether the results of both of the automated techniques for cell count correspond with each other, and whether the results of the automated viability assessment conform those of the 7-AAD technique during the manufacturing processes of cellular therapy products intended for clinical use. Even though we have the opportunity to use the hemocytometer in our laboratory setting, the automated trypan blue exclusion technique gives cell count results in concordance within the range of the expectations of our Quality Management System (QMS)

    Turkish Primary Care Patients’ Overviews and Attitudes About Traditional and Complementary Medicine: A Cross-Sectional Study

    Aim:Primary care physicians need to be aware of the types of traditional and complementary medicine (TCM) used by patients to be able to guide patients away from harmful approaches and to aim them toward useful or at least seeming benign approaches. This study aimed to determine the prevalence of the usage of TCM methods in Turkey and the level of knowledge and attitudes of applicants about these methods.Methods:The universe of this cross-sectional study consisted of individuals aged 18 and over who applied to the Family Health Centers and agreed to participate in the study in 12 provinces in different regions of Turkey between July and December 2016. The questionnaires were applied to the volunteer participants by the researchers using face-to-face interview techniques. The questionnaire had 25 questions.Results:80.6% of the participants benefited from TCM, and 86.4% stated they believed in the effectiveness of TCM. Women, university graduates of college who had private insurance used TCM (respectively; p=0.031; p=0.004; p=0.000), and women (83.3%) found TCM more useful than men (p=0.005). The most frequent reason for using TCM was “heard that it was useful”.Conclusion:The results of our study indicated that a large portion of Turkish primary care patients use TCM and recommend it to their relatives. Therefore, health policies and academic knowledge should be developed in this sense

    A Role for Thrombospondin-1 Deficits in Astrocyte-Mediated Spine and Synaptic Pathology in Down's Syndrome

    Down's syndrome (DS) is the most common genetic cause of mental retardation. Reduced number and aberrant architecture of dendritic spines are common features of DS neuropathology. However, the mechanisms involved in DS spine alterations are not known. In addition to a relevant role in synapse formation and maintenance, astrocytes can regulate spine dynamics by releasing soluble factors or by physical contact with neurons. We have previously shown impaired mitochondrial function in DS astrocytes leading to metabolic alterations in protein processing and secretion. In this study, we investigated whether deficits in astrocyte function contribute to DS spine pathology.Using a human astrocyte/rat hippocampal neuron coculture, we found that DS astrocytes are directly involved in the development of spine malformations and reduced synaptic density. We also show that thrombospondin 1 (TSP-1), an astrocyte-secreted protein, possesses a potent modulatory effect on spine number and morphology, and that both DS brains and DS astrocytes exhibit marked deficits in TSP-1 protein expression. Depletion of TSP-1 from normal astrocytes resulted in dramatic changes in spine morphology, while restoration of TSP-1 levels prevented DS astrocyte-mediated spine and synaptic alterations. Astrocyte cultures derived from TSP-1 KO mice exhibited similar deficits to support spine formation and structure than DS astrocytes.These results indicate that human astrocytes promote spine and synapse formation, identify astrocyte dysfunction as a significant factor of spine and synaptic pathology in the DS brain, and provide a mechanistic rationale for the exploration of TSP-1-based therapies to treat spine and synaptic pathology in DS and other neurological conditions

    Uterine Rupture with Massive Late Postpartum Hemorrhage due to Placenta Percreta Left Partially In Situ

    Placental adhesive disorders involve the growth of placental tissue into or through the uterine wall. Among these disorders, placenta percreta is the rarest one. However, it may cause significant complications. This report aimed to report a neglected patient with placenta percreta who developed uterine rupture with life-threatening late postpartum intra-abdominal hemorrhage. On admission, the patient had acute abdomen with moderate abdominal distention and was subjected to emergency laparotomy. A full-thickness defect of the anterior uterine wall involving the hysterotomy site was seen. Placental tissues occupied both sides of the incision and posterior bladder wall was also invaded by placenta. Total abdominal hysterectomy with partial resection of the posterior bladder wall was performed
